While the Coronavirus disease 2019 (COVID-19) pandemic is gradually turning into an endemic disease, concerns about post COVID-19 conditions (Long COVID) are emerging. Obesity is a major risk factor for severe complications of COVID-19, and COVID-19 has a wide range of effects on obesity and metabolic function. This paper aims to examine the interaction between COVID-19 and obesity, the effects and mechanisms of long COVID on obesity, and the role of Korean medicine on long COVID-related obesity. Obesity may worsen with cardiometabolic damage and psychosocial insecurity during COVID-19 and long COVID-induced neuroinflammation, systemic inflammation, mitochondrial dysfunction, and hypoxia also may aggravate obesity. Korean Medicine treatments, which have been widely used to treat obesity, have the potential to improve obesity in the era of long COVID by intervening in these mechanisms.
Although it has recently been reported that Acanthopanax senticosus has a protective effect against the development of metabolic disorders such as diabetes and hyperlipidemia, its underlying molecular mechanisms have remained to be elucidated. In this study, we observed that treatment of an extract of A. senticosus (ASE) reduced body weight and adiposity and improved glucose tolerance in high-fat diet (HFD)-induced obese mice. HFD-induced weight gain in white adipose tissues and liver was also significantly reduced in ASE-treated HFD-fed mice, which was found to be contributed by suppression of mRNA expression of genes involved in fatty-acid uptake and fat synthesis in white adipose tissue. ASE treatment also attenuated M1 macrophage activation, increased regulatory T ($T_{reg}$) cell population, and modulated leptin/adiponectin profile that might alleviate chronic inflammation to protect against HFD-induced insulin resistance. Our findings suggest a therapeutic potential of ASE for prevention and treatment of type 2 diabetes without adverse effects such as weight gain and dyslipidemia.
Obesity-induced inflammation plays a crucial role in obesity-related pathologies such as type II diabetes and atherosclerosis. Adipose tissue macrophages and the cell-derived proinflammatory chemokines are key components in augmenting inflammatory responses in obesity. Anthocyanins such as cyanidin and $cyanidin-3-O-{\beta}-D-glucoside$ (C3G) are known to elicit anti-inflammatory activities by suppressing the production of proinflammatory mediators such as tumor necrosis factor alpha and nitric oxide in LPS-stimulated macrophages. In the present study, we investigated whether cyanidin and C3G have the potential to suppress the inflammatory responses of adipose cells. Cyanidin and C3G not only suppressed the migration of RAW 264.7 macrophages induced by mesenteric adipose tissue-conditioned medium, but also inhibited the activation of the cells to produce inflammatory chemokines such as monocyte chemoattractant protein-1 (MCP-1) and macrophage inflammatory protein-related protein-2 (MRP-2) in a dose-dependent manner. Cyanidin and C3G also inhibited the release of MCP-1 and MRP-2 from adipocytes and/or macrophages. These findings suggest that cyanidin and C3G may suppress the inflammatory responses of adipose tissue in obesity.
Objectives: Recent data have revealed that the plasma concentration of inflammatory mediators is increased in the insulin-resistant states of obesity and type 2 diabetes. The purpose of this study was to investigate the antidiabetic and anti-obesity effect of Massa Medicata Fermentata on obese type 2 diabetes mice. Methods: In order to examine the effects of Massa Medicata Fermentata, obese type 2 diabetes mice induced by Surwit's high fat, high sucrose diet. Mice were divided into 4 groups of ND (normal diet), HFD (high fat and high sucrose diet), Met (high fat and high sucrose diet with metformin) and MMF (high fat and high sucrose diet with Massa Medicata Fermentata) and investigated over 8 weeks. Diabetic and obese clinical markers, including body weight, glucose level, lipid level, leptin concentration, epididymal fat pad and liver weights and adipose tissue macrophage (ATM) were determined. Results: Compared with the HFD group, body weight, fructosamine, triglyceride, epididymal fat pad weight and ATM were significantly reduced in the MMF group. Conclusions: From the above results, the intake of Massa Medicata Fermentata may be effective in anti-hyperglycemia and anti-obesity by the attenuation of glucose and lipid levels and also inflammation state. Massa Medicata Fermentata may be beneficial for controlling diabetes mellitus type 2 in humans.
본 연구는 고지방식이를 섭취한 난소절제 쥐의 지방조직에서 제니스테인에 의한 지방조직감소와 항염증에 대한 운동의 영향을 조사하였다. 고지방식이를 섭취한 쥐(C)에 비해 제니스테인 단독처리(G)와 운동 단독처리(Ex)는 지방조직무게, 지방세포 크기 및 혈청 속 포도당과 유리 지방산 수치를 감소시켰으며, 이러한 현상은 제니스테인과 운동을 동시처리(G/Ex) 하였을 경우 더 효과적으로 감소하였다. 지방조직에서 C에 비해 G와 Ex의 지방생성 유전자와 염증성 사이토카인 유전자의 발현은 모두 감소하였고, 특히 이러한 유전자의 감소 현상은 G와 Ex 각각 보다 G/Ex에서 더욱 효과적이었다. 따라서 본 연구는 고지방식이를 섭취한 난소절제 쥐에서 제니스테인과 운동의 동시처리는 제니스테인 단독처리에 비해 더 효과적으로 비만과 지방조직 염증을 개선하였으며 이것은 지방생성 유전자를 긍정적으로 조절한 결과임을 밝혔다. 본 연구는 제니스테인의 항비만과 지방조직 항염증에 대해 운동이 유익한 영향을 미친다는 것을 제안한다.
Objectives: There is a steady increase in the prevalence of obesity worldwide and obesity is often accompanied by inflammation. Although much emphasis has been placed on the adipose tissue inflammation in obesity, a study with herbal medicine is few. This study aimed to investigate the antidiabetic and anti-inflammatory effect of a complex herbal medicine (CHM) composed of Cornus officinalis, Dioscorea rhizoma, Aurantii fructus, and Mori Foliumon on obese type 2 diabetes mice. Methods: Type 2 diabetes mellitus and obesity were induced by Surwit's high fat, high sucrose diet for 8 weeks. Mice were divided into ND (normal diet, n=10), HFD (high fat and high sucrose diet, n=10), CHM (high fat and high sucrose diet with complex herbal medicine, n=10) and Met (high fat and high sucrose diet with metformin, n=10) groups. The body weight, fructosamine and OGTT (oral glucose tolerance test) were measured. After 8 weeks the blood samples of all mice were taken from the heart, and lipid profiles were measured. Epididymal fat pad, histological size of the adipocyte tissue and liver weights were measured. Inflammatory markers such as leptin and adipocyte tissue macrophage were measured to evaluate the effect of CHM on adipocyte tissue inflammation. Results: Compared with the HFD group, there was an improvement in OGTT and epididymal fat decreased in the CHM group. White adipocyte size and adipocyte tissue macrophage decreased in CHM group. Conclusions: These results suggest that CHM has antidiabetic and anti-inflammatory effects in high fat, high sucrose diet induced obese mice.

Sirtuin 1 (SIRT1) is a mammalian $NAD^+$-dependent protein deacetylase that regulates cellular metabolism and inflammatory response. The organ-specific deletion of SIRT1 induces local inflammation and insulin resistance in dietary and genetic obesity. Macrophage-mediated inflammation contributes to insulin resistance and metabolic syndrome, however, the macrophage-specific SIRT1 function in the context of obesity is largely unknown. C57/BL6 wild type (WT) or myeloid-specific SIRT1 knockout (KO) mice were fed a high-fat diet (HFD) or normal diet (ND) for 12 weeks. Metabolic parameters and markers of hepatic steatosis and inflammation in liver were compared in WT and KO mice. SIRT1 deletion enhanced HFD-induced changes on body and liver weight gain, and increased glucose and insulin resistance. In liver, SIRT1 deletion increased the acetylation, and enhanced HFD-induced nuclear translocation of nuclear factor kappa B (NF-${\kappa}B$), hepatic inflammation and macrophage infiltration. HFD-fed KO mice showed severe hepatic steatosis by activating lipogenic pathway through sterol regulatory element-binding protein 1 (SREBP-1), and hepatic fibrogenesis, as indicated by induction of connective tissue growth factor (CTGF), alpha-smooth muscle actin (${\alpha}$-SMA), and collagen secretion. Myeloid-specific deletion of SIRT1 stimulates obesity-induced inflammation and increases the risk of hepatic fibrosis. Targeted induction of macrophage SIRT1 may be a good therapy for alleviating inflammation-associated metabolic syndrome.
Objective: This study was undertaken to investigate the effects of Mori folium on insulin resistance and adipose tissue inflammation in an experimental mouse model of obesity.Methods: Obesity was induced in C57BL/6 mice by feeding them a high-fat diet. The mice were divided into four groups (n=6): a normal diet, high-fat diet, high-fat diet with 40 mg of Mori folium, and high-fat diet with 800 mg of Mori folium groups. After 13 wk, the body weights, fasting blood glucose and fasting serum insulin levels, insulin resistance (homeostatic model assessment) levels, oral glucose tolerance test levels, epididymal fat and liver weights, and gene expression of tumor necrosis factor-α, interleukin-6, and interferon-γ were measured. In addition, adipose tissue macrophages were analyzed by fluorescence-activated cell sorting.Results: Mori folium significantly reduced blood glucose levels, oral glucose tolerance levels, and liver weights. It also reduced adipose tissue macrophage numbers and tumor necrosis factor receptor-α gene expression.Conclusions: These results show that Mori folium has insulin resistance reduction and anti-inflammatory effects in an experimental mouse model of obesity.
27-Hydroxycholesterol (27OHChol), a prominent cholesterol metabolite present in the bloodstream and peripheral tissues, is a kind of immune oxysterol that elicits immune response. Recent research indicates the involvement of 27OHChol in metabolic inflammation (meta-inflammation) characterized by chronic responses associated with metabolic irregularities. 27OHChol activates monocytic cells such that they secrete pro-inflammatory cytokines and chemokines, and increase the expression of cell surface molecules such as pattern-recognition receptors that play key roles in immune cell-cell communication and sensing metabolism-associated danger signals. Levels of 27OHChol increase when cholesterol metabolism is disrupted, and the resulting inflammatory responses can contribute to the development and complications of metabolic syndrome, including obesity, insulin resistance, and cardiovascular diseases. Since 27OHChol can induce chronic immune response by activating monocyte-macrophage lineage cells that play a crucial role in meta-inflammation, it is essential to understand the 27OHChol-induced inflammatory responses to unravel the roles and mechanisms of action of this cholesterol metabolite in chronic metabolic disorders.
Objectives : Obesity is a public health concern associated with chronic diseases including hyperlipidemia, diabetes, fatty liver, atherosclerosis and cancer. As several anti-obesity drugs have been limited owing to their side effects, the development of new anti-obesity drugs through herbal medicines has been increasing. Cynanchum Wilfordii Radix (CW) traditionally is consumed for various health benefits including immune enhancing, anti-inflammation and anti-tumor activities. The aim of the present study is to evaluate the effects of CW on High fat diet (HFD)-induced obese mice. Methods : The mice were randomly divided into four groups (n=7). The mice were respectively fed a normal diet (ND), a high-fat diet (HFD), HFD plus CW (50 mg/kg/day), HFD plus CW (100 mg/kg/day). All groups were assayed for body weights, food efficiency ratio, blood biochemistry parameters, and organic tissue weights. Results : HFD-fed mice showed an increase in the body weight and serum biochemistry parameters levels (total cholesterol and triglycerides) as well as organic tissue weights. However, the administration of CW to obese mice induced a reduction in their body weight, food efficiency ratio, blood biochemistry parameters and weight of liver and fat compared with the HFD fed mice. Additionally, we observed that CW inhibited the lipid accumulation in liver and serum lipid parameter induced by HFD. Conclusions : Taken together, the findings of this study suggest that CW may be a potential agent for use in the treatment of obesity and obesity-related metabolic diseases.
