Gastroprotective Activity of Curcumae Longae Rhizoma against Gastric Ulcer in Mice (위궤양 유발 마우스모델에서 강황(薑黃) 추출물의 위 보호 효과)

  • Objectives : The objective of this study was to evaluate the gastric protective effect of Curcuma Longae Rhizoma (CLR) in 150 mM HCl/60% ethanol induced gastric ulcer (GU) in mice. Methods : Forty ICR mice were divided into five groups (n=8/Group): Nor group; Normal, Veh group; GU control, SC group; GU + sucralfate 10 mg/kg, CL; GU + CLR 30% ethanol extract 100 mg/kg, CH group; GU + CLR 30% ethanol extract 200 mg/kg. Then, mice were orally administered with 150 mM HCl/60% ethanol and caused GU. After 1 hr, mice were sacrificed, and blood and stomach tissue were collected. Results : CLR showed significance scavenging effects in 1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azinobis-3-ethyl-benzothiazoline-6-sulfonic acid (ABTS) radical scavenging activities (DPPH IC50; 78.18 ± 0.60 ㎍/㎖, ABTS IC50; 55.91 ± 1.86 ㎍/㎖). CLR significance reduce inflammatory-related factors such as cyclooxygenase-2 (COX-2), inducible nitric oxide synthase (iNOS), tumor necrosis factor alpha (TNF-α), interleukin 1 beta (IL-1β), and interleukin-6 (IL-6) via nuclear factor kappa B (NF-κB) inactivation. In addition, the activation of nuclear factor erythroid2-related factor 2 (Nrf2) significantly led to up-regulation of anti-oxidant enzymes including factors heme oxygenase-1 (HO-1), super oxide dismutase (SOD), and glutathione peroxidase-1/2 (GPx-1/2). Conclusions : Our discovery provides that CLR possesses anti-oxidant and anti-inflammatory effects. Hence, CLR may ameliorate the development of gastric ulcer though the inhibition of NF-κB inflammatory pathway and the elevation of Nrf2 anti-oxidant pathway.

Antioxidative Activities and Protective Effects on Alcohol-Induced Oxidative Stress in the Human Hepatic HepG2 Cells of Undaria pinnatifida and Costaria costata Extracts (미역과 쇠미역 추출물의 항산화 및 간 보호 효과)

  • We investigated the antioxidant and hepatoprotective effects of extracts from the Undaria pinnatifida and Costaria costata against ethanol-induced oxidative damage. The total polyphenol and flavonoid contents were highest in the 70% ethanol extract from Undaria pinnatifida and Costaria costata. Also, the radical scavenging activity of DPPH (IC50 0.33± 0.21, 0.48±0.47 mg/ml) and ABTS (IC50 0.34±0.30, 0.47±0.17 mg/ml) in the 70% ethanol extract was higher than that of the hot water and 10% ethanol extracts. To determine the hepatoprotective effects of extracts in ethanol-induced oxidative damage, cell viability was measured using an MTT assay. In the pre-treatment of Undaria pinnatifida and Costaria costata hot water extracts, the concentration-dependent increased the cell viability compared with the ethanol treated cells (73.95%) by 89.91~97.63% and 84.99~90.54%, respectively. The data suggests that 70% ethanol extracts have antioxidant activity and hot water extracts exhibit hepatoprotective effects. Therefore, Undaria pinnatifida and Costaria costata may be considered potential agents for control ethanol-induced liver damage.

Antioxidant and Anti-inflammatory Activities of Atractylodes japonica According to Extract Methods (백출 용매추출 방법에 따른 항산화 활성 및 항염증 효과)

  • Atractylodes japonica has been widely used in a traditional Korean herbal medicine exerting various pharmacological activities such as diauretic action, asriction, anti-allergy, neuroprotective activity, anti-cancer, immunomodulation and gastrointestinal protective effect. This study was to investigate the antioxidant, nitric oxide and inflammatory cytokines production of A. japonica extract by water and 70% ethanol. DPPH and ABTS free radical scavenging activity were increased in a dose-dependent manners with both extracts and there was no difference with extract solvents. 70% ethanol extract of A. japonica showed a very strong inhibitory effect on NO production. Both extracts of A. japonica significantly reduce the expression of iNOS and COX-2 proteins involved in NO prodction. A. japonica extract by water and 70% ethanol inhibited LPS-induced proinflammatory cytokines such as IL-6 and IL-1b. In this study, 70% ethanol extract of A. japonica significantly suppresses LPS-induced NO and inflammatory cytokine production. Therefore it can be widely used to treat and improve inflammatory diseases.

Antioxidant and Whitening Activities of Chlorogenic Acid, Quercetin, and Quercitrin from the Fruit of Vaccinum oldhami (정금나무 열매(Fruit of Vaccinum oldhami)의 분리 정제물(클로로겐산, 퀘르세틴 및 퀘르시트린)에 관한 항산화 및 미백활성 검증)

  • The fruit of Vaccinum oldhami was separated and purified to obtain the compounds chlorogenic acid (CA), quercetin (QT), and quercitrin (QR). The electron-donating abilities of CA, QT, and QR at 1,000 ㎍/ml were 91.9%, 89.9%, and 77.4%, respectively QT and QR showed 99.5% and 91.4% ABTS+ radical scavenging ability at a 1,000 ㎍/ml concentration, respectively, and CA showed a 95% ability or higher at 100 ㎍/ml. Regarding tyrosinase inhibitory activity, CA, QT, and QR exhibited 29.5%, 34.7%, and 23.7% efficacy, respectively, at 1,000 ㎍/ml. Regarding the cell viability for melanoma cells (B16F10) assessed through MTT assay, CA, QT, and QR showed cell a viability of 80% or more at 100 ㎍/ml. To measure the deterrent of protein expression, CA affected TRP-1 and TRP-2 in accordance with increases in concentration. The protein expression inhibition rate of QT was excellent for TRP-1, TRP-2, and tyrosinase. CA was confirmed to have an excellent mRNA expression inhibitory effect against MITF, and the amount of mRNA expression of TRP-1, TRP-2, and tyrosinase decreased with an increase in the CA concentration. As the concentration of QT increased, the mRNA expression of MITF, TRP-2, and tyrosinase decreased. QR decreased the amount of mRNA as the QR concentration increased. The excellent antioxidant and whitening effects of CA, QT, and QR were thus confirmed.

Antioxidant, anti-inflammatory, and antibacterial activities of a 70% ethanol-Symphyocladia linearis extract

  • Research on the potential biological activity of red alga Symphyocladia spp. has been limited to Symphyocladia latiuscula, which is widely used as a food ingredient in Korea. Here, we examined the biological activity of another species, Symphyocladia linearis, which is found in Korea and was reported as a new species in 2013. The aim of this study was to evaluate the antioxidant, anti-inflammatory, and antibacterial properties of a 70% ethanol extract of S. linearis. Antioxidant activity, which was evaluated using radical scavenging assays, revealed half maximal inhibitory concentration values for 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) and 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) of 34.57 and 11.70 ㎍/mL algal extract, respectively. Anti-inflammatory activity of the S. linearis ethanolic extract was evaluated using RAW 264.7 cells by measuring the inhibition of lipopolysaccharide-induced nitric oxide (NO) and prostaglandin E2 (PGE2) production. The potential cytotoxicity of NO and PGE2 was first examined, confirming no toxicity at concentrations ranging from 10-100 ㎍/mL. NO production was inhibited 61.1% and 78.0% at 50 and 100 ㎍/mL S. linearis extract, respectively; and PGE2 production was inhibited 69.1%, 83.2%, and 94.8% at 25, 50, and 100 ㎍/mL S. linearis extract, respectively. Thus, the S. linearis extract showed very strong efficacy against PGE2 production. The cellular production of reactive oxygen species, measured using 2',7'-dichlorofluorescin diacetate fluorescence, was inhibited 48.8% by the addition of 100 ㎍/mL S. linearis extract. Antibacterial activity was evaluated using the disc diffusion method and minimum inhibitory concentration (MIC). S. linearis was effective only against gram-positive bacteria, exhibiting antibacterial activity against Staphylococcus aureus with a MIC of 256 ㎍/mL extract and against Bacillus cereus with a MIC of 1,024 ㎍/mL extract. Based on these results, we infer that a 70% ethanolic extract of S. linearis possesses strong anti-inflammatory properties, and therefore has the potential to be used in the prevention and treatment of inflammatory and immune diseases.

Effect of Pt-Co/C Cathode Catalyst on Electrochemical Durability of Membrane in PEMFC (PEMFC에서 Pt-Co/C Cathode 촉매가 고분자막의 전기화학적 내구성에 미치는 영향)

  • As a PEMFC (Polymer Exchange Membrane Fuel Cell) cathode catalyst, Pt-Co/C has recently been widely used because of its improved durability. In a fuel cell, electrodes and electrolytes have a close influence on each other in terms of performance and durability. The effect on the electrochemical durability of the electrolyte membrane when Pt-Co/C was replaced in the Pt/C electrode catalyst was studied. The durability of Pt-Co/C MEA (Membrane Electrode Assembly) was higher than that of Pt/C MEA in the electrochemical accelerated degradation process of PEMFC membrane. As a result of analyzing the FER (Fluorine Emission Rate) and hydrogen permeability, it was shown that the degradation rate of the membrane of Pt-Co/C MEA was lower than that of Pt/C MEA. In the OCV (Open Circuit Voltage) holding process, the rate of decrease of the active area of the Pt-Co/C electrode was lower than that of the Pt/C electrode, and the amount of Pt deposited on the membrane was smaller in Pt-Co/C MEA than in Pt/C MEA. Pt inside the polymer membrane deteriorates the membrane by generating radicals, so the degradation rate of the membrane of Pt/C MEA with a high Pt deposition rate was higher than Pt-Co/C MEA. When the Pt-Co/C catalyst was used, the electrode durability was improved, and the amount of Pt deposited on the membrane was also reduced, thereby improving the electrochemical durability of the membrane.

Performance Indicators Analysis and Development Plans of K University's Professional Graduate School for the Training Professionals in the Human Resources (인적자원 분야 전문인력 양성을 위한 K대 전문대학원의 성과지표 분석 및 발전방안 연구)

  • The purpose of this study is propose performance indicators analysis and development plans for K University professional graduate school for the training of human resource professionals. Specifically, the main research includes the derivation of performance indicators for K University's professional graduate school, PR plan, academic establishment plan, name change, vision proposal, and curriculum improvement plan for each major. To this end, a survey was conducted on a total of 132 current and former students of K University's professional graduate school. FGI was also conducted with 29 participants. The results of the research are as follows. In the case of key performance indicators, 15 indicators were proposed, taking into account previous research, the characteristics of professional graduate school, surveys, and FGI surveys. Promotion should be continuous, not one-time, and the advantages and benefits of the graduate school should be actively informed to target customers. The proposed name of the professional association is the Korean Association for Employment and Vocational Competency Development. It was found that operating as a convergence society and focusing on convergence research were appropriate. The names of K University's professional graduate school were HR (Human Resources), HRD, and employment and vocational competency development graduate school. As for the vision, it was suggested that a balance between identity and differentiation is needed to flexibly respond to new changes while maintaining existing strengths. As for the proposed improvement of the curriculum by major, it is proposed to reform in a stepwise and gradual manner while maintaining the existing framework to some extent rather than being radical.

Antioxidant Activity of Goat Meat Hot Water Extract and Effect of Extract on Expression of Apoptosis-Related Proteins (염소고기 열수추출물 처리에 따른 항산화 활성 및 암 세포주에서 항암 관련 단백질 발현량 확인)

  • This study was conducted to evaluate in vitro antioxidant activity of goat meat hot water extracts and the changes in apoptosis-related protein expression levels in the cancer cells treated with these extracts. Goat meat hot water extracts were prepared using different cuts of goat meat, including foreleg, hindleg, loin, and rib. Among these extracts, the foreleg and hindleg extracts displayed higher (P<0.05) ABTS radical scavenging activity than the other two extracts. Protein expression levels of BAX, p53, and p21 were not different in the cells treated with the extracts from different cuts, regardless of the cell type. Only p53 expression in HT-29 cells was elevated (P<0.05) after loin extract treatment. These results suggest that antioxidant activity and apoptosis-related effects of goat meat hot water extract varied with cut of meat under in vitro conditions. Because all data was obtained from the in vitro experiment, the ability to generalize conclusions is limited. Additional in vivo studies are necessary.

Reducing the Test Time for Chemical/Mechanical Durability of Polymer Electrolyte Membrane Fuel Cells (고분자연료전지의 화학적/기계적 내구성 평가 시간 단축)

  • A chemical/mechanical durability test of polymer membrane evaluation method is used in which air and hydrogen are supplied to the proton exchange membrane fuel cell (PEMFC) and wet/dry is repeated in the open circuit voltage (OCV) state. In this protocol, when wet/dry is repeated, voltage increase/decrease is repeated, resulting in electrode degradation. When the membrane durability is excellent, the number of voltage changes increases and the evaluation is terminated due to electrode degradation, which may cause a problem that the original purpose of membrane durability evaluation cannot be performed. In this study, the same protocol as the department of energy (DOE) was used, but oxygen was used instead of air as the cathode gas, and the wet/dry time and flow rate were also increased to increase the chemical/mechanical degradation rate of the membrane, thereby shortening the durability evaluation time of the membrane to improve these problems. The durability test of the Nafion 211 membrane electrode assembly (MEA) was completed after 2,300 cycles by increasing the acceleration by 2.6 times using oxygen instead of air. This protocol also accelerated degradation of the membrane and accelerated degradation of the electrode catalyst, which also had the advantage of simultaneously evaluating the durability of the membrane and the electrode.
