Metabolism of Ginseng Saponins by Human Intestinal Bacteria (Park II) (사람의 장내세균에 의한 인삼 사포닌의 대사(제2보))

  • Following ginsenoside-Rb1-hydrolyzing assay, strictly anaerobic bacteria were isolated from human feces and identified as Prevotella oris. The bacteria hydrolyzed ginsenoside Rb1 and Rd to $20-O-{\beta}-D-glucopyranosyl-20(S)-protopanaxadiol$ (I), ginsenoside Rb2 to $20-O-[{\alpha}-L-arabinofuranosyl (1{\rightarrow}6)-{\beta}-D-glucopyranosyl] - 20(S)-protopanaxadiol$ (ll) and ginsenoside Rc to $20-O-[{\alpha}-L-arabinofuranosyl (1{\rightarrow} 6){\beta}-D-g1ucopyranosyl]-20(S)-protopanaxadiol$ (III) like fecal microflora, but did not attack ginsenoside Re nor Rgl (Protopanaxatriol-type). Pharmacokinetic studies of ginseng saponins was also performed using specific pathogen free rats and demonstrated that the intestinal bacterial metabolites I-111, 20(S)- protopanaxatriol(IV) and 20(S)-protopanaxadiol(V) were absorbed from the intestines to $blood(0.4-5.1\;{\mu}g/ml)$ after oral administration with total saponin(1 g/kg/day).

Inhibition of LPS Induced iNOS, COX-2 and Cytokines Expression by $Genistein-4'-O-{\alpha}-L-Rhamnopyranosyl-(1-2)-{\beta}-D-Glucopyranoside$ through the $NF-{\kappa}B$ Inactivation in RAW 264.7 Cells ($Genistein-4'-O-{\alpha}-L-rhamnopyranosyl-(1-2)-{\beta}-D-glucopyranoside$의 RAW 264.7 세포에서 $NF-{\kappa}B$ 불활성화를 통한 LPS에 의해 유도되는 iNOS, COX-2 그리고 cytokine들의 발현 저해효과)

  • This study were designed to evaluate the anti-inflammatory effects of $genistein-4'-O-{\alpha}-L-rhamnopyranosyl-(1-2)-{\beta}-D-glucopyranoside$ (GRG) isolated from Sophora japonica (Leguminosae) on the lipopolysaccharide (LPS)-induced nitric oxide (NO) and prostaglandin ($PGE_2$) production by RAW 264.7 cell line. GRG significantly inhibited the LPS-induced NO and $PGE_2$ production. Consistent with these observations, GRG reduced the LPS-induced expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) at the protein and mRNA levels in a concentration-dependent manner. In addition, the release and the mRNA expression levels of tumor necrosis $factor-{\alpha}\;(TNF-{\alpha})$ and interleukin-6 (IL-6) were also reduced by GRG. Moreover, GRG attenuated the LPS-induced activation of nuclear factor-kappa B ($NF-{\kappa}B$), a transcription factor necessary for pro-inflammatory mediators, iNOS, COX-2, $TNF-{\alpha}$ and IL-6 expression. These results suggest that the down regulation of iNOS, COX-2, $TNF-{\alpha}$, and IL-6 expression by GRG are achieved by the downregulation of $NF-{\kappa}B$ activity, and that is also responsible for its anti-inflammatory effects.

The Occurrence of Laminarionema elsbetiae (Phaeophyceae) on Rhodymenia pseudopalmata (Rhodophyta) from the Patagonian Coasts of Argentina: Characteristics of the Relationship in Natural and Experimental Infections, and Morphology of the Epi-endophyte in Unialgal Free Cultures

  • The occurrence of Laminarionema elsbetiae (Ectocarpaceae, Phaeophyceae), as epi-endophyte of Rhodymenia pseudopalmata (Rhodymeniales, Rhodophyta), described from Santa Isabel, Rawson, Argentina. L. elsbetiae grows in the host tissues forming epi-endophytic relationship in the epidermal, cortical and medullar layers. Epiphytic thalli of L. elsbetiae were unbranched filaments emerging from hostis surface. Reproductive structures of L. elsbetiae on the host were absent. On the contrary, free cultured individuals formed different reproductive structures. Macrozoosporangia containing a single large motile zoospore originated from vegetative cells, they were conical to cylindrical in shape, 30-50 ${\mu}m$ in length and 18-20 ${\mu}m$ in wide. Uniseriate plurilocular zoosporangia were cylindrical shape, 40 ${\mu}m$ in length and 10-13 ${\mu}m$ in wide. Sexual fusion was not seen. In mixed cultures of L. elsbetiae with R. pseudopalmata fronds, L. elsbetiae infected the host, grew as in natural host and, formed macrosporangia between host subcortical cells. Gametophytes of L. elsbetiae were filaments with diffuse growth, branched with a branch pattern alternate or opposite. Gametangia were plurilocular, uni or biseriate and lateral. When mature they contained 2 to 6 isogametes. The presence L. elsbetiae on R. pseudopalmata could be defined as an epi-endophytic relationship. The percentage of infection of R. pseudopalmata thalli by L. elsbetiae was 34%.A25% of the infected thalli presented a low, non-symptomatic level infection, whereas a 62% and a 13% of them exhibited respectively moderate and high indexes of infection.

Cytotoxic Anthraquinones and Stilbenes from Reynoutria sachalinensis (Fr. Schm.) Nakai

  • Five known anthraquinones, physcion (1), I-O-methylemodin (2), emodin (3), $physcion-8-O-{\beta},-D-glucopyranoside$ (5), $emodin-8-O-{\beta},-D-glucopyranoside$ (6) and two known stilbenes, trans-resveratrol (4), $trans-resveratrol-3-O-{\beta},-D-glucopyranoside$ (7) were isolated from MeOH extract of Reynoutria sachalinensis (Polygonaceae). All structures were unambiguously established by 1D and 2D NMR and MS data and the compounds were evaluated for their cytotoxicity against L1210, HL-60, BI6F10 tumor cell lines in MTT assay. Among the compounds, trans-resveratrol (4) exhibited significant cytotoxic activity with $IC_{50}$ values of 9.2, 6.7 and $9.8\;{\mu}g/ml$, against the test cell lines respectively, but compounds 1-3 exhibited the moderate cytotoxic activity.

Anion-Dependent Exocyclic Mercury(II) Coordination Polymers of Bis-dithiamacrocycle

  • Synthesis and structural characterization of mercury(II) halides and perchlorate complexes (1-4) of bis-$OS_2$-macrocycle (L) are reported. L reacts with mercury(II) chloride and bromide to yield an isostructural 2D coordination polymers with type $[Hg(L)X_2]_n$ (1: X = Cl and 2: X = Br). In 1, each Hg atom which lies outside the cavity is six-coordinate with a distorted octahedral geometry, being bound to four adjacent ligands via monodentate Hg-S bonds and two remaining sites are occupied by two terminal chlorido ligands to form a fishnet-like 2D structure. When reacting with mercury(II) iodide, L afforded a 1D coordination polymer $\{[Hg_2(L)I_4]{\cdot}CHCl_3\}_n$ (3) in which each exocyclic Hg atom is four-coordinate, being bound to two sulfur donors from different ligands doubly bridging the ligand molecules in a head-to-tail mode. The coordination sphere in 3 is completed by two iodo terminal ligands, adopting a distorted tetrahedral geometry. On reacting with mercury(II) perchlorate, L forms solvent-coordinated 1D coordination polymer $\{[Hg_2(L)(DMF)_6](ClO_4)_4{\cdot}2DMF\}_n$ (4) instead of the anion-coordination. In 4, the Hg atom is five-coordinate, being bound to two sulfur donors from two different ligands doubly bridging the ligand molecules in a side-by-side mode to form a ribbon-like 1D structure. The three remaining coordination sites in 4 are completed by three DMF molecules in a monodentate manner. Consequently, the different structures and connectivity patterns for the observed exocyclic coordination polymers depending on the anions used are influenced not only by the coordination ability of the anions but also by anion sizes.

Spitzer and Herschel observations of protostellar outflows in L1251B

  • L1251B is an excellent example of a small group of pre- and protostellar objects in low-mass star-forming region. Previous interferometer data with a high angular resolution resolved the outflows associated with L1251B into a few components. To understand the physical conditions of the protostellar outflows in L1251B, we mapped this region spectroscopically with Spitzer/IRS and obtained spectral line data from Herschel/PACS. Spitzer/IRS provides the S(0)-S(7) pure rotational lines of H2 as well as fine-structure emissions produced in shocks such as S, [Si II], and [Fe II] and it is a powerful tool for studying shocked interstellar gas. In addition, [O I] lines observed with Herschel/PACS are described well by J-type shock models expected in the outflows from protostars. We will present an analysis of the L1251B protostellar outflow in the H2 pure rotational lines and fine-structure emissions.

  • The authors report on the fabrication of thin film transistors (TFTs) that use amorphous indium-gallium-zinc oxide (a-IGZO) channel and have the channel length (L) and width (W) patterned by dry etching. To prevent the plasma damage of active channel, a 100-nm-thckness $SiO_{x}$ by PECVD was adopted as an etch-stopper structure. IGZO TFT (W/L=10/50${\mu}m$) fabricated on glass exhibited the high performance mobility of $35.8\;cm^2/Vs$, a subthreshold gate voltage swing of $0.59V/dec$, and $I_{on/off}$ of $4.9{\times}10^6$. In addition, 4.1” transparent QCIF active-matrix organic light-emitting diode display were successfully fabricated, which was driven by a-IGZO TFTs.

Effects of Dipsacus asper on Free Radical Scavenging and Lowdensity Lipoprotein Oxidation (속단의 유리기소거활성 및 저밀도지방단백질 산화에 미치는 영향)

  • The roots of Dipsacus asper are used in traditional Chinese medicine as an analgesic, enhancement of liver activity, an anti-inflammatory agent and the treatment of fractures. In order to investigate the antioxidative activity, Dipsaci Radix was extracted with MeOH and fractionated with $CHCl_3$, EtOAc, BuOH and water. MeOH ex. and its solvent fractions were determined on the free radical scavenging activity by DPPH method and antioxidative activity on low density lipoprotein oxidation. The EtOAc and BuOH fractions showed antioxidative activities and from their fractions, two compounds were isolated and identified as hederagenin 3-O-${alpha}$-L-arabino pyranoside (compound 1) and O-${\alpha}$-L-arabinopyranosyl hed-eragenin 28-O-${\beta}$-D-glucopyranosyl ($1{\to}6$)-${\beta}$-D-glucopyranosyl ester (compound 2). Saponin glycoside, compound I showed anti-oxidative activity.

Resonant Properties of $Pb(La,Ce)TiO_3$ System Ceramics (레조네이터용 $Pb(La,Ce)TiO_3$ 계 세라믹스의 공진특성)

  • In this study, Pb(La Ce)$TiO_3$ composition ceramics were manufactured for 20 MHz resonator application. Electromechanical coupling factor, mechanical quality factor and dynamic range of thickness vibration mode were measured as the variations of L/T(length/thickness) ratio of ceramic substrats. Mechanical quality factor and dynamic range of third overtone thickness vibration mode showed the highest value of 2,773 and 52.22 dB at specimen S4(l/t=12), respectively. The excellent temperature stability of resonant frequency suitable for resonator application was shown, regardless of thermal shock.

  • The characteristics of the type I and type II SHG in LiB305 crystals grown by TSSG method have been investigated using 1064 nm beam from a Q-switched Nd:YAG laser. The measured phase matching angles and angular acceptance bandwidths were $\theta_m=90^{\circ}, \phi_m=11.6^{\circ}$, <$\delta\theta_{int}L_{1/2}=3.3^{\circ}-cm^{1/2}, \theta\phi_{int}L=0.27^{\circ}-cm^{1/2}$ for type I SHG and $\theta_m=20^{\circ}, \phi_m=90^{\circ}$, TEX>$\delta\theta_{int}L_=0.65^{\circ}-cm, \theta\phi_{int}L^{1/2}=3.5^{\circ}-cm^{1/2}$ for type II SHG, respectively. Thp. type I NCPM temperature of 1064 nm beam was found to be $149^{\circ}C$ with the temperature bandwidth $\DeltaTL$of $4.8^{\circ}C-cm$. An energy conversion efficiency of about 1.8% with 2.6 mm thick LBO crystal at an incident power of TEX>$171 MW/\textrm{cm}^2$ was demonstrated. The measured $d_{32} was 0.74\pm0.05 pm/V$..
