무 유식물에서의 니켈내성과 히스티딘의 작용 (Nickel Tolerance and the Complexing Role of Histidine in Raphanus sativus)

  • The effect of nickel (Ni) on growth and some tolerance strategies with regard to heavy metal tolerance mechanism was investigated in radish (Raphanus sativus) seedlings. The protective effect of histidine on nickel stress conditions was also monitored. The seedling growth decreased with an increase in metal concentrations. The inhibitory effect was more pronounced in the root elongation than in the shoot elongation. Increasing Ni supply showed a progressive increase of Ni concentrations in the roots and shoots. Ni content was higher in the shoots than in the roots. In the presence of nickel, radish exhibited an antioxidative defense mechanism, as evidenced by the elevated malondialdehyde(MDA), showing that nickel is an efficient inducer of lipid peroxidation. Exposure of radish to elevated concentrations of nickel was accompanied by an increase in the proline content. Supplemental histidine in the presence of Ni ameliorated metal-induced growth inhibition and lipid peroxidation. Combinations of Ni and histidine resulted in a significant decline in proline content compared with Ni stress alone, indicating that histidine may provide protection against the adverse effect of Ni stress. From the results it is suggested that histidine is an efficient chelator by complexing metal ion within the plant and may playa role in nickel tolerance implicated in metal detoxification.

Alteration of macronutrients, metal translocation and bioaccumulation as potential indicators of nickel tolerance in three Vigna species

  • Macronutrients ($Na^+$, $K^+$, $Ca^{2+}$, $Mg^{2+}$), yield and yield components, bioaccumulation and translocation of metal in plant parts of three Vigna species (V. cylindrica, V. mungo, V. radiata) were evaluated at 0, 50, 100 and $150mgkg^{-1}$ soil of Nickel (Ni). A marked inhibition (p < 0.001) in the distribution of various macronutrients was noticed in these Vigna species except for $Mg^{2+}$ content of the shoot and leaves. Similarly, all species retained more $Ca^{2+}$ in their roots (p < 0.05) as compared to the aerial tissues. Ni induced a drastic decline (p < 0.001) for various yield and yield attributes except for 100 seed weight. Toxicity and accumulation of Ni in plant tissues considerably increased in a concentration dependent manner. Vigna species signify an exclusion approach for Ni tolerance as both bioaccumulation factor (BF) and translocation factor (TF) were less than 1.0. The Ni content of plants being root > shoot > leaves > seeds. Scoring for percentage stimulation and inhibition (respective to control) at varying levels of Ni revealed tolerance of the species in an order of V. radiata > V. cylindrica > V. mungo. The acquisition of Ni tolerance in V. radiata seems to occur through an integrated mechanism of metal tolerance that includes sustainable macronutrients uptake, stronger roots due to greater deposition of $Ca^{2+}$in the roots, restricted transfer of Ni to above ground tissues and seeds as well as exclusion capacity of the roots to bind appreciable amount of metal to them. Thus, metal tolerant potential of V. radiata could be of great significance to remediate metal contaminated soil owing lesser impact of Ni on macro-nutrients, hence the yield.

토양측정망 운영 결과 분석 연구 (Analysis on Monitoring Results of Korean Soil Monitoring Network)

  • Usability of soil quality monitoring network for ascertaining soil quality changes was evaluated by analysing soil quality monitoring results. Tolerance limits of soil quality monitoring results from 1997 to 2007 were calculated and compared with Korean soil quality standards. This study determined that soil quality was changed if the upper 95% tolerance limit value was greater than the soil quality standard. Fluoride most frequently exceeded the soil quality standard and nickel, zinc, arsenic, copper, lead and cadmium were followed. Analysis on land use showed that tolerance limits of industrial land use most frequently exceeded the soil quality standards and residential, road and various land uses then frequently exceeded. Tolerance limits of land uses expecting high contaminant loads frequently exceeded the soil quality standards. This fact imply that the soil quality monitoring network generates reasonable data to represent change in Korean soil quality. This study also suggested that representative sampling from well identified points should be done to improve data reliability and accurately ascertain soil quality changes.

TgMTP1 과발현 애기장대에서 Nickel 흡수 연구 (Studies on nickel uptake in transgenic Arabidopsis thaliana introduced with TgMTP1 gene encoding metal tolerance protein)

  • 본 연구는 토양에 오염된 중금속을 제거하기 위한 식물정화공정에 사용할 식물체를 개발하기 위해 $TgMTP_1$ 유전자를 CaMV35S 상시 발현 할 수 있도록 Ti-plasmid 벡터를 구축하여 형질전환식물을 육성하였다. 유전자가 도입된 형질전환 애기장대에서 TgMTP유전자를 과발현하는 호모 TG-116 (T3 generation) 계통을 육성하여 그 특성을 조사하였다. 호모계통으로 육성한 TG-116 계통은 callus 및 식물체에서 중금속에 대한 저항성을 보였다. 특히 RT-PCR 및 Western 분석에서 유전자의 발현은 잎에서 높게 나타났으며, Ni 흡수 및 축적이 많이 일어났다. 따라서 MTP1 유전자가 발현되어 액포에 중금속을 축적하는 실험결과를 활용한다면 식물정화공정에 사용할 수 있는 다양한 유전자원으로 기대할 수 있다.

  • Metal induced crystallization of a-Si with a source of Ni/Si oxide was studied. Its mechanism to induce crystallization was discussed. It was found that new source behaves an effect of self-released nickel and reducing nickel residua, so can provide a wider process tolerance; improve the uniformity and stability of TFTs.

아연 저항성 갖는 인위적으로 유도된 효모 돌연변이체의 특성 (Characterization of artificially induced zinc-tolerant yeast mutants)

  • 생물학적정화는 미생물을 이용하여 중금속을 포함한 오염물질을 정화하는 방법이다. 효모는 다양한 중금속을 흡착하여 정화하는 능력이 우수하다고 잘 알려졌다. 따라서 본 연구는 여러 중금속 중 하나인 아연에 저항성인 큰 효모를 인위적인 돌연변이를 통해서 얻는 것에 초점을 두었다. 대조구인 효모를 1 mM 농도의 아연이 포함된 배지에서 키운 후 점차 아연의 농도를 높여 최종적으로 80 mM이 되는 지점까지 키웠으며 이 농도는 효모가 점진적 적응을 거쳤어도 성장이 불가능한 한계점 근처이며 이렇게 유도 분리된 효모를 ZnR이라 명명하였다. ZnR은 대조구 효모보다 아연에 대한 저항성이 현저히 증가되었으며 이 외에 카드뮴과 니켈에 대한 저항성도 증가되었지만 구리에 대한 저항성은 오히려 감소 되었다. ZnR이 보여준 아연에 대한 저항성 증가는 유전자 돌연변이에 의한 영구적인 획득 결과이다. 단 기간 내에 아연에 대한 저항성 큰 효모의 인위적인 유도 및 분리는 생물학적정화 연구에 유용하게 쓰여질 정보이다.

수인성 니켈 급성 노출이 붕어, Carassius carassius의 반수치사농도, 혈액학적 성상 및 혈장 성분에 미치는 독성 영향 (Toxic Effects on Lethal Concentration 50%, Heamatological Parameters and Plasma Components of Crucian carp, Carassius carassius by Acute Waterborne Nickel Exposure)

  • 본 실험은 붕어(Crucian carp, Carassius carassius)(무게 39.7±3.1 g, 전장 14.8±0.5 cm)의 수인성 니켈 0, 10, 20, 40, 80 및 160 mg Ni2+/l 농도로 96시간 급성 노출을 실시하였다. 수인성 니켈에 노출된 붕어의 반수치사농도(LC50)는 117.69 mg Ni2+/l으로 나타났다. 혈액학적 성상에서 RBC count는 수인성 니켈 96시간 급성 노출 중 48시간에서 유의적으로 증가한 반면, 96시간에서 유의적 감소가 나타났다. MCV와 MCH는 96시간에서 80 mg Ni2+/l 농도에서 유의적으로 증가했다. Calcium, magnesium, glucose, cholesterol, total protein, AST, ALT 및 ALP 와 같은 혈장 성분은 수인성 니켈 노출에 의해 유의적 변화가 나타났다. 이 연구의 결과는 수인성 니켈 노출에 따른 붕어의 생존율, 혈액학적 성상 및 혈장 성분의 변화를 확인하고 이는 수인성 니켈의 독성에 의한 것으로 판단했다.

  • Micro engine that includes Micro scale combustor is fabricated. Design target was focused on the observation of combustion driven actuation in MEMS scale. Combustor design parameters are somewhat less than the size recommended by feasibility test. The engine structure is fabricated by isotropic etching of the photosensitive glass wafers. Electrode formed by electroplating of the Nickel. Photosensitive glass can be etched isotropically with almost vertical angle. Bonding and assembly of structured photosensitive glass wafer form the engine. Combustor size was determined to be 1 mm scale. Movable piston is engraved inside the wafer. Ignition was done by nickel spark plug which was electroplated with thickness of 40 ${\mu}{\textrm}{m}$. The wafers were bonded by epoxy that resists high temperature. In firing test due to the bonding method and design tolerance pressure buildup by reaction was not confirmed. But ignition, flame propagation and actuation of micro structure from the reaction was observed. From the result basement of design and fabrication technology was obtained.

Removal of mid-frequency error from the off-axis mirror

  • Manufacturing of lens and mirror using Diamond Turning Machine (DTM) offers distinct advantages including short fabrication time and low cost as compared to grinding or polishing process. However, the DTM process can leave mid-frequency error in the optical surface which generates an undesirable diffraction effect and stray light. The mid-frequency error is expected to be eliminated by mechanical polishing after the DTM process, but polishing of soft surface of ductile aluminum is extremely difficult because the polishing process inevitably degrades the surface form accuracy. In order to increase its surface hardness, we performed electroless nickel plating on the surface of diamond-turned aluminum (Al-6061T6) off-axis mirrors, which was followed by the 6-hour-long baking process at $200^{\circ}C$ for improving its hardness. Then we polished the nickel plated off-axis mirrors to remove the mid-frequency error and measured polished mirror surfaces using the optical surface profilometer (NT 2000, Wyko Inc.). Finally, we ascertained that the mid-frequency error on the mirror surface was successfully removed. During the whole processes of nickel plating and polishing, we monitored the form accuracy using the ultra-high accurate 3-D profilometer (UA3P, Panasonic Corp.) to maintain it within the allowable tolerance range (< tens of nm). The polished off-axis mirror was optically tested using a visible laser source and a pinhole, and the airy pattern obtained from the polished mirror was compared with the unpolished case to check the influence of mid-frequency error on optical images.
