Isoliquiritigenin Inhibits Ovalbumin-induced Cyclooxygenase-2 and Inducible Nitric Oxide Synthase Expression

  • Egg allergy has been raised as the most prevalent food hypersensitivity in children. Ovalbumin (OVA), which is one of the major egg allergens, is the major protein in egg white. Isoliquiritigenin (ILG) is an active component present in licorice and has been used to treat many chronic diseases for many years. In this study, we present biochemical evidence that ILG inhibits OVA-induced nuclear factor-${\kappa}B$ (NF-${\kappa}B$) activation. ILG also inhibits OVA-induced cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) expression. These data suggest that ILG has anti-allergy effects.

과잉배란처치면양에 있어서 수술적방법에 의한 난회수법의 검토 (Studies on Flushing Ova from the Sheep Uterus)

  • A comparative study on the two techniques of uterine flushing in superovulated sheep was conducted. The first method was carried out by flushing of the horn through a Foley catheter inserted into the uterine horn (method I ). The second method involved flushing of the horn with the medium injected into uterine tip nea. the utero-tubal junction through a Foley catheter inserted into uterine horn(method II). Superovulation was carried out on day 8 of the estrous cycle by a FSH-PG method. Ova were flushed on 6 or 7 days after mating. The recovery rates in methods I and II were 68.0%(51/75) and 43.6%(17/39), respectively(p<0.05). It indicated that method I is effective technique for flushing of the sheep ova from the uterus.

황금이 천식모릴 생쥐의 혈청 사이토카인에 미치는 영향 (Effect of Scutellariae Radix Extract on Serum Cytokines in OVA-induced Asthmatic Mice)

  • The purpose of this research is to examine the effects of Scutellariae Radix(SR) extract on cytokines in ovalbumin (OVA)-induced asthmatic mice. In vivo, C57BL/6 mice were sensitized and handicapped by OVA for 12 weeks. During this experiment, the one group was then treated with SR extract for the later 8 weeks (3 times per week) and analyzed by ELISA. There were significant decreases in IL-4(p<0.05), IL-5(p<0.05), IL-13(p<0.01), histamine(p<0.05) in serum of SR group. IgE also decreased, but was not significant compared with that of control group. The results of this study support a role for SR as an effective treatment for asthma in its experimental success in significantly decreasing inflammation and asthma reactions.

  • Astragali Radix(AR), is a popular tonic herb prescribed for 'insufficient qi' in Korea, Japan and China. The present study examined the effect of AR ethanol extract on ovalubumin induced allergic mouse model. AR administration reduced levels of IFN-gamma, Interleukin(IL)-4, IL-5 and total IgE in the OVA induced allergic inflammation. It also protected the upper airway respiratory epithelium from being damaged by the OVA induced inflammation. Taken together, our results showed that the use of AR alone proved to down-regulate Th1 and Th2 cytokine production and play a protective role in tissue damage in allergic disease.

  • To demonstrate the effect of formulation conditions on the controlled release of protein from poly(lactide-co-glycolide) (PLGA) microspheres for use as a parenteral drug carrier, ovalbumin (OVA) microspheres were prepared using the W/O/W multiple emulsion solvent evaporation and extraction method. Methylene chloride or ethyl acetate was applied as an organic phase and poly(vinyl alcohol) as a secondary emulsion stabilizer. Low loading efficiencies of less than 20% were observed and the in vitro release of OVA showed a burst effect in all batches of different microspheres, followed by a gradual release over the next 6 weeks. Formulation processes affected the size and morphology, drug content, and the controlled release of OVA from PLGA microspheres.

Selonsertib, an ASK1 Inhibitor, Ameliorates Ovalbumin-Induced Allergic Asthma during Challenge and Sensitization Periods

  • Apoptosis signal-regulating kinase 1 (ASK1) is an upstream signaling molecule in oxidative stress-induced responses. Because oxidative stress is involved in asthma pathogenesis, ASK1 gene deficiency was investigated in animal models of allergic asthma. However, there is no study to investigate whether ASK1 inhibitors could be applied for asthma to date. Selonsertib, a potent and selective ASK1 inhibitor, was applied to BALB/c mice of an ovalbumin (OVA)-induced allergic asthma model. Selonsertib suppressed antigen-induced degranulation of RBL-2H3 mast cells in a concentration-dependent manner. The administration of selonsertib both before OVA sensitization and OVA challenge significantly reduced airway hyperresponsiveness, and suppressed eosinophil numbers and inflammatory cytokine levels in the bronchoalveolar lavage fluid. Histopathologic examination elucidated less inflammatory responses and reduced mucin-producing cells around the peribronchial regions of the lungs. Selonsertib also suppressed the IgE levels in serum and the protein levels of IL-13 in the bronchoalveolar lavage fluid. These results suggest that selonsertib may ameliorate allergic asthma by suppressing immune responses and be applicable to allergic asthma.

기니픽에서 Ovalbumin으로 유발된 즉시형 기관지 수축반응에 대한 Pranlukast의 효과 (Effects of Pranlukast on Ovalbumin Induced Early-Phase Bronchoconstriction in Guinea Pigs)

  • 연구배경 : 기관지 천식에서 기관지 수축 및 호산구 화학 주성 효과를 나타내는 Leukotrienes (LTs)의 역할이 최근 잘 알려져 있으며 이러한 LTs의 작용을 억제하는 치료제에 대한 관심이 대두되고 있다. 저자들은 비마취, 비결박 상태의 기니픽 기관지 천식 모델에서 즉시형 기관지 수축 반응 시 기도저항의 변동을 측정하고, pranlukast 투여가 기도저항, 기관지 조직내 호산구의 침윤 정도, 혈중 $LTC_4$의 농도의 변화에 미치는 영향을 평가하고자 본 연구를 실시하였다. 방 법 : 기니픽 44마리 (치료군 16마리, 대조군 19마리, 정상군 9마리)를 대상으로 하였으며 치료군과 대조군 기니픽에 ovalbumin(OVA)을 감작하고 1주후 OVA을 연무기를 이용하여 흡입시켜 즉시형 기도수축 모델을 만든 뒤 실험 동물용 체적 기록 상자를 이용하여 기도저항을 측정하였다. 정상군은 감작하지 않은 상태에서 기도저항을 측정하였다. 치료군은 pranlukast 10mg/kg/day를, 대조군과 정상군은 생리식염수 1ml/day를 7일간 경구 투여한 후 치료군, 대조군, 정상군 모두 indomethacine 10mg/kg 및 mepyramine 10mg/kg를 복강내 투여한 다음 OVA 분무로 즉시형 기관지 수축반응을 일으켜 기도저항을 측정하였다. 기도저항 측정 후 혈중 $LTC_4$의 농도를 측정하고 한 쪽 폐장을 적출하여 파라핀 포매조직을 만들어 H-E 염색으로 세기관지를 포함하는 400배 현미경 시야내에 존재하는 호산구의 수로써 호산구의 침윤 정도를 확인하였다. 결 과 : 천식 모델로 확인된 기니픽은 치료군 8마리, 대조군 16마리로 57.1%의 발생율을 나타내었고 기관지 천식이 발생되지 않은 기니픽은 모두 본 연구 대상에서 제외시켰다. OVA으로 감작 시키지 않은 정상군의 기도저항의 변동은 기저치의 50% 내외였다. OVA 흡입으로 즉시형 기관지 수축 반응을 나타낸 상태에서 측정한 기도저항은 치료군에서는 흡입 후 3분부터 24분까지 3분 간격으로 측정 했을때 3분, 6분에서 pranlukast 투여 후가 투여 전에 비하여 통계적으로 유의하게 낮았고 24분까지 통계적 유의성은 없었지만 기도저항이 투여전에 비하여 감소된 경향을 나타내었다. 대조군에서는 OVA 흡입후 3분부터 30분까지 생리식염수 투여전파 후에 기도저항의 차이가 없었다. 혈중 $LTC_4$ 농도는 치료군 348.4 pg/ml, 대조군 373.9 pg/ml였으며, 정상군에서는 364.4 pg/ml로 치료군과 대조군(p=0.232), 치료군과 정상군(p=0.501), 사이에 통계적으로 유의한 차이가 발견되지 않았고, 치료군, 대조군 및 정상군의 세 구간의 비교에서도 통계적으로 의미 있는 차이가 없었다(p=0.456). 기관지 조직내 호산구의 침윤은 400배 현미경 시야당 치료군, 대조군, 정상군 각각 7.06, 19.2, 4.50개로 치료군, 대조군, 정상군 사이의 분산분석에서는 세군간에 의미 있는 차이가 있었고(p=0.008) 치료군이 대조군에 비하여 통계적으로 의미 있는 감소를 나타내었다(p=0.001). 곁 론 : OVA으로 감작된 기니픽 천식 모델에서 즉시형 기관지 수축 반응 시 cys-LTs 수용체 억제제인 pranlukast 경구 투여는 대조군과 비교하여 기도저항의 감소 및 기관지, 폐 조직 내 호산구 침윤의 감소를 나타내어 기관지 수축 억제 효과가 있음을 알 수 있었다. 혈중 $LTC_4$의 농도는 대조군과 차이가 없음이 관찰되어 전신의 순환 cys-LTs에는 영향을 미치지 않음을 알 수 있었다. 이와 같은 기니픽에 대한 결과를 바탕으로 향후 기관지 천식 환자에 대한 pranlukast의 효과에 관하여 더 많은 연구가 필요할 것으로 사료된다.

가감양격산(加減凉膈散)이 RBL-2H3 비만세포와 OVA/alum에 감작된 생쥐에 미치는 항알레르기 효과 (Anti-allergic Effects of Gagam-YangGyeokSan on RBL-2H3 Mast Cells and OVA/alum Sensitized Mice)

  • Objectives: Gagamyanggyeoksan (G-YGS) has been used to suppress allergic reaction, however, the cellular target of G-YGS and its mode of action remain unclear. The present study was designed to investigate the effect of extracted G-YGS on the PMA and lonomycin (PI)-induced activation of RBL-2H3. Methods: For this investigation, We examined IL-4, IL-13 mRNA expression by Real-Time PCR, IL-4, IL-13 production by ELISA analysis and manifestations of GATA-1, GATA-2, NF-AT1, NF-AT2, AP-1 and NF-${\kappa}B$ p65 transcription factors by western blotting, OVA-specific IgE, IL-4, IL-13 by mouse be sensitive to OVA. Results: Here we showed that treatment of RBL-2H3 mast cells with G-YGS, suppressed PI-induced production of Th2 cytokines including IL-4 and IL-13 in a dose dependent manner. The mRNA expression of IL-4 were completely abolished by G-YGS at the concentration of $100{\mu}g/ml$. Data from a stable cell lines consistently expressing IL-4. And the mRNA expression of IL-13 were abolished by G-YGS at the $200{\mu}g/ml$. But there is no difference between the $50{\mu}g/ml$, the $100{\mu}g/ml$ and the comparison. Results from the western blot analysis of transcription factors involving IL-4 and IL-13 expression indicated that it prominently decreased the expression of mast cell specific transcricption factors including GATA-1, GATA-2, NF-AT2, c-Jun, NF-${\kappa}B$ p65 but not c-Fos. And G-YGS suppressed IgE, IL-4, IL-13 in mouse be sensitive to OVA. Conclusions We suggested the anti-allergic activities of G-YGS might be mediated by down-regulation of Th2 cytokines such as IL-4 and IL-13 through the regulation of transcription factors as GATA-1, GATA-2, NF-AT2, c-Jun, NF-${\kappa}B$ p65.

마행감석양가감방(麻杏甘石揚加減方)이 천식모델생쥐의 면역세포 및 사이토카인에 미치는 영향 (Effects of Mahaenggamseok-tang-gagambang on Immune Cells and Cytokines in OVA-Induced Asthmatic Mice)

  • The purpose of this research is to evaluate the effect of Mahaenggamseok-tang-gagambang (MGTG) on airway hyper- responsiveness (AHR), immune cells, cytokines and lung tissue in OVA-induced asthmatic mice. C578L/6 mice were injected, inhaled and sprayed with OVA for 12 weeks (3times a week) for asthma sensitization and challenge. Two experimental groups were treated with different concentrations of MGTG (400 mg/kg and 200 mg/kg) extract and cyclosporin A (10 mg/kg) for the later 8 weeks. Enhanced pause (Penh) levels were measured by whole body plethysmography. Immune cells were analyzed by flow cytometer in peripheral blood monocyte cell (PBMC) and lung cells. The IL-1b, IL-12, IFN-${\gamma}$, OVA-lgE, IL-4, IL-5, TNF-${\alpha}$ were analyzed by ELISA kit in serum and splenocyte+a-cCDS/a-CD28. Enhanced pause (Penh) levels of the MGTG groups (400 mg/kg and 200 mg/kg) were decreased significantly compared with that of control group. The numbers of MGTG groups (400 mg/kg and 200 mg/kg) on lung total cells were decreased significantly compared with that of control group. The numbers of MGTG groups (400 mg/kg and 200 mg/kg) on $CD3^+/CD69^+$, $B220^+/CD22^+$, $B220^+/CD23^+$, $B220^+/lgE^+$, $CCR3^+$ cells were decreased significantly compared with that of control group. The number of MGTG group (400 mg/kg) on $CD3^+/CD49b^+$ cells was decreased significantly compared with that of control group. The level of MGTG groups (400 mg/kg and 200 mg/kg) on IL-4, IL-5, IL-12, TNF-${\alpha}$, OVA-lgE were decreased significantly compared with that of control group. The level of MGTG group (400 mg/kg) on IL-1b, IL-1S, OVA-lgE were decreased significantly compared with that of control group. These results demonstrate that MGTG could be a desirable alternative therapy for allergic asthma by inhibiting the expression of immune cells, the activation of inflammatory mediator.

삼자양친탕(三子養親湯) 물 추출물(抽出物)이 마우스 Th1/Th2 분화 및 알레르기 염증반응 조절에 미치는 효과 (Effect of Samjayangchin-tang on Cytokine Levels of Mouse Th1/Th2 Cells and Anti-allergic Activity in Ovalbumin-sensitized Allergic Inflammation Model)

  • This study was to evaluate the effect of Samjayangchin-tang (STYCT) on mouse Th1 and Th2 cells' differentiation and ovalbumin (OVA)-induced allergic inflammation. The proliferation of mouse CD4 T cells and the secretion of Th1/Th2 cytokines under the influence of STYCT extract were measured as well as the amount of ${\beta}-hexosaminidase$ in RBL-2H3 cells and the levels of $TNF-{\alpha}$ and IL-6 secretion in Raw264.7 cells. BALB/c mice were orally administered with STYCT extract and simultaneously inoculated with OVA to induce allergic reaction and measure the level of total IgE, OVA-specific IgE and the production of IFN- g, IL-4, IL-5 by the spleen cells. When mouse CD4 T cell were stimulated with anti-CO3 and anti-CD28 for 48 hours in various concentrations of STYCT extract, it decreased proliferation of CD4 cells. CD4 T cells under Th1/Th2 polarizing conditions for 3 days with STYCT resulted in mild decrease of IFN- g in Th1 cells and significant decrease of IL-4 in Th2 cells. STYCT extract had a dose-dependent inhibitory effect on antigen-induced release of ${\beta}-hexosaminidase$ in RBL-2H3 cells. Treatment of STYCT extract on LPS stimulated Raw 264.7 cells showed dose-dependent decrease in IL-6 production. Oral administration of STYCT extract on OVA-induced allergic mice showed an inhibitory effect on the levels of total serum IgE and OVA-specific IgE by 53% and 44%, respectively. Culture of spleen cells with OVA resulted in significant increase of IFN- g by 54% and significant decrease of IL-4 and IL-5 by 42%, and 29%, respectively. The results show that STYCT does not strongly induce mouse T cells to transform into Th1 or Th2 but it has an anti-allergic effect in vitro, and that it also corrects the unbalance between the reactions of Th cells in allergic diseases.
