The objective of this experiment was to investigate the effect of Newcastle disease virus (NDV) and infectious bursal disease virus (IBDV) vaccination on performance of broiler chicks for five weeks. Two types of poultry houses and three patterns of vaccination ($NDV^-/IBDV^-$, $NDV^+/IBDV^-$ and $NDV^+/IBDV^+$) were factorially assigned to six treatments. NDV, B1 strain and IBDV, Bursin-2 vaccine were orally administered at 5, 14 and 7, 18 days, respectively. Forty eight hundred chicks were grouped into four replications with two hundnyd hybro $\times$ hybro chicks per each treatment. Weight gain, feed conversion ratio (FCR), mortality and product index were surveyed at the end of experiment. Bursa index and IBDV antibody titer of chicks were weekly measured. Weight gain of chicks vaccinated with $NDV^+/IBDV^+$ was significantly increased compared to that of other treatments at both window and windowless poultry houses (p<0.05). Chicks vaccinated with $NDV^+/IBDV^+$ also showed significantly improving the FCR and mortality compared to those of other treatments at both poultry houses (p<0.05). The bursa indecies of both poultry houses were high from one-day- to three-weeks-old, but were low for the rest of two weeks. IBDV antibody of all chicks was detected 100% by agar gel precipitation (AGP) test at one day old, but was not detected in $NDV^-/IBDV^-$ and $NDV^+/IBDV^-$ treatments at four weeks old. However, it showed 100% in $NDV^+/IBDV^+$ treatment. Antibody titer using ELISA showed similar trend to that of AGP test. The results of this experiment confirmed that IBDV and NDV combined vaccine significantly improved the performance of broiler chicks.
The immune responses of commercial layer chickens against Newcastle disease (ND) were compared among different administration methods and times of vaccination during 4 weeks of age. A total of 372 day-old chickens were devided into 4 groups of 93 birds each. Each of 3 groups was received a commercially available B$_1$ live vaccine via drinking water, eye instillation or spray method at one, 14 and 28 days of age. One group was used as an unvaccinated control. At two and 4 weeks after each time of vaccination, 15 birds from each group were challenged with virulent ND virus at the dose of 10$^{5}$ EID$_{50}$ per bird to examine the pretection rate. Ten to 15 birds from each group were bled at two weeks intervals from day old to 8 weeks of age to determine hemagglutination inhibition antibody titer. The protection rate was generally low regardless of the times of vaccination although two or more times vaccination gave higher protection than once vaccination. The low protection was considered due to low titer of the vaccine used since the vaccine titer was less than 10$^{2.5}$ EID$_{50}$ per bird. Spray method gave better protection compared to eye instillation of drinking water method which resulted in lowest response. When birds were challenged majority showed clinical signs on ND between 3 and 6 days after challenge. Death occured one or two days after onset of symptoms. Major clinical signs observed were depression (96%), drowsy(90%), anorexia (84%), diarrhoea (29%), difficult breath (15%) and torticollis (10%). Hemorrhagic lesions on post mortem were seen in duodenum (51%), trachea(36%), illeum (13%), ceacal tonsil (11%), proventriculus (10%) and some other organs. When birds were challenged majority showed clinical signs on ND between 3 and 6 days after challenge. Death occured one or two days after onset of symptoms. Periodic outbreaks of Newcastle disease (ND) caused by velogenic viscerotropic ND virus (vvNDV) has become a major concern in Korea nowadays. Throughout last epidemic, the winter season in 2000, most chicken flocks infected early, under 2-4 weeks of age, showed high mortality up to 50-100%. Serum samples collected from 201 breeder, 284 layer and 112 broiler chicken flocks were examined to evaluate the efficacy of various vaccination methods and programs routinely used for mass vaccination in the field poultry farms. Despite repeated live vaccination, most poultry flocks vaccinated by drinking water route using nipple water supply system failed to produce solid active immune response to NDV during the growing time. In the present study, we applied the spray vaccination technique using Ulvavac or Desvac sprayer to the experimental poultry flocks and examined the efficacy of live vaccination effects induced by it under field condition. Measurable antibody to NDV as well as early protection against vvNDV challenge were found in poultry flocks vaccinated by spray route. Further, we did not found significant post vaccination reactions caused by spray vaccination if properly administered. These data indicate that the spray vaccination will be safe and reliable mass vaccination method for the prevention of ND.

Newcastle disease (ND) is an infectious poultry disease that caused high mortality and reduced egg production. NDVs are regularly present in the domestic duck population. And ducks play a possible role in the maintenance and transmission of NDVs. While we were monitoring the Avian Influenza, NDVs were isolated from field samples by accident. So we analysed the biological and genetic characteristics of these viruses. Lentogenic NDVs were isolated from two farms among twenty breeder duck farms. The ages of ducks were 39 weeks old in the 'A' farm and 3~72 weeks old in the 'B' farm. And they were not inoculated with the NDVs vaccine. In the biological characteristics, the both viruses which separated from the farm 'A' and 'B' were thermostable. The amino acid sequence of a site from 112 to 119 in the fusion (F) protein was 'GKQGRLIG' which has monobasic motif in the samples of both farms. And this means the separated NDVs are lentogenic. Phylogenetic analysis was performed by entire nucleotide sequence of F protein. The virus strains from the A farm (MN095239) and the B farm (MN095240) belonged to class II genotype I. Using the analysis of whole F protein nucleic acid sequence, the MN095239 (GenBank) had homology with Ulster strain about 99.95% and the MN095239 (GenBank) had homology with KR/CK/KU_LBM255/09 strain about 99.89%. NDV surveillance is needed to investigate epidemiological relationship of domestic breeder duck isolates in Korea.
To confirm the effect of food and water deprivation prior to Newcastle disease(ND) virus vaccination, three hundred chicks were divided into five groups with three replications. ND vaccine were sprayed to at 1 -day old chicks at commercial hatchery. Secondary and third vaccination was conducted at 2-week old and 24-day old chicks by LaSota strain. Control was conventionally vaccinated without withdrawing the food and water before or after vaccination. In group 2(G2) and 3(G3), LaSota strain was vaccinated to chicks before and after fasting the food and water for 3 and 2 hours, respectively. Group 4(G4) has the same fasting time of group 2, but supplemented the skim milk in vaccin dilution water. In group 5(G5), skim milk was added into group 3. Weight gain, feed intake and feed conversion were weekly measured for 5 weeks. Blood was collected from wing vein at 24 and 35 days of age. Each serum antibody level were measured by hemagglutination inhibition(HI) test. The average weight gain, feed intake, feed conversion of all group were not significantly different. Weight gain of each groups was 1910.30(control), 1875.28(G2), 1952.12(G4) and 1896.05(G5), respectively. Feed intake of all group was recorded at 3160.67(control), 3167.07(G2), 3189.48(G3), 3157.85(G4) and 3178.16(G5), respectively. The feed conversion of each groups was 1.655(control), 1.688(G2), 1.633(G3), 1.699(G4) and 1.676(G5), respectively. The HI titer of G4 was $ 5.50{\Pm}$1.40 and significantly higher than the other groups (p<0.05)(control : $4.36{\Pm}$1.87 , G2 : $5.18{\Pm}$2.14, G3 : $4.51{\Pm}$2.19, G : $5.28{\Pm}$1.58 at 35 days old. The results of this experiment indicated that two or three hours of fasting time before or after vaccination would be able to show the higher antibody level against ND virus.
Reciprocal crosses were conducted between three strains of Taiwan Country chickens, developed in the National Chung-Hsing University, and two strains of Single Comb White Leghorns, developed in the Taiwan Livestock Research Institute. Traits studied were growing performances, laying performances, egg quality traits and traits concerning disease resistance, including resistance to Marek's disease virus and immune responses to Newcastle disease virus vaccine and to sheep red blood cell. Results indicated that laying performances of Taiwan country chickens were much inferior to White Leghorns, but they matured earlier, their eggs had better shell strength and larger proportion of yolk, and their general disease resistance was much better than White Leghorns. Heterosis were found in laying performances and egg quality traits. The heterosis in laying traits was so large that the hybrid laid as many eggs and as large eggs as did pure strains of White Leghorns. Strategies on the improvement of native chickens and the utilization of genetic merits of native chickens were also discussed.
To compare of serum antibody titers using ELISA and HI, serum samples were collected from 100 breeders and their progeny 550 broilers. The breeders and broilers were vaccinated with infectious bronchitis(IB)- and Newcastle disease(ND)-viruses according to general vaccination program. The antibodies in serum samples against IB and ND viruses were detected by enzyme-linked immunosorbent assay(ELISA) using commercial ELISA kit and hemagglutination inhibition(HI) test. Geometric mean titer(GMT) of ELISA and In titers were monitored from 1-day-old to 35-day-old broilers and compared to those of breeder chickens. The antibody titers of breeders vaccinated with ]B virus showed 47,800, ELISA and 7.2, HI, respectively. Progeny chicks, 1-day-old, vaccinated with IBV showed high antibody titers than those of breed chickens. Those chicks were maintained protective antibody levels until 11-day-old. From 14-day-old, the antibody level decreased below protective levels. In ND, breeders serum antibody titers ELISA and Eiu were 30,200 GMT and 8.7 HI titer, respectively. On 1-day-old chicks, antibody levels was decreased to half in ELISA(16,270) compared with those of breeders, but In titers was 7.4. Progeny broilers, protective antibody level was maintained until 14- day-old by ELISA, but at 11-day-old by HI titers. After then, ND antibody titer was continuously decreased underdefense level. These result indicated that the ELISA method be more sensitive than HI titration to detect serum antibody level for IBV and NDV.
This study was conducted from May to December 2016 to investigate the prevalence of antibody titers for Newcastle disease (ND), Infectious Bronchitis (IB), Egg drop syndrome (EDS), Avian pneumovirus (APV), Fowl Typhoid (FT), Mycoplasma gallisepticum (MG), Mycoplasma synoviae (MS) in laying hens in Incheon area. In ND, 418 (99.5%) of the 420 outbreaks were positive. IB showed 330 (78.5%) positive antibody out of 420 cases. EDS showed 419 (99.7%) positive seropositive rates among 420 cases. In the case of APV, 357 cases (85%) were positive in 420 cases. In the case of PT, the positive rate of antibody was 128 (30.4%) out of 420 cases. Among the 420 cases of MG, 222 cases (52.8%) were positive for antibodies. In the case of MS, 395 out of 420 cases (94.0%) were positive. This result suggests that the positive rate of antibody for PT in Incheon area was low. And it was also clear that MS without vaccine is now rampant in Incheon.

In this report, fifteen monoclonal antibodies (MAbs) against an avian influenza virus (H9N2 subtype) were newly produced and characterized. These MAbs proved to react to the epitopes of nucleocapsid protein (NP), hemagglutinin (HA), neuraminidase (NA) and non-structural protein 1 (NS1) of Korean H9N2 strain, respectively. Two HA-specific MAbs showed the ability to inhibit the hemagglutination activity of H9N2 subtype avian influenza virus when tested by hemagglutination inhibition (HI) assay. All MAbs did not cross-react with other avian-origin viruses (Newcastle disease virus, infectious bursal disease virus, infectious bronchitis virus and avian rotavirus) by immunofluorescence test or enzyme-linked immunosorbent assay. The MAbs produced in this study could be useful as the materials for diagnostics and therapeutics against Korean-lineage H9N2 virus infections.
Male broiler chicks were fed graded levels of organic zinc (zinc-methionine) supplementation to investigate the effects of partial or complete substitution of the organic zinc source for inorganic ones on the development of lymphoid organs and immunological responses. A total of 450 day-old male broilers were distributed into groups of 10 chicks and randomly assigned to nine experimental diets during a 42-day feeding trial. Dietary treatments consisted of two basal diets supplemented with 40 mg/kg added zinc as feed-grade Zn sulfate or Zn oxide in which, Zn was replaced with that provided from zinc-methionine (ZnMet) complex at the levels of 25, 50, 75 or 100%. Two randomly-selected birds from each pen replicate were bled and then slaughtered by cervical cutting on the final day of the trial to measure leukocyte subpopulations and relative weights of lymphoid organs. Among lymphoid organs, only thymus weight was affected (p<0.05) by dietary treatments. The sulfate-supplemented birds were heavier (p<0.01) in relative weight of thymus than oxide-supplemented birds. The 10 days of age-assessed cutaneous hypersensivity reaction was stronger in chicks fed ZnMet-containing diets. Dietary ZnMet supplementation caused (p<0.05) an increase in proportion of lymphocytes and consequently a decrease in heterophil to lymphocyte ratio. Diet fortification by zinc-methionine complex increased (p<0.01) Newcastle antibody titer at 19 days of age. Also, a similar response was observed in antibody titers at 6 and 12 d after infectious bronchitis vaccine administration. There was no significant effect of replacement of dietary zinc on antibody titer against infectious bursal disease virus (IBDV) at the 6th d post-vaccine inoculation; however, at d 12 after vaccination, ZnMet-fortified diets improved antibody titer against IBDV. Although dietary inclusion of ZnMet had no marked effect on primary antibody titer against sheep erythrocytes, effective responses were observed during secondary reaction from the viewpoint of both total antibody and immunoglobulin Y (IgY) titers. From the present findings, it can be concluded that dietary supplementation with organic zinc improves both cellular and humoral immune responses. It is necessary to replace 75% of supplemental inorganic zinc with organic ZnMet complex to achieve the optimum immunological responses in broiler chicks.
