Cu Accumulation and Elimination in the Tissues of the Olive Flounder Paralichthys olivaceus

  • Olive flounder, Paralichthys olivaceus were exposed to waterborne Cu (control, 50, 80, 150, and 320 ${\mu}g$/L) for 30 days and then depurated for 20 days to investigate the effects of waterborne Cu exposure on growth, accumulation, and elimination. The weight-specific growth rate was significantly negatively related to waterborne Cu concentrations at 150 and 320 ${\mu}g$/L. The order of Cu accumulation in different tissues of exposed fish was liver>intestines>gills>kidneys>muscle, suggesting that the liver is more important than other tissues for the storage of Cu in olive flounder. The accumulation factor for the gills, intestines, liver, and muscle increased with increasing exposure time, and accumulation was negatively related to exposure concentration for the gills, kidneys, and muscle. Cu concentrations in the gills, intestines, and liver continuously decreased for 20 days of depuration. The fastest elimination rate occurred in the intestines at all exposure concentrations, and the order of Cu elimination in the different tissues was intestines>liver>gills.

  • This study was made to find the compound of it's taste and properties of texture and also it's color desired and optimal conditions when the natural spices garlic, celery, parsley, bay leaves, pepper com, nutmeg, shallot, onion, tarragon, lemon juice-are added to reduction of the spices if the liquid egg yolk and hot virgin olive oil. Extracted by white wine and vinegar and then baked on salamander and gas oven in emulsified properties of sabayon and hot virgin olive oil. It may be indicated the pasteurization that during that in all treatment of the liquid egg yolk was achieved with whisk holding water in double boiling pen 95$^{\circ}C$ for 1min with the object of removing salmonella possibly contained in the yolk.   • Two infectious species of Streptococcosis pathogens were detected by multiplex PCR assay. Detection rates of Streptococcus iniae and S. parauberis could reach 44.9% and 55.1% respectively for one year during 2004 to 2005 in Jeju island. These findings showed that S. parauberis strains were important pathogen with streptococcosis of olive flounder in Jeju island. These findings showed that S. parauberis strains were important pathogen with streptococcosis of olive flounder in Jeiu island. In the present study we have investigated the interspecific relationship of all Jeju area of S. parauberis by RAPD analysis. Represent strains divided to four groups by RAPD fingerprints. The important differences observed between the olive flounder isolates suggest that they could constitute a well-differentiated group or a separate clonal line within this bacterial species. Though, serological research of S. parauberis strains in Jeju island not exist yet. These strains doing the serological evolution.

Effects of Different Feeding Regimes on the Compensatory Growth of Olive Flounder Paralichthys olivaceus

  • The effects of different feeding regimes on the compensatory growth of olive flounder Paralichthys olivaceus were investigated. Seven treatments with triplicates of different feeding regimes were conducted, as follows: (1) fed twice a day 6 days a week for 8 weeks; (2) starved for 1 week, then fed twice a day 6 days a week for 3 weeks; (3) starved for 2 weeks, then fed twice a day 6 days a week for 6 weeks; (4) starved for 5 days, then fed twice a day for 9 days; (5) starved for 10 days, then fed twice a day for 18 days; (6) starved for 2 days, then fed twice a day for 5 days, starved for another 3 days, and fed twice a day 4 days; and (7) starved for 4 days, then fed twice a day for 10 days, starved for another 6 days, and fed twice a day for 8 days. Forty fish averaging 12.5 g, were hand-fed to satiation according to the designated feeding schedule. Fish from the control group gained more weight than those from any other group. Feeding efficiency did not vary among treatments. In summary, olive flounder subjected to fasting with different feeding regimes did not fully compensate growth compared to fish fed for 8 weeks without fasting. In addition, the less that fasted groups were subsequently fed, the lower their compensatory growth.

Multiple Drug Resistance and Transferable R-plasmid of Streptococci Isolated from Diseased Olive Flounder (연쇄상구균의 약제내성과 전이성 R-plasmid의 특성)

  • MICs of 8 chemotherapeutic agents against forty streptococcal isolates were determined. These strains were isolated from diseased olive flounder, Paralichthys olivaceus, and showed 2-5 multiple drug resistance against different antibacterial agents including ampicillin, doxycycline (DOXY), erythromycin, florfenicol, flumequine, norfloxacin, oxolinic acid, and oxytetracycline (OTC). In conjugation experiment, we found transferable R plasmids carrying OTC and DOXY resistance determinant in 3 drug resistance strains analyzed. Six out of 40 isolates showed positive signal in colony hybridization with the R plasmid DNA (pST9) as a probe. It suggests that other types R plasmid different from pST9 is also involving in multiple drug resistance of streptococci isolated from olive flounder.

  • The effect of dexamethasone injection on the burden of marine birnavirus (MABV) in asymptomatically infected olive flounder (Paralichthys olivaceus) fingerlings was investigated. In real time PCR analysis, the threshold cycle (Ct) value of the fish injected with dexamethasone was significantly lower than that of the fish in the PBS-injected and no-handling groups. The higher amplification of the MABV gene in the dexamethasone-injected group than the 2 control groups was confirmed also by semi-quantitative RT-PCR. The results indicate an increase of MABV burden in olive flounder fingerlings after a single injection with dexamethasone.

Effect of Intermittent Feeding on Growth, Feed Utilization and Body Composition of Subadult Olive Flounder Paralichthys olivaceus in Suboptimal Temperature

  • The effect of intermittent feeding on growth, feed utilization and body composition of subadult olive flounder fed extruded pellet (EP) in suboptimal temperature ($13.5{\pm}2.10^{\circ}C$) was determined. Two hundred twenty five subadult fish averaging 272 g were randomly distributed into fifteen of 300 L circular flow-through tanks (fifteen fish per tank). Five treatments with different days of feeding a week were prepared in triplicate: fish were hand-fed with EP to apparent satiation once a day, seven days a week (7DF), which was used as the control group or consecutive six, five, four and three days a week, for 12 weeks, referred to as 6DF, 5DF, 4DF and 3DF treatments, respectively. Weight gains and specific growth rates of fish in 6DF treatment were higher than those of fish in 7DF, 4DF and 3DF treatments, but not different from those of fish in 5DF treatment. Daily feed intake of fish in 6DF and 7DF treatments was higher than that of fish in 5DF, 4DF and 3DF treatments. Feed efficiency and protein efficiency ratio of fish were not different among treatments. There was no difference in the chemical composition of fish between treatments. Results of this study demonstrated that feeding five days per week could be recommended for subadult olive flounder fed extruded pellet in suboptimal temperature.

  • This study was investigated the survival and hatching rate of fertilized eggs in olive flounder Paralichthys olivaceus at water temperature, salinity, duration of transport and different stock densities during commercial transport. The observed optimal temperature during transport was 17.5℃ similar to the natural environmental conditions. The proper salinity was observed to be at 33 psu (practical salinity unit) in both surviving and hatching rate. In terms of the duration of transport, there were no differences until 12 hours between survival and hatching rate, but shorter time of transport would be better. With the best conditions during transport, 20,000 eggs/L were handled in both survival and hatching rate. These results can be essential in the production and transport of healthy olive flounder seeds.

Effect of Quercetin on the Activity and mRNA Expression of Antioxidant Enzymes and Physiological Responses in Olive Flounder (Paralichthys olivaceus) Exposed to Cadmium

  • We investigated the antioxidant efficacy of quercetin (0% Diet 1, 0.25% Diet 2, and 0.5% Diet 3) pretreatment for 30 and 60 days in response to cadmium (Cd) toxicity in the olive flounder, and measured the plasma lysozyme activity to understand the immune effects of quercetin. The lysozyme activity with Diets 2 and 3 was higher than with Diet 1. Based on this result, to examine the immune ability and antioxidant role of quercetin, we exposed olive flounder fed quercetin to Cd and then measured the expression and activity of antioxidant enzymes (superoxide dismutase (SOD) and catalase (CAT)) and lipid peroxidation (LPO). With Diets 2 and 3, the expression and activity of antioxidant enzymes and the $H_2O_2$ concentration were lower than with Diet 1. In addition, the LPO levels were lower than with Diet 1, which protected the cell membrane. Therefore, quercetin removed the reactive oxygen species (ROS) produced by Cd, indicating that quercetin has antioxidant ability. In addition to its antioxidant ability, quercetin has immune effects.

Effect of PCBs on the Skin and Gill Development in the Larval Stage of Olive Flounder, Paralichthys olivaceus (넙치 (Paralichthys olivaceus) 자어의 피부 및 아가미 발달에 미치는 PCBs의 영향)

  • Effect of PCBs on the skin and gill development of olive flounder, Paralichthys olivaceus were investigated by histological methodology and morphometric data. The olive flounder were exposed to limit concentration of effulent of PCBs (3.0 ${\mu}g/L$) for 60 days. Skin development can be classified into four stages: SSEL (simple squamous epithelial layer), MCA (mucous cell appearance) & CCA (club cell appearance) DLA (dermal layer appearance), and SEL (startification of epidermal layer) stages. Gill development had five stages: GFA (gill filament appearance), IGFE (identification of early gill filament epithelial cell) MCA (mucous cell appearance), PCA (pillar cell appearance), and FGL (formation of gill lamella) stages. The periods of structural completion of skin and gill were 22-30 days and 23-30 days after hatching in the exposure group respectively. The process of development of skin and gill of the exposure group was very similar to that of the control group. Therefore, PCBs (3 ${\mu}g/L$) have no influence on the development of skin and gill in the larval stage of olive flounder.
