Study on the Chemical Properties of Korean Local Tobacco (Nicotiana tabacum L.) (한국 재래종 연초(Nicotiana tabacum L.)의 화학적 특성)

  • Field experiment was conducted to find out the chemical characteristics of 12 Korean local varities, Local varities were compared with oriental, flue-cured and burley tobacco. Among tested varities, Hyangcho and Matskawa were desirable in respect of flavor, leaf color and texture. Hyangcho and Matskawa were higher in the content of nicotine and petroleum ether extract. Gajacho, Byeolcho and Kukjuo were higher in the content of reducing sugar, but lower nicotine, total nitrogen and petroleum ether extract. The nicotine and total nitrogen content of Kwangcho, Daruma and Catterton were lower than that of Br. 21. In the content of volatile organic acid which were major characteristic component of oriental tobacco, the Korean local varities were lower in the content of 3-methyl butanoic and 3-methyl pentanoic acid, but hirities, Hyangcho, Matskawa and Daruma were higher in the content of volatile organic acid. Korean local varities, especially small leaf group, were higher in the content of benzyl alcohol and furfural than any other type tobacco.

Inheritance of White Flower of Mutant Line KF 8832-85 in Flue-cured Tobacco (황색종 연초 돌연변이 계통 KF 8832-85의 흰꽃 유전)

  • Cultivars of Nicotiana tabacum L. normally have pink flowers, but the flue-cured tobacco mutant line, BU 8832-85, had white flower. The mutant line was crossed with five normal varieties of KF 109, NC 82, TC 499, NC 567 and Coker 176. All Fl plants showed pink flower. The progenies of F2 generations were segregated with the phenotypic ratio 9 : 3 : 4 with pink, varigated(a recombinant type) and white flower, respectively. Test-cross populations showed 1 : 1 : 2 ratios. These results showed that the white flower character was controlled by two recessive genes. The genes were designated as FFCC for pink and ffcc for white flower. The recessive gene ff was epistatic to C and c. Therefore, white flower had a recessive epistasis gene.

  • The anthers of Yellow Special A, a leading tobacco variety, were cultured on the modified RM-1964 medium. Approximately three weeks after culture, plantlets emerged out of the anthers, and they reached blooming stage in six months. 24 univalent chromosomes were most frequently observed at M1 of PMC. Number of bivalent was 0.8 per PMC.

  • Two Tobamoviruses showing different local lesion types on Nicotiana glutinosa was isolated from commercial pepper seeds. These viruses were designated Tobamovirus-6 (T-6) and Tobamovirus-19 (T-19). The biological and serological assays revealed that T-6 and T-19 were closely related to Pepper mild mottle virus (PMMoV) and Tomato mosaic virus (ToMV), respectively, The isolates also had low similarity in the array of viral coat protein gene sequences, of which T-19 was most identical to known strains of ToMV, while T-6 was closely related to PMMoV.

Correlation, Regression, and Path Analysis between Yield and its Components in Tobacco (Nicotiana tabacum L.) (담배의 수량과 수량구성요소의 상관, 회귀 및 경로분석)

  • Data for this study were obtained from Burley 21 (Nicotiana tabacum L.) grown under various densities on the field in 1978 and 1979 at the Jeonju Experiment Station, Korea Ginseng & Tobacco Research Institute. Interrelations between yield and its components were statistically studied by correlation, regression, and pathway analysis. Correlation of yield with plant population was significant and positive. Quadratic functions for yield vs. plant population and the length of the largest leaf were fitted to the data. Multiple recession equation between yield and its components (leaf number ($X_1$), a leaf area ($X_5$), weight per unit leaf area ($X_9$), plant population ($X_14$)), was significant at the 5% level. Measuring the relative importance of its components on yield, plant population was 49.5%, weight per unit leaf area 25.3%, a leaf 15.6%, and leaf number 9.8%.

  • Productivity of secreted recombinant protein depends largely on its stability in the extracellular environment with protease. Most hGM-CSF produced by transgenic tobacco cell cultures and secreted to the medium was confirmed to be rapidly degraded by protease in medium. To increase the productivity, therefore, various protein stabilizers such as gelling agents such as carrageenan and alginate, polymers, polyols, and amino acids have been tested. The stability of hGM-CSF in spent medium without cells was improved by the presence of gelling agents. However, the reason for the enhanced production by the addition of gelling agents may be due to the increased expression level and permeability rather than stability. The addition of DMSO inhibited the cell growth, but improved specific yield. The others were not effective for stability as well as hGM-CSF production.

Transgenic tobacco plants overexpressing the Nicta; CycD3; 4 gene demonstrate accelerated growth rates

  • D-type cyclins control the onset of cell division and the response to extracellular signals during the G1 phase. In this study, we transformed a D-type cyclin gene, Nicta;CycD3;4, from Nicotiana tabacum using an Agrobacterium-mediated method. A predicted 1.1 kb cyclin gene was present in all of the transgenic plants, but not in wild-type. Northern analyses showed that the expression level of the Nicta;CycD3;4 gene in all of the transgenic plants was strong when compared to the wild-type plants, suggesting that Nicta;CycD3;4 gene driven by the CaMV 35S promoter was being overexpressed. Our results revealed that transgenic plants overexpressing Nicta;CycD3;4 had an accelerated growth rate when compared to wild-type plants, and that the transgenic plants exhibited a smaller cell size and a decreased cell population in young leaves when compared to wild-type plants.

  • Production of haploids in vitro anther culture of Nicotiana tabacum L. was oriented in a large number on a chemically defined culture medium. The haploids were screened for T.M.V. resistance and the segregating ratio in F$_1$ were in good agreement with the expected ratio.

NMMP1, a Matrix Metalloprotease in Nicotiana benthamiana Has a Role in Protection against Bacterial Infection

  • Plant matrix metalloproteases (MMPs) are a family of apoplastic metalloproteases closely related to human matrilysins. Up-regulation of Nicotiana benthamiana matrix metalloprotease 1 (NMMP1) expression by treatment with pathogens, ethephon and aging indicates that the gene is related to plant defense and the aging process through ethylene signaling. NMMP1 expression was higher than in normal growth leaves following infection with an incompatible pathogen Pseudomonas syringae pv. tomato T1 or a compatible pathogen P. syringae pv. tabaci and in aged leaves. Transient overexpression of NMMP1 in N. benthamiana leaves lowered the growth of P. syringae pv. tabaci. However, NMMP1-silenced leaves showed increased growth of P. syringae pv. tabaci. These data strongly suggest that NMMP1 in N. benthamiana is a defense related gene, which is positively regulated by ethylene.

  • The present experiments were conducted to investigate some of the factors affecting the number of haploids derived from anther culture of Nicotiana tabacum. Anther - donor plants grown under controlled environment room at 3$0^{\circ}C$ yielded more haploid than room at 18, 25 and 26-22-18$^{\circ}C$ in anther culture. Donor plants starved of fertilizer yielded more haploids as compared to those of the well fed with fertilizer in anther culture. Pretreatment of exercised flower bud at 5$^{\circ}C$ was shown to be more effective in anther culture than pretreatment at 7 and 1$0^{\circ}C$, and the optimum temperature and period of pretreatment were 4 or 6 days at 5$^{\circ}C$.
