Diels-Alder type adducts from the fruits of Morus alba L. (오디(Morus alba fruit)로 부터 Diels-Alder형 부가체 화합물의 분리 및 동정)

  • The fruits of Morus alba L. were extracted with 80 % aqueous MeOH, and the concentrated extract was partitioned into EtOAc, n-butyl alcohol, and water fractions. The repeated silica gel ($SiO_2$) and octadecyl silica gel column chromatographies for the EtOAc and n-butyl alcohol fractions led to isolation of two phenolic compounds. The chemical structures of the compounds were determined as Diels-Alder type adducts, mulberrofuran E (1) and chalcomoracin (2) based on spectroscopic data analyses including nuclear magnetic resonance, mass spectrometry, and infrared spectrometry. Compounds 1 and 2 were isolated for the first time from the fruits of M. alba L. in this study.

  • The leaves of eggplant (Solanum melongena L.) were extracted with 80 % aqueous MeOH, and the concentrated extract was partitioned with n-hexane, EtOAc, n-BuOH, and water fractions. From the n-BuOH fraction, five compounds were isolated through the repeated silica gel, octadecyl silica gel, and Sephadex LH-20 column chromatographies. On the basis of physic-chemical and spectroscopic data including mass spectrometry, infrared spectroscopy, and nuclear magnetic resonance, they were identified to be caffeic acid (1), chlorogenic acid (2), cryptochlorogenic acid (3), panasenoside (4), and (6R,7E,9R)-4,7-megastigmadien-3-one-9-${\beta}$-${\small{D}}$-glucopyranoside (5). Compounds 3 and 4 were isolated for the first time from the leaves of S. melongena L. in this study.

Phenolic Compounds from the Fruit Body of Phellinus linteus Increase Alkaline Phosphatase (ALP) Activity of Human Osteoblast-like Cells

  • Secondary metabolites from the fruit body of Phellinus linteus were evaluated for their proliferative effect on human osteoblast-like cells. 3-[4,5-Dimethylthiazole-2-y1]-2,5-diphenyl-tetraxolium bromide (MTT) assay and alkaline phosphatase (ALP) activity assay were used to assess the effect those isolates on the human osteoblast-like cell line (Saos-2). Activity-guided fractionation led to the isolation of ALP-activating phenolic compounds through the extraction of P. linteus, solvent partitioning, and repeated silica gel and octadecyl silica gel (ODS) column chromatographic separations. From the result of spectroscopic data including nuclear magnetic resonance (NMR), mass spectrometry (MS), and infrared spectroscopy (IR), the chemical structures of the compounds were determined as 4-(4-hydroxyphenyl)-3-buten-2-one(1), 2-(3',4'-dihydroxyphenyl)-1,3-benzodioxole-5-aldehyde (2), 4-(3,4-dihydroxyphenyl)-3-buten-2-one (3), 3,4-dihydroxybenzaldehyde (4), and protocatechuic acid methyl ester (5), respectively. This study reports the first isolation of compounds 1-3 and 5 from P. linteus. In addition, all phenolic compounds stimulated proliferation of the osteoblast-like cells and increased their ALP activity in a dose-dependent manner ($10^{-8}$ to $10^{-1}\;mg/mL$). The present data demonstrate that phenolic compounds in P. linteus stimulated mineralization in bone formation caused by osteoporosis. The bone-formation effect of P. linteus seems to be mediated, at least partly, by the stimulating effect of the phenolic compounds on the growth of osteoblasts.

  • Repeated $SiO_2$ and octadecyl silica gel (ODS) column chromatographies of the EtOAc fraction from Magnolia obovata fruits, 10 neolignans, named magnolol (1), honokiol (2), isoobovatol (3), isomagnolol (4), obovatol (5), obovatal (6), 9-methoxyobovatol (7), magnobovatol (8), obovaaldehyde (9), and 2-hydroxyobovaaldehyde (10) were isolated and identified. All isolated compounds were evaluated for in vitro cytotoxicity against seven human cancer cell lines.

  • The root bark of Morus alba L. were extracted with 80% aqueous MeOH, and the concentrated extract was partitioned with EtOAc, n-BuOH, and $H_2O$ fractions. The repeated silica gel ($SiO_2$), octadecyl $SiO_2$ (ODS), and Sephadex LH-20 column chromatographies of the EtOAc fraction led to isolation of 12 phenolic compounds. The chemical structures of the compounds were determined as sanggenol Q (1), sanggenol A (2), sanggenol L (3), kuwanon T (4), cyclomorusin (5), sanggenon F (6), sanggenol O (7), sanggenon N (8), sanggenon G (9), mulberrofuran G (10), mulberrofuran C (11), and moracin E (12). All isolated compounds were evaluated for inhibit lipopolysaccharide-induced nitric oxide production in RAW 264.7 macrophages.

  • Present study describes the optimization of disk type sorptive extraction using monolithic material (Mono Trap) for the analysis of volatile aroma compounds from Lantana camara L. in combination with gas chromatography/mass spectrometry (GC/MS). Monolithic material sorptive extraction (MMSE) is a new sampling technique using a monolithic hybrid adsorptive disk (O.D. 10 mm, 1 mm thickness) made of high purity silica and activated carbon having a large surface area chemically bonded with octadecyl silane (ODS). The experimental parameters that may influence the MMSE efficiency have been optimized. Linearity, accuracy, precision and detection limits were evaluated to assess the performance of the proposed method. The method was validated with real plant samples of Lantana camara L. Twenty eight compounds including the main representative compounds of ${\alpha}$-curcumene and ${\beta}$-caryophyllene were found in analyzed samples. Results proved that proposed method could be used as a good alternative for the analysis for such volatile aroma compounds in plant samples.

  • This study was conducted to investigate the extraction method for the determination of organochlorine, organophosphorus and synthetic pyrethroid pesticide residues in beef fat by gas chromatography-mass selective detector(GC-MSD). Twenty one pesticide residues in fat were determined using a simple and rapid procedure based on solid- phase extraction(SPE) clean-up cartridges with octadecyl $(C_{18})-bonded$ porous silica, florisil, $10\%$ deactivated florisil. a tandem $C_{18}$ and florisil and a tandem $C_{18}$ and $10\%$ deactivated florisil. Solvent-solvent extraction using acetonitrile was not satisfied to eliminate fat interference for pesticide residue analysis by GC-MSD, and the recoveries of the method in fat ranged from 16.2 to $57.3\%$ except DDT$(83.2\%)$. The recoveries of SPE methods using a tandem $C_{18}$ and Florisil was $59.6\~123.8\%$ except fenitrothion $(135.2\%)$. the SPE method was verified the satisfactory performance of pre-treatment for pesticide residues analysis in fat by GC-MSD. The efficiency of florisil deactivated with $10\%$ water has been not proved significantly on recoveries of pesticide residues in fat.

Isolation and Identification of Antioxidant Compound from the Lythrum Salicaria L. Roots (털부처꽃(Lythrum Salicaria L.) 뿌리로부터 항산화 물질의 분리 및 구조동정)

  • The roots of Lythrum salicaria L. were extracted in 80% aqueous MeOH and the concentrated extract was fractionated with EtOAc, n-BuOH, and $H_2O$, successively. The repeated silicagel and octadecyl $SiO_2$ column chromatographies of the EtOAc fractions led to isolation of an antioxidant compound and two major compounds. From the results of spectral data and the chemical characteristics including nuclear magnetic resonance, MS, and IR, the structures of compounds were determind as myricetin-3-O-${\beta}$-D-glucopyranoside (1), oleanolic acid (2), betulinic acid (3). This is the first reported isolation of compounds (1, 2) from L. salicaria. Compound 1 as well as EtOAc, n-BuOH, and $H_2O$ solvent fractions were evaluated for 2,2-dipicryl-1-phenylhydrazyl radical scavenging activity.

Analysis of volatile aroma compounds from vanilla perfume using headspace disk type monolithic material sorptive extraction (시료상층부 원판 형태 단일 다공성 물질을 이용한 바닐라 향수의 휘발성 아로마 성분 추출 분석)

  • In this study, headspace disk type monolithic material sorptive extraction (HS-MMSE) was developed, validated and applied to the analysis of volatile aroma compounds from vanilla perfume by gas chromatography -mass spectrometry (GC/MS). HS-MMSE uses monolithic material (MonoTrap) based on silica bonded with octadecyl silane (ODS) and activated carbon as a sorbent. Aroma compounds was adsorbed onto the MonoTrap in headspace and extracted by only 100 ${\mu}L$ of solvent. Total 12 volatile compounds from vanilla perfume were successfully analyzed using HS-MMSE. The influence of extractive parameters was investigated and optimized, using benzyl acetate, linalyl acetate, vanillin, ethyl vanillin as target compounds. Under the optimum condition, the limit of detection (S/N = 3) and the limit of quantification (S/N = 10) of proposed method for the target compounds were obtained within the range of 8.35~13.76 ng and 27.82~45.88 ng, respectively. The method showed good linearity with correlation coefficient more than 0.9888, satisfactory recovery and reproducibility. These results showed that HS-MMSE using disk type MonoTrap is a new promising technique for the analysis of volatile aroma compounds from vanilla perfume.

  • Sajabalssuk (Artemisia princeps PAMPANINI) was extracted with 80% aqueous MeOH, and the concentrated extract was partitioned with EtOAc, n-BuOH and $H_{2}O$, successively. From the EtOAc fraction, two phenylpropanoids were isolated through the repeated silica gel and ODS column chromatographies. From the results of physico-chemical data including NMR, MS and IR, the chemical structures of the phenylpropanoids were determined as eugenol (1) and (-)-sesamin (2). They were the first to be isolated from Sajabalssuk (Artemisia princeps PAMPANINI, Sajabalssuk). Also, eugenol inhibited LDL-oxidation with the inhibitory activity of 87.8 ${\pm}$ 1.0% at a concentration of 40 ${\mu}g/ml$.
