Newcastle disease virus: the past and current situation in Indonesia

  • The Newcastle disease virus (NDV) outbreak was first reported in Java Island, Indonesia, in 1926, which was then reported further in Newcastle-upon-Tyne, England. Nevertheless, the NDV is still endemic in Indonesia, with outbreaks occurring in free-range and commercial chicken farms. The dynamic evolution of the NDV has led to the further development of vaccines and diagnostic tools for more effective control of this virus. This paper discusses the history of the NDV occurrence, vaccines, the development of diagnostic tools, and the epidemiological condition of the NDV in Indonesia. Indonesia, which has the largest poultry population in the world after China, has challenges in preventing and controlling this virus that causes economic losses to the farmers and has an impact on the welfare of the poultry farming community in Indonesia.

뉴캣슬병 $B_1$ 생독백신의 효능비교 (Comparison of Efficacy of Newcastle Disease Virus $B_1$ Vaccine by Different Administration Method in Commerical Layer Chickens)

  • The immune responses of commercial layer chickens against Newcastle disease(ND) were compared among different administration methods and times of vaccination during 4 weeks of age. A total of 372 day-old chickens were divided into 4 groups of 93 birds each. Each of 3 groups was received a commercially available B$_1$live vaccine via drinking water, eye instillation or spray method at one, 14 and 28 days of age. One group was used as an unvaccinated control. At two and 4 weeks after each time of vaccination, 15 birds from each group were collected randomly out and challenged with virulent ND virus at the dose of $10^5E1D_{50}$ per bird. Ten to 15 birds from each group were bled at two weeks intervals from day old to 8 weeks of age for hemagglutination inhibition antibody titer, The protection rate was generally low regardless of the times of vaccination although two or more times vaccination gave higher protection than once vaccination. The low protection was considered due to low titer of the vaccine used since the vaccine titer was less than $10^{3.5}EID_{50}$ per bird. Spray method gave better protection compared to eye instillation or drinking water method which resulted in lowest response. Majority of birds showed clinical signs of ND between 3 and 6 days after challenge. Death occured one or two days after onset of symptoms. Major clinical signs observed were depression(94%), anorexia(84%), diarrhoea(29%), difficult breath(15%) and torticollis(10%). Hemorrhagic lesions on post mortem were seen in duodenum(51%), trachea(35%), illeum(13%), ceacal tonsil(11%), proventriculus(10%) and some other odrgans.

미량영양소들의 추가급여가 뉴캣슬 또는 전염성 기관지염 백신 접종시 산란계의 면역성에 미치는 효과 (Effects of Excess Dietary Supplementation of Several Micronutrients on Immune Response in Layers Inoculated with Newcastle Disease and Infectious Bronchitis Vaccines)

  • This study was conducted to investigate the immune response of layers fed diets supplemented with excess micronutrients, i.e., vitamin A, methionine, Zn, Cu, and Fe to the inoculation of Newcastle disease vaccine(NDV) or infectious bronchitis vaccine(IBV). The antibody titer against the NDV increased immediately after the inoculation and stayed high during the next 6 wk. On the other hand, The antibody titer against the IBV increased after 4 wk of inoculation The IgM level increased rapidly after 1 wk of NDV inoculation, however, it decreased after 5 wk of inoculation. The IgA displayed similar pattern to that of IgM in response to NDV inoculation. The pattern of IgM change after IBV inoculation was similar to that when layers were treated with NDV. However, IgA level changed earlier than did IgM. The IgG response to the NDV and IBV was very weak compared to the other immune responses. The excess supplementation of micronutrients to the diets of layers inoculated with NDV elicited favorable antibody titer and immune response compared to the layers fed the control diet. The excess Zn, however, allowed the layers to have higher antibody titer for the 4-wk period after NDV injection: after that they showed no effect of extra-Zn. The immune responses of layers fed excess vitamin A, Cu, methionine, and Fe were markedly higher in IgA and IgG than the control layers. The excess Zn, however, did not bring about any favorable result. No difference was detected in IgG level between control and micronutrients-treated groups.

Molecular differentiation of Korean Newcastle disease virus (NDV) by restriction enzyme analysis and pathotype-specific RT-PCR

  • Newcastle disease virus (NDV) is a single-stranded negative sense RNA virus, which has been classified as a member of the Avulavirus genus of the Paramyxoviridae family. It is also one of the most important pathogens in the poultry industry. The glycoproteins, fusion (F) and hemagglutinin-neuraminidase (HN), determine the virulence of NDV, and the relevant molecular structures have already been determined. NDV isolates differ in terms of virulence, and at least 2 of 9 genotypes (I-IX) have been shown to co-circulate. Therefore, it is clearly important to differentiate between vaccine strains and field isolates. In vivo pathogenicity tests have been the standard protocol for some time, but molecular methods appear preferable in terms of the rapidity of diagnosis, as well as animal welfare concerns. In this study, we have designed primer sets from HN gene for phylogenetic analysis and restriction enzyme analysis, and from F gene for pathotype-specific RT-PCR. Via the combination of 2 methods, 106 Korean NDV isolates obtained from 1980 to 2005 were differentiated into vaccine strains, and virulent genotypes VI and VII. The genotype VI viruses were only rarely isolated after 1999, and genotype VII, after it was initially isolated from poultry in 1995, recurred in 2000, and then became the main NDV constituting a threat to the Korean poultry industry.

Binary Ethylenimine으로 불활화(不活化)한 Newcastle Disease Virus의 항원성(抗原性)과 면역원성(免疫原性)에 관한 연구(硏究) (Studies on the Antigenicity and Immunogenicity of Newcastle Disease Virus Inactivated with Binary Ethylenimine)

  • Effects of binary ethylenimine (BEI) treatment on the inactivation of infectivity and hemagglutinin of Newcastle disease virus (NDV) were studied in comparison with those of formalin treatment. Immune responses of chickens vaccinated with BEI-inactivated NDV vaccines were also investigated. The results were summarized as followings; 1. Complete loss of infectivity of NDV (Bl) was observed at 3, 7, and 24 hours after the treatment at $37^{\circ}C$ with BEI concentrations of 0.01M, 0.005M and 0.001M, respectively. 2. The hemagglutinin activity of NDV (Bl) remained constant when treated with 0.01M BEI at $37^{\circ}C$. However, it gradually decreased when treated with 0.1% or 0.2% formalin at $37^{\circ}C$. 3. When 4-week-old chickens were vaccinated with NDV vaccines prepared from Bl or Miyadera strains of NDV, inactivated with 0.1M BEI and adsorbed to aluminium hydroxide gel, favorable immune responses were observed throughout the 8 weeks of observation period. 4. When these chickens were revaccinated at 8 weeks after the first vaccination, strong anamnestic responses were evoked and the immunity maintained for 4 weeks of the observation. Though slightly bettor immune responses were observed after primary vaccination in chickens vaccinated with Bl vaccine compared with those vaccinated with Miyadera vaccine, the differences were not significant. 5. On the electron microscopy, BEI (0.01M) gave least effect to the envelope as well as capsid of NDV.

한국산 꿩으로부터 분리한 열 안정성 뉴캣슬병 바이러스의 순화와 방어효과 (Attenuation and Protective Effects of a Thermostable Newcastle Disease Virus Isolated from Korean Pheasants)

  • The objective of these experiments was to develop an attenuated thermostable Newcastle disease virus(NDV), CBP-1 strain isolated from infected pheasants. Safety, pathogenicity and protective effects against velogenic NDV were also investigated to evaluate if the attenuated NDV, CBP-1 strain could be a candidate for a new NDV vaccine strain. CBP-1 strain was passaged up to the 173 times by nine days old embryonated eggs and chicken embryo fibroblast(CEF) cell cultures. Its sensitivitly to lipid solvents and low pH, thermostability, mean death time(MDT), intracerebral pathogenicity index(ICPI) of one day old chicks and intravenous pathogenicity index(IVPI) of four weeks old chicks were examined. Safety, boosting and protective effects were tested by chicks mortality. CBP-1 NDV strain had significant thermostability at 56$\^{C}$ for 30 minutes. by hemagglutinin activity and egg infectivity test, but was not resistant to lipid solvent. It showed possibility to use as a feed or water vaccine because of the resistance to low pH. MDT, ICPI and IVPI of CBP-1 were attenuated from 51.5, 1.96, 2.60 to 112.4, 1.12, 1.45. These results implied that the 173rd passages in embryonated egg and CEF cell cultures induced a substantial attenuation of the pathogenicity of the parent virus, changing the virulence from velogenic to intermediate between mesogenic and lentogenic. After vaccination with CBP-1 at one day old by drinking water mortality was 17.5%. However, spray vaccination with B1 at one day old, CBP-1 at two weeks ild and challenge with velogenic Kyojeongwon strain at four weeks old showed 93.5% survival rate. Mortality of chicks, vaccination with 173rd passaged CBP-1 strain at one day old, two weeks old and challenge with Kyokeongwon strain at four weeks old, was 20.0%. The results of these studies indicated that partial attenuated CBP-1 strain tended to be a low safety for ND of broiler chicks and would need to be more successive attenuation.

Validation of a Real-Time RT-PCR Method to Quantify Newcastle Disease Virus (NDV) Titer and Comparison with Other Quantifiable Methods

  • A method for the rapid detection and quantification of Newcastle disease virus (NDV) produced in an animal cell culture-based production system was developed to enhance the speed of the NDV vaccine manufacturing process. A SYBR Green I-based real-time RT-PCR was designed with a conventional, inexpensive RT-PCR kit targeting the F gene of the NDV LaSota strain. The method developed in this study was validated for specificity, accuracy, precision, linearity, limit of detection (LOD), limit of quantification (LOQ), and robustness. The validation results satisfied the predetermined acceptance criteria. The validated method was used to quantify virus samples produced in an animal cell culture-based production system. The method was able to quantify the NDV samples from mid- or late-production phases, but not effective on samples from the early-production phase. For comparison with other quantifiable methods, immunoblotting, plaque assay, and tissue culture infectious dose 50 ($TCID_{50}$) assay were also performed with the NDV samples. The results demonstrated that the real-time RT-PCR method is suitable for the rapid quantification of virus particles produced in an animal cell-culture-based production system irrespective of viral infectivity.

모계의 전염성기관지염, 전염성 F낭병 및 뉴캣슬병 백신투여에 따른 모체이행 항체의 변동 (Changes of maternal antibodies in chicks vaccinated to breeder against infectious bronchitis, infectious Bursal disease, and Newcastle disease virus)

  • Serum samples collected from 30 breeders and their progeny 30 chicks. The antibodies against infectious bronchitis(IB), infectious bursal disease (IBD) and Newcastle disease(ND) viruses were detected by ELISA using commercial ELISA kit. The breeders were vaccinated against IB, IBD and ND viruses according to general vaccination program. Geometric mean titers(GMT) of ELISA were monitored from 1-day old to 17-day old chicks and compared with breeder chickens. The GMT of ELISA to IB, IBD and ND were declined half level of the breeder antibody titer at 6-, 8- and 7-day old. And, the GMT of ELISA to IB, IBD and ND were declined than that of protective titer at 6-, 1-, and 4-day old. Thereafter, the GMT of ELISA was declined and disappeared according to ages of chicks. Taken together, this study led to conclusion that time-course of maternal antibody titers of chicks from vaccinated breeders, and this is very important data for vaccination to chicks.

Serological responses and protection levels in chickens administered with Newcastle disease vaccines

  • Vaccination against Newcastle disease (ND) is the most effective means of controlling the disease, and these vaccines are commercialized only after their safety and effectiveness have been verified through tests that comply with Korean Standards of National Lot Release for Veterinary Biologics. This study investigated whether a relatively convenient and safe serological test can be used in place of the challenge test using highly virulent ND virus. Hemagglutination inhibition (HI) assay and enzyme-linked immunosorbent assay (ELISA) were considered positive of log2 2 or more and cutoff value of 200 or more, respectively, in both live and inactivated vaccines. However, when the antibody levels of the live and inactivated vaccines induced using the Ulster 2C, KBNP-C4152R2L, and K148/08 strains were compared, the antibody titers for inactivated vaccines were significantly higher than those for live vaccines in both the HI assay and ELISA. A strong positive correlation was observed between HI and ELISA antibody titers. The live vaccines corresponded to a survival rates of ≥ 80% and the inactivated vaccines corresponded to 100% survival rates. This study confirmed that standard efficacy tests can serve as serological tests, and can replace the challenge test and that the vaccine approval process can be improved.

육용계 초생추에 대한 뉴켓슬병 생독 및 사독 백신의 동시접종 효과 (Efficacy of Concurrent Administration of Live and Killed Newcastle Disease Vaccines in Day - Old Broiler Chicks)

  • 모체이행항체가 있는 병아리에 생독과 사독 ND 백신을 동시접종 했을때 면역효과를 시험하기 위하여 국내에서 유통되고 있는 $B_1$과 LaSota의 2가지 생독백신과 Gel, Oil의 2가지 사독백신을 구입 1 일령에 각각 단독접종하거나 생독과 사독을 서로 다르게 조합하여 동시에 접종하였다. 생독백신은 백신제조회사에서 추천하는 1수분을 25${\mu}\ell$ 씩 한쪽눈에 적하 하였으며 사독은 0.3$m\ell$씩 경부피하에 접종하였다. HI 가를 주별로 8주까지 조사하였으며 방어율을 측정하기 위하여 4주와 8주에 강독으로 수당 $10^{5.3}$$ELD_{50}$씩 공격 접종하였다. 8개 시험군 중에서 .사독Oil백신과 생독을 동시에 접종한 군이 항체가와 방어율이 가장 높았다. 생독백신이나 Gel백신만 단독접종한 군은 방어율이 50%이하로 낮았으며 항체가도 4~5주 때 $2^2$이하로 하강하였다. 사독 Oil과 동시 접종한 생독백신으로서 $B_1$과 LaSota간에 는 방어율이나 항체가에서 차이를 인정할 수 없었다. 2가지 생독이 모두 Oil백신과 동시 접종했을 때 4주에서 방어율이 100%였으며 8주 때는 90-100%이었고 항체가도 $2^4$ 이상 8적까지 지적되었다. 동시접종해도 호흡기증상등의 부작용이나 증체율 저하 등의 현상은 볼 수 없었다.
