Optical Coherence Tomography with Sinusoidal-Wave Drive an Optical Delay Line using Piezoelectrics Strecher (정현파로 구동되는 PZT 광경로 지연기를 이용한 광 간섭 단층촬영시스템)

  • We fabricated and characterized an optical delay line for optical coherence tomography (OCT). The delay line was composed of a cylindrical piezoelectric transduce (PZT) and a single mode optical fiber. The OCT system used a duplex scanning optical delay line which was symmetrically driven in the reference and sample arms. We showed that the sinusoidal-wave was superior to a triangular-wave for driving the optical delay line for scanning depth and repeatability.

  • We report a technique that improves the spatial resolution of optical coherence tomography (OCT) by utilizing fiber-based spectral filtering. The proposed technique improves the resolution by filtering out the erbium’s characteristic peak from the amplified spontaneous emission (ASE) source spectrum, and reshaping the spectrum to Gaussian-like. We used a long period fiber grating (LPG) and an erbium doped fiber (EDF) absorber for the spectral filtering. An in-house made ASE source as well as a commercial ASE source [ASE-FL7002] was used as the OCT sources to study the proposed technique. The resolution of the OCT based on an in-house made ASE source is enhanced from 200 to 40 ㎛ with an LPG. While, the resolution of the OCT based on a commercial ASE source is enhanced from 25 to 19 ㎛ with the aid of an EDF absorber. However, sidelobes still exist in the interferogram due to imperfect spectral filtering, which limited the resolution. Further enhancement in the spatial resolution of the OCT system using the ASE source is possible with the aid of cascaded LPGs and/or carefully designed EDF absorber.

  • We report an application of full-field optical coherence tomography (FF-OCT) for identifying counterfeit bank notes. The depth-resolved imaging capability of FF-OCT was used for tomographic identification of superficially-identical objects. By retrieving the internal structures of the security feature (cash hologram) of an original banknote, we could demonstrate the feasibility of FF-OCT to identify counterfeit money. The FF-OCT images showed that the hologram consisted of micron scale multi-coated layers including an air gap. Therefore, it is expected that FF-OCT has potential as a new non-invasive tool to discern imitation of currency, and it would find applications in a wide field of counterfeit sciences.

  • In this study, we developed an optical coherence tomography(OCT) using a sweep-source laser whose center wavelength is 1310 nm and a probe of hand-held type. The developed hand-held probe targets to diagnose the middle ear, so it is miniaturized for ease and convenience of control. For the first time, we performed in-vivo clinical experiments on tympanic membrane(TM) perforation patients not reported from previous studies about OCT. The high-resolution sectional images of tympanic membrane perforation can be obtained. There aren't many studies about diagnostic instrument of the middle ear and diagnosis with sectional image of the middle ear, so the developed OCT system and hand-held probe are applicable to tympanic membrane perforation and other pathologic diagnosis in the middle ear.

Development of Real-time Screening System for Superior Melon Seeds Using Optical Coherence Tomography (광간섭 단층촬영법을 이용한 우량 참외 종자 실시간 감별 시스템 개발)

  • We developed a real-time screening system using optical coherence tomography (OCT) to distinguish the fruitful melon seeds efficiently. Cross-section images of melon seeds infected with Cucumber green mottle mosaic virus (CGMMV) showed an additional layer that did not appear in normal seeds. Additional layer appeared under $100{\sim}300{\mu}m$ from the surface of the seed. OCT can visualize the micro-structural and morphological changes of the internal seed structure. Real-time OCT seed screening system provided the real-time, non-destructive, cross-section image and quantitative information such as A-scan analysis of selected region in the cross-section image. We can distinguish the viral infection seeds while monitoring the averaged A-scan analysis graph in real-time by considering the second peak value of the graph which refers to the layer that occurred owing to the virus. Real-time OCT seed screening system could assist to distinguish the disease caused by CGMMV.

( Partitioning of Carbon and Nitrogen Reserves During Winter Adaptation and Spring Regrowth III.   • The objective of this study is to obtain the basic data for investigating the effects of organic reserves on winter survial or regrowth yield. Forage rape(Brassica napus L.) was sown on Sep. 1, 1994. Experimental plots were divided into three replicates under 6 different cutting dates(l0 days interval from Oct. 15 to Dec. 4). Field-grown palnts were sampled on the each cutting date and on the wintering period (Jan. 16) to analyze the nitrogen and non-structural cahohydrate reserves. The rate of winter survival and regrowth yield were also measured in the spring of next year. On the before wintering, dry matter yields were 152, 274, 500, 718, 776 and 981 kg/lOa, respectively, from the cutting date on Oct. 15, Oct. 25, Nov. 4, Nov. 14, Nov. 24, and Dec. 4. Cmde protein yield significantly increased as cutting date was later until Nov. 14, thereafter a significant increase did not occured. Nitrogen and starch contents per plant significnatly increased as the cutting date was later. The increasing rate of starch was greatly higher than that of nitrogen. On the wintering period, nitrogen reserves in mts were 85.3, 68.8, 47.6, 28.3, 44.3, and 55.3 mglplant, and starch reserve were 11 1.3, 75.3, 39.3, 19.6, 26.4 and 34.6 mglplant, respectively, in the plots cut on Oct. 15, Oct. 25, Nov. 4, Nov. 14, Nov. 24, and Dec. 4. It showed that carbohydrate reserves were much highly utilized than nitrogen reserves during wintering period. The rates of winter survival were 91, 83, 46, 22, 35 and 43% and regrowth yields were 692, 545, 316, 84, 127 and 140 kgD.M/lOa, respectively, in each plots. The highly significant correlation (p<0.01) between the level of organic reserves and the rate of winter survival or regrowth yield were obtained.

Expression of CD133, CD44, CK7, and OCT4 in Animal Cancers

  • Cancers are mainly sustained by a small pool of neoplastic cells, known as cancer stem cells or tumorinitiating cells. These cells possess the ability to self-renew and proliferate, and are thus able to form the tumor. In the present study cells that correspond to cancer stem cells in mammary and liver cancers in animals were identified by the expression of CD133, CD44, CK7, and OCT4 using immunochemistry. As a result, we found with CD133+ and CD44+ cancer stem cell-like phenotypes in mouse and canine hepatocellular carcinoma and canine mammary gland tumors. However, CK7+ and OCT4+ cells were not identified in animal mammary and liver cancer. CD133+ and CD44+ cells are wellknown stem cell lines and play key roles in development and metastasis in human cancer. These findings suggest that cancer stem cells are involved in animal tumorigenesis and may provide insight into mechanisms in cancer development as well as cancer diagnostics.

Inter-device agreement between spectral domain optical coherence tomography, ultrasound biomicroscopy, and gonioscopy in evaluating the iridocorneal angle in normotensive dogs

  • Background: There has not been a thoroughly reported study of the comparison between spectral domain-optical coherence tomography (SD-OCT) with both ultrasound biomicroscopy (UBM) and gonioscopy on the evaluation of the iridocorneal angle (ICA) in dogs. Objectives: To investigate the diagnostic value of SD-OCT for the early detection of narrowing ICA by comparing and assessing inter-device agreement in anterior chamber angle (ACA) measurements obtained by SD-OCT and UBM, and ICA evaluations by gonioscopy. Methods: A total of 28 eyes from 28 client-owned dogs with normal intraocular pressure were included for examination. The ACA and angle opening distance (AOD) were measured from the SD-OCT and UBM images, and gonioscopy images were analyzed using the ICA grade and ZibWest angle index. Results: The mean ACA and AOD for SD-OCT were 28.31° ± 5.37° and 658.42 ± 219.90 ㎛, and for UBM, 28.34° ± 5.82° and 859.29 ± 221.80 ㎛, respectively. The mean difference in ACA between the average values of SD-OCT and UBM measurements was 0.03° with a 95% limit of agreement (LoA) span of 16.2°, indicating positive agreement; that in AOD was 200.85 ㎛ with a 95% LoA span of 1,110.95 ㎛, indicating poor agreement. The Pearson correlation coefficient of the ACA of SD-OCT and ZibWest indices of gonioscopy was 0.624, indicating strong agreement; that of UBM and gonioscopy was 0.43, indicating moderate agreement. Conclusions: SD-OCT is well tolerated by canine patients due to its non-contact method and might be an alternative option for early screening of ICA narrowing in clinical settings.

  • Recently, pluripotency induction or cellular reprogramming by introducing critical transcription factors has been extensively studied, but has been demonstrated only in vitro. Based on reports that Oct4 is critically involved in transforming neural stem cells into pluripotent cells, we used the lentiviral vector to introduce the Oct4 gene into the hippocampal dentate gyrus (DG) of adult mice. We examined whether this manipulation led to cellular or behavioral changes, possibly through processes involving the transformation of NS cells into pluripotent cells. The Oct4 lentivirus-infused group and the green fluorescent protein lentivirus-infused group showed a similar thickness of the DG and a comparable level of synaptophysin expression in the DG. Furthermore, our behavioral analyses did not show any differences between the groups concerning exploratory activity, anxiety, or memory abilities. This first trial for pluripotency induction in vivo, despite negative results, provides implications and information for future studies on in vivo cellular reprogramming.
