종의 포플라수종에서 리그닌생합성에 관계된 OMT유전자의 발현 (Seasonal Expression of OMT Gene in Relation to Lignin biosynthesis in two Poplar Species)

  • 리그닌함량이 감소한 펄프소재의 육성을 위해 OMT유전자와 리그닌생합성 간의 관계를 2종의 포플라 수종을 이용하여 실험하였다. OMT유전자의 발현은 리그닌생합성이 가장 많이 일어나는 developing secondary xylem에서 높앗다. 이 사부조직에서의 리그닌함량의 증가는 봄에 성장을 개시할 때부터 증가하기 시작하여 8월 말 경에 가장 높았다. 이러한 리그닌의 증가는 OMT유전자의 발현 증가와 일치하였으며, 이는 OMT유전자가 리그닌생합성에서 매우 중요한 역할을 함을 나타내는 것이다.

Isolation 특성이 좋은 Orthogonal Mode Transducer 설계와 제작 (Design and Fabrication of a Simple Orthogonal Mode Transduce with Good Iolation)

  • 이 논문에서는 Z-밴드용 간단한 구조의 OMT (orthogonal mode transducer)가 소개되다. 이 OMT는 몇 개의 가는 유도 봉을 이용하여 한 편파를 주어진 방향으로 유도하고. 또한 다른 직각 편파를 유도 봉을 이용하여 다른 방향으로 유도하도록 설계되었다. 이 OMT는 유도 봉들이 있는 T자 형태의 도파관 부분과 두 도파관 연결부분으로 구성되어 있고, 한 안테나와 두 개의 어댑터들과 연결되도록 세 플랜지를 갖고 있다. 유도 봉의 최적개수와 최적 위치는 CAD인 HFSS를 이용하여 수치적으로 얻었다. 제작된 OMT에서 두 폄파간의 isolation은 9.5GHz와 10.5GHz 주파수 밴드에서 42.8dB보다 높았으며, 반사계수는 -17.5dB보다 낮았다. 이 OMT는 polarimetric scatterometer 시스템에 사용되는 polarimetric 안테나에 사용되어질 수 있다.

Oxymatrine Causes Hepatotoxicity by Promoting the Phosphorylation of JNK and Induction of Endoplasmic Reticulum Stress Mediated by ROS in LO2 Cells

  • Oxymatrine (OMT) often used in treatment for chronic hepatitis B virus infection in clinic. However, OMT-induced liver injury has been reported. In this study, we aim to investigate the possible mechanism of OMT-induced hepatotoxicity in human normal liver cells (L02). Exposed cells to OMT, the cell viability was decreased and apoptosis rate increased, the intracellular markers of oxidative stress were changed. Simultaneously, OMT altered apoptotic related proteins levels, including Bcl-2, Bax and pro-caspase-8/-9/-3. In addition, OMT enhanced the protein levels of endoplasmic reticulum (ER) stress makers (GRP78/Bip, CHOP, and cleaved-Caspase-4) and phosphorylation of c-Jun N-terminal kinase (p-JNK), as well as the mRNA levels of GRP78/Bip, CHOP, caspase-4, and ER stress sensors (IREI, ATF6, and PERK). Pre-treatment with Z-VAD-fmk, JNK inhibitor SP600125 and N-acetyl-l-cysteine (NAC), a ROS scavenger, partly improved the survival rates and restored OMT-induced cellular damage, and reduced caspase-3 cleavage. SP600125 or NAC reduced OMT-induced p-JNK and NAC significantly lowered caspase-4. Furthermore, 4-PBA, the ER stress inhibitor, weakened inhibitory effect of OMT on cells, on the contrary, TM worsen. 4-PBA also reduced the levels of p-JNK and cleaved-caspase-3 proteins. Therefore, OMT-induced injury in L02 cells was related to ROS mediated p-JNK and ER stress induction. Antioxidant, by inhibition of p-JNK or ER stress, may be a feasible method to alleviate OMT-induced liver injury.

3D Structure of Bacillus halodurans O-Methyltransferase, a Novel Bacterial O-Methyltransferase by Comparative Homology Modeling

  • Bacillus halodurans O-methyltransferase (BhOMT) is a S-adenosylmethionine (SAM or AdoMet) dependent methyltransferase. Three dimensional structure of the BhOMT bound to S-adenosyl-L-homocysteine (SAH or AdoHcy) has been determined by comparative homology modeling. BhOMT has 40% sequence identity with caffeoyl-CoA 3-O-methyltransferase (CCoAOMT) from alfalfa. Based on x-ray structure of CCoAOMT, three dimensional structure of BhOMT was determined using MODELLER. The substrate binding sites of these two proteins showed slight differences, but these differences were important to characterize the substrate of BhOMT. Automated docking study showed that four flavonoids, quercetin, fisetin, myricetin, and luteolin which have two hydroxyl groups simultaneously at 3'- and 4'-position in the B-ring and structural rigidity of Cring resulting from the double bond characters between C2 and C3, were well docked as ligands of BhOMT. These flavonoids form stable hydrogen bondings with K211, R170, and hydroxyl group at 3'-position in the Bring has stable electrostatic interaction with Ca2+ ion in BhOMT. This study will be helpful to understand the biochemical function of BhOMT as an O-methyltransferase for flavonoids.

A Broadband W-band Orthomode Transducer for KVN Polarization Observations

  • A W-band Orthomode Transducer (OMT) has been developed for Korean VLBI Network (KVN) polarization observation. The OMT design was based on E-plane split-block technique using septum structure. 3-dimensional electromagnetic simulation was fully employed to optimize the performance of the OMT. Measurements of the fabricated OMT show that the return losses for the vertically and horizontally polarized modes are better than -20 dB across 80 ~ 108 GHz and the insertion losses for the both modes are less than 0.47 dB. The cross-polarization level of the OMT is less than -30 dB. The bandwidth of the developed OMT is estimated as around 30%.

O-Methyltransferases from Arabidopsis thaliana

  • O-methylation mediated by O-methyltransferases (OMTs) is a common modification in natural product biosynthesis and contributes to diversity of secondary metabolites. OMTs use phenylpropanoids, flavonoids, other phenolics and alkaloids as substrates, and share common domains for S-adenosyl-L-methionine (AdoMet) and substrate binding. We searched Arabiposis genome and found 17 OMTs genes (AtOMTs). AdoMet- and substrate-binding sites were predicted. AdoMet binding domain of AtOMTs is highly conserved, while substrate-binding domain is diverse, indicating use of different substrates. In addition, expressions of six AtOMT genes in response to UV and in different tissues were investigated using real-time quantitative reverse transcriptase-polymerase chain reaction. All the AtOMTs investigated were expressed under normal growth condition and most, except AtOMT10, were induced after UV illumination. AtOMT1 and AtOMT8 were expressed in all the tissues, whereas AtOMT10 showed flower-specific expression. Analysis of these AtOMT gene expressions could provide some clues on AtOMT involvement in the cellular processes.

한방음악치료가 사상체질에 따라 분류된 뇌경색환자에 마치는 효과에 관한 연구 (The Study on the Effect of Oriental Music Therapy in Ischemic Stroke Patients Classified by Sasang Constitutional Medicine)

  • 1. Objectives The purpose of this study was to examine the regional cerebral blood-flow(r-CBF) difference between OMT(Oriental Music Therapy) group and control group using brain SPECT, to evaluate the effect of OMT in ischemic stroke patients Classified by Sasang Constitutional. 2. Methods We selected 10 ischemic storke patients, classified groups into OMT group or control group. r-CBF of two groups were examined by brain SPECT. in all group we applied SCM(Sasang constitutional medicine)treatment for 4-5 weaks, and additionally in OMT group We applied OMT for 4-5 weaks. after that all the r-CBF of two groups took brain SPECT again. r-CBF difference and change before and after SCM treatment and OMT were quantified and analyzed using SPM. 3. Results r-CBF of OMT groups were increased compared to control groups. 4. Conclusions This study demonstrated that Sasang Constitutional OMT increased regional cerebral blood perfusion to the corresponding brain areas in ischemic stroke patients.

  • PURPOSE; The main purpose of this article is to document correctly of the orthopedic manual therapy's history in Korea. Therefore this study aimed not only to know the root of the historical Korean OMT, but also to develope of the Korean manual therapy advanced in future. RESULT; The results of this study were as following; 1. The first introduction for OMT in Korea is the '1st International Orthopedic Physical Therapy Seminars' at Seoul National University Hospital by Prof. Russell M. Woodman etc(Quinnipiac college, USA), 1991. 1.16-1.21. 2. The first OMT study group in Korea is to establish the 'Korean Study Group of OMT at Hallym University Kangdon Sacred Heart Hospital, 1992.2.22. 3. Crystallization and Preliminary X-Ray Diffraction Analysis of BcOMT2 from Bacillus cereus: A Family of O-Methyltransferase

  • O-Methyltransferases (OMTs), one of the ubiquitous enzymes in plants, bacteria, and humans, catalyze a methyl-transfer reaction using S-adenosylmethionine and a wide range of phenolics as a methyl donor and acceptor, respectively. Substrates for most bacterial OMTs have largely remained elusive, but recent investigation using BcOMT2, an OMT from Bacillus cereus, suggested that ortho-dihydroxyflavonoids could serve as substrates. To elucidate the functional and structural features of BcOMT2, we expressed, and purified BcOMT2, and crystallized an apoenzyme and its ternary complex in the presence of a flavonoid and S-adenosylhomocysteine. Each crystal diffracted to $1.8{\AA}$ with its space group of C2 and $P2_{1}2_{1}2_{1}$, respectively. Structural analysis of apo-BcOMT2 and its ternary complex will provide the structural basis of methyl transfer onto (iso)flavonoids in a regiospecific manner.

Characterization of an O-Methyltransferase from Streptomyces avermitilis MA-4680

  • A search of the Streptomyces avermitilis genome reveals that its closest homologs are several O-methyltransferases. Among them, one gene (viz., saomt5) was cloned into the pET-15b expression vector by polymerase chain reaction using sequence-specific oligonucleotide primers. Biochemical characterization with the recombinant protein showed that SaOMT5 was S-adenosyl-L-methionine-dependent Omethyltransferase. Several compounds were tested as substrates of SaOMT5. As a result, SaOMT5 catalyzed O-methylation of flavonoids such as 6,7-dihydroxyflavone, 2',3'-dihydroxyflavone, 3',4'-dihydroxyflavone, quercetin, and 7,8-dihydroxyflavone, and phenolic compounds such as caffeic acid and caffeoyl Co-A. These reaction products were analyzed by TLC, HPLC, LC/MS, and NMR spectroscopy. In addition, SaOMT5 could convert phenolic compounds containing ortho-dihydroxy groups into O-methylated compounds, and 6,7-dihydroxyflavone was known to be the best substrate. SaOMT5 converted 6,7-dihydroxyflavone into 6-hydroxy-7-methoxyflavone and 7-hydroxy-6-methoxyflavone, and caffeic acid into ferulic acid and isoferulic acid, respectively. Moreover, SaOMT5 turned out to be a $Mg^{2+}$-dependent OMT, and the effect of $Mg^{2+}$ ion on its activity was five times greater than those of $Ca^{2+}$, $Fe^{2+}$, and $Cu^{2+}$ ions, EDTA, and metal-free medium.
