Antioxidant Effects and Nitrite Scavenging Ability of Extract from Acanthopanax cortex Shoot (오가피순 추출물의 항산화 효과 및 아질산염 소거능)

  • This study was conducted to examine the antioxidative effect and nitrite scavenging ability of extract from Acanthopanax cortex shoot. The total phenolic compound and flavonoids contents of extract from Acanthopanax corex shoot were $116.33{\pm}6.09mg\;GAE/g$ and $65.07{\pm}4.10mg\;RE/g$, respectively. Antioxidative activities were measured by various in vitro models such as DPPH radical scavenging activity, FRAP, reducing power, ABTS radical scavenging activity, ORAC assay. This results showed that the extract of Acanthopanax cortex shoot was effective in scavenging radicals and protecting oxidation when assessed various in vitro systems. Similarly, the nitrite scavenging ability of extract was increased in a dose-dependent manner. Moreover, ORAC value at a concentration of $0.1mg/m{\ell}$ was $103.4{\pm}5.6{\mu}M\;TE/g$. Considering high consumer demand beneficial health effects, Acanthopanax cortex shoot can be utilized to develop functional food health-promoting and natural antioxidant agents.

Antioxidant Activity According to Each Kind of Natural Plant Pigments (식물유래 천연색소의 항산화 활성)

  • The objective of this study was to determine the effect of antioxidant enzyme activity and radical scavenging activities of thirteen kinds of natural plant pigments. The analytic method of antioxidant activities were measured by estimating DPPH free radical scavenging and nitrite scavenging ability activity. The free radical scavenging activity by method using stable free radical DPPH was the highest in the red cabbaged pigment. Addition of ethanol extract 1mg/ml from onion peel pigment displayed remarkable effect on nitrite scavenging ability about 91.9%. Antioxidative enzyme activity was evaluated in terms of superoxide dismutase(SOD), catalase(CAT), and ascorbate peroxidase(APX) activity. The bitter melon pigment had the highest SOD activity of 87.3%. The activities of CAT and APX were higher in the mulberry leave pigment compared with other natural plant pigments. In contrast, CAT activity of plant pigment samples were unaffected. These results suggest that natural plant pigment had the potent biological activities such as antioxidant enzyme activities, and that their activities exhibited differently depending on each kind of pigments.

Effect of Garlic (Allium sativum L.) Extracts on Formation of N-Nitrosodimethylamine (마늘 추출물이 N-Nitrosodimethylamine의 생성에 미치는 영향)

  • This study was conducted to investigate the effect of garlic extracts on electron donating ability nitrite-scavenging activity and N-nitrosodimethylamine (NDMA) formation. Content of total phenolic compounds was the highest in ethyl acetate extract. Electron donating ability was significantly increased with increasing sample concentration, as $88.0{\pm}1.53%$ from ethyl acetate extract and $77.5{\pm}0.8%$ from butanol extract in concentration of 10 mg/mL. Butanol extract from garlic with highest nitrite scavenging activity and inhibition effect of NDMA formation exhibited $96.9{\pm}0.75%$ and $68.6{\pm}1.15%$ in concentration of 10 mg/mL, respectively. The butanol extract was fractionated into 5 fractions using prep-LC. Nitrite-scavenging activity and inhibition effect of NDMA formation were excellent in fraction III, which further fractioned into 3 subfractions (III-a, III-b and III-c) using prep-LC. Of the 3 subfractions, its activity was the highest in the subfraction III-c. Therefore butanol extract of garlic was suggested to be effective inhibitor of NDMA formation.

Effect of hot-air drying temperature on antioxidative activity of sweetpotato leaves (열풍건조 온도에 따른 고구마 잎 메탄올 추출물의 항산화 효과)

  • To provide the basic data to dry vegetate the sweetpotato leaves, the total contents of tannin, flavonoid and polyphenol in the methanol extract from the hot-air dried sweetpotato leaves were analyzed and DPPH radical scavenging activity, ABTS radical scavenging ability, nitrite scavenging ability, and others were comparatively analyzed. The total tannin content was decreased from Shinmi of 10.87 mg/g at $40^{\circ}C$ to 7.28 mg/g at $70^{\circ}C$ and the total flavonoid was decreased from Hayanmi of 55.37 mg/g at $40^{\circ}C$ to 39.63 mg/g at $70^{\circ}C$. That is, the low temperature drying contained more of these substances than in the high temperature drying. The DPPH radical scavenging activity was the highest in Shinmi and Hayanmi of 84.33% and 85.25% at $40^{\circ}C$, and the ABTS radical scavenging ability was a high value of over 80% in the treatment plot at $40^{\circ}C$. The nitrite scavenging ability was highest in Shinmi and Hayanmi of 76.15% and 73.74% at $40^{\circ}C$ but low at $70^{\circ}C$. That is, the antioxidant effect of the hot-air dried sweetpotato leaves was high in the sample of $40^{\circ}C$ and low in the sample of $70^{\circ}C$. Affected by the drying temperature, the high antioxidant effect is resulting from the little decrease of active ingredient when drying at low temperatures.

Antioxidant Activities of Water Extracts from Different Parts of Lycopus lucidus Turcz. ex Benth. (쉽싸리 부위별 물추출물의 항산화 활성)

  • Objectives : This study was performed to investigate the antioxidant activity of water extracts from Lycopus lucidus Turcz. ex Benth. leaves, stems and roots at the $100{\mu}g/m{\ell}$ concentration. Methods : The different part of Lycopus lucidus Turcz. ex Benth. extract was prepared using water. The antioxidant activities of polyphenol contents, total flavonoid contents, DPPH(1,1-diphenyl-2-picrylhydrazyl) free-radical scavenging activity, SOD like activity, hydroxyl radical, ABTS(2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid), $Fe^{2+}$ chelating, and nitrite scavenging activity. Results : The total polyphenol and total flavonoid content of leaves were the highest at $221.85{\mu}g/mg$ and $794.13{\mu}g/mg$, respectively. Electron donating ability was the 79.68% in the water extract from leaves. The ABTS radical scavenging activity of the hot extracts, leaves ${\gg}$ roots > stems was higher in the order. It was shown the highest at 94.53% in the water extract from leaves, which showed a value equal to 94.7% of ascorbic acid. The hydroxyl radical scavenging activity was the highest at 8.07% in the water extract from leaves. SOD like activity and $Fe^{2+}$chelating activity were leaves of 12.3% and 27%, respectively, which were much higher than those of any other parts. The nitrite scavenging ability of extracts was increased at pH 2.5, and those was the highest in leaves of 83.03%. Its were more than twice the 41.61% of BHT. Conclusion : The results suggest that Lycopus lucidus Turcz. ex Benth. can be used as nutraceutical foods and natural antioxidant.

Effect of Germinated Brown Rice Extract Powder on Free Amino Acid Content, Antioxidant and Nitrite Scavenging Ability of the Korean Cabbage Kimchi (동결건조 발아현미 추출분말이 배추김치의 유리아미노산 함량, 항산화성 및 아질산염 소거작용에 미치는 영향)

  • This study investigated the effect of the addition of germinated brown rice extract powder (0, 0.67, 3.35 and 6.70%) to the fermentation of Korean cabbage kimchi. During the fermentation of Korean cabbage kimchi with GBREP, content of total free amino acid and essential amino acid was showed higher value by increasing the addition of GBREP. In particular, ${\gamma}$-aminobutyric acid (GABA) content of Korean cabbage kimchi, prepared with 6.70% GBREP was 2 times higher than control Korean cabbage kimchi. By increasing the content of GBREP, total phenol content was showed high value and the content was more increased during fermentation except control Korean cabbage kimchi. By increasing the content of GBREP, electron donating activity and superoxide radical scavening activity were showed higher value and then showed maximum scavening activity on palatable period. Nitrite-scavenging ability of Korean cabbage kimchi with GBREP was higher than control Korean cabbage kimchi and showed the highest scavenging ability at pH 1.2.

Antimicrobial Effects and Antioxidative Activities of the Cosmetic Composition Having Natural Plant Pigments (천연색소 함유 화장료 조성물의 항균효과 및 항산화활성)

  • This study was conducted to clarify the antimicrobial effects and radical scavenging activities of the cosmetic compositions having the natural plant pigments, and to enhance the natural materials utilization of cosmetics. The antimicrobial activities of the fifteen kinds of cosmetic composition having natural plant pigments were evaluated using the agar diffusion method. Most of the cosmetic composition having the natural pigments showed the clear zone formation of growth inhibition against Bacillus subtilis, Staphylococcus aureus, Listeria monocytogenes, Vibrio parahaemolyticus and Aspergillus flavus. Especially, purple sweet potato, bitter melon, mulberry leaf and gromwell showed the higher antimicrobial activities. All the cosmetic compositions were evaluated for their antioxidant activity using DPPH radical scavenging capacity and nitrite scavenging ability activity. Both of the free radical DPPH and nitrite scavenging ability was the highest in the cosmetic compositions of onion peel, and these antioxidant activity was significant differences according to different plant pigments. In this study, we conjectured that the plant pigment had the potent biological activities, therefore these plant resources having functional components could be a good materials for development into source of natural cosmetics.

Functional Properties of Pine Needle Extract and Its Antioxidant Effect on Soybean Oil (솔잎 추출물의 기능적 특성과 대두유 산화억제 효과에 관한 연구)

  • To investigate the functional properties and the antioxidant effect of pine needle(Pinus densiflora), the pine needle extract was obtained with methanol and its functionality was measured by spectrophotometric method, and the antioxidant experiment on soybean oil was carried out by the active oxygen method. The extraction yield of pine needle with 99% methanol was about 19%, the total flavonoid content of the pine needle-methanol extract was 11.32 mg/g on dry basis and the superoxide dismutase-like activity was 94.3%. The nitrite scavenging ability of the extract was pH dependent with the values of 77.44% at pH 1.2, 48.45% at pH 3.0 and 11.04% at pH 6.0, respectively. The peroxide value was 92.6 meq/kg at 5% dosage, 138.4 meq/kg at 2% dosage of the extract on 8 oxidation days. The period of the peroxide value to be 100 mg/kg was 4.9, 6.3 and 8.5 days at control, 2% and 5% dosage of extract, respectively. And the relative antioxidant effectiveness of the extract was 27.9% and 72.3% increase at 2% and 5% dosage, respectively, compared to control. The thiobarbituric acid value showed few differences within 4 oxidation days, but with the dosage of the extract it fairly decreased with considerable antioxidant effect to control above 4 days.

Physiological Activities of Roots Extracts from Calystegia japonica

  • This study was investigated to evaluate the contents of polyphenols and flavonoids, and physiological activities of various extracts from Calystegia japonica roots for making good use of their functional materials. The roots of C. japonica were extracted with water (WE), ethanol (EE) and hot water (HWE) by different methods. Among these extracts, the highest extracting yield was 30.30% of HWE, while the highest contents of total polyphenols and flavonoids were 40.85% and 6.40% of WE, respectively. The nitrite scavenging abilities were ranged from 31.31% (HWE) to 37.46% (EE) at pH 1.2 and 1.0 mg/ml concentration. In the measurements of electron donating abilities, EE showed the highest effect as 91.83% at 0.3 mg/ml assay concentration, and the electron donating ability was decreased as the extract concentration was increased. In the results of superoxide dismutase (SOD)-like activity, HWE showed the highest effect as 7.15% at 1.0 mg/ml. The tyrosinase inhibition activities of WE and EE were 15.28% and 14.97%, respectively. The xanthine oxidase inhibitory effects were ranged from 97.50 to 99.28% at 1.0 mg/ml. These results indicate that C. japonica extract has a good antioxidant effects and could be useful for developing functional products.

Extractive Optimization of Functional Components for Processing of Onion Health Promotion Drink (양파 음료 제조를 위한 기능성 성분 추출 최적화)

  • Onion, licorice, abgelia root, Chinese date, pine needle, and mulberry leaf were used to extract functional components far onion complex drink. No differences were observed between water extraction at room temperature and methanol extraction in electron-donating ability (EDA), thiosulfinate content (TSC), and ascorbic acid content (AAC), whereas water extraction resulted in higher nitrite-scavenging ability at pH 1.2 and 3.0, and lower superoxide dismutase-like ability than methanol extraction. Level of water extracts prepared by hard-boiling for 3 hr at above $100^{\circ}C$ was higher in all functional abilities except TSC than those prepared at room temperature. Optimal conditions for water extraction and storage were $100-120^{\circ}C$ for 6 hr and low-temperature storage, respectively.
