Insulin Receptor Substrate Proteins and Diabetes

  • The discovery of insulin receptor substrate (IRS) proteins and their role to link cell surface receptors to the intracellular signaling cascades is a key step to understanding insulin and insulin-like growth factor (IGF) action. Moreover, IRS-proteins coordinate signals from the insulin and IGF receptor tyrosine kinases with those generated by proinflammatory cytokines and nutrients. The IRS2-branch of the insulin/IGF signaling cascade has an important role in both peripheral insulin response and pancreatic $\beta$-cell growth and function. Dysregulation of IRS2 signaling in mice causes the failure of compensatory hyperinsulinemia during peripheral insulin resistance. IRS protein signaling is down regulated by serine phosphorylation or protea-some-mediated degradation, which might be an important mechanism of insulin resistance during acute injury and infection, or chronic stress associated with aging or obesity. Under-standing the regulation and signaling by IRS1 and IRS2 in cell growth, metabolism and survival will reveal new strategies to prevent or cure diabetes and other metabolic diseases.

  • This study was designed to observe the effects of nutritional supplementation of general Korean diet on protein metabolism in growing rats. Seventy weanling Sprague-Dawely male rats weighing 76.8$\pm$1.45g were blocked into ten groups and fed ten different experimental diets for eight weeks. The rats in cereal-vegetable group were fed ad libitum for all the experimental period and those in other experimental groups were pairfed following the feed intake of rats in C-V gp. Control group was given 72% cornstarch-29% casein diet : C-V gp was fed cerealvegetable diet composed of rice, barley, soybean and Chinese cabbage : the other eight groups were fed C-V diets supplemented with casein, vitamin B\sub 2\ calcium, vitamin A, vitamin B\sub 2\ and vitamin A, vitamin A and calcium, vitamin B\sub 2\ and calcium, vitamin A and vitamin B\sub 2\ and calcium, respectively, on the basis of each nutrients content of standard diet.

Combined analysis of transcriptome and proteome for high cell density cultivation of Escherichia coli

  • For understanding physiology and metabolism under various culture conditions, combined analysis of transcriptome and proteome is attractable way. We have manufactured DNA microarray containing 2,850 genes including all functionally known and putative ones. In this study, we report analysis of transcriptome and proteome during the high cell density culture of E. coli by using DNA microarray and 2-DE. Fed-batch fermentation of E. coli was carried out by exponential feeding of nutrients until the maximum cell density reached 74 g dry cell weight/L (g DCW/L). Changes in transcriptome and proteome during the HCDC are analyzed qualitatively and quantitatively to provide their physiological and metabolic meanings.

Autophagy Dysregulation and Obesity-Associated Pathologies

  • Autophagy is one of the major degradative mechanisms that can eliminate excessive nutrients, toxic protein aggregates, damaged organelles and invading microorganisms. In response to obesity and obesity-associated lipotoxic, proteotoxic and oxidative stresses, autophagy plays an essential role in maintaining physiological homeostasis. However, obesity and its associated stress insults can often interfere with the autophagic process through various mechanisms, which result in further aggravation of obesity-related metabolic pathologies in multiple metabolic organs. Paradoxically, inhibition of autophagy, within specific contexts, indirectly produces beneficial effects that can alleviate several detrimental consequences of obesity. In this minireview, we will provide a brief discussion about our current understanding of the impact of obesity on autophagy and the role of autophagy dysregulation in modulating obesity-associated pathological outcomes.

  • Special formulas are those in which one of the basic nutrients (usually the protein and/or carrbohydrate) has been changed to an alternative nutrient that an individual baby may better tolerate. The following are specialized formulas : Soy formulas, hypoallergenic formulas, Lactose free formulas, Premature baby formulas, Low phosphate formulas, Formulas for inborn errors of metabolism. Less is known about the long-term effects of feeding babies these special formulas and each formula has its unique properties and indications. The pediatrician must acquaint themselves with all commercially available infant formulas so as to provide the parent with reliable and unbiased information about them.

Mechanisms of amino acid sensing in mTOR signaling pathway

  • Amino acids are fundamental nutrients for protein synthesis and cell growth (increase in cell size). Recently, many compelling evidences have shown that the level of amino acids is sensed by extra- or intra-cellular amino acids sensor(s) and regulates protein synthesis/degradation. Mammalian target of rapamycin complex 1 (mTORC1) is placed in a central position in cell growth regulation and dysregulation of mTOR signaling pathway has been implicated in many serious human diseases including cancer, diabetes, and tissue hypertrophy. Although amino acids are the most potent activator of mTORC1, how amino acids activate mTOR signaling pathway is still largely unknown. This is partly because of the diversity of amino acids themselves including structure and metabolism. In this review, current proposed amino acid sensing mechanisms to regulate mTORC1 and the evidences pro/against the proposed models are discussed.

  • The prevalence of obesity has been increasing worldwide. Several dietary treatments have been suggested to control weight, and recent guidelines recommend individualizing the composition of macronutrients. Carbohydrates are the most important nutrients in meals, and carbohydrate restriction is a dietary strategy that promotes weight loss. A low-carbohydrate diet is effective for short-term weight loss and can help improve glycated hemoglobin, systolic blood pressure, diastolic blood pressure, and triglyceride levels; however, the long-term effects and safety of this diet remains doubtful. In the short term, there is a risk of gastrointestinal symptoms such as vomiting, diarrhea, constipation, and gastroesophageal reflux, and type 1 diabetes patients are at risk of severe hypoglycemia, while in the long term, it can lead to malnutrition and decreased exercise capacity. Thus, rather than limiting the intake of carbohydrates, it is important to limit the intake of refined grains, sugar, honey, syrup, and sweetened beverages while maintaining the planned carbohydrate intake rate and improving meal quality.

고지혈증 환자의 웹기반 식사관리 및 영양평가 프로그램 (A Web-based Internet Program for Nutritional Assessment and Diet Management of Patient Having Hyperlipidemia)

  • 본 연구는 임상영양 분야의 전문 웹사이트로서 고지혈증 환자를 위한 식사관리 및 영양평가 프로그램을 개발하기 위하여 수행되었다 프로그램은 고지혈증을 고콜레스테롤혈증과 고중성지방혈증으로 분류하고 이들에 대한 식사관리, 영양상담 및 영양평가 프로그램으로 구성하였다. 프로그램은 표준체중 및 열량 필요량 파일, 열량별 식단 및 일일 식단표 파일, 식사요법 및 영양관리 파일, 식사력 조사 및 평가파일, 식품 및 영양소 데이터베이스 파일, 식품의 지방산 파일, 음식 영양소 함량 및 영양소별 20순위 식품 파일 등을 데이터베이스로 하여 웹 페이지 형식으로 만들어졌다. 사용자는 인터넷 사이트로 들어가 자신의 정보를 입력하면 그 결과로 1일 열량 필요량 및 기타 영양소 필요량이 제시되고 그에 알맞은 식단을 보고 이용할 수 있도록 하였다. 식사관리 프로그램에서는 열량별 식단 자료실을 두어 1200 kcal에서 2500kcal까지 100 kcal 단위로 일주일간의 식단을 볼 수 있게 하였고, 식단계획시 고려사항, 식품의 선택방법, 식품교환표, 고지혈증 식사지침, 식사요법 및 영양관리, 영양소별 20순위식품, 용어설명 및 관련 사이트를 소개하였다 영양상담 프로그램으로는 고지혈증 예방을 위한 식습관 평가와 질환에 바른 식사력조사 및 상담 평가, on-line 영양상담, FAQ 등이 있다. 영양평가프로그램으로는 섭취음식을 입력함으로서 영양소 섭취 상태, 열량 영양소, 지방산, 식사별, 식품군별 영양소 섭취상태, 동ㆍ식물성 식품 영양소 섭취 상태, 기간별 영양소 섭취 변화 등을 평가받을 수 있도록 하였다. 영양소 섭취상태는 환자의 필요량과 비교하여 그래프로 제시되고, 열량영양소, 식사별 영양소, 지방산 섭취 상태는 아침, 점심, 저녁 ,간식으로 구분하여 결과를 볼 수 있고, 동ㆍ식물성 식품 섭취 및 기간별 영양소 변화 등은 각각의 영양소별로 그 섭취 상태를 비교할 수 있도록 하였다.

Folate-Related Nutrients, Genetic Polymorphisms, and Colorectal Cancer Risk: the Fukuoka Colorectal Cancer Study

  • One-carbon metabolism plays an important role in colorectal carcinogenesis. Meta-analyses have suggested protective associations of folate and vitamin $B_6$ intakes with colorectal cancer primarily based on studies in Caucasians, and genetic polymorphisms pertaining to the folate metabolism have been a matter of interest. Less investigated are the roles of methionine synthase (MTR) and thymidylate synthetase (TS) polymorphisms in colorectal carcinogenesis. In a study of 816 cases and 815 community controls in Japan, we investigated associations of dietary intakes of folate, methionine, vitamin $B_2$, vitamin $B_6$, and vitamin $B_{12}$ with colorectal cancer risk. The associations with MTR 2756A>G, MTRR 66A>G, and TSER repeat polymorphism were examined in 685 cases and 778 controls. Methionine and vitamin $B_{12}$ intakes were inversely associated with colorectal cancer risk, but the associations were totally confounded by dietary calcium and n-3 fatty acids. The other nutrients showed no association with the risk even without adjustment for calcium and n-3 fatty acids. The TSER 2R allele was dose-dependently associated with an increased risk. The MTR and MTRR polymorphisms were unrelated to colorectal cancer risk. There was no measurable gene-gene or gene-nutrient interaction, but increased risk associated with the TSER 2R allele seemed to be confined to individuals with high folate status. This study does not support protective associations for folate and vitamin $B_6$. The TSER 2R allele may confer an increased risk of colorectal cancer. The role of the TSER polymorphism in colorectal carcinogenesis may differ by ethnicity.

Lipogenesis와 adipose expansion의 전사조절 (Transcriptional Regulation of Lipogenesis and Adipose Expansion)

  • PPARγ와 C/EBPα는 adipose tissue 발생에 필요한 핵심 adipogenic TFs이다. 두 TFs는 adipose tissue의 배아 발생에 있어 초기 adipogenesis와 adipocytes 유전자 발현을 조절한다. Adipose expansion은 adipose tissue가 태어난 뒤에도 계속 팽창이 지속되는 것을 말한다. 이러한 adipose expansion은 후기 adipogenesis의 과정인 lipogenesis가 요구된다. 특히 간과 adipose tissue은 탄수화물이 기본적으로 fatty acids으로 전환되는 de novo lipogenesis (DNL)의 주요 장기이다. Fatty acids는 이어서 glycerol-3-phosphate에 에스테르화되어 adipocytes의 lipid droplets 생성을 위해 triglycerides로 전환된다. 간의 DNL이 활발하게 연구가 된 반면, in vivo에서 adipocytes의 DNL은 아직 잘 알려져 있지 않다. 그러므로, adipose expansion과 DNL에 대한 이해는 비만, 2형 당뇨 그리고 대사성 질환을 위한 치료제 개발에 도움을 줄 수 있다. Adipocytes에서 DNL 유전자 발현은 ChREBP나 SREBP1a와 같은 lipogenic TFs 뿐 아니라, lipogenesis coactivator에 의해 전사 수준에서 조절된다. 최근 in vivo 연구에 의해 lipogenesis 유전자 발현과 adipose expansion의 새로운 측면이 밝혀졌다. 향후 구체적인 분자기전 연구는 어떻게 영양분이 DNL과 adipose expansion을 조절하는 지를 규명해 줄 것이다. 본 리뷰논문은 전사조절 관점에서 adipocytes와 adipose expansion에 있어 DNL에 대한 최신 연구결과를 요약정리 할 것이다.
