Isolation of a Korean Domestic Microalga, Chlamydomonas reinhardtii KNUA021, and Analysis of Its Biotechnological Potential

  • A freshwater microalga, Chlamydomonas reinhardtii KNUA021, was characterized for its potential as a biochemical feedstock. Its optimal growth was observed when the culture was incubated at $25^{\circ}C$ and pH 9.4. However, the isolate was capable of survival and growth under a variety of temperatures (10-$30^{\circ}C$) and pH (pH 4.0-12.0) conditions. The total lipid content of the isolate was 21.7% of dry weight and it was found that a high-value fatty alcohol, hexadecenol ($C_{20}H_{40}O$), was autotrophically produced by strain KNUA021. In addition, a nutritionally important $C_{18:3}{\omega}3$ (${\alpha}$-linolenic acid, ALA) was also identified in this photosynthetic microorganism as one of the major fatty acids. Hence, C. reinhardtii KNUA021 appears to show promise for use in the production of microalgae-based biochemicals.

간세포 암화과정에서 p/s 고정비율과 $\omega3$ 지방산 급원에 따른 전암성 병변과 Glutathione 의존 해독화 효소계에 미치는 영향 (Different Sources of $\omega3$ Fatty Acids at the Fixed Ratio of p/s Affect Glutathione Dependent Enzymes in Rat Hepatocarcinogenesis)

  • This study is conducted to determine the effects of dietary source of $\omega$3 fatty acids on preneoplastic foci and the glutathione dependent enzymes in rat hepatocarcinogenesis initiated by diethylnitrosamine (DEN). Male Sprague-Dawley rats were fed one of three diets containing 10% (w/w) fats fixed p/s = -1.0 and $\omega$6/$\omega$3 ratio = -0.4 or 4.0 ; fish oil-com oil blended (FC), com oil-beef tallow-fish oil blended (CF), com oil-beef tallow-perilla oil blended (CP), from gestation period. At 10 weeks, animals of experimental groups were injected intraperitoneally with DEN (200 mg/kg body weight) and two-thirds partial hepatectomy was carried out 3 weeks later and were sacrificed 8 weeks after DEN initiation. The area and number of glutathione S-transferase placenta (GST-P) positive foci were significantly decreased in rats fed diets containing fish oil (FC and CF) than those fed perilla oil diet (CP). Fish oil feeding significantly increased the activities of glutathione dependent enzymes. Rats fed diets containing fish oil (FC and CF) significantly increased the glutathione (GSH) content and the activities of glutathione peroxidase (GPx), glutathione reductase (GR), and glutathione S-transferase (GST). Glutathione dependent enzymes had significantly negative correlation with GST-P positive foci. Glucose 6-phosphatase (G6Pase) was increased in rats feeding fish oil. Thiobarbituric acid reactive substances were not different among groups. Therefore, the preventive effect against hepatocarcinogenesis might be explained by induction of the glutathione dependent enzymes and G6Pase. (Korean J Nutrition 36(8): 785∼792, 2003)

고등어(Scomber japonicus) 부산물 가수 분해물로부터 분리된 유지의 정제 및 특성 (Purification and Characterization of Fish Oil Obtained from Hydrolysates of Mackerel (Scomber japonicus) By-products)

  • 본 연구에서는 고등어 부산물의 활용을 위해 가수분해된 고등어 부산물을 사용하였고, 분리된 유지를 시료로 하여 정제조건과 그 특성을 분석하였다. 탈검은 6% citric acid 수용액으로 시행하였을 때, 인의 함량이 45 mg/kg에서 5.8 mg/kg으로 감소하여 가장 효과적이었으며, 탈산은 8% sodium hydroxide수용액으로 처리하여 산가와 과산화물가가 각각 18.03 mgKOH/g에서 1.19 mg KOH/g과 30.25 meq/kg에서 3.18 meq/kg으로 감소하였다. 탈색은 activated charcoal을 5%로 사용하였을 때 산가와 과산화물가가 각각 0.17 mgKOH/g과 1.21 meq/kg으로 감소하였다. 색도의 변화는 탈산을 거쳤을 때 0.488 nm에서 0.057 nm로 감소하였는데 이후 탈색과정에서는 큰 변화를 보이지 않았다. 불포화 지방산의 조성 증가를 위해 냉침을 행하여 docosahexaenoic acid (DHA)와 eicosapentaenoic acid (EPA)의 함량의 합이 228.83 mg/g으로 측정되었으며, 오메가-3 지방산 함유 유지의 기준에 적합하게 정제하여 제품으로 활용이 가능함을 나타내었다.

식이지방의 종류와 수준이 흰쥐의 Lecithin : Cholesterol Acyltransferase 활성 및 콜레스테롤대사에 미치는 영향 (Effects of Dietary Lipid Sources and Levels on Lecithin : Cholesterol Acyltansferase Activity and Cholesterol Metabolism in Rats)

  • 본 실험은 식이내 지방의 종류와 수준이 흰쥐의 LCAT활성 및 콜레스테롤 대사에 어떻게 영향을 미치는가를 알아보기 위하여 실시되었다. 식이지바의 종류는 정어리유($\omega$3 linolenic acid) 및 옥수수기름 ($\omega$6 linolic acid)의 네 처리로 나누고 각 처리마다 지방의 수준을 총 에너지의 10%, 20%, 40%등 세수준으로 모두 12군을 두었으며, 식이지바의 무첨가군을 대조구로 두었다. 총 78마리의 Sprague-Dawley 계통 흰쥐 수컷에게 시험식이를 6주간 자유급여하였다. 사양시험 종료 후 간조직에서의 콜레스테롤 합성능력을 알아보기 위해 흰쥐의 간조직을 채취하여 in vitro 배양시험을 실시하였다. 본 시험에서 얻어진 결과를 요약하면 다음과 같다. 1) 혈장 총 콜레스테롤, HDL-콜레스테롤. LDL-콜레스테롤농도 및 HDL-C/T-C비율은 식이지방의 종류에 의해 유의하게(p<0.001) 영향을 받았다. ($\omega$3지방산을 많이 함유한 군(정어리유와 들기름)이 ($\omega$6지방산과 포화지방산을 많이 함유한 군(옥수수기름과 우지)에 비해 혈장내 HDL-콜레스테롤농도가 유의성있게(p<0.001)증가하였고, LDL-콜레스테롤농도는 유의성있게(p<0.05) 감소하였다. 혈장내 콜레스테롤농도는 우지군이 다른지방군에 비해(p<0.001),무지방식이군이 식이지방첨가군에 비해 유의하게(p<0.05)영향을 받았다. 2) 혈장 lecithiin : cholesterol acyltransferase(LCAT) 활성은 식이지방첨가군이 무지방식이군보다(p<0.001), 식물성유지군이 동물성지방군보다(p<0.001), 정어리유군이 우지군보다(p<0.001), 들기름군이 옥수수기름군보다(p<0.05), 정어리유군이 들기름군보다 유의성있게(p<0.05) 증가하였다. 저지방식이군이 중지방식이군과 고지방식이군에 비해 혈장내 LCAT 활성이 유의하게9p<0.001) 증가하였다. 3) 간의 콜레스테롤합성 능력은 정어리유군이 다른지방군보다(p<0.001), 무지방식이군이 식이지방첨가군보다(p<0.001), 동물성지방군의 식물성유지군보다 유의하게(p<0.001) 증가하였으나, 식이 지방의 수준에 의한 차이는 나타나지 않았다.

Evaluation of Meat from Native Chickens: Analysis of Biochemical Components, Fatty Acids, Antioxidant Dipeptides, and Microstructure at Two Slaughter Ages

  • This study examined biochemical components, fatty acids, antioxidant dipeptides, and muscle fiber density of breast and thigh muscles from Korean new native chicken strains (A and B) at two slaughter ages, compared with white semi-broiler (W) or broilers. The pH values were different by chicken breed. The new native strains had the lowest fat content in the breast at 12 wk (p<0.05). Regardless of the muscles, A and B at 12 wk had higher levels of arachidonic acid (ARA; C20:4), docosahexaenoic acid (DHA; C22:6), and nervonic acid (C24:1) than broilers (p<0.05). A similar result was observed for the polyunsaturated fatty acids (PUFAs) and polyunsaturated and saturated fatty acids ratio (P/S) content in the breast. Irrespective of the muscles, A and B enriched with omega-3 fatty acids had a lower ω-6/ω-3 PUFA ratio than broilers (p<0.05) at 12 wk. Of the antioxidant di-peptides, the anserine contents were highest in A and B than in the W or broilers (p<0.05), regardless of the muscles and slaughter ages. Furthermore, the breast meat from A and B contained a higher muscle fiber density for both slaughter ages than the W and broilers (p<0.05). Based on these findings, even if the commercial birds (broilers or W) are raised under the similar environmental conditions as A and B, the new native chicken strains have distinct meat quality attributes, particularly higher ARA and DHA levels, lower ω-6/ω-3 PUFA ratio, and higher anserine contents.

Effect of Biodegradable Mulch Film on Soil Microbial Community

  • The biodegradable film application can escape from plastic environmental pollution. This experiments studied the effect of biodegradable mulch film on the soil microbial community using fatty acid methyl ester method in soybean production field. The soil $NO_3$-N content in polyethylene mulch film (PE) soil was significantly higher than biodegradable mulch film soil (p < 0.05). The soil microbial community of Gram negative bacteria showed significantly higher in biodegradable mulch film soil than PE mulch film soil (p < 0.05). In addition, biodegradable mulch film soil had significantly low ratio of cy17:0 to $16:1{\omega}7c$ and cy19:0 to $18:1{\omega}7c$ compared with those of PE mulch film soil (p < 0.05), indicating that microbial stress decreased. The ratio of cy17:0 to $16:1{\omega}7c$ and cy19:0 to $18:1{\omega}7c$ should be considered as a potential responsible factor for the obvious differentiation that was observed between the biodegradable mulch film soil and PE mulch film soil in a upland field. The results of this experimentation show the potential of using biodegradable mulch film in place of PE.

Comparisons of fatty acid accumulation patterns of two filter feeders, Branchinella kugenumaensis and Daphnia magna in a controlled environment

  • Background: Filter-feeding zooplankton has limited food resources owing to their habitat. Consequently, it is crucial for them to acquire all essential compounds, such as fatty acids (FAs) and amino acids, from confined diets. To elucidate the trophic transfer of FAs to filter feeders, the primary consumers in freshwater ecosystems, we compared the FA accumulation patterns of two species of filter-feeding zooplankton, Daphnia magna and Branchinella kugenumaensis, in a laboratory experiment. Experimental neonates and nauplii preyed on a single phytoplankton species (Selenastrum capricornutum) for three days after hatching prior to diet switching. Five replicates per feeding group in each species were fed on six different types of mixed phytoplankton diet for 10 days after diet switching. Subsequently, the consumers and diets were harvested and FAs were extracted. Results: Principal component analysis showed that the FA profiles of zooplankton were well-grouped by species and diet. Although diet affects the FA profiles of consumers, they exhibit different FA accumulation patterns. D. magna had a higher 18C-ω3 content and ω3/ω6 ratio than did B. kugenumaensis. In contrast, B. kugenumaensis had higher contents of 18:1ω7 and 20:5ω3 (eicosapentaenoic acid), 22:6ω3 (docosahexaenoic acid), and a higher ratio of ∑18C monounsaturated FAs to ∑18C-ω3 polyunsaturated FAs than did D. magna. Conclusions: This study showed that two primary consumers, D. magna and B. kugenumaensis, fed the same diet had different assimilation patterns of FAs under controlled environments. Specific FA accumulation patterns in filter feeders can provide information on the transfer process of various FAs to high-trophic organisms.

Eicosanoid 유도체가 흰쥐 혈압 변화에 미치는 영향 (The Effect of Eicosanoid Analogues on the Change to Blood Pressure in Rat)

  • Arachidonic acid (AA, C20 : 4, $\omega$-6) and eicosapentanoic acid (EPA,C20 : 5, $\omega$-3), which are polyunsaturated fatty acids forming eicosanoids, were tested for their effects on blood pressure in Wistar rats and SHR. AA is the most important precursor for the biosynthesis of eicosanoids which include the prostaglandins, prostacyclin (PGI$_2$), thromboxane $A_2$ (TXA$_2$) and the leukotriens. TXA$_2$is a potent vasoconstrictor and a powerful inducer of platelet aggregation causing myocardial infarction and hypertention. In contrast, PGI$_2$ induces vasodilation and inhibits platelet aggregation. In this study, AA markedly increased blood pressure, but its effect was antagonized by both EPA, a structural analog of AA, and dazmegrel, a TX synthetase inhibitor. Also, AA enhanced the antihypertensive effects of hydralazine and captopril, and EPA reduced TXA$_2$ production. These results indicate that the hypotensive effects of EPA might be closely related to the decrease in TXA$_2$ biosynthesis due to competitive inhibition by structural similarity of the EPA to the AA, the precursor of TXA$_2$.

인슐린 비의존성 당뇨환자의 탄수화물과 지질섭취비율이 혈당과 지질성상에 미치는 영향 (Serum glucose and lipid profiles according to dietary carbohydrate and lipid intake ratio in NIDDM patients)

  • This study aimed at the effect on serum glucose and lipid profiles according to carbohydrate / lipid intake ratio in Korean 191 NIDDM patients. Most of NIDDM patients belong to under 50-64 years. As carbohydrate intake level increased, lipid intake level decreased. The group of higher carbohydrate / lipid intake ratio shows significantly high in fasting glucose level and postprandial glucose in NIDDM patients. The higher carbohydrate / lipid intake ratio the higher dietary linoleic and $\omega$-3 fatty acid level but not in dietary $\omega$3/$\omega$6 ratio. Serum HDL decrease inhigher carbohydrate lipid intake ratio group. Serum total lipid and PUFA level decrease according to ditary total lipid intake decrease in men but not significantly different.

참치가공 부산물의 부위별 성분 분석 (Analysis of Chemical Compounds on Tuna Processing By-products)

  • 수산물 제조를 위한 가공공정 중 발생하는 부산물 양은 전체 원료의 30~35%정도를 차지하나 일부만이 식품원료로 재 이용되고 대부분이 사료로 이용되거나 폐기물로 버려지고 있는 실정이다. 따라서 이들 부산물의 이용성을 증대시키기 위한 일환으로 기능성 향상 및 제품화를 위 한 기초적인 자료를 확보하고자 하였다. 먼저, 참치 가공 부산물을 어피살, 어피, 꼬리, 혈합육, 복육으로 분류하여 일반성분을 조사한 결과에서 조단백질은 26.31%로 어피에서 비교적 많았으며 나머지 부위에서는 21% 내외였다. 조지방도 어피에서 15.58%로 가장 높게 함유되어 있었으나 혈합육에서는 0.75%로 낮게 함유되어 있었다. 특수성분 중 vitamin C의 경우에 복육(175.56 mg/ kg)에서, vitamin B$_1$, B$_2$의 경우는 혈합육(4.26 mg/ kg, 3.09 mg/kg)에 많이 함유되어 있었고 불포화 지방산은 어피살과 꼬리에 많이 함유되어 있었고 특히, 고도 불포화지방산인 DHA(C22 : 6 $\omega$ -3)와 EPA(C22 : 6 $\omega$ -3)는 어피살과 어피에 많았다. 이들 유용 성분에 대하여 참치의 일반적인 가공처리 공정인 자숙 후 지방산 함량 변화는 어피살과 혈합육의 15-methylhexadecanoic acid(i-17 : 0)가 증가되었으며 어피에서는 3-hydroxytetra-decanoic acid (3-OH 14 : 0), heptadecanoic acid(17 : 0), 2-hydroxyhexadecanoic acid (2-OH 16 : 0), nonadeca-noic acid(19 : 0) 등이 새로이 검출되었고 혈합육은 DHA와 EPA가 변화가 적었고 어피에서는 DHA의 변화가 적었으나 EPA가 검출되지 않았고, 꼬리에서는 DHA와 EPA가 검출되지 않았다.
