Microbiological Safety of Commercial Salt-fermented Shrimp during Storage (새우젓 저장 유통 중 위해 미생물학적인 안전성 연구)

  • Microbiological safety investigation of 36 commercial salt-fermented shrimps revealed presence of coliform and Gram(+) cocci, whereas pathogenic bacteria such as Escherichia coli O26, Salmonella, Shigella, Staphylococcus aureus, Vibrio parahaemolyticus, and V. cholerae were not detected. When pathogenic bacteria were inoculated into 9, 18, and 27% salted shrimps, Salmonella so., E. coli O26, and S. aureus were not detected up to 13, 80, and 90 days of fermentation at $20^{\circ}C$, respectively, whereas up to 15 day in commercial salt-fermented shrimps.

  • A 2-day-old male (Premie, Large for gestational age(LGA), Intrauterine period(IUP) 33 weeks, birth weight 2,955 gram) was transferred with marked abdominal distention, bilious return via the orogastric tube, respiratory difficulty, and generalized edema (hydrops fetalis). He was born by cesarean section to a 36 year-old mother. Antenatal ultrasonogram at IUP 31 weeks demonstrated multiple dilated bowel loops suggestive of intestinal obstruction. There was no family history of cystic fibrosis. Simple abdominal films disclosed diffuse haziness and suspicious fine calcifications in the right lower quadrant. Barium enema demonstrated a microcolon. Sweat chloride test was not available in our institution. At laparotomy, there noted 1) a segmental volvulus of the small bowel with gangrenous change, associated with meconium peritonitis, 2) an atresia of the ileum at the base of the volvulus, and 3) the terminal ileum distal to the volvulus was narrow and impacted with rabbit pellets-like thick meconium. These findings appeared to be very similar to those of a complicated meconium ileus. In summary, the ileal atresia and meconium peritonitis seemed to be caused by antenatal segmental volvulus of the small intestine in a patient with probable meconeum ileus.

Production of taxadiene from cultured ginseng roots transformed with taxadiene synthase gene

  • Paclitaxel is produced by various species of yew trees and has been extensively used to treat tumors. In our research, a taxadiene synthase (TS) gene from Taxus brevifolia was used to transform the roots of cultured ginseng (Panax ginseng C.A. Meyer) to produce taxadiene, the unique skeletal precursor to taxol. The TS gene was successfully introduced into the ginseng genome, and the de novo formation of taxadiene was identified by mass spectroscopy profiling. Without any change in phenotypes or growth difference in a TS-transgenic ginseng line, the transgenic TSS3-2 line accumulated $9.1{\mu}g$ taxadiene per gram of dry weight. In response to the treatment of methyl jasmonate for 3 or 6 days, the accumulation was 14.6 and $15.9{\mu}g$ per g of dry weight, respectively. This is the first report of the production of taxadiene by engineering ginseng roots with a taxadiene synthase gene.

  • The effects of crude extract and bioactive fractions from Brachyglottis monroi on biological activity were investigated. The crude ethanol extract inhibited the growth of the Gram positive bacterium Bacillus subtilis (ATCC Strain number 19659, 1 mm zone at 150 μg/disk) and the dermatophyte Trichophyton mentagrophytes (ATCC 28185, 2 mm zone at 150 μg/disk), and toxic to P388 tumor cells (IC50 23.96 μg/ml at 75 μg/disk). Cytotoxic activity was strongly showed by Fr. 6 (P388 IC50 19.67 μg/ml at 75 μg/disk).

  • TLR4 together with CD14 and MD-2 forms a pattern recognition receptor that plays an initiating role in the innate immune response to Gram-negative bacteria. Here, we employed the surface plasmon resonance technique to investigate the kinetics of binding of LPS to recombinant CD14, MD-2 and TLR4 proteins produced in insect cells. The dissociation constants ($K_D$) of LPS for immobilized CD14 and MD-2 were $8.7{\mu}m$, and $2.3{\mu}m$, respectively. The association rate constant ($K_{on}$) of LPS for MD-2 was $5.61{\times}10^3M^{-1}S^{-1}$, and the dissociation rate constant ($K_{off}$) was $1.28{\times}10^2S^{-1}$, revealing slow association and fast dissociation with an affinity constant $K_D$ of $2.33{\times}10^6M$ at $25^{\circ}C$. These affinities are consistent with the current view that CD14 conveys LPS to the TLR4/MD-2 complex.

Antimicrobial Effect of Acidified Sodium Chlorite (ASC) on Whole Croaker

  • The antimicrobial effect of acidified sodium chlorite (ASC) solution on whole croaker skin was evaluated. Whole croaker skin was treated with ASC (50, 100, 200, 400, and 600 ppm) and distilled water. After 10-minute exposure to 600 ppm ASC, 8% of Gram-negative bacteria survived on the whole croaker sample. Treatment with 50 ppm ASC eliminated all coliforms in the initial load. Immersion treatment with 600 ppm ASC resulted in $1.3\;log\;CFU/cm^2$ greater kill of the initial mesophile loads of control ($2.8\;log\;CFU/cm^2$) than distilled water. Fifty ppm ASC solution produced a 1.6-log reduction of psychrotrophic bacteria. ASC treatment was an effective method for reducing naturally occurring microflora on whole croaker skin.

  • The effects of chloroform extract from Lepidolaena tayiorii (L. tayiorii) on antifungal activity were investigated. The crude chloroform extract of L. tayiorii inhibited the growth of the Gram positive bacteria Bacillus subtilis ATCC 19659, (5 mm inhibition zone at $150{\mu}g/disc$) and the dermatophytic fungus Trichophyton mentagrophytes ATCC 28185, (6 mm inhibition zone at $150{\mu}g/disc$), and cytotoxic to P388 murine leukaemia cells ATCC CCL 46 P388D1, ($IC_{50}\;405.0{\mu}g/mL\;at\;150{\mu}g$/disc) and cytotoxic to BSC monkey kidney cells (@ 5 mg/mL, $150{\mu}g/disc;$ +++: 100% activity). We suppose that this crude chloroform extract of L. tayiorii is the strong antimicrobial and cytotoxic activities.

Encephalitic listeriosis in two Korean native goats in Jeju (제주지역 흑염소에서 뇌염형 리스테리아증 발생 증례 보고)

  • In February 2013, two Korean native goats with circling, dysphagia, sialorrhea, and death were submitted for necropsy to the Jeju Self-Governing Provincial Veterinary Research Institute. Histopathologically, multifocal to confluent micro-abscess composed of neutrophils and macrophages and severe multifocal perivascular cuffing were observed in pons, medulla oblongata, and cerebellum. Isolated bacteria from the brain sample were Gram-positive coccobacilli and were confirmed as Listeria (L.) monocytogenes by VITEK and PCR analysis. Based on clinical sign, histopathology and bacterial isolation, this case was diagnosed as encephalitic listeriosis. In our best knowledge, this is the first report of encephalitic listeriosis caused by L. monocytogenes in Korean native goats in Jeju.

Effect of Asthma Management Education Program on Stress and Compliance of Patients with Allergic Asthma to House Dust Mite

  • Purpose. This study was designed to examine the effect of asthma management education program applied to allergic asthma patients receiving immunotherapy due to house dust mite on their stress and compliance with health care regimens. Methods. A quasi experimental design with non-equivalent control group and non-synchronized design was used. The subjects of this study were 61 patients who were receiving immunotherapy at intervals of a week after their symptoms were diagnosed as house dust mite allergic asthma at the pulmonary department of a university hospital in Seoul. They were divided into an experimental group of 29 patients who received asthma management education and a control group of 32 patients. The asthma management education pro-gram was composed of group education (once) and reinforcement education (three times) with environmental therapy and immunotherapy to house dust mite. Results. Stress significantly decreased in the experimental group compared to that in the control group. Compliance with health care regimens significantly increased in the experimental group compared to that in the control group. Conclusions. The results suggested that the asthma management education program is effective for the management of stress and the improvement of compliance in patients with allergic asthma to house dust mite.

Primary Pneumococcal Peritonitis in a Healthy Child (건강한 소아에서 발생한 원발성 폐렴구균성 복막염 1례)

  • Primary peritonitis usually refers to a bacterial infection of the peritoneal cavity without a demonstrable intra-abdominal source. Most cases occur in children with ascites resulting from nephrotic syndrome or cirrhosis. Rarely, it may occur in previously healthy children less than 7years of age, usually a girl. Distinguishing primary peritonitis from appendicitis may be impossible in patients without a history of nephrotic syndrome or cirrhosis. Accordingly, the diagnosis of primary peritonitis is made only at laparotomy. We report one case of primary pneumococcal peritonitis in a 27-month-old female who underwent explorative laparotomy to discover the cause of suspicious intestinal perforation and mechanical ileus. Later, pneumococci were cultured in blood and gram-positive diplococci were isolated from the pus of peritoneal cavity.
