방사선피폭관리시스템를 위한 D-Shuttle 선량계의 방사선 선량측정 (Radiation Dose Measurement of D-Shuttle Dosimeter for Radiation Exposure Management System)

  • D-Shuttle (Chiyoda Technol Corporation, Tokyo, Japan) 선량계를 이용하여 개인피폭관리 및 자연방사선량의 모니터링을 위한 기초자료를 제공하는데 연구의 목적이 있다. D-Shuttle을 이용하여 선량을 산출하였다. 선량보고서에서 400 일 노출되었을 때에 1.346 mSv 이었고, 연간선량 (annual dose per year)은 1.228 mSv/year, 평균시간선량 (average dose per hour)은 $0.014{\mu}Sv/hr$ 이었다. 국내의 개인 외부피폭선량 (1.295 mSv/year =Korea average natural individual external dose), 국내의 연간부가선량 (additional dose per year)은 -0.0663 mSv/year 이다. D-Shuttle은 방사선모니터링을 위한 개인선량계로 방사선의 검출성능 우수한 기능, 실시간 방사선 피폭관리, 방사선 작업의 경보 기능, 효율적이고 사용이 편리한 개인 방사선선량의 피폭관리로 ALARA에 매우 유용한 선량계로 사용할 수 있다. 방사선작업종사자와 지역주민의 방사선모니터링 측정기기로 병원, 산업, 의료현장, 원전사고 지역과 비파괴 분야의 위험한 지역에서 방사선모니터링으로 활용될 수 있다.

Platynosomum fastosum (Trematoda: Dicrocoeliidae) from Cats in Vietnam: Morphological Redescription and Molecular Phylogenetics

  • The present study was performed to reveal the morphological characteristics and molecular phylogenetic position of Platynosomum fastosum Kossack, 1910. A total 167 specimens of P. fastosum were collected in 8 (4.9%) out of 163 sets of gall-bladders and bile ducts of cats. The number of worms was 1-105 per infected cat. This species was characterized by having a long and slender body, slightly larger ventral sucker than the oral sucker, indistinct prepharynx, small pharynx, short esophagus, bifurcation midway between 2 suckers, and ceca extending to the posterior end of the body. The length of the partial sequences of ITS1 and 5.8S rDNA of P. fastosum were 990 bp, GC-rich. AT/GC ratio was 0.9, there were 9 polymorphic sites, and intraspecific variations ranged from 0.1% to 0.9%. Phylogenetic analyses by neighbor-joining phylogram inferred from ITS1 rDNA sequences revealed that the genetic distance between P. fastosum specimens ranged from 0.3 to 1.5% while the smallest interspecific distance among dicrocoeliid species was 20.9 %. The redescription and genetic characters of P. fastosum are taxonomically important to recognize future different species of the genus Platynosomum showing high intraspecific and morphological variability.

Genetic diversity of Kalopanax pictus populations in Korea based on the nrDNA ITS sequence

  • $Kalopanax$ $pictus$ is a long-lived deciduous perennial tree in the family Araliaceae mainly distributed in the East Asia. In Korea, this species is of ecological and medical importance. Because typical populations of this species are small and distributed in patches, $K.$ $pictus$ has been considered as a narrow habitat species. To understand the genetic diversity and population structure of this species, the sequence variation of the nuclear ribosomal DNA (nrDNA) internal transcribed spacer (ITS) region was analyzed among 18 different $K.$ $pictus$ populations in the present investigation. The nrDNA ITS sequences of Korean populations investigated in this study showed identical of 616 bp in length, and no any nucleotide variation was found in the entire nrDNA ITS region sequence. This result suggested that the $K.$ $pictus$ populations in Korea might belong to the same isolate, and no mutation was found in the nrDNA ITS region. Compared with other known ITS sequence sources from $K.$ $pictus$ populations, only four variable nucleotide sites were found within the entire ITS region. Very narrow genetic diversity appearing in the population level of $K.$ $pictus$ makes us hypothesize that their relatively isolated habitats. The long-lived traits might be one main reason. However, another probability was that the nr-DNA ITS region might be noneffective in classifying populations of $K.$ $pictus$. Thus, to further understand the phylogenetic relationship of $K.$ $pictus$ populations, more samplings should be performed based on more DNA sequences.

NMR Studies on the Structure of Human Annexin I

  • Human annexin I is a member of annexin family of calcium dependent phospholipid binding proteins, which have been implicated in various physiological roles including phospholipase A$_2$ (PLA$_2$) inhibition, membrane fusion and calcium channel activity. In this work, the structure of N-terminally truncated human annexin I (Δ-annexin I) and its interactions with Ca$\^$2+/, ATP and cAMP were studied at atomic level by using $^1$H, $\^$15/N, $\^$l3/C NMR (nuclear magnetic resonance) spectroscopy. The effect of Ca$\^$2+/ binding on the structure of Δ-annexin I was investigated, and compared with that of Mg$\^$2+/ binding. The addition of Ca$\^$2+/ to Δ-annexin I caused some changes in the high field and low field regions of $^1$H NMR spectra. Whereas, upon addition of Mg$\^$2+/ to Δ-annexin I, almost no change could be observed. Also we found that the binding ratio of ATP to Δ-annexin I is 1. Because Δ-annexin I is a large protein with 35 kDa molecular weight, site-specific (carbonyl-$\^$l3/C, amide-$\^$15/N) labeling technique was used to determine the interaction sites of Δ-annexin I with Ca$\^$2+/ and ATP. Assignments of all the histidinyl carbonyl carbon resonances have been completed by using Δ-annexin I along with its specific 1,2-subdomain. The carbonyl carbon resonances originating from His52 and His246 of Δ-annexin I were significantly affected by Ca$\^$2+/ binding, and some Tyr and Phe resonances were also affected. The carbonyl carbon resonances originating from His52 is significantly affected by ATP binding, therefore His52 seems to be involved in the ATP binding site of Δ-annexin I.

중성자 방사화분석법과 Gent SFU 샘플러를 이용한 도시의 농촌지역의 대기분지($PM_{10}$)관측 연구 (Study on Airborne Particulate Matter ($PM_{10}$) Monitoring in Urban and Rural Area by Using Gent SFU Sampler and Instrumental Neutron Activation Analysis)

  • The aim of this research is to collect and characterize fine particles (FPM:$\leq$2.5${\mu}{\textrm}{m}$) and coarse particles (CPM: 2.5~10${\mu}{\textrm}{m}$) using a low volume air sampler provided by the IAEA, at urban (Taejon) and rural area(Wonju) for a period of about two years(April 1996 to May 1998) and to promote a use of nuclear analytical techniques for air pollution studies. For the collection of airborne particulate matter (PM(sub)10), the Gent stacked filter unit sampler and polycarbonate membrane filters were employed. The concentration of trace elements in collected APM samples were determined byu instrumental Neutron Activation Analysis. For validation of the analytical data, internal quality control were implemented by using both the comparison of the analytical results of standard reference materials(NIST SRM 1648) and interlaboratory comparison for proficiency test (NAT-3). The standard uncertainty was less than 15% and Z-score of two samples were within $\pm$1. The monitoring of (PM(sub)10) mass concentration and elemental concentrations were carried out weekly. The average mass concentration of (PM(sub)10) in urban and rural areas were 59.2$\pm$36.5$\mu\textrm{g}$/㎥ and 41.4$\pm$23.7$\mu\textrm{g}$/㎥, respectively. To investigate the emission source, the enrichment factors were calculated for the fine and coarse particle fractions at two sites, respectively and these values were classified for anthropogenic and soil origin elements.

과산화수소를 이용한 V2O5 Xerogel의 합성 및 전기화학적 특성 (The Electrochemical Properties and Synthesis of V2O5 Xerogel using H2O2)

  • $V_2O_5$ 분말을 과산화수소에 용해시켜 $V_2O_5$ 겔을 졸-겔법으로 합성한 후 물성과 전기화학적 특성을 NMR, 선형전압전류법 등을 이용하여 조사하였다. $V_2O_5$ xerogel에 삽입된 $Li^+$ 이온의 량에 따라 NMR 스펙트라의 화학적이동값이 다르게 나타났으며 xerogel 구조 내에서 다른 환경의 리튬이온 자리가 존재함을 알 수 있었다. 합성에 이용한 $V_2O_5$의 초기농도 변화와 pH 변화에 따른 xerogel의 전기화학적 특성은 큰 차이가 나타나지 않았으며, 전지용량은 이온교환수지법으로 제조한 xerogel과 비슷한 140 mAh/g으로 나타났다.

흑운모의 인위적 풍화와 우라늄 수착 특성 (Artificial Weathering of Biotite and Uranium Sorption Characteristics)

  • 신선한 흑운모 및 인위적으로 풍화시킨 흑운모에 대한 우라늄 수착 실험을 실시하였다. 실험 이후, 원심분리하여 상등액은 유도결합플라즈마분광분석(ICP-MS)으로 우라늄 농도를 분석하였고, 고체시료는 X-선 회절분석(XRD) 및 주사전자현미경(SEM) 분석을 실시하였다. 3 mm 이하의 흑운모 시료를 7개월 간 실험실에서 풍화시킨 후, 신선한 흑운모와 동일한 조건으로 pH 변화에 따른 우라늄 수착량을 구하였다. 낮은 pH(pH 3)에서는 흑운모에 대한 우라늄의 수착이 크지 않았지만, pH가 증가함에 따라 점차 우라늄 수착이 증가하는 경향을 보였다. 풍화된 흑운모의 우라늄 수착량은 신선한 흑운모에 비해 작았고 높은 pH(pH 11)에서는 그 차이가 더 심하였다. 풍화된 흑운모의 낮은 우라늄 수착특성은 광물 표면의 변화 및 주위 용존이온들의 화학적 거동에 의한 것으로 보인다. 풍화된 흑운모 시료에는 광물 표면에서 선호되는 흡착자리가 파괴되거나 용해되고 더불어 용존 이온들의 콜로이드화에 의해 우라늄-광물 표면 수착반응이 감소되는 것으로 보인다.

일본의 과학적 특성 지도 개발에 대한 고찰 (A Review on Development of Nationwide Map of Scientific Features for Geological Disposal in Japan)

  • 일본에서는 2000년 고준위방사성폐기물의 심층처분을 위한 「특정방사성폐기물의 최종 처분에 관한 법률」을 제정하고 부지선정을 착수하였으나, 부지선정 절차에 참여를 원하는 지자체가 존재하지 않았다. 따라서, 일본 정부는 2015년 문헌조사 단계에 새로운 부지선정을 절차를 개발하고, 지자체의 공모를 촉진하고자 2017년 6월 28일 심층처분을 위한 전국규모 과학적 특성 지도를 발간하였다. 이 지도는 심층처분장 초기 혹은 개념단계에 고려되는 요건 및 기준 등을 제공하고 심층처분을 위한 적합성을 분석함으로써, 공공의 이해도 증진과 지자체와의 의견교환 등을 위해서 유용하게 활용되고 있다.

Characterization of Copper/Zinc-Superoxide Dismutase (Cu/Zn-SOD) Gene from an Endangered Freshwater Fish Species Hemibarbus mylodon (Teleostei; Cypriniformes)

  • Gene structure of copper/zinc-superoxide dismutase (Cu/Zn-SOD; sod1) was characterized in Hemibarbus mylodon (Teleostei; Cypriniformes), an endangered freshwater fish species in Korean peninsula. Full-length cDNA of H. mylodon SOD1 consisted of a 796-bp open reading frame sequence encoding 154 amino acids, and the deduced polypeptide sequence shared high sequence homology with other orthologs, particularly with regard to metal-coordinating ligands. Genomic structure of the H. mylodon sod1 gene (hmsod1; 1,911 bp from the ATG start codon to the stop codon) was typical quinquepartite (i.e., five exons interrupted by four introns); the lengths of the exons were similar among species belonging to various taxonomic positions. The molecular phylogeny inferred from sod1 genes in the teleost lineage was in accordance with the conventional taxonomic assumptions. 5'-flanking upstream region of hmsod1, obtained using the genome walking method, contained typical TATA and CAAT boxes. It also showed various transcription factor binding motifs that may be potentially involved in stress/immune response (e.g., sites for activating proteins or nuclear factor kappa B) or metabolism of xenobiotic compounds (e.g., xenobiotic response element; XRE). The hmsod1 transcripts were ubiquitously detected among tissues, with the liver and spleen showing the highest and lowest expression, respectively. An experimental challenge with Edwardsiella tarda revealed significant upregulation of the hmsod1 in kidney (4.3-fold) and spleen (3.1-fold), based on a real-time RT-PCR assay. Information on the molecular characteristics of this key antioxidant enzyme gene could be a useful basis for a biomarker-based assay to understand cellular stresses in this endangered fish species.

Conformational Change of Human Annexin I by the Binding of $Ca^{2+}$, ATP and cAMP

Carbonyl carbon resonances of some histidines were significantly broadened by Ca2+ binding. However, in the case of methionine, phenylalanine, and tyrosin, small changes could be observed. We found that ATP and cAMP bind {{{{ DELTA }}-annexin I, and the binding ratio of ATP to {{{{ DELTA }}-annexin I is 1. These results are well consistent with the report that cAMP and ATP interact with annexin I, and affect the calcium channels formed by annexin I. Because {{{{ DELTA }}-annexin I is a large protein with 35 kDa molecular weight, site-specific (carbonyl-13C) labeling technique was used to study the interaction sites of {{{{ DELTA }}-annexin I with Ca2+. NMR study was focused on the carbonyl carbon resonances of tyrosine, phenylalanine, methionine and histidine residues of {{{{ DELTA }}-annexin I because the number of these amino acids is small in the amino acid sequence of {{{{ DELTA }}-annexin I.
