Sequence Analysis of $\beta$-Xylosidase Gene from Bacillus stearothermophilus (Bacillus stearothermophilus $\beta$-Xylosidase 유전자의 염기 서열 결정 및 분석)

  • The neucleotide sequences of the xylA gene encoding $\beta $-xylosidase of Bacillus stearothermophilus and is its flanking regions were datermined. Three open reading frame(ORFs) were found, one of which(ORF1) appeared to code for the $\beta $-xylosidase. The 1830 base pair ORF1 encoded 609 amino acids starting from a TTG initiation codon. The molecular weight deduced from the nucleotide sequence(68 KD) was in agreement with that estimated by SDS-polyacrylamide gel electrophoresis of the purified enzyme(66 KD). The Shine-Dalgarno sequence(5'-AGGAGG-3') was found 11 bp upstream of the initiation codon. Further 15 bp upstream, there observed a potential transcription initiation signals. The putative -10 sequence(CATAAT) and -35 sequence(TTGTTA) coresponded closely to the consensus sequences for Bacillus subtilis RNA polymerase with major sigma factor. The guanine-plus-cytosine content of the coding region of the xylA gene was 56mol% while that of the third position of the codons was 63 mol%. Based on the comparison with the amino acid sequences of several other carbohydrate degrading enzymes, two conserved regions, possibly participating in the catalytic mechamism of $\beta $-xylosidase xylA, were identified in 278-298 and 329-350 regions of the translated xylA gene. The nucleotide sequence of the xylA was found to exhibit no homology to any other genes so far reproted.

Toxicity of Tomato Spotted Wilt Virus Glycoprotein Signal Peptide and Promoter Activity of th 5' UTR

  • Cloning of the 5'untranslated region (5' UTR) and Nterminus of the glycoprotein precursor (G2G1) open reading frame of tomato spotted wilt virus has been problematic, possibly because of the toxicity of a signal peptide at the beginning of th G2G1 protein precursor. The toxicity of the signal peptide to bacterial growth and the reason for the expression of the peptide gene in Escherichia coli were investigated by cloning the 5' UTR and the signal peptide sequence separately. Cells transformed with the plasmid containing both the first 30 amino acids of the glycoprotein and the 5' UTR showed a severe growth inhibition whereas transformants harboring either the plasmid with the signal sequence or the 5'UTR alone did not show any ingibition. An E. coli promoter-like sequence was found in the 5'UTR and tis promoter acivity was confirmed with a promoter-less GUS gene cloned downstream of the 5'UTR. In the cloning of the Tomato spotted wilt virus (TSWV) glycoprotein G2G1 open reading frame all the recovered plasmids contained stop codons in the signal sequence region. However, clones containing no stop codon were recovered when the signal sequence and the 5'UTR were cloned separately.

Analogical Transfer: Sequence and Connection

  • The issue of connection between entities has a lengthy history in educational research, especially since it provides the necessary bridge between base and target in analogical transfer. Recently, the connection has been viewed through the application of technology to bridge between sequences in order to be cognitively useful. This study reports the effect of sequence type (AT vs. TA) and connection type (fading vs. popping) on the achievement and analogical transfer in a multimedia application. In the current research, 10th -grade and 11th -grade biology students in Korea were randomly assigned to five groups to test the effects of presentation sequence and entity connection type on analogical transfer. Consistent with previous studies, sequence type has a significant effect: analogical transfer performance was better when base representations were presented first followed by target representations rather than the reverse order. This is probably because presenting a familiar base first helps in understanding a less familiar target. However, no fully significant differences were found with the entity connection types (fading vs. popping) in analogical transfer. According to the Markman and Gentner's (2005) spatial model, analogy in a space is influenced only by the differences between concepts, not by distance in space. Thus connection types fail on the basis of this spatial model in analogical transfer test. The findings and their implications for sequence and connection research and practice are discussed. Leveraging on the analogical learning process, specific implications for scaffolding learning processes and the development of adaptive expertise are drawn.

  • Lately, a large amount of textual data have been poured out of the Internet and the technology to refine them is needed. Most of these data are long text and often have no title. Therefore, in this paper, we propose a technique to combine the sequence-to-sequence model of RNN and the REINFORCE algorithm to generate the title of the long text automatically. In addition, the TextRank algorithm was applied to extract a summarized text to minimize information loss in order to protect the shortcomings of the sequence-to-sequence model in which an information is lost when long texts are used. Through the experiment, the techniques proposed in this study are shown to be superior to the existing ones.

  • The tire is considered as one of the Important noise sources having an influence on vehicle's performance. The Pattern noise of a tire is the transmission sound of airborne noise. On smooth asphalt road, Pattern noise is amplified with the velocity. In recent, the study on the reduction of Pattern noise is energetically processed. Pattern noise is strongly related with pitch sequence. To reduce the pattern noise, tire's designer has to randomize the sequence of pitch. The FFT is a traditional method to evaluate the level of the randomization of the pitch sequence, but gives no information on time-varying, instantaneous frequency. In the study, we found that Time-Frequency transform is a useful method to non-stationary signal such as tire noise.

Analysis of Leuconostoc citreum strains using multilocus sequence typing

  • The objective of this study was to perform genetic diversity analysis of 13 strains isolated from South Korean foods by multilocus sequence typing (MLST). For typing, seven housekeeping loci (atpA, dnaA, dnaK, gyrB, pheS, pyrG, and rpoA) were selected, amplified and analyzed. Fifty-one polymorphic sites varying from 1 to 22 in each species were identified. Thirteen sequence types were generated with allele numbers ranged from 2 to 10. The overall relationship between strains was assessed by unweighted pair group method with arithmetic mean dendrogram and minimum spanning tree. In addition, combined spits tree analysis revealed intragenic recombination. No clear relationship was observed between the isolation sources and strains. The developed MLST scheme enhanced our knowledge of the population diversity of Leu. citreum strains and will be used further for the selection of industrially important strain.

  • For any non-negative real number ${\epsilon}_0$, we shall introduce a concept of the ${\epsilon}_0$-dense subset of $R^m$. Applying this concept, for any sequence {${\epsilon}_n$} of positive real numbers, we also introduce the concept of the {${\epsilon}_n$}-attainable sequence and of the points of {${\epsilon}_n$}-attainable ace in the open subset of $R^m$. We also study the characteristics of those sequences and of the points of {${\epsilon}_n$}-dense ace. And we research the conditions that an {${\epsilon}_n$}-attainable sequence has no {${\epsilon}_n$}-attainable ace. We hope to reconsider the social consideration on the ace in social life by referring to these concepts about the aces.

An Improved Pseudorandom Sequence Generator and its Application to Image Encryption

  • This paper proposes an improved Pseudorandom Sequence Generator (PRSG) based on the concept of modular arithmetic systems with non-integral numbers. The generated random sequence use in various cryptographic applications due to its unpredictability. Here the mathematical model is designed to solve the problem of the non-uniform distribution of the sequences. In addition, PRSG has passed the standard statistical and empirical tests, which shows that the proposed generator has good statistical characteristics. Finally, image encryption has been performed based on the sort-index method and diffusion processing to obtain the encrypted image. After a thorough evaluation of encryption performance, there has been no direct association between the original and encrypted images. The results show that the proposed PRSG has good statistical characteristics and security performance in cryptographic applications.
