  • The contents of organic acid in Korean apple juice were analyzed by HPLC using YMC-peak ODS-AQ column and enzymatic assay. Model apple juices were prepared at the laboratory and commercial apple juices were purchased from the market. Individual organic acid contents were as follows: DL-malic acid 62~402mg%, L-malic acid 48~360mg%, citric acid 1.81~15.74mg%, fumaric acid nd~0.50mg%. Together, these tests gave useful information about the quality and authenticity of a particular apple juice smaple. The presence of D-malic acid was a clear indication of adulteration because this isomer did not occur naturally. Fumaric acid and citric acid levels above trace amounts were also inconsistent with pure apple juice.

  • Ginseng saponins were analyzed by thermospray (TSP) LCMS method using ODS column and with acetonitrile/ammonium acetate solution. Optimal condition for TSP Lchfs was found as follows: capillary temperature: 33$0^{\circ}C$ repelled voltage: 200 V, and concentration of ammonium acetate: 0. 05 M. Panaxadiol and panaxatriol type saponins showed characteristic fragment ions. The calibration curve of ginseng saponin showed good linearity with a correlation coefficient of 0.99. Detection limits using selected ion monitoring (SIM) technique were improved by 10~200 times compared to conventional HPLCnnr detection method.

  • In continued studies on cultivated Korean rhubarb rhizomes, three additional stilbene compounds were isolated from water extract by column chromatographic separation using Sepha dex LH-20, ODS-gel. Compound I, desoxyrhapontigenin(3,5-dihydroxy-4`-methoxystilbene), Compound II, rhapontigenin(3,3`,5-trihydroxy-4`-methoxystilbene), and compound III, piceatannol(3,3`,4`,5-tetrahydroxystilbene) were elucidated based on physico-chemical and spectroscopic evidences(UV, IR, $^1H$-NMR, $^13C$-NMR and Mass).

  • Rhus verniciflua Stokes (RVS) is a widely used herbal plant with various biological properties. Our previous study using in vitro platelet aggregation in whole blood showed that ethyl acetate layer of RVS had strong anti-aggregatory activity. In this study. to investigate the anti-aggregatory activity and antioxidative effects of RVS ethyl acetate layer. the layer was subsequently fractionated by ODS columm chromatograph (50% MeOH). As a result. the fraction 3 was most inhibited the aggregation of platelet in rat whole blood induced by thrombin and all fraction of RVS was detected strong antioxidative effect. (omitted)

  • In continuing search for BChE-inhibitory compounds from Korean marine algae. we found a highly potent inhibitory activity in the methanolic extract of Sargassum species. After partition of the MeOH extract between $CHCl_3$ and 30% MeOH. the former layer was subjected to a series of ODS flash chromatography. silica column chromatography. and preparative TLC to afford three compounds (1-3). Detailed structural elucidation of them is in progress. Compound 1 showed potent BChE-inhibitory activity with $IC_{50}$ values of 11 ng/mL. (omitted)

  • In the course of search for various bioactive compounds from marine algae, we found strong antioxidant activity of the methanolic extract of the brown alga G. elliptica. Chromatographic purification [ODS flash, gel-filtration on Sephadex LH-20, HPLC] of the BuOH layer of the methanolic extract afforded two known polyphenolic compounds, 6,6'-bieckol (1) and dieckol (2). Compound 1 showed acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE) inhibitory activities, free radical scavenging activity on DPPH (1,1-diphenyl-2-picryl-hydazyl) with IC$\_$50/ values of 91.2, 45.6 and 15.5 $\mu\textrm{g}$/$m\ell$, respectively. Their structures were identified by comparison with the literature data. Compounds 1 and 2 showed moderate acetylcholinesterase and butyrylcholinesterase inhibitory activities with IC$\sub$50/ values of 65.0∼48.0 ${\mu}$M and 34.0∼23.0 ${\mu}$M, respectively. (omitted)

  • This paper describes the concept and basic technique of measuring torsional operational deflection shapes using a laser-based torsional vibration meter, a dual-channel FFT analyzer and operational deflection shapes software running on a PC. Torsional Operational Deflection Shapes (TODS) is defined similar to ODS (Operational Deflection Shapes), with the exception the TODS designates the operational deflection shapes of structures vibrating in a rotational, or angular, degree of freedom. Thus the TODS measurements can be applied to rotating shafts and the results of such a measurement are shown. In some cases it may be great benefit to apply order tracking and/or synchronous time domain averaging techniques in order to avoid smearing and reduce noise problems.

  • Antibiotics are substances produced by microorganisms that kill or inhibit and are essential for infectious diseases management. This study aimed to provide basic data for overcoming antibiotic resistance in the marine bacterium LJ-18. The API 20NE and API 50CH kits were used to identify this microorganism. Morphological, physiological, and biochemical properties were investigated using MacFaddin's manuals. Subsequently, isolated LJ-18 was found to belong to a genus of Streptomyces that forms mycelia. LJ-18 also grew well at 28-32℃ on modified Bennett's agar. To isolate and purify the antibacterial compound, LJ-18 culture was divided into ethyl acetate and distilled water fractions. Considerable antimicrobial activity against various pathogenic microorganisms, including methicillin-resistant Staphylococcus aureus (MRSA), was confirmed in the C18 ODS open column fractions. Peak 2 compound was obtained using reversed-phase HPLC. As a result, this compound had a significant antimicrobial activity against various pathogenic microorganisms. In particular, it showed strong activity against MRSA, Mycobacterium smegmatis, Bacillus subtilis, Bacillus cereus, and Staphylococcus aureus.
