Human Intersectin 2 (ITSN2) binds to Eps8 protein and enhances its degradation

  • Participates in actin remodeling through Rac and receptor endocytosis via Rab5. Here, we used yeast two-hybrid system with Eps8 as bait to screen a human brain cDNA library. ITSN2 was identified as the novel binding factor of Eps8. The interaction between ITSN2 and Eps8 was demonstrated by the in vivo co-immunoprecipitation and colocalization assays and the in vitro GST pull-down assays. Furthermore, we mapped the interaction domains to the region between amino acids 260-306 of Eps8 and the coiled-coil domain of ITSN2. In addition, protein stability assays and immunofluorescence analysis showed ITSN2 overexpression induced the degradation of Eps8 proteins, which was markedly alleviated with the lysosome inhibitor NH4Cl treatment. Taken together, our results suggested ITSN2 interacts with Eps8 and stimulates the degradation of Eps8 proteins.

Effects of Reduced Glutathione and Ginseng Extract on Non-Protein Sulfhydryl, and Non-Protein Disulfide of Mouse Liver and Blood Following Whole Body X-Irradiation (Reduced glutathione 및 인삼추출액(人蔘抽出液)이 X-선전신조사(線全身照射)를 입은 마우스 간조직(肝組織) 및 혈중(血中) NP-SH 및 NP-SS에 미치는 영향(影響))

  • In an attempt to better understand the radioprotective effect of reduced glutathione(GSH), and to observe a possible radioprotective effect of Ginseng extract, whole body X-irradiation of 1,200 r was administered to the mouse either independently or immediately following the injection of GSH or Ginseng extract to the mouse intraperitoneally. The non-protein sulfhydryl (NP-SH) and non-protein disulfide (NP-SS) levels of the liver, and NP-SH level of NP-SH of the blood of the mouse were measured at 30, 60 and 120 minutes, and results were compared with the normal. The results thus obtained are summarized as follows; 1) The normal values of NP-SH and NP-SS of the mouse liver were $5.90{\pm}0.46\;{\mu}\;mol/gm\;wet\;wt.,\;and\;3.02{\pm}0.42\;{\mu}\;mol/ml$ wet wt., respectively, and the normal value of NP-SH of NP-SH of the mouse blood was $3.98{\pm}1.29\;{\mu}\;mol/ml$ 2) The injection of both GSH and Ginseng extract produced the highest values of NP-SH in the liver at 30 minutes, but a gradual decrease to the normal was observed thereafter. When X-irradiation alone was applied, the liver NP-SH value was lower than the normal at 60 minutes post-irradiation and thereafter. When Ginseng extract was injected immediately prior to X-irradiation, the liver NP-SH was lower than the normal throughout the experiment with the lowest value at 60 minutes. However, the combination of GSH and X-irradiation produced higher than the normal values throughout the entire experiment. 3) The liver NP-SS value was most significantly elevated at 30 minutes after the injection of GSH, hut the recovery to the normal was observed thereafter. The injection of Ginseng extract produced slightly higher liver NP-SS values at 30 and 60 minutes, but the value at 120 minutes was similar to the normal. The single application of X-irradiation resulted in the lower then normal liver NP-SS values throughout the entire experiment. When GSH was injected price to X-irradiation, the liver NP-SS values were higher than the normal at 30 and 60 minutes followed b the recovery to the normal at 120 minutes. The combination of Ginseng extract and X-irradiation showed generally lower liver NP-SS values throughout the experiment. 4) The blood NP-SH showed the higher than the normal values in all the experimental groups except when GSH was injected prior to X-irradiation alone produced e significantly elevated blood NP-SS value at 30 minutes post-irradiation.

The Effect of Exercise Therapy on Serum Lipid Level and Antioxidants of obese College Female Students (운동요법이 비만여대생의 혈청지질 및 항산화계에 미치는 영향)

  • The purpose of this study is to analyze the effects of exercise therapy on serum lipid levels and antioxidants of obese and normal college female students. The subject group composed of ten normal-weight students(below 30% body fat ratio) and ten obese students(above 30% body fat ratio). After a pilot test, the subjects were given an eight-weeks exercise program. Before and after the exercise program, the subjects were given test for serum lipid and antioxidants were analyzed. The SAS program was used in the data analysis. The statistical measurements employed here were T-test, Wilcoxon signed rank test, and Wilcoxon rank sum test. The results of this research are as follows. 1) The effects of exercise therapy on serum lipid levels : Before the exercise therapy, the levels of Total-cholesterol, Triglyceride and LDL-cholesterol of the obese group were highier than those of the normal-weight group. However, the HDL-cholesterol levels were highier in the normal-weight group than in the obese group, but these differences were not significant. With the exercise therapy, the levels of Total-cholesterol increased gradually. The HDL-cholesterol increased gradually, the LDL-cholesterol level decreased in both groups, However, the Triglyceride level decreased in the obese group and increased in normal group, but the difference was not significant. 2) The effects of the exercise therapy on serum antioxidants ; Before exercise therapy, the serum FR and GSSG levels of the obese group were significantly highier than those of the normal-weight group(p=0.00, p=0.04). The serum GSH level of the normal-weight group was highier than that of the obese group, and the serum MDA level of the obese group was highier than that of the normal-weight group. Again these differences were not significant. With exercise therapy, serum FR level was reduced and serum GSSG level significantly increased in both group(obese group p=0.01, normal-weight group p=0.01), The serum GSH level of the obese group significantly increased (p=0.01), and serum MDA level significantly increased in the obese group(p=0.01), but the difference in the normal-weight group was not significant. These results show that regular exercise therapy reduces serum FR levels and activation of antioxidant systems, and suppress oxidative stress. These effects were slightly highier in the obese group than in the normal-weight group. The regular exercise therapy decreased the serum Triglyceride levels more in the obese group than in the normal-weight group. However the improvement of the serum lipid profile may require a longer exercise period than eight weeks. The results show that the exercise therapy was overall more effective in the obese group than the normal-weight group.

ZNF552, a novel human KRAB/C2H2 zinc finger protein, inhibits AP-1- and SRE-mediated transcriptional activity

  • In this study, we report the identification and characterization of a novel C2H2 zinc finger protein, ZNF552, from a human embryonic heart cDNA library. ZNF552 is composed of three exons and two introns and maps to chromosome 19q13.43. The cDNA of ZNF552 is 2.3 kb, encoding 407 amino acids with an amino-terminal KRAB domain and seven carboxyl-terminal C2H2 zinc finger motifs in the nucleus and cytoplasm. Northern blotting analysis indicated that a 2.3 kb transcript specific for ZNF552 was expressed in liver, lung, spleen, testis and kidney, especially with a higher level in the lung and testis in human adult tissues. Reporter gene assays showed that ZNF552 was a transcriptional repressor, and overexpression of ZNF552 in the COS-7 cells inhibited the transcriptional activities of AP-1 and SRE, which could be relieved through RNAi analysis. Deletion studies showed that the KRAB domain of ZNF552 may be involved in this inhibition.

  • Zinc finger-containing transcription factors are the largest single family of transcriptional regulators in mammals, which play an essential role in cell differentiation, cell proliferation, apoptosis, and neoplastic transformation. Here we have cloned a novel KRAB-related zinc finger gene, ZNF424, encoding a protein of 555aa. ZNF424 gene consisted of 4 exons and 3 introns, and mapped to chromosome 19p13.3. ZNF424 gene was ubiquitously expressed in human embryo tissues by Northern blot analysis. ZNF424 is conserved across species in evolution. Using a GFP-labeled ZNF424 protein, we demonstrate that ZNF424 localizes mostly in the nucleus. Transcriptional activity assays shows ZNF424 suppresses transcriptional activity of L8G5-luciferase. Overexpression of ZNF424 in HEK-293 cells inhibited the transcriptional activity of NFAT and p21, which may be silenced by siRNA. The results suggest that ZNF424 protein may act as a transcriptional repressor that suppresses NFAT and p21 pathway to mediate cellular functions.

KBTBD7, a novel human BTB-kelch protein, activates transcriptional activities of SRE and AP-1

  • In this study, a novel member of BTB-kelch proteins, named KBTBD7, was cloned from a human embryonic heart cDNA library. The cDNA of KBTBD7 is 3,008 bp long and encodes a protein product of 684 amino acids (77.2 kD). This protein is highly conserved in evolution across different species. Western blot analysis indicates that a 77 kD protein specific for KBTBD7 is wildly expressed in all embryonic tissues examined. In COS-7 cells, KBTBD7 proteins are localized to the cytoplasm. KBTBD7 is a transcription activator when fused to GAL4 DNA-binding domain. Deletion analysis indicates that the BTB domain and kelch repeat motif are main regions for transcriptional activation. Overexpression of KBTBD7 in MCF-7 cells activates the transcriptional activities of activator protein-1 (AP-1) and serum response element (SRE), which can be relieved by siRNA. These results suggest that KBTBD7 proteins may act as a new transcriptional activator in mitogen-activated protein kinase (MAPK) signaling.

  • Subquadratic space complexity multipliers for optimal normal bases (ONBs) have been proposed for practical applications. However, for the Gaussian normal basis (GNB) of type t > 2 as well as the normal basis (NB), there is no known subquadratic space complexity multiplier. In this paper, we propose the first subquadratic space complexity multipliers for the type 4 GNB. The idea is based on the fact that the finite field GF($2^n$) with the type 4 GNB can be embedded into fields with an ONB.

  • Owing to the development of the notion of normal adjacency of a digital product [9], product properties of digital topological properties were studied efficiently. To equivalently represent a normal adjacency of a digital product, the present paper proposes an S-compatible adjacency of a digital product. This approach can be helpful to understand a normal adjacency of a digital product. Finally, using an S-compatible adjacency of a digital product, we can study product properties of digital topological properties, which improves the presentations of the normal adjacency of a digital product in [9] and [5, 6].

  • The temperature distribution on the superconducting thin-film is analyzed as moving constant field is applied above upper critical field. The distribution of magnetic field is derived in the normal spot. Governing equation is obtained with the help of the equation of conservation of energy. The temperature distribution and the heat dissipation are obtained through computer simulation by the method of numerical analysis. Maximum temperature is occured in the most right side inside normal spot. The temperature is increased abruptly inside the normal spot, and decreased more gradually outside normal spot in the direction of moving field as velocity is increased. Increasing the velocity rather than increasing magnitude of the normal spot and the applied field makes maximum temperature larger. Heat dissipation is affected by the velocity rather than the magnitude of normal spot and the applied field.

Blood Biochemical Changes during Periparturient Period in the Holstein Cattle (젖소에서 분만기의 혈액화학치의 변화: 지방간을 중심으로)

  • This study examined the incidence of disease and blood biochemical values in 68 Holstein cattle during the a month period after parturition. In order to compare the blood biochemical values, the animals were divided into 2 groups of prepartum and 4 groups of postpartum according to the blood collecting day, respectively. The results compared postpartum with prepartum were obtained as follows : the incidences of displacement of abomasum, culled and retained placenta were 11.8%. 13.2% and 8.8%, respectively. In the normal group(pre and post parturition group), NEFA and bilirubin were increased. Triglyceride and cholesterol were significantly lower than In the pre-parturition group, and total lipid was decreased. In the disease group, displacement of abomasum was significantly higher than in the normal group in the levels of NEFA, triglyceride and bilirubin. Culled was significantly higher than in the normal group in the levels of NEFA, AST and bilirubin. but lower than in the normal group in the levels of triglyceride and cholesterol. Retained placenta was significantly higher than in the normal group in the levels of NEFA and bilirubin, but lower than in the normal group in the levels of triglyceride.
