Comparison of Biochemical and Immunological Properties Between Rat and Nicotiana glutinosa Ornithine Decarboxylase

  • Ornithine decarboxylase (EC 4.1.1.17) is an essential enzyme for polyamine synthesis and growth in mammalian cells and plants. We compared the biochemical and immunological properties of rat and Nicotiana glutinosa ODC by cloning and expressing the recombinant proteins. The primary amino acid sequence between rat and N. glutinosa ODC had a 40% homology The molecular weight of the overexpressed rat ODC was 53 kDa, and that of N. glutinosa was 46.5 kDa. Adding 1 mM of putrescine to the enzyme reaction mixture inhibited both rat and N. glutinosa ODC activity to 30%. Agmatine had an inhibitory effect only on N. glutinosa ODC. Cysteine and lysine modifying reagents reduced both ODC activities, verifying the key roles of cysteine and lysine residues in the catalytic mechanism of ODC. ELISA was performed to characterize the immunological difference between the rat and plant ODC. Both the rat and N. glutinosa ODC were recognized by the polyclonal antibody that was raised against purified N. glutinosa ODC, but the rat ODC was 50-fold less sensitive to the antibody binding. These results indicate that even though both ODCs have the same evolutionary origin, there seems to be a structural distinction between the species.

Mutation of Cysteine-115 to Alanine in Nicotiana glutinosa Ornithine Decarboxylase Reduces Enzyme Activity

  • Ornithine decarboxylase (ODC, EC 4.1.1.17) is the first and key enzyme in eukaryotic polyamine biosynthesis. The cDNA encoding ornithine decarboxylase from Nicotiana glutinosa was cloned ($GeBank^{TM}$ AF 323910) and expressed in E. coli. Site directed mutagenesis were performed on several highly conserved cysteine residues. Among the mutants, C115A showed significant changes in the kinetic properties. The $K_m$ value of the C115A mutant was $1790\;{\mu}M$, which was 3-fold higher than that of the wild-type ODC. There was a dramatic decrease in the $k_{cat}$, values of the C115A mutant, compared to that of the wild-type ODC, which had a $k_{cat}$ value of $77.75\;s^{-1}$. C115A caused a shift in the optimal pH from 8.0 to 8.4. Considering these results, we suggest that cys-115 is involved in the catalytic activity of N. glutinosa ODC.

  • Nicotiana ($NR^{-}/SR^{+}$)과 N. glutinosa간 전기적 (0.5 MHz의 AC와 2 ms 동안 1 kV DC) 원형질체 융합에서 선발은 1.2mg/ml의 streptomycin이 첨가된 $MSNO_3$ 배지를 통하여 가능하였으며, 이 융합세포의 분열이 지속되어 형성된 녹색 세포괴를 얻었다. 이 녹색 세포괴에서 재분화된 4개의 식물 계통들은 꽃과 잎에서 모식물체의 특징을 보였으며, 엽조직의 peroxidase 동위효소 유형 분석에서도 양쪽 모식물체의 특성을 갖고 있었다. XhoI의 제한효소 절단에 의해 융합식물체 계통의 엽록체 DNA의 분석에서도 융합체 식물계통들은 N. tabacum과 N. glutinosa에서 공통되는 단편과 모식물체 유래 단편을 보였고, 이 융합식물계통들 중 한 계통은 9개의 공통 단편에 N. tabacum의 1개 단편, N. glutinosa의 2개 단편이 추가되어 있는 유형을 보여 N. tabacum과 N. glutinosa의 엽록체 게놈을 모두 소유하고 있다는 것을 보여주었다.

담배 야생종의 원형질체 배양에 미치는 무기영양소의 효과 (Evaluation of Nutrients for the Protoplast Culture of Genus Nicotiana)

  • Leaf mesophy11 protoplast cultures from six Nicotiana species, N. debneyi, N. rustica, N. amplexicaulis, N. glauca, N. glutinosa, and N. sylvestris were carried out. When we reduced the NH4NO3 and Fe.EDTA concentration to 1/3(7 mM) and 1/10(10$\mu$M) from the Murashige and Skoog medium respectively, cell division of the protoplasts was efficiently induced in four Nicotiana species, N. debneyi, N. rustica, N. amplexicaulis and N. glauca. However, other two species, N. glutinosa and N. sylvestris were failed in inducing cell division at the same culture condition. The protoclone calluses derived from four Nicotiana species were consequently regenerated on a MS basal medium supplemented with the appropriate auxin and cytokinin.

  • Nicotiana glutinosa를 이용한 단병반분리를 통해 국내에 시판되고 있는 상용고추종자로부터 괴사국부병반의 크기가 다른 두 종류의 Tobamovirus를 분리하고 이들을 각각 Tobamovirus-6(T-6)과 Tobamovirus-19(T-19)로 명명하였다. 기주범위와 혈청학적 연관성 조사에서 T-6은 pepper mild mottle vitus(PMMoV)와, T-19는 tomato mosaic virus (ToMV)와 유사한 특성을 보였다. 또한 외피단백질 염기서열 분석에서 T-6은 PMMoV와 T-19는 ToMV와 매우 유사하였다.

  • 모자익병징을 나타내는 호박, 박, 오이, 참외, 수박의 박과작물에서 Watermelon Mosaic Virus(WMV)를 분리동정하였다. 이병주로 부터 분리된 WMV는 복숭아혹진딧물(Myzus persicae, Sulzer)에 의해 쉽게 충매전염되었고 종자전염은 되지 않았다. 지표식물검정결과 명아주에서는 접종엽에 대형의 국부병반이, 호박, 박, 오이, 참외, 수박에서는 접종상엽에서 모자익병반이 나타났고 담배(Nicotiana tabacum, Bright yellow), 담배 (Nicotanan glutinosa), 동부, 페추니아, 독말풀에서는 반응이 나타나지 않았다. WMV isolate의 물리적 성질은 내열성 $55\~65^{\circ}C$, 내희석성 $10^{-4}\~10^{-5}$배, 내보존성 $7\~8$일간이었다. WMV isolate의 항혈청반응은 미량침강법에서 양성으로 나타났고 바이러스립자의 형태는 전자현미경검경결과 대부분이 750nm의 사상입자이었다. 이상의 시험결과로 박과 작물에서 분리한 바이러스가 WMV 임이 확인되었다.

Isolation of Defense-Related Genes from Nicotiana glutinosa Infected by Tobacco Mosaic Virus Using a Modified Differential Screening

  • Many of plant defense responses are consequence of transcriptional activation of related genes. We have developed a modified differential screening procedure to isolate tobacco genes that are involved in the defense responses against TMV infection. A cDNA library was constructed from Nicotiana glutinosa leaves infected by TMV under temperature shift conditions. Each of plasmid DNA in the library was hybridized on a set of slot blots to a pool of cDNA probes prepared from either TMV-infected or mock-treated tobacco leaves. Among 900 plasmid DNAs, 81 clones exhibiting significantly enhanced or reduced level of hybridization to either probe were selected for nucleotide sequencing. The clones were listed into 61 genes considering redundancy between the sequences. The genes were identified to be defense-related genes including PR-genes and genes involved in primary or secondary metabolisms. This results supports the implication that plant defense process entails a major shift in total cellular metabolisms rather than activation of a limited number of defense-related genes. Expression patterns of a number of defense-related genes. Expression patterns of a number of selected genes were examined in northern blot analyses. It is notable that the clone 630 of unknown function exhibits expression pattern similar to those of previously known PR-genes. Experiments to elucidate the roles in defense mechanism of a couple of genes newly identified in this study are in progress.

시금치 바이러스병에 관한 연구 II. 시금치에 발생하는 Broad Bean Wilt Virus (BBWV)의 분류동정 (Investigations on the Virus Diseases in Spinach (Spinacia oleracea L.) II. Identification of Broad Bean Wilt Virus Occuring Spinach)

  • 짙은 연색의 모자익병징을 나타내는 이병 시금치를 채집하여 Broad bean wilt virus(BBWV)를 분리동정 하였다. 분리된 Broad bean wilt virus (BBWV)를 지표식물에 즙액접종한 결과, 명아주(Chenopodium amaranticolor), 명아주(Chenopodium quinoa) ,잠두(Vicia faba)에서는 접종엽에 국부병반이 나타났고 접종상엽에서는 모자익(전신감염)병징이 나타났으며 꽈리(Physalis floridana), 시금치, 담배 (W.B), 담배 (Nicotiana glutinosa)에는 모자익 병징이 나타났다. 이병시금치로부터 분리한 병원바이러스와 BBWV의 항혈청을 한천내확산법(Agar gel-diffusion test)으로 반응시킨 결과 양성 반응이 나타났다. 이병엽을 Dip법으로 시료를 제작하여 전자현미경에서 검경한 결과 구형의 입자가 관찰되었으며 직경은 25nm이었다. 시금치 재배포장에서 BBWV의 발생분포는 수원, 안양, 대구 진주 등 거의 전국적으로 발생하였다.
