Antioxidant and free radical scavenging activities of Cleome rutidosperma

  • The study was aimed at evaluating the antioxidant and free radical scavenging activities of ethanolic extract and its fractions of Cleome rutidosperma. The antioxidant activity, reducing power, total phenolic content, total flavonoid content, superoxide anion scavenging activity, nitric oxide anion scavenging activity, in vitro antilipid peroxidation activity and in vitro non-enzymatic hemoglobin glycosylation were studied. The results obtained in the study indicate that Cleome rutidosperma is a potential source of natural antioxidant. All the parameters were found to be concentration dependent and increased with increasing amounts of sample. Flavonoids, phenolic compound like tannins, terpenoids may be responsible for the antioxidant activity of the plant. Variation of solubility parameters in various models may be attributed to non-linearity of activity of ethanol extract fractions models. Further investigation on the isolation and identification of antioxidant component(s) in the plant may lead to chemical entities with potential for clinical use.

Effectiveness of Silver Thiosulfate Treatment in Reducing Ozone Injury to Tomato Plants (Silver Thiosulfate 처리가 토마토의 오존피해경감에 미치는 효과)

  • This study was conducted to determine the effectiveness of silver thiosulfate(STS) in reducing $O_3$ injury to tomato plants(Lycopersicon esculentm Mill. 'Pink Glory'). Two days prior to $O_3$ fumigation, plants were given a foliar spray of STS solution at concentrations of 0, 0.2, 0.4, 0.6 mM contained with 0.05% Tween-20. STS concentrations below 0.6 mM were significantly effective in providing protection aginst $O_3$ exposure(16 h at 0.3 ppm). STS reduced leaf injury rate, defoliation of cotyledons, ethylene production and degree of epinasty induced by $O_3$ injury. STS slightly increased ethylene production in non-$O_3$-fumigated plants, but changes of chlorophyll content and transpiration rate on a whole plant basis were not observed. In $O_3$-fumigated plants, STS treatment reduced chlorophyll destruction but did not affect transpiration rate. STS treatment seemed not to affect peroxidase(POD) and superoxide dismutase (SOD) activity in non-fumigated plants but reduced increasing activity of POD by $O_3$ fumigation. However, such an effect as above was not found in SOD activity. Even though enzymatic protection effects were not confirmed, the fact that reduction of acute injury rate was attained for 16 h fumigation indicates that the phytoprotective effects of STS are not necessarily related to blocking the action of strees-induced-ethylene as an anti-ethylene agent.

Effect of Cellulase on Characteristics of Denim (중성 셀룰라제 처리에 의한 데님의 물성)

  • Recently, eco-friendly processing has been focused in the textile industry in order to reduce environmental pollutions. Applications of enzyme technology to the textile industry are an example of more environmentally compatible processes. However, there is not enough quantity of referring to denim fabric subjected to enzymatic treatment. In this study, depending on pH, temperature, cellulase concentration, and treatment time, the weight loss of denim fabrics was examined. Characteristics of enzyme-treated fabrics were measured by tearing strength, stiffness, and K/S values. The effect of a non-ionic surfactant (Triton X-100) on characteristics of the enzyme-treated fabrics was evaluated. The cellulase treatment condition on the cotton fabric were optimized to pH 6.0, $50^{\circ}C$, 1%(o.w.f.), and 60minutes. Characteristics of denim fabrics by cellulase treatment in the presence of Triton X-100 did not improve because Triton-X inhibited the activity of enzyme.

Potential of proteolytic enzyme treatment for production of Korean red ginseng extract (홍삼 추출물의 제조에서 단백질 분해효소의 활용)

  • In this study, proteolytic enzymatic treatment conditions for Korean red ginseng were examined to increase the extraction yield. Commercially available proteases were screened to obtain high protein and carbohydrate yield. The optimal dosage and reaction time for Alcalase, the chosen protease, were found to be 2.0% (w/w) and 1.5 h, respectively. Treatment with optimal conditions of Alcalase increased solid yield, total phenolic content and gensenosides content by 57.6, 81.8, and 33.8%, respectively, over levels in non-treated Korean red ginseng. Antioxidative activities evaluated by free radical scavenging activity, cation radical scavenging activity and reducing power were exactly similar between Alcalase-treated and non-treated extracts.

Model System Study for the Mutagenicity of Sugar-Glycine Systems

  • The mutagenicity after heating of different sugars (glucose, fructose, galactose, and tagatose) on the non-enzymatic browning reaction in different sugars and glycine model system was investigated. The model system containing 0.2 M glycine and 0.2 M of different sugars in 10 mL water was heated at $150{\pm}5^{\circ}C$ for 30 min. After heating, degree of non-browning reaction intensity and mutagenicity using Salmonella typhimurium TA 98 were examined. Heated glycine model systems containing different sugars increased their mutgenicity ranged from 30 to 372 revertant colonies. After heating for 40 min, mutagenicity was achieved with glycine model systems containing 4 different sugars with by 145, 356, 206, and 369 revertants per plate, respectively. The glycine model systems containing fructose or tagatose were significantly (p<0.05) higher mutagenic activity than glycine model systems containing glucose or galactose after 40 min of heating. The linear regression between Maillard reaction intensity and mutagenic activities (slope=32.38, $R^2=0.93$) indicates that mutagenicity could be fully ascribed to Maillard reaction products.

Physiological Role of a Multigrain Diet in Metabolic Regulations of Lipid and Antioxidant Profiles in Hypercholesteremic Rats -Multigrain diet in hyperlipemia-

  • Objectives: The objective of the present study was to investigate the lipid and the antioxidant regulatory potential of a multigrain diet in laboratory animals with reference to lipid profiles, tissue lipid peroxidation and antioxidant status. Methods: Two types of diets, with or without addition of cholesterol, were used in the study - a commercial diet and a formulated multigrain diet (with Sorghum vulgare, Avena sativa, Pennisetum typhoideum, Oryza sativa, Eleusine coracana and Zea mays grains). After a 10-week period of feeding the diets to albino rats the plasma, liver and fecal lipid profiles and the hepatic and renal antioxidant status of the animals that were fed the commercial and the formulated diets (with and without cholesterol addition) were assessed. Results: The commercial diet supplemented with cholesterol elevated the levels of plasma total lipids, total cholesterol, triglycerides, low-density lipoprotein cholesterol (LDL-C), and very low-density lipoprotein cholesterol (VLDL-C), as well as the atherogenic index (AI). The high-density lipoprotein cholesterol (HDL-C) content and the antioxidant profiles (total ascorbic acid, superoxide dismutase, catalase, glutathione peroxidase reduced glutathione) declined along with increases in lipid peroxidation. The formulated diet (with and without addition of cholesterol) was found to be more efficient than the commercial diet in controlling plasma, hepatic and fecal lipid profiles, as well as hepatic and renal lipid peroxidation and antioxidant status, than of the hypercholesteremic animals. Conclusion: The multigrain diet used in the present study is effective in countering the hyperlipidemia and oxidative stress caused by high cholesterol intake.

  • This study was carried out to invastigate the researches of Snake Venom and snakes which used in treatment 1. The fist literature that used the snake for treatment is Shin Nong Ben Cao Jing 2. Composition of Snake Venom is consist of Enzymatic proteins ; Phospholipase A(A1-2), Protease, L-amino acid oxidase etc, and Non-enzymatic proteins ; Crotamine(Cytolysin), Proteolytic factor(Hematoxin), Crotoxin(Neurotoxin) etc. 3. Main toxins in Snake Venom are Hematoxin, Cytolysin, Neurotoxin and Cardiotoxin. Lethal dose 50 value of Agkistrodon brevicaudus is $45.87{\mu}g$/18g, Agkistrodon saxatilis is $10.28{\mu}g$/18g, Agkistrodon ussuriensis is $8.68{\mu}g$/18g, therefore Agkistrodon ussuriensis has strongist Snake Venom of all in Korea. 4. Pharmacological actions of Snake Venom are anticoagulation, thrombolytic function, hypotensor etc. 5. Systemic syndromes and signs after snakebite are Dizziness(25.7%), Vomitting(23.1%), Fever(22%), Visual disturbance(18%), Headache(17.7%) and Dyspnea(17.6%), etc. 6. Local syndrome and sign after snakebite is Discoloration(54.2%), Bleeding(20.2%), Bullae(10.7%), Skinulcer(10.8%), etc. 7. Pathological syndromes after snakebite are WBC increase, Urine protein, Urine sugar, Haematuria and elevation of S-GDT, S-GPT etc. These syndromes are leaded by Hematoxin and Cytolysin. 8. Complication signs after snakebite are Cellulitis, Gastritis, Lympoma, Abscess etc. 9. Common function of Viperidae(Agkistrodon acutus or Zaocys dhumnades etc) is expelling the wind(祛風), removing obstruction in the channels(通絡), antipastic function(止痙). And it is used in order to cure hemiparesis, hemiplegia, facial palsy and CVA disease, etc. 10. Using way of snake for medical treatment is various like Herbal alchol therapy, pill, powder and injection etc. The Study on the Snake Venom should be carried out continuously for using of medical treatment.

  • Chitin deacetylase (CDA; EC 3.5.1.41) catalyzes the hydrolysis of N-acetamide bonds of chitin, converting it to chitosan. Chitosan has several applications in areas such as biomedicine, food ingredients, cosmetics, pharmaceuticals, and agriculture. In this paper, occurrence, assay and purification protocols, enzymatic characteristics, substrate specificity, and mode of action of microbial CDAs have been described. Several lines of evidence have substantiated the biological roles involved in cell wall formation and plant-pathogen interactions for fungal CDAs. The gene structure of CDAs has been compared with other family 4 carbohydrate esterases which deacetylate a wide variety of acetylated poly/oligo-saccharides. The use of CDAs for the conversion of chitin to chitosan, in contrast to the presently used chemical procedure, offers the possibility of a controlled, non-degradable process, resulting in the production of well-defined chitosan oligomers and polymers. Insect pathogen that can secrete high levels of chitin-metab­olizing enzymes including CDA can be a possible alternative for new pest management tools.

Effect of Enzyme Treatments on the Extraction Efficacy and Antioxidant Activity of Haematococcus Extract from Haematococcus pluvialis (Haematococcus pluvialis로부터 Haematococcus 추출물 제조 공정에서 효소 처리가 추출 효율과 항산화 활성에 미치는 영향)

  • An efficient production method of food-grade heamatococcus extract was developed based on stepwise enzymatic hydrolysis. In the first step, Haematococcus pluvialis cells hydrolysis carried out with commercially available exopeptidase(Flavourzyme) and endopeptidase (Alcalase), resulted in increased astaxanthin content. In the second step, proteolytic hydrolyzed H. pluvialis cells treated with hetero-polysaccharides hydrolytic enzyme (Viscozyme). By two-stage treatments using Alcalase and Flavourzyme and Viscozyme, the highest astaxanthin content was obtained. The astaxanthin content was remarkably enhanced by 320% $(529{\mu}g/g\rightarrow2,256{\mu}g/g)$ than that of the non-treated extract. And then, antioxidative activities determined by DPPH method were increased with increasing the astaxanthin content in haematococcus extract prepared by enzymatic hydrolysis.

Effects of Culture Condition on Solubilization of Coal by Microorganisms (배양 조건의 변화가 미생물에 의한 석탄의 액화에 미치는 영향)

  • Biosolubilization of an Australian lignite was investigated by using Streptomyces viridosporus and Poria cocos. In order to solubilize coals effectively they were pretreated by nitric acid both in surface and liquid cultures. The optimum growth pH was 7.5 for S. viridosporus and 4.5 for P. cocos. The effects of various carbon, nitrogen and metal sources on overall solubilization were also studied. Solubility increased with the addition of urea for S. viridosporus, and peptone and tryptone for P. cocos. However carbon and metal sources had little or negative effects on solubilization. Maximum amount of coal solubilized was 85%(w/w) in a batch fermentation culture. Extracellular materials produced by micro-organism were found to be responsible for the coal solubilization. Approximately 70 to 80% of coal solubilization was determined to be the result of non-enzymatic reactions, and the rest to be the result of enzymatic reactions. Characteristics of the solubilized coal were compared with those of original coal and pretreated coal by the approximate and ultimate composition analysis, and IR-spectrum analysis. The spectroscopic results showed that the mechanism of coal solubilization was caused by continuous oxidation.
