A Comparative Analysis of Monofunctional Biosynthetic Peptidoglycan Transglycosylase (MBPT) from Pathogenic and Non-pathogenic Bacteria

  • Monofunctional biosynthetic peptidoglycan transglycosylase (MBPT) catalyzes the formation of the glycan chain in bacterial cell walls from peptidoglycan subunits: N-acetylglucosamine (NAG) and acetylmuramic acid (NAM). Bifunctional glycosyltransferases such as the penicillin binding protein (PBP) have peptidoglycan glycosyltransferase (PGT) on their C terminal end which links together the peptidoglycan subunits while transpeptidase (TP) on the N terminal end cross-links the peptide moieties on the NAM monosaccharide of the peptide subunits to create the bacterial cell wall. The singular function of MBPT resembles the C terminal end of PBP as it too contains and utilizes a similar PGT domain. In this article we analyzed the infectious and non infectious protein sequences of MBPT from 31 different strains of bacteria using a variety of bioinformatic tools. Motif analysis, dot-plot comparison, and phylogenetic analysis identified a number of significant differences between infectious and non-infectious protein sequences. In this paper we have made an attempt to explain, analyze and discuss these differences from an evolutionary perspective. The results of our sequence analysis may open the door for utilizing MBPT as a new target to fight a variety of infectious bacteria.

Characterizations of Pt-SPE Electrocatalysts Prepared by an Impregnation-Reduction Method for Water Electrolysis (함침-환원법으로 제조된 수전해용 Pt-SPE 전극촉매의 특성)

  • Solid polymer electrolyte(SPE) membrane with electrodes embedded on both faces offer unique possibilities for the electrochemical cells like water electrolyzer with fuel cell. The Nafion 117 membrane was used as the SPE, and $Pt(NH_3)_4Cl_2$ and $NaBH_4$ as the electrocatalysts and reducing agent, respectively. The 'impregnation-reduction(I-R) method' has been investigated as a tool for the preparation of electrocatalysts for water electrolyzer by varying the concentration of reducing agent and reduction time at fixed concentration of platinum salt, 5 mmol/L. Pt-SPE electrocatalysts prepared by non-equilibrium I-R method showed the lowest cell voltage of 2.17 V at reduction time, 90 min and with concentration of reducing agent 0.8 mol/L and the cell voltage with those by equilibrium I-R method was 2.42 V at reduction time, 60 min and with concentration of reducing agent 0.8 mol/L. The cell voltage were obtained at a current density $1\;A/cm^2$ and $80^{\circ}C$. In water electrolysis, hydrogen production efficiency by Pt-SPE electrocatalyst is 68.2% in case of non-equilibrium I-R method and 61.2% at equilibrium I-R method.

  • There are more than 2,900 different species of termites in the world, and just there are 23 species in Japan. They caused more severe infestation compared with Korea. When a structure has become infested with termites, it is important that appropriate action must be taken: the chemical pest control (fumigation, soil termiticide treatment, bait system etc.) or the non-chemical pest control (low oxygen treatment, carbon dioxide treatment, high/low temperature treatment etc.). Especially, there were attempts to make practical protocol of various alternatives since the fumigant(methyl bromide) had been phased out in 2005 in Japan, and practically non-chemical methods would be effective alternatives for some cases, where the scale of infestation is small and limited and when long treatment is possible. But most of all, it is important that the process of pest control is made according to each species of termites with consideration for different characteristics of termites.

Optimum Condition for Non-Aqueous Dispersion Polymerization of Environmentally-friendly Acrylic Resin (비수계 분산중합에 의한 환경친화적 아크릴 수지 합성의 최적화 연구)

  • Optimum synthesis conditions were investigated for the non-aqueous dispersion (NAD) polymerization of environmentally-friendly acrylic resin. The optimum amount of stabilizer was observed to be 20 wt% to prevent flocculation of the synthesized polymer particles. The optimum ratio of aliphatic solvents to aromatic solvents turned out to be 80 to 20 in order to obtain a stabilized NAD resins. The viscosity of NAD resins was affected not only by the reaction time but also by the amount of initiator and the impeller rpm. Also, the particle size and the size distribution were influenced by the amount of stabilizers and initiators. Since the stability of NAD resin was devastated by the increase in particle size when all the monomers were fed at the beginning of polymerization, the monomers need to be put into the reactor step by step throughout the entire polymerization process.

Curcumin Analogue A501 induces G2/M Arrest and Apoptosis in Non-small Cell Lung Cancer Cells

  • Curcumin and its analogues have been reported to exert anti-cancer activity against a variety of tumors. Here, we reported A501, a new curcumin analogue. The effect of A501 on cell viability was detected by MTT assay, the result showed that A501 had a better inhibiting effect on the four non-small cell lung cancer (NSCLC) cells than that of curcumin. Moreover, Colony forming experiment showed A501 significant restrained cell proliferation. Flow cytometry displayed A501 can cause G2/M arrest and induce apoptosis. Western blotting showed that A501 decreased the expression of cyclinB1, cdc-2, bcl-2, while increased the expression of p53, cleaved caspase-3 and bax. In conclusion, curcumin analogues A501 played antitumor activity by inhibiting cell proliferation and inducing apoptosis of NSCLC cells. And it was likely to be a promising starting point for the development of curcumin-based anticancer drugs.

A Comparison Study on the Sterilization Effect of Ballast Water with Cylinder Type and Groove Type (선박평형수 처리장치의 cylinder type과 groove type의 살균효과 비교 연구)

  • Current ballast water treatment technologies are applying chemical or electrical treatment technology which are not free from secondary environmental pollution. The purpose of this study is to treat the ballast water by shear stress without an additional environmental pollution and to find out the optimal treatment apparatus. We tried to treat ballast water by applying shear stress with two different type of combination of inner and outer cylinder, such as non-pattern type and groove type. In the case of non-pattern type of inner and outer cylinder, sterilization effect was comparatively low because of a slip between inner and outer cylinder. But in the case of groove type of inner and outer cylinder, sterilization effect was superior to the non-pattern type. With a same revolutional speed of 8000rpm, an extinction effect was acquired in the gap of 1 mm of inner and outer cylinder at non-pattern type, but 3mm of that of groove type.

Physico-chemical Properties of leaf tobacco by expansion processes. (잎담배의 팽화방법에 따른 팽화율 및 내용성분 특성)

  • Flue-cured tobaccos(cv. NC82, grade AB3O-1) expanded by super steam, volatile organic solvents(tricuorofluoromethane, propane), carbon dioxide, nitrogen, and by freeze-drying were examined for their physico-chemical properties. The expanding rate by using the organic solvents or carbon dioxide at the impregnated pressure was 70% or more, whereas the rate by super steam and freeze-drying was 49.4 and 42.9%, respectively. The contents of total sugar, alkaloid and ether extracts of the expanded tobacco by organic solvents or carbondioxide were decreased to 27.6, 20% or more, and 13%, respectively, when compared to non-expanded one. The contents of citric acid of the tobacco expanded by using trichlorofluoromethane and carbon dioxide were increased to 19.1, 14.0%, respectively. Linoleic acid in the tobacco expanded by super steam or trichlorofluoromethane was also increased to 17.3%, but malonic acid in the tobacco expanded by super steam was 18.5% less than that in the non-expanded one. Nineteen essential oil among 22 assayed in this experiment were decreased in the expanded tobaccos. Key words : expanding method, tobacco, physico-chemical properties.

Quantum Chemical Designing of Novel Organic Non-Linear Optical Compounds

  • In the present study, ten metal free non-linear optical (NLO) compounds have been designed. These compounds have designed by structural modification of (2-cyano-5-(4-(phenyl(4-vinylphenyl)amino)phenyl) penta-2,4-dienoic acid (TC4). Density functional theory was used for structure optimization and determination of photo-physical properties. These compounds contain triphenylamine as electron-donor and cyanoacrylic acid as acceptor. Five ${\pi}$-spacers are used to connect the donor and acceptor. Two auxiliary donors are also used to assist the donor. Results of this study indicate that stronger electron-donating auxiliary groups and longer ${\pi}$-conjugation enhance NLO response. Major absorption peaks of all systems were in the visible region. These absorption peaks are associated with the ${\pi}-{\pi}^*$ transitions of the entire molecule. From calculations it is clear that all system will be good NLO material. The present calculations will provide new ways for experimentalists to synthesize high-performance NLO material.

A Non-Pt Catalyst for Improved Oxygen Reduction Reaction in Microbial Fuel Cells

  • Fe-tetramethoxyphenylporphyrin on carbon black (Fe-TMPP/C) is examined and compared with carbon (C) and Pt-coated carbon (Pt/C) for oxygen reduction reaction in a two chambered microbial fuel cell (MFC). The Fe-TMPP/C is prepared by heat treatment and characterized using SEM, TEM, and XPS. The electrochemical properties of catalysts are characterized by voltammerty and single cell measurements. It is found that the power generation in the MFC with Fe-TMPP/C as the cathode is higher than that with Pt/C. The maximum power of the Fe-TMPP/C is 0.12 mW compared with 0.10 mW (Pt/C) and 0.02 mW (C). This high output with the Fe-TMPP/C indicates that MFCs are promising in further practical applications with low cost macrocycles catalysts.

  • This paper proposes a new analytical formulation for computing the seismic input at various levels of a structure in terms of floor response spectra. The approach, which neglects the dynamic interaction between primary structure and secondary element, is particularly useful for the seismic assessment of secondary and non-structural elements. The proposed formulation has a robust theoretical basis and it is based on few meaningful dynamic parameters of the main building. The method has been validated in the linear and nonlinear behavior of the main building through results coming from both experimental tests (available in literature) and parametric numerical analyses. The conditions, for which the Floor Spectrum Approach and its simplified assumptions are valid, have been derived in terms of specific interval ratios between the mass of the secondary element and the participant mass of the main structure. Finally, a practice-oriented formulation has been derived, which could be easily implementable also at code level.
