Effects of treadmill exercise on the regulatory mechanisms of mitochondrial dynamics and oxidative stress in the brains of high-fat diet fed rats

  • [Purpose] The purpose of this study was to investigate the effects of treadmill exercise on oxidative stress in the hippocampal tissue and mitochondrial dynamic-related proteins in rats fed a long-term high-fat diet (HFD). [Methods] Obesity was induced in experimental animals using high fat feed, and the experimental groups were divided into a normal diet-control (ND-CON; n=12), a high fat diet-control (HFD-CON; n=12) and a high fat diet-treadmill exercise (HFD-TE; n=12) group. The rats were subsequently subjected to treadmill exercise (progressively increasing load intensity) for 8 weeks (5 min at 8 m/min, then 5 min at 11 m/min, and finally 20 min at 14 m/min). We assessed weight, triglyceride (TG) concentration, total cholesterol (TC), area under the curve, homeostatic model assessment of insulin resistance, and AVF/body weight. Western blotting was used to examine expression of proteins related to oxidative stress and mitochondrial dynamics, and immunohistochemistry was performed to examine the immunoreactivity of gp91phox. [Results] Treadmill exercise effectively improved the oxidative stress in the hippocampal tissue, expression of mitochondrial dynamic-related proteins, and activation of NADPH oxidase (gp91phox) and induced weight, blood profile, and abdominal fat loss. [Conclusion] Twenty weeks of high fat diet induced obesity, which was shown to inhibit normal mitochondria fusion and fission functions in hippocampal tissues. However, treadmill exercise was shown to have positive effects on these pathophysiological phenomena. Therefore, treadmill exercise should be considered during prevention and treatment of obesity-induced metabolic diseases.

한국 비만남성들의 외모 관련 삶의 질에 대한 탐색적 연구 (An Exploratory Study on Appearance Related Quality-of-Life of Obese Korean Men)

  • This study explored various issues related to the quality-of-life for obese Korean men. In-depth interviews were conducted with thirty men over BMI 25 whose age ranged from 20's to 50's. Projective techniques along with semi-structured interviews were used. The results of interviews were analyzed based on grounded theory. "Stress" was the main phenomenon resulting from the causal conditions of obesity and stigma. Stress consisted of social, psychological, physical and environmental factors. Contextual conditions mediating the relationship between causal conditions and main phenomenon were obesity level, pressure, and public appearance. Avoidance, change of personal traits, and insensitivity were the action/reactions to the main phenomenon. Various factors due to obesity disappeared and quality-of-life increased when they succeed in weight reduction; however, factors related to obesity and lowered condition of quality-of-life continued if they experienced a weight gain/loss yo-yo.

Can antioxidants be effective therapeutics for type 2 diabetes?

  • The global obesity epidemic and the growing elderly population largely contribute to the increasing incidence of type 2 diabetes. Insulin resistance acts as a critical link between the present obesity pandemic and type 2 diabetes. Naturally occurring reactive oxygen species (ROS) regulate intracellular signaling and are kept in balance by the antioxidant system. However, the imbalance between ROS production and antioxidant capacity causes ROS accumulation and induces oxidative stress. Oxidative stress interrupts insulin-mediated intracellular signaling pathways, as supported by studies involving genetic modification of antioxidant enzymes in experimental rodents. In addition, a close association between oxidative stress and insulin resistance has been reported in numerous human studies. However, the controversial results with the use of antioxidants in type 2 diabetes raise the question of whether oxidative stress plays a critical role in insulin resistance. In this review article, we discuss the relevance of oxidative stress to insulin resistance based on genetically modified animal models and human trials.

사상체질(四象體質)과 비만(肥滿)에 관한 임상적(臨床的) 연구(硏究) (A Clinical study on the Sasang Constitution and Obesity)

  • Objectives To study on the relation between Sasang constitution and obesity and that of obese patient's clinical symptom. Methods The subjects were 129 female patients to visit to diet. The subjects were 129 female patients to diet who answered questionnaire QSCC II and were diagnosed by the Sasang constitution from May to December in 2001 at Nara oriental medical clinic. Results 1. Of obese patients 79.1% were Taeumin, 11.6% Soeumin, and 9.3% Soyangin. 2. A relation of a grade of obesity and Sasang constitution. 2-1) 45.8% of the obese patients were high level obesity who had over 35% of body fat. Out of these people, 91.5% were Taeumin. 2-2) 40.3% of the obese patients weight hip ratio was between 0.84 to 0.89 and 84.6% of these people were proved to be Taeumin. 2-3) 39.2% of the people who had a minimum weight hip ratio of 0.90 were significantly Taeumin, 6.7% Soeumin and 16.6% Soyangin. 2-4) 65.7% of Taeumin were obese patients. 3. Obesity treatment with constitution. 3-1) 59.8% of Taeumin were Pyo-zng patients. 3-2) As treatment periods tended to increase, Taeumin and Soeumin patients showed a large decling of body fat, whereas Soeumin patients showed the opposite reaction. 3-3) The weight hip ratio was led by Taeumin with 0.03 and then came Soeumin and Soyangin with a 0.02 ratio. As treatment was taken into process longer Taeumin patients showed a great progress in decreasing their weight hip ratios. 4. Costitutional related symptom with Obesity. 4-1) Taeumin was followed by Soyangin and then Soeumin in family history. 4-2) Soeumin was followed by Taeumin, then Soyangin when complain of physical fatigue was taken into consideration. 4-3) The ordinary exercise was led by Soyangin, Taeumin then Soeumin. 4-4) People who receive stress turned out to be 72.9% much higher than the people who don't. 4-5) Overeating due to stress was found in 83.3% of Soyangin.

간호대학생의 비만스트레스와 건강신념이 체중조절행위에 미치는 영향 (Effects of Obesity Stress and Health Belief on Weight Control Behavior among Nursing Students)

  • 본 연구는간호대학생의 비만스트레스와 건강신념이 체중조절행위에 미치는 영향을 파악하기 위해 시도되었다. 자료 수집은 2016년 9월 1일부터 10월 10일까지 U시에 소재한 대학교에 재학 중인 간호대학생을 대상으로 설문조사를 실시하였으며, 수집된 자료는 SPSS 23.0프로그램을 이용하여 t-test, ANOVA, Scheffe test, Person's correlation coefficients와 위계적 다중회귀분석(Hierarchical multiple regression analysis)으로 분석하였다. 대상자의 일반적 특성 중 성별(t=2.30, p=.044), 건강상태(F=8.03, p<.001)가 체중조절행위에 유의하게 영향을 주었다. 체중조절행위의 식이요법(r=-.26, p<.001), 운동요법(r=-.25, p<.001)과 행동수정요법(r=-.29, p<.001)이 지각된 장애성과 유의한 음의 상관관계가 있으며, 체중조절행위의 전문지식획득은 비만스트레스(r=.42, p<.001), 지각된 민감성(r=.25, p<.001)과 지각된 유익성(r=.19, p<.001)이 유의한 양의 상관관계가 있는 것으로 나타났다. 또한, 간호대학생의 체중조절행위에 유의한 영향을 준 것은 건강신념 중 지각된 장애(${\beta}=-.30$, p<.001)와 비만스트레스(${\beta}=.20$, p<.05) 순으로 나타났고, 이들 변인들의 총 설명력은 17.2%이었다. 이상의 결과를 토대로 간호대학생의 체중조절행위에 영향을 주는 성별, 건강상태 및 비만스트레스와 건강신념의 변인들을 이용하여 체중조절행위를 실천할 수 있는 효율적인 교육 및 프로그램의 개발과 적용이 요구된다.

성인여성의 성격유형이 건강통제행동 및 비만스트레스에 미치는 영향 (A Study on the Effect of Adult Female's Personality Type on Health Control Behavior and Obesity Stress)

  • 본 연구는 성인여성들을 대상으로 성격특성을 알아보고 건강통제행동 및 비만스트레스에 미치는 영향에 대하여 알아보기 위해 인천지역 소재 여성 257명을 대상으로 설문조사를 실시하였는데 그 결과를 요약해 보면 다음과 같다. 첫째, 조사대상자의 BMI (체질량 지수)를 살펴보면 저체중 30명 (11.7%), 정상체중 170명 (66.1%), 과체중 33명 (12.8%), 비만 24명 (9.4%)으로 나타나 대체적으로 정상체중 범위가 많은 것으로 볼 수 있다. 이에 따라 건강 통제행동 및 비만스트레스에 차이가 있는지를 분석한 결과 식생활관련 통제행동, 비만스트레스에 대해 통계적으로 유의미한 차이가 나타났으며 (p <.05), 저체중이나 정상체중에 비해 과체중이나 비만의 경우 식생활관련 통제행동이 강한 것으로 볼 수 있으며, 이에 따른 비만스트레스도 높게 나타나는 경향을 보였다. 둘째, 성인여성의 성격유형 (친화성, 성실성, 외향성, 신경증, 개방성)이 건강통제행동에 미치는 영향을 검증한 결과 자신에게 주어진 일에 대해서 성실하게 철저하게 이행하며, 성취욕구가 강하고, 항상 어떤 일을 해내고자 하는 성실한 성향이 강하고 새로운 것에 개방적이고, 지적으로 민감한 특성을 지니는 개방적 성향이 강할수록 건강이나 식생활을 통한 통제행동이 적극적인 것으로 나타난 반면, 정서적으로 안정되어 있지 못하고, 자신의 충동을 잘 조절하지 못하며, 스트레스에 잘 대처하지 못하는 신경증 성향이 강할수록 건강이나 식생활을 통한 통제행동은 약해지는 경향을 나타내었다. 셋째, 성인여성의 성격유형 (친화성, 성실성, 외향성, 신경증, 개방성)이 비만스트레스에 미치는 영향을 검증한 결과 정서적으로 안정되어 있지 못하고, 자신의 충동을 잘 조절하지 못하며, 스트레스에 잘 대처하지 못함을 나타내는 신경증 성향이 강할수록 비만스트레스는 높아지는 것으로 볼 수 있으며, 타인을 신뢰하고 관심을 가지며, 솔직하고 순응적인 태도를 나타내는 친화적 성향이 강할수록 비만스트레스 또한 적은 것으로 나타났다. 전체적으로 건강통제행동은 성격유형 중 내적 성향이 강한 성인여성이 비만스트레스에 유의한 영향을 미치는 것을 알 수 있으며, 주관적 규범과 지각된 행동통제로 이어져 이는 정서적으로 안정되지 못 할수록 자신의 충동을 잘 조절하지 못해 스트레스 관리에 잘 대처하지 못하는 것으로 나타났다.

고지방식이에 의한 말초 생체시계 유전자 발현 변화 (High-fat Intake is Associated with Alteration of Peripheral Circadian Clock Gene Expression)

  • Purpose: Recent studies demonstrated disruption of the circadian clock gene is associated with the development of obesity and metabolic syndrome. Obesity is often caused by the high calorie intake, In addition, the chronic stress tends to contribute to the increased risk for obesity. To evaluate the molecular mechanisms, we examined the expression of circadian clock genes in high fat diet-induced mice models with the chronic stress. Methods: C57BL/6J mice were fed with a 45% or 60% high fat diet for 8 weeks. Daily immobilization stress was applied to mice fed with a 45% high fat for 16 weeks. We compared body weight, food consumption, hormone levels and metabolic variables in blood. mRNA expression levels of metabolic and circadian clock genes in both fat and liver were determined by quantitative RT-PCR. Results: The higher fat content induced more severe hyperglycemia, hyperlipidemia and hyperinsulinemia, and these results correlated with their relevant gene expressions in fat and liver tissues. Chronic stress had only minimal effects on metabolic variables, but it altered the expression patterns of metabolic and circadian clock genes. Conclusion: These results suggest that the fat metabolism regulates the function of the circadian clock genes in peripheral tissues, and stress hormones may contribute to its regulation.

비만아동과 비 비만아동 간 스트레스저항 차이와 비만도 집단 간 스트레스저항 차이 분석 (Differences in Stress Resistance Level Felt by Obese and Normal Child, and Their Level of Obesity)

  • 본 연구는 아동들의 스트레스의 적절한 조절을 통하여 비만 현상을 개선하거나 예방을 할 수 있다는 이론에 의거하여 2017년 7월부터 2017년 9월까지 K시 K센터에 내방한 240명의 아동들을 대상으로 체성분 분석과 뇌파분석을 실시하였다. 분석한 자료를 토대로 비만 아동과 비비만 아동 간 스트레스저항 차이를 분석하고, 비만아동의 비만도 집단 간 스트레스 저항의 차이를 보고자 하였다. 뇌파측정을 통한 뇌기능은 시계열 선형분석 방법을 사용하였다. 연구의 결과로는 첫째, 비만아동이 비비만아동보다 스트레스 저항이 낮았다. 둘째, 비만도가 높을수록 항 스트레스지수가 낮았다. 이 결과는 비만으로 인한 질병이나 심리적인 증상 등이 스트레스의 원인이 되며, 비만과 스트레스 사이 밀접한 관계가 있다는 점을 시사한다. 또한, 과학적이고 객관적인 신경생리학적 지표인 뇌기능분석을 통해 유의함을 발견한 결과 도출 과정이 연구의 신뢰성을 주고 있다고 본다. 비만아동들의 비만 해결과 예방적 차원에서 본 연구의 결과가 체내의 생리, 생화학적 변화, 정신활동에 긍정적인 효과를 제공하는데 중요한 지표가 되었다고 사료된다. 제언으로 지속적인 효과 검증이 요구된다.

청소년의 신체활동 및 운동 실태와 관련 요인 (The State and Factors Associated with the Level of Physical Activity and Exercise in Adolescents)

  • Purpose: The purpose of this study was to investigate the state and factors associated with level of physical activity and exercise in adolescents. Method: The subjects in this study included 528 adolescents registered at two junior high schools and two senior high schools in J City. The data were collected between October 20 and 30, 2006. Result: The study results showed that 23.8% of the adolescents engaged in vigorous activities for at least 20 minutes, three times a week, while 3.8% of the adolescents participated in moderate activities for at least 30 minutes, five times a week. The subjective level of health was determined to be $3.51\pm.91$ points on a 5-point scale. The level of stress was $2.58\pm.83$ points on a 4-point scale. The obesity index was $4.26\pm20.84$ points. Significant correlations were found between the subjective level of health and level of stress (r=-.201, p=.000), the subjective level of health and the obesity index (r=-.135, p=.004), and between the level of stress and the obesity index (r=.107, p=.024). Conclusion: These results showed that developing a health promotion program that includes an exercise program specific to gender and level of education, individualized strategies of stress control and optimal weight control is required.

The Relationship between Oxidative Stress and Oxidative Markers

  • Metabolic syndrome such as, hypertension, diabetes, obesity, dyslipidemia etc are well known lifestyle diseases which threaten a health state. The purpose of this study is to evaluate antioxidant status and reactive oxygen species (ROS) in various stressful conditions. A group of persons (n=28) were enrolled in this study and stress status was evaluated using questionnaire of psychological well-being index (PWI-SF) and hematologic analysis of free oxygen radical defense (FORD), free oxygen radical test (FORT), lipid profiles and serum cortisol level. Analysis was done according to stress index, BMI status and overtime working. Blood samples from diabetics were used for control. Results are as in followings: Higher PWI-SF group showed higher FORT. Obese persons with BMI > $25kg/m^2$ showed statistically lower FORD (p<0.05) and higher FORT level (p<0.01). When compare with diabetics, an overweight group had similar levels in FORD and FORT while a normal body weight group showed quite different with diabetics in FORD (p<0.001). Higher ROS group has higher triglyceride, CRP and insulin levels but lower in FORD and HDL-cholesterol levels. Overtime working at night showed no meaningful result against our expectations. These results suggested that obesity showed most unfavorable correlations in an antioxidant status in various situations.
