  • To search and development a new material with superior melanogenesis inhibitory activity, the bioactivities (obs. pl$_{50}$) of alkyl-3,4-dihydroxybenzoyl esters and N-alkyl-3,4-dihydroxybenz-oyl amides as substrate molecules were measured in mouse melanoma cells. And also, we have studied that 3-D QSARs (3 dimensional Quantitative Structure-Activity Relationships) between molecular interaction field of substrates and the bioactivities were analyzed using CoMFA (Comparative Molecular Field Analyses) method. When cross-validation value (q$^2$) is 0.68 at 3 components, the Pearson correlation coefficient ($r^2$) is 0.900. From the basis on the findings, the model was appeared by the contour map such as steric field and electrostatic field relationships between quantitative structure and the bioactivity of the various substrate derivatives. Measured bioactivities (obs. pl$_{50}$) of unknown compounds are very similar to predicted activity (pred. pl$_{50}$) according to the CoMFA model. As the results of prediction, we could conclude that the bioactivities were increased by creation of R$_1$ substitution of 5,5-dime-thylhexoxy, 6,6-dimethylheptyl, 1-amino-6,6-dimethylheptyl group etc and R$_2$ substitution of hydroxy, methyl, methoxy group etc.p etc.

  • As increasing the growth of the Internet in medical area, a new technology to transmit effectively massive medical data is required. In optical internet, all OBS nodes have fiber delay lines, hardware components. These components are calculated under some optimal traffic conditions, and this means that if the conditions change, then the components should be altered. Therefore, in this article a new service differentiation algorithm using the previously installed components is proposed, which is used although the conditions vary. When traffic conditions change, the algorithm dynamically recalculates the threshold value used to decide the length of data bursts. By doing so, irrelevant to changes, the algorithm can maintain the service differentiation between classes without replacing any fiber delay lines. With the algorithm, loss sensitive medical data can be transferred well.

Kinetics and Mechanism of the Aminolysis of Aryl N-Benzyl Thiocarbamates in Acetonitrile

  • The aminolysis reactions of phenyl N-benzyl thiocarbamate with benzylamines in acetonitrile at $50.0^{\circ}C$ are investigated. The reactions are first order in both the amine and the substrate. Under amine excess, pseudo-first coefficient ($k_{obs}$) are obtained, plot of $k_{obs}$ vs free amine concentration are linear. The signs of ${\rho}_{XZ}$ (< 0) are consistent with concerted mechanism. Moreover, the variations of $\rho_X$ and $\rho_Z$ with respect to the sustituent in the substrate and large ${\rho}_{XZ}$ value indicate that the reactions proceed concerted mechanism. The normal kinetic isotope effects ($k_H/k_D$ = 1.3 ~ 1.5) involving deuterated benzylamine nucleophiles suggest a hydrogen-bonded, four-centered-type transition state. The activation parameters, ${\Delta}H^\ddagger$ and ${\Delta}S^\ddagger$, are consistent with this transition state structure.

Impaired Osteogenesis in Human Induced Pluripotent Stem Cells with Acetaldehyde Dehydrogenase 2 Mutations

  • Acetaldehyde dehydrogenase 2 (ALDH2) is the second enzyme involved in the breakdown of acetaldehyde into acetic acid during the process of alcohol metabolism. Roughly 40% of East Asians carry one or two ALDH2*2 alleles, and the presence of ALDH2 genetic mutations in individuals may affect the bone remodeling cycle owing to accumulation of acetaldehyde in the body. In this study, we investigated the effects of ALDH2 mutations on bone remodeling. In this study, we examined the effects of ALDH2 polymorphisms on in vitro osteogensis using human induced pluripotent stem cells (hiPSCs). We differentiated wild-type (ALDH2*1/*1-) and ALDH2*1/*2-genotyped hiPSCs into osteoblasts (OBs) and confirmed their OB characteristics. Acetaldehyde was administered to confirm the impact caused by the mutation during OB differentiation. Calcium deposits formed during osteogenesis were significantly decreased in ALDH2*1/*2 OBs. The expression of osteogenic markers were also decreased in acetaldehyde-treated OBs differentiated from the ALDH2*1/*2 hiPSCs. Furthermore, the impact of ALDH2 polymorphism and acetaldehyde-induced stress on inflammatory factors such as 4-hydroxynonenal and tumor necrosis factor α was confirmed. Our findings suggest that individuals with ALDH2 deficiency may face challenges in acetaldehyde breakdown, rendering them susceptible to disturbances in normal bone remodeling therefore, caution should be exercised regarding alcohol consumption. In this proof-of-concept study, we were able to suggest these findings as a result of a disease-in-a-dish concept using hiPSCs derived from individuals bearing a certain mutation. This study also shows the potential of patient-derived hiPSCs for disease modeling with a specific condition.

Chemical Reactivity between Ni(II)-Macrocycle Complex Ions ($NiL_m{^{2+}}$) and $CN^-$ (Ni(II)-거대고리 리간드 착이온 ($NiL_m{^{2+}}$) 과 $CN^-$ 이온간의 반응성)

  • The Chemical reactions between $NiL_m{^{2+}}\{$Ni(rac-1[14]7-diene)^{2+},\;Ni(meso-1[14]7-diene)^{2+},\;Ni(1[14]4-diene)^{2+},\;{\alpha}-Ni(rac-[14]-decane)^{2+},\;{\beta}-Ni(rac-[14]-decane)^{2+},\;and\;Ni(meso-[14]-decane)^{2+}$}\and\ CN^-$ ion were studied by the spectrophotometric method. The equilibrium constants (K_1$) for the 1:1 complex ion, $[NiL_m(CN)]^+\;with\;NiL_m{^{2+}}\;and\;CN^-$ ion were determined in the range of 3 to $25^{\circ}C$. The $K_1\;for\;Ni(rac-1[14]7-diene)^{2+},\;Ni(meso-1[14]7-diene)^{2+},\;Ni(1[14]4-diene)^{2+},\;{\alpha}-Ni(rac-[14]-decane)^{2+},\;{beta}-Ni(rac-[14]-decane)^{2+},\;and\;Ni(meso-[14]-decane)^{2+}\;at\;15^{\circ}C$ was 4.7, 5.3, 6.2, 7.5, 9.4, and 9.8, respectively. The values of $K_1$ decreased with increasing temperature. From the temperature effect on equilibrium constant ($K_1$), thermodynamic parameters $({\Delta}H^{\circ},\;{\Delta}S^{\circ},\;{\Delta}G^{\circ})$ for reaction were evaluated and the reaction of $NiL_m{^{2+}}\;and\;CN^-$ ion was exothermic. $NiL_m{^{2+}\;reacts\;with\;CN^-$ ion to give $Ni(CN)_4{^{2-}}$ ion and macrocyclic ligand $(L_m)$. The kinetics of formation of the $Ni(CN)_4{^{2-}}$ ion of varying the $[CN^-],\;[HCN],\;and\;[OH^-]$ have been investigated at 3∼$25^{\circ}C\;and\;0.5M\;NaClO_4$. Maintaining a constant $[CN^-],\;k_{obs}/[CN^-]^2$ increases linearly with increasing [HCN]. In the presence of large quantities of $[OH^-],\;k_{obs}/[CN^-]^2$ also increases linearly with $[OH^-]$. From the temperature effect on kinetic constant (k_{obs})$, parameter of activation $({\Delta}H^{\neq},\;{\Delta}S^{\neq})$ of reaction of $NiL_m{^{2+}}\;with\;CN^-$ ion were determined. For the $Ni(rac-1[14]7-diene)^{2+},\;Ni(meso-1[14]7-diene)^{2+},\;{\alpha}-Ni(rac-[14]-decane)^{2+},\;{\beta}-Ni(rac-[14]-decane)^{2+},\;and\;Ni(meso-[14]-decane)^{2+}\;series\;{\Delta}H^{\neq}$ gradually decrease as the d-d transition energy, $ν(cm^{-1})$ decrease. And the reaction of the five $NiL_m{^{2+}}\;with\;CN^-$ ion take place by way of equal paths.

Effects of feed intake and water hardness on fluralaner pharmacokinetics in layer chickens

  • Background: Fluralaner is a novel drug belonging to the isoxazoline class that acts on external parasites of domestic animals. It is used systemically via drinking water, especially against red poultry mite in layer chickens. Fluralaner is frequently used in layers infected with D. gallinae. However, no study to date has investigated the effects of feed intake and water hardness. Objectives: This study aimed to investigate the effects of variable water hardness and feed intake on the pharmacokinetic profile of fluralaner. Methods: Layer chickens were divided into four groups (n = 8): fed + purified water (Group 1), feed restricted + purified water (Group 2), feed restricted + hard water (Group 3), and feed restricted + soft water (Group 4). After administering a single dose of the drug with drinking water, the blood samples were collected for 21 days. Fluralaner concentrations in plasma samples were determined by liquid chromatography/tandem mass spectrometry. The maximum plasma concentration (Cmax), time to reach maximum plasma concentration (tmax), area under the concentration-time curve values (AUC0-21d), half-life (t1/2), and other pharmacokinetic parameters were calculated. Results: Although the highest maximum plasma concentration (Cmax) was determined in Group 1 (fed + purified water), no statistically significant difference was found in the Cmax, tmax, t1/2, MRT0-inf_obs, Vz/Fobs, and Cl/F_obs parameters between the experimental groups. Conclusions: It was concluded that the feed intake or water hardness did not change the pharmacokinetic profile of fluralaner in layer chickens. Therefore, fluralaner could be used before or after feeding with the varying water hardness in poultry industry.

  • 연안유역으로 유입되는 소하천의 유량 및 오염물질의 농도는 유역의 기상상태(강우량 및 증발량), 토지피복도, 인공구조물의 운영규clr 등에 의하여 변화한다. 특히, 강우에 의한 오염부하량 변화는 평상시의 오염부하량에 비하여 그 변화량이 매우 크기 때문에 평상시의 측정간격으로는 보다 정확한 오염부하량 산정에 한계가 있다. 따라서, 본 연구에서는 충청남도 보령시를 관통하는 대천천을 대상으로 강우에 의한 오염부하량 변화를 연속적으로 측정하였다. 대천천은 상류지역에 청천저수지가 형성되어 있으며, 보령시내를 관통하는 과정에서 도시 오염물질이 유입되는 양상을 보이고 있으며, 천수만 입구지역으로 유입되는 지방하천이다. 유역오염부하량 계산에 필요한 유량은 보(weir)에서 측정한 수위자료를 이용하여 환산하였으며, 수온, 탁도 및 염도는 OBS-3A 장비를 이용하여 연속측정하였다. 또한, 영양염류의 오염물질 농도는 30분 간격으로 채수하여 분석하였다. 분석결과, 오염물질 항목에 따라 시간에 따른 오염부하량 변화양상이 매우 상이하게 나타났으며, 변화 폭도 매우 크게 나타나서 강우시의 오염부하량을 추정하기 위해서는 1시간 간격 이내의 관측자료를 이용하여 추정하는 방법이 타당한 것으로 파악되었다.

A Study on Organic Brain Syndrome in the Elderly at the Home for the Aged (양로원 재원노인의 정신건강에 관한 연구 I. 기질성 뇌증후군을 중심으로)

  • The authors studied organic brain syndrome, using organic brain syndrome scale of the Diagnostic Interview Schedule for DSM-II, in the subjects of 65 males and 231 females at the homes for the aged in the Taegu and Kyong-buk areas. The authors collected the data during the period from June to August, 1986, and applied ANOVA and $x^2$-test in order to compare various psychologic factors in relation to the organic brain syndrome. The results could be summarized as follows : The elderly males(15%) showed high OBS scores of 12 and over, while ninety elderly females(39%) showed the same scores. Among those psychosocial factors, age, education, birth place, occupation, religion, history before institutionalization are significantly related to the OBS scores.

  • A digital data logging system has been developed for the purpose of a compact offline Ocean Bottom Seismometer(OBS). The Digital Data Logger(DDL) consists of A/D system, Micom with storage memory and firmware managing data files. The A/D system acquires data of 16bit/4ch with sampling rate of 250Hz per channel. The Micom, a micro controller board with T33521 processor of 8051 class, was equipped with 8 flash memories of 128MB for data storage capacity of 1GB. The firmware stores the acquiring data in form of binary files. The DDL was designated to be compact and light and to consume low energy as possible. The DDL is to interface with PC through USB(Universal Serial Bus). The performance of the DDL has been validated through tests with respect to a 3-axis seismometer.

  • The Wallach rearrangement of 4'-halogenated azoxybenzenes in strong sulfuric acid was reinvestigated by UV-Vis spectrophotometric method. Interestingly an excellent linearity $(logk_{obs}=-0.61\;{\sigma}_R{^+} -3.29,\;{\gamma = 0.998)$ was obtained on plotting log (rate) vs. substituent constant, ${\sigma}_R{^+}$, which is known as the Pi delocalization parameter. Thus B-H mechanism via a charged intermediate $(DC^{++})$, which can be largely stabilized by resonance, is far more favored for the Wallach rearrangement.
