Compositional differences of Bojungikgi-tang decoctions using pressurized or non-pressurized extraction methods with variable extraction times

  • Objectives : In other to determine the optimal extraction conditions, the various Bojungikgi-tang (BJIGT) decoctions prepared by different pressure levels and different extraction times were compared and evaluated in terms of the extract yield and the total soluble solid content. Methods : Decoctions were prepared by the pressure levels of 0 (non-pressurized) and 1 $kgf/cm^2$ (pressurized) for 60, 120 and 180 min. The extract yield and the total soluble solids content of decoctions were measured, and the amounts of the reference compounds in decoctions were investigated by the analysis using high performance liquid chromatography. Results : The extract yield and the total soluble solid content were higher in decoctions extracted by the pressurized method than those from decoction with non-pressurized method. The patterns of yield and contents showed a proportional increase to the extraction time. In analysis of the linear regression for four reference compounds such as liquiritin, nodakenin, hesperidin, and glycyrrhizin, the good linearity with the correlation coefficient more than 0.9999 was observed. The highest contents for four reference compounds were observed at 180 min of both the pressurized method and the non-pressurized method. Conclusions : This study suggests that the pressure in extraction method and the extraction time affect the compositional constituents in BJIGT decoctions. The extraction time of 180 min could be chosen in both pressurized and non-pressurized method as optimal extraction condition.

두개천골 약침의 항산화 활성 및 표준화 연구 (Study on Antioxidant Activity and Standardization of Craniosacral Pharmacopuncture)

  • Objectives The craniosacral therapy is closely related to the herbal meridians, so we try to explore, compare and develop pharmacopuncture that can have a synergistic effect. Methods The craniosacral pharmacopuncture, Hominis placenta pharmacopuncture, jungseongohhyeol pharmacopuncture, bamboo salt pharmacopuncture 1.8%, bamboo salt pharmacopuncture 3.0%, and normal saline, which are used with craniosacral therapy in clinical practice, were all made and prepared by ourselves. In order to compare antioxidant activity, 2,2-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid), 2,2-diphenyl-1-picrylhydrazyl, ferric reducing antioxidant powder, total flavonoids, total phenolics methods were all measured. Method validation such as specificity, linearity, precision, and accuracy were performed for craniosacral pharmacopuncture. Results In all antioxidant methods, craniosacral pharmacopuncture showed the highest activity and followed by a Hominis placenta pharmacopuncture. The rest of the pharmacopunctures were measured to have low antioxidant activity. Nodakenin and glycyrrhizin were suitable as index compounds of craniosacral pharmacopuncture and they contained 0.82±0.01 ㎍/mL and 2.56±0.01 ㎍/mL respectively. Conclusions Craniosacral pharmacopuncture has the highest activity in all antioxidant activity experiments, which will help activate craniosacral therapy and quality control is possible through standardized research. Such research will contribute to the development of the oriental medicine industry.

새로운 생약복합제제(HPL-4)의 품질관리를 위한 분석법 개발 (Development of Analytical Method for Quality Control from New Herbal Medicine(HPL-4))

  • HPL-4 is a new herbal formulation developed for the treatment of osteoarthritis. In this study, we took HPL-4 to develop a method for simultaneous determination of nine marker compounds (protocatechuic acid, chlorogenic acid, liriodendrin, nodakenin, ${\beta}$-$\small{D}$-(3-O-sinapoyl)frucofuranosyl-${\alpha}$-$\small{D}$-(6-O-sinapoyl)glucopyranoside, quercitrin, 6-gingerol, decursin and decursinol angelate) present in the formulation. The analytes were separated by UPLC system consisting of diode array detector at 205 nm and RP-amide column, and solvent system of $ACN/0.1%H_3PO_4$. Analytical method was validated to evaluate its linearity, limit of detection (LOD), limit of quantification (LOQ), precision, and accuracy. All standards showed good linearity ($R^2$ >0.99) in the rage of $0.25-400.0{\mu}g/mL$. The LOD and LOQ were within the range of 0.021-0.148 and $0.070-0.448{\mu}g/mL$, respectively. Intra- and inter-day precision was less than RSD 4.0% and the accuracy was range from 92.00-104.81% with RSD<4.2%. The results suggest that the developed UPLC method is precise, accurate and effective, and could be applied for quality control of HPL-4 formulation.

고온단시간 처리에 따른 저장 중 참당귀의 품질 특성 (Quality Characteristics of Angelica gigas Nakai in Response to High-Temperature-Short-Time Treatment during Storage)

  • Angelica gigas Nakai (A. gigas) easily changes its color during storage, and appropriate thermal treatment can improve storage stability through inactivation of enzymes such as polyphenol oxidase. Therefore, this study was performed to determine quality characteristics of dried A. gigas in response to high-temperature-short-time (HTST) treatment during storage. Dried A. gigas were treated at 120-180℃ for 10 min, the samples were stored at 4℃ and 50℃ for 10 weeks, and used for the analysis of qualities. Concerning the color values, the sample treated at 120℃ was similar to the control, and the color change was large when treated above 180℃. However, color difference (ΔE⁎ab) was lower in treated samples than in control. Browning index was similar for all the samples except for the sample treated at 180℃. Functional qualities (phenolics content, antioxidant activities, and level of major components) showed a slight difference according to storage periods in all samples without control, and nodakenin content was observed in control. The results of this study showed that HTST treatment improved storage stability such as stability of colors and browning index in dried A. gigas during storage, and the appropriate treatment temperature was 120℃ in terms of stability in color and browning index.
