Comparison of Essential Oil Composition Extracted from Agastache rugosa by Steam Distillation and Supercritical Fluid Extraction (수증기 증류법과 초임계유체 추출법으로 분리한 배초향의 정유성분 조성 비교)

  • In order to compare the extraction patterns of main components from the raw material between the extraction methods, the aerial parts (dried stem, leaves, and flowers) of Agastache rugosa were extracted by SDE simultaneous steam distillation & extraction) and SFE (supercritical fluid extraction). Volatile components of essential oil and extract were identified by GC and GC-MSD. The contents of essential oil extracted by SDE were 0.49% in aerial part of Agastache rugosa on dry basis. Major components were methyl chavicol(27.2%), isomenthone(24.6%), hexadecanoic acid(13.0%). menthone (5.5%) among 32 kinds of components confirmed in essential oil. On the other hand, the contents of SFE extracts revealed 3.21% on dry basis, 6 times higher than those of SDE. Major components were isomenthone(15.3%), hexadecanoic acid(13.7%), methyl chavicol(12.6%), benzoic acid(3.8%) among 33 kinds of components identified in extract.

Extraction of Deasphalted Oil from Vacuum Residue (감압잔사유로부터 탈아스팔트오일의 추출)

  • In order to maximize the utility of vacuum residue, supercritical solvent extraction technique where n-pentane was used as a supercritical solvent was applied to obtain deasphalted oil from vacuum residue. Oil-extraction yield at various temperatures and pressures and the contents of metal complex and sulfur of extracted oil were investigated. In supercritical state, extraction yield of deasphalted oil was found to be strongly dependent on the n-pentane density, and the metal complex content of extracted oil was effectively lowered when compared with that of vacuum residue. However, the sulfur content of extracted oil showed little difference when compared with that of vacuum residue.

Extraction Method of Antioxidants in Soybean Oil (Sweep Co-Distillation 법에 의한 산화방지제의 추출법)

  • This study was conducted to evaluate the extraction methods for the determination of antioxidants in soybean oil. Recovery rates of various antioxidants in soybean oil showed similar rates as 80.4~102.1% by solvent/solvent extraction method and 89.9~106.4% by sweep co-distillation method except 46.6~61.2% of PG at corresponding spiked concentractions. The maximun recovery rates of antioxidnts were obtained when extraction time and extraction temperature used in UNITREX were 20min and 21$0^{\circ}C$ respectively. In the recovery rates with the activation of florisil when 2% ofwater was added to florisil the highest recovery rates for TBHQ, BHA, BHT were obtained by sweep co-distillation met-hod. Therefore sweep co-distillation method showed less solvent simple operation and high recovery rate compared with solvent/solvent extraction method.

Quality and characteristics of fermented ginseng seed oil based on bacterial strain and extraction method

  • Background: In this study, the fermentation of ginseng seeds was hypothesized to produce useful physiologically-active substances, similar to that observed for fermented ginseng root. Ginseng seed was fermented using Bacillus, Pediococcus, and Lactobacillus strains to extract ginseng seed oil, and the extraction yield, color, and quantity of phenolic compounds, fatty acids, and phytosterol were then analyzed. Methods: The ginseng seed was fermented inoculating 1% of each strain on sterilized ginseng seeds and incubating the seeds at $30^{\circ}C$ for 24 h. Oil was extracted from the fermented ginseng seeds using compression extraction, solvent extraction, and supercritical fluid extraction. Results and Conclusion: The color of the fermented ginseng seed oil did not differ greatly according to the fermentation or extraction method. The highest phenolic compound content recovered with the use of supercritical fluid extraction combined with fermentation using the Bacillus subtilis Korea Food Research Institute (KFRI) 1127 strain. The fatty acid composition did not differ greatly according to fermentation strain and extraction method. The phytosterol content of ginseng seed oil fermented with Bacillus subtilis KFRI 1127 and extracted using the supercritical fluid method was highest at 983.58 mg/100 g. Therefore, our results suggested that the ginseng seed oil fermented with Bacillus subtilis KFRI 1127 and extracted using the supercritical fluid method can yield a higher content of bioactive ingredients, such as phenolics, and phytosterols, without impacting the color or fatty acid composition of the product.

  • Various single solvents were tested to find the effective solvent for the extraction of algae oil from wet-form Chlorella minutissima. In the case of single solvents, their extractabilities for algae oil were increased with their polarity because the water in wet algae cell is to form a solvent shell around the lipids. Based on these results, the wet-form algae samples were treated with a polar alcohol solvent and then a nonpolar solvent was added in algae residue. In the algae oil extraction by ethanol/n-hexane, total lipid contents were 40-50% and composition of triglyceride in extracted oil was 46.50%. Considering solvent toxicity of conventional solvent mixture such as chloroform and methanol for algae oil extraction, the ethanol/n-hexane system was identified as the effective one for the oil extraction from wet-form Chlorella minutissima.

Optimization of fish oil extraction from Lophius litulon liver and fatty acid composition analysis

  • The Lophius litulon liver was used as raw material for the extraction of fish oil via various extraction methods. The extraction rate by water extraction, potassium hydroxide (KOH) hydrolysis and protease hydrolysis were compared and the results revealed the protease hydrolysis extraction had a higher extraction rate with good protein-lipid separation as observed by optical microscope. Furthermore, subsequent experiments determined neutrase to be the best hydrolytic enzyme in terms of extraction rate and cost. The extraction conditions of neutrase hydrolysis were optimized by single-factor experiment and response surface analysis, and the optimal extraction rate was 58.40 ± 0.25% with the following conditions: enzyme concentration 2,000 IU/g, extraction time 1.0 h, liquid-solid ratio 1.95:1, extraction temperature 40.5℃ and pH 6.5. The fatty acids composition in fish oil from optimized extraction condition was composed of 19.75% saturated fatty acids and 80.25% unsaturated fatty acids. The content of docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) were 8.06% and 1.19%, respectively, with the ratio (6.77:1) surpassed to the recommendation in current researches (5:1). The results in this study suggest protease treatment is an efficient method for high-quality fish oil extraction from Lophius litulon liver with a satisfactory ratio of DHA and EPA.

Supercritical $CO_2$ Extraction of Whole Berry Oil from Sea Buckthorn ($Hippopha\ddot{e}$ rhamnoides var. sp) Fruit

  • The whole berry, pulp, and seed of sea buckthorn fruit were extracted with supercritical $CO_2$ to produce edible oils. The effects of extraction pressure, temperature, and $CO_2$ flow rate on the oil yield and extraction rate were investigated, and the fatty acid composition, tocopherol, and carotenoid contents of the oils were compared. The results showed that the extraction rate was affected by pressure, temperature, and $CO_2$ flow rate and, in general, the yield increased with a rise in any of the 3 variables. Fatty acids in the whole berry and pulp oil were dominated by monounsaturated fatty acids (>64%), followed by saturated fatty acids (about 30%). In contrast, fatty acids in the seed oil consisted mainly of polyunsaturated (>60%) and monounsaturated fatty acids (>24%). The seed oil had a slightly higher content of tocopherols, but a much lower content of carotenoids, compared with the whole berry or pulp oil.

Quality Characteristics of Ginseng Seed Oil Obtained by Different Extraction Methods (추출방법에 따른 인삼씨유의 품질특성)

  • This study aimed to determine the quality characteristics of ginseng seed oil as well as evaluate the efficacy of ginseng seed oil as a food resource. Ginseng seed oil was obtained by different extraction methods; from solvent extraction oil, supercritical fluid extraction oil, and screw pressed extraction oil. Total unsaturated fatty acids were present at 97.72~97.92%. Oleic acid (80.13~81.16%) was the highest, followed by linoleic acid (14.98~15.69%). The total phenol content (mg gallic acid equivalent/100 g oil) was higher in screw pressed extraction oil ($56.32{\pm}1.47$) compared to others. ${\gamma}$-Tocopherol was only present in ginseng seed oil and screw pressed extraction oil showed the highest levels of ${\gamma}$-tocopherol ($5.95{\pm}0.25$ mg/100g oil) among the tested samples. Screw pressed extraction oil showed the greatest oxidative stability with an induction time of 16.58 hours. Acid values and peroxide values of ginseng seed oil increased with increasing storage period. The total phenol and ${\gamma}$-tocopherol contents were higher in screw pressed extraction oil than in other ginseng seed oils, which suggests that screw pressed extraction oil has the greatest oxidative stability.

Comparison of Volatile Compounds of Chamaecyparis obtusa Essential Oil and its Application on the Improvement of Atopic Dermatitis (추출방법에 따른 편백 정유의 향기 성분 비교 및 아토피 개선에의 응용)

  • Volatile flavor compounds of Chamaecyparis obtuse essential oil were extracted by simultaneous steam distillation extraction (SDE) and supercritical fluid extraction (SFE) and analyzed by GC-MS. A total of 48 and 50 components were identified in essential oil by SDE and SFE, respectively. Monoterpenes, oxygenated monoterpenes, sesquiterpenes, oxygenated sesquiterpenes, and diterpenes in essential oil by SDE were 37.24, 10.9, 9.61, 0.22, and 0.22%, respectively. In the case of SFE, they were 19.1, 23.3, 22.66, 1.31, and 10.57%, respectively. Antioxidant activities were increased with the increase of essential oil up to $80{\mu}L/mL$, irrespective of extraction method. Especially, when the essential oil concentration extracted by SDE was increased from 20 to $80{\mu}L/mL$, the antioxidant activity was increased from 10.5 to 55.1%. However, over $80{\mu}L/mL$ of essential oil, an equilibrium state was maintained. In the case of essential oil extracted by SFE, it was decreased compared to that of SDE. For the improvement of atopic dermatitis, various cosmetics such as an ato-cide soap, ato-cide spray, and ato-cide lotion containing essential oil extracted by SFE were tested. About over 90% was useful for the improvement of atopic dermatitis after 4 weeks of clinical trial targeting 40 female adults. These results demonstrate that ato-cide soap, ato-cide spray, and ato-cide lotion containing essential oil extracted by SFE could be used in functional cosmetics.

  • Grape seed oil made by press or organic solvent extraction does not contain resveratrol, a bioactive compound. Supercritical carbon dioxide could extract oil containing resveratrol from grape seed. The extraction efficiency was mainly dependent on the water content in grape seed. More resveratrol was contained in the oil extracted with un-dried grape seed. No resveratrol was extracted with dried grape seed. Time course changes of grape seed oil extraction also resulted that resveratrol could be extracted by supercritical carbon dioxide with the positive influence of water.
