Nitrogen Assimilation and Carbohydrate Concentration as Affected by the N Supply Form and Their Level in Shoot of Perennial ryegrass (Lolium perenne L.) (페레니얼 라이그라스에서 질소공급형태 및 수준에 따른 질소동화와 탄수화물 대사산물의 변화)

  • To investigate the short-term effects of N-supply form ( $NO_3\;^-$ or $NH_4\;^+$ ) and their level (0.2, 1.0 and 6.0 mM) on N assimilation and C metabolism were examined in perennial ryegrass (Lolium perenne L.). The increase in shoot fresh for $NH_4\;^+$-fed plants much less than $NO_3\;^-$ fed ones. Nitrate concentration in $NO_3\;^-$ -fed plants tended to increase with increasing the supply level, while that of $NH_4\;^+$-fed plants was nearly stable. Nitrate reductase activity (NRA) responded much quickly, showing a proportional increase within 24 h of feeding. NRA in $NO_3\;^-$ -fed plants at 72 h increased by 13.7, 40.3 and 84.0% in 0.2, 1.0 and 6.0 mM $NO_3\;^-$ -fed, but it was not changed in$NH_4\;^+$-fed plants regardless of the supply level. After 72 h of treatment the sugar accumulation in the plants supplied with 0.2 and 1.0 mM -$NH_4\;^+$fed was remarked. After 72 h of feeding, fructan hydrolysis was observed in all levels fur $NH_4\;^+$-fed plants, but only in 6.0 mM for $NO_3\;^-$ -fed plants.

  • In order to improve effectiveness of rhizobia- legume symbiotic nitrogen fixation, ecological and physiological characteristics of indigenous rhizobia distributed in Korea, that is, symbiotic effectiveness of indigenous soybean rhizobia, nitrate reductase activities of the soybean bacteroid from five different soils, and differences of host-infection abilities among the soybean cultivars under population densities of the same indigenous soybean rhizobia, were investigated. The results were summarized as follows: 1. The number of indigenous soybean rhizobia was ranged from $9.2{\times}10^2$ cells per gram of soil in calcareous soil II to $42.4{\times}10^3$ cells per gram of soil in calcareous soil I in Danyang. 2. The symbiotic effectiveness of indigenous soybean rhizobia from five different soils was high in the case of soybean continuously cultivated, and calcareous soil I that population densities of indigenous soybean rhizobia were observed highly. 3. Inverse relationship was observed between total nitrogenase activity (TNA) and nitrate reductase activity (NRA) from the soybean bacteroids ($r=-0.502^*$), but the correlation between nitrate reductase and specific nitrogenase activities (SNA) could be devided into two groups. It was classified into group I which is high in SNA and low in NRA, and group II which is low in SNA and high in NRA. 4. The infection ability of the indigenous soybean rhizobia in the same soil conditions showed the reciprocal difference among each soybean cultivars. In Kwangkyo and Jangyeup, the symbiotic effectiveness appeared by infection of indigenous soybean rhizobia was higher than it of the other soybean cultivars.

Changes of Nitrogen Uptake, Growth and Activities of Nitrogen Metabolizing Enzymes by Different Source of Nitrogen in Tobacco (담배에서 질소 형태에 따른 흡수 양상 및 생육과 질소대사 효소의 활성 변화)

  • Tobacco plant was grown for 40 days hydroponically in nutrient solutions composed of different forms of nitrogen, like NO$_3$$^{[-10]}$ -N, NH$_4$$^{+}$-N, and a mixed formulation of NO$_3$$^{[-10]}$ -N and NH$_4$$^{+}$-N. Uptake response, nitrate reductase, and glutamine synthetase activity at growth stage were investigated to understand the basic knowledge of nitrogen metabolism. The better growth of shoot and root was observed in the mixed nutrient solution than NO$_3$$^{[-10]}$ -N or NH$_4$$^{+}$-N, alone. The plant growth in NH$_4$$^{+}$-N nutrient solution was poor due to ammonium toxicity. The pH of nutrient solution containing NO$_3$$^{[-10]}$ -N increased up to 40 days after transplanting. But the pH of solution containing NO$_3$$^{[-10]}$ -N decreased drastically to 3.42 at 20 days after transplant. The pH in the mixed formulation dropped to pH 3.64 at 30 days after transplant and showed re-increase. It is assumed that nitrogen of NH$_4$$^{+}$-N form was taken up preferentially at early stage and NO$_3$$^{[-10]}$ -N form was taken up preferentially at middle stage in the treatment with the mixed solution. The result indicates that the relative proportion of nitrogen forms affected the uptake patterns at each growth stages. The contents of chlorophyll and soluble protein were high with the mixed solution. Total nitrogen content was the highest in NH$_4$$^{+}$-N solution and the content also increased by the application of the mixed type of nitrogen. The amount of nitrate in leaves was high in NO$_3$$^{[-10]}$ -N treatment and the amount of ammonium was high in NH$_4$$^{+}$-N treatment. The activity of nitrate reductase or glutamine synthetase was highest in the leaves grown in mixed nutrient solution than in those with any other single of nitrogen form.

  • The effect of oxidation-reduction potential (ORP) level on the denitrification by Ochrobactrum anthropi SY509 was investigated under nongrowing condition. The maximum ORP level of nitrate-containing buffer solution was -70∼-80 mV, under which the denitrification took place. By decreasing the initial ORP level, denitrifying enzyme activity was greatly enhanced, which led to higher denitrification efficiency.

  • The excretion of ammonia and glutamine synthetase activities were measured in aposymbiotic Paramecium and symbiotic Paramecium. The uptake of nitrate and ammonia, and specific enzyme activities of nitrate reductase, glutamate dehydrogenase and glutamine synthetase were investigated in symbiotic Chlorella from Paramecium bursaria and Chlorella ellipsoidea. The ammonia concentration in the culture media of aposymbiotic Paramecium was increased according to the growth of the Paramecium but it was not changed in symbiotic Paramecium. Nitrate, the major nitrogen source, was taken up at a rate of 0.635 nmol/ 106 Chlorella/hr in Chlorella ellipsoidea. Most of ammonia was assimilated to glutamine by glutamine synthetase, of which acitivty was 1,467 $\mu$mol/mg protein/min in Chlorella elliposidea. Contrary to Chlorella ellipsoidea, ammonia and glutamine transported from the Paramecium were the nitrogen source of symbiotic Chlorella and ammonia was taken up at a rate of 3.854 nmo./106 Chlorella/hr into synmbiotic Chlorella. Most of ammonia were assimilated to glutamate by glutamate dehydrogenase in symbiotic Chlorella. The glutamate dehydrogenase (GDH/NADH) activity was 0.851 $\mu$mol/mg protein/min.

Sulfur Deficiency Effects on Nitrate Uptake and Assimilatory Enzyme Activities in Rape Plants (유채에서 황 결핍이 질산염의 흡수 및 동화관련 효소활력에 미치는 영향)

  • Rape plants, especially forage type, are known as one of high S-demanding plants. Their productivity and quality have often limited under S-deficient condition. To investigate the effect of S-deficiency on N uptake and its assimilation, $NO_3^-$ absorption, nitrate reducatse (NR) and glutamine synthetase (GS) activity in leaf and root tissues as affected by different S-supplied level was determined. $NO_3^-$ uptake was not significant between control and S-deficient treated plants, while significantly depressed in S-deprived plants for the early 8 h. NR activity decreased as S-availability decreased, especially in young and middle leaves, representing more than 35% of decrease in S-deficient and 70% in S-deprived plants when compared with control. In roots, a significant decrease (-29%) in NR was observed only in S-deprived plants. Relatively higher GS activity was found in young leaves for three all treatments. As a whole leaf tissue, S-limited conditions resulted in a reduction of GS activity. In root which showed the lowest activity, a significant decrease (-30%) was observed only in S-deprived plants.

  • In order to understand more clearly the integration between N-assmilation and C-metabolism in relation to N fertilization, a pot experiment with 5 different level of N fertilization(0, 5, 10, 25, 50 mM NO$_3$$_{[-10]}$ ) was conducted in Manchester, U.K. The peas (Pisum sativum L., cv. Early Onward) were sown in vermiculate (5 cm depth) and cultivated for 6 days under temperature controlled dark room conditions ($25^{\circ}C$). The plants received frequent irrigation with a nutrient solution: it was fertilized every 2 days, 3 times a day at 10h, 13h, 16h respectively. Elevated NO$_3$$^{[-10]}$ concentration, the activity levels of NR, NiR, total GS(crude extract), GS$_2$(plastid) in both root and shoot were increased and reached the peak in 5~25 mM, except NiR specific activity which increased its activity continually until 50 mM NO$_3$$^{[-10]}$ treatment. Total activities of GS (crude extract) in both root and shoot became higher than those of GS$_2$(Plastid), and the activity ratios of total GS in the crude extract and GS$_2$ in the plastids were 3.0 to 4.3 in root, but 3.2 to 10.6 in shoot. It was concluded that the reductants and A TP from OPPP itself should be enough to achieve the high rate of NR, NiR, GS$_1$, GS$_2$ in plant root and shoot for reduction or assimilation of nitrogen, but these enzyme activities might be inhibited by an excess of NO$_3$$^{[-10]}$ influx over the reduction capacity.

N Uptake and Assimilation of Barley Seedlings as Affected by N Availability, Temperature and Water Potential (질소량, 온도, 수분포텐셜 조절에 따른 보리유묘의 질소흡수 및 체내동화)

  • Water culture experiments were carried out to elucidate the effects of N availability, temperature and water potential of culture solution on the uptake and assimilation of N and dry matter accumulation by barley seedlings. N assimilation and dry matter accumulation at 3 to 4 leaves stage in barley plants were maximized at about 3.4 % of N concentration in leaf. N assimilation by barley plants increased with increase of nitrate concentration up to 80ppm in the solution. Over this level nitrate began to accumulated in the leaves and stems proportionally to the N availability in culture solution. Nitrate reductase activity increased in parallel with the increase in the concentration of reduced N in leaves. N uptake by barley plants decreased markedly when water potential reduced below -2 bar or when temperature dropped below 5$^{\circ}C$. These results suggest that the basal application rate of N, 60kg per hectare, for the barley crop needs to be re-examined under the concept of N use efficiency with taking into consideration of temperature and soil N availability because about a half of N accumulated in the leaves of barley plant before wintering is known to be lost by winter killing of above-ground part of the plant.

Abundance and expression of denitrifying genes (narG, nirS, norB, and nosZ) in sediments of wastewater stabilizing constructed wetlands

  • As expected, the expression of denitrifying genes in a Typha wetland (relatively stagnant compared to other ponds), showing higher nitrogen removal efficiency in summer, was affected by temperature. The abundance and gene transcripts of nitrate reductase (narG), nitrite reductase (nirS), nitric oxide reductase (norB), and nitrous oxide reductase (nosZ) genes in seasonal sediment samples taken from the Acorus and Typha ponds of free surface flow constructed wetlands were investigated using quantitative polymerase chain reaction (Q-PCR) and quantitative reverse transcription PCR (Q-RT-PCR). Denitrifying gene copy numbers ($10^5-10^8$ genes $g^{-1}$ sediment) were found to be higher than transcript numbers-($10^3-10^7$ transcripts $g^{-1}$ sediment) of the Acorus and Typha ponds, in both seasons. Transcript numbers of the four functional genes were significantly higher for Typha sediments, in the warm than in the cold season, potentially indicating greater bacterial activity, during the relatively warm season than the cold season. In contrast, copy numbers and expression of denitrifying genes of Acorus did not provide a strong correlation between the different seasons.

The Effect of Acidic Deposition on Nitrate Reductase Activity and Photosynthesis of Hypnum plumaeforme (산성강하물이 털깃털이끼(Hypnum plumaeforme)의 질산 환원효소와 광합성에 미치는 영향)

  • The acidic deposition (wet) was collected at three different locations; representative industrial (Kun-Jang industrial areas), semiindustrial (Kunsan City) and nonindustrial (Daeya areas) region between March, 1991 and June, 1993. Samples were analyzed for major chemical components along with pH. Also the responses of Hypnum filunzaeforme to the acidic deposition adjusted to pH 4.6, 3.6 and 2.6 with 1:4 molar ratio of nitric to sulfuric acid were investigated. Seasonal variation of pH showed the lowest in early spring and the highest in summer. The annual mean concentrations of $S0_4^{2-},\;NO_3^{-}\;and\;NH_4^{+}$ were 2.94 (81.0%), 0.48 (13.2%) and 0.21 (5.78%) mg/l in industrial region and 0.61 (64.21%), 0.25 (26.3%) and 0.9 (9.5%) mg /I in nonindustrial region, respectively. $S0_4^{2-}$ contents of industrial region was 4 times higher than those of nonindustrial region. The ratio of $S0_4^{2-}\;to\;NO_3^{-}$ was 4.75~7.35 in industrial region and 0.69-6.36 in nonindustrial region. The acidic deposition near industrial region was associated with excess ions, expecially $S0_4^{2-}$. Nitrate reductase activity(NRA) of mosses was inhibited by acidic deposition in both regions. The time required for the maximum NRA induction by 2mM $KNO_3$ was shortened in mosses collected from industrial region. Photosynthesis of H. plumaejorme began to decrease with acidic deposition by pH below 4.6, and was completely inhibited at pH 2.6 in both regions. Water potential of mosses treated with acidic deposition was almost constant during 18 days, however after withholding the supply of acidic deposition, it was rapidly decreased.
