Current status on applications of conventional breeding techniques and biotechnological system in ornamentals

  • Flower industry is now growing due to the development of economy in many countries. Simultaneously, needs from consumers in flower market are varied widely. To satisfy the needs from consumers and deal with a variety of diseases from a lots of pathogens as well as climate change, new elite flower cultivars should be released in flower market. For this purpose, conventional and biotechnological techniques can be employed to make good cultivar. Therefore, this review describes the general overview of flower breeding techniques including cross-hybridization, mutation breeding and genetic transformation systems. Also, breeding systems for ornamentals derived from plant tissue culture such as embryo culture, in vitro fertilization, ovary/ovule culture and haploid production were reviewed. Furthermore, in this study recent development of the generation of new flower cultivars using marker-assisted breeding, plant transformation including particle bombardment and Agrobacterium tumefaciens as well as genome-editing technology were described. This review will be contributed to the development and releasement of new flower cultivars with horticulturally useful traits in the future.

벼 이삭도열병 발생정도 및 병원균 레이스의 분포 변동(1993~1994)과 새로운 레이스의 병원성 (Incidence of Panicle Blast, Race Distribution During 1993??¡?1994 and Pathogenicity of New Races of Pyricularia grisea in Korea)

  • Severity of panicle blast disease on the leading cultivars of rice was investigated at nine locations of Korea during summer in 1993 with frequent raining and low temperature conditions. Incidence of panicle blast was 26% on Jinmibyeo, 23% on Chuncheongbyeo and 40% on Odaebyeo. Race distribution of Pyricularia grisea was examined from 1993 to 1994. A total of 1,098 isolates of the fungus obtained from the blast nurseries and framers; fields were screened using Korean differential varieties. Twenty one races were identified in 1993 and 19 races in 1994. KI-181, KJ-103, KJ-104, KI-241 and KI-209 were identified as new races, and KJ-301 (15%), KJ-201 (13%), KJ-105 (12%) and KI-409 (12%) were major races during 1993 and 1994. Race KI-197, which has a wide spectrum of virulence on rice cultivars, was isolated in a ratio of 8.3% and evenly distributed in the whole country. Some of the major and newly identified races during 1993∼1994 were used for spray-inoculation on leading cultivars and differential varieties in a greenhouse. We found that most of the cultivars were resistant to the major races, KJ-301 and KI-409, but were very susceptible to the new races, KI-241, KI-209, KI-181 and KJ-103.

수박 품종들의 덩굴쪼김병균에 대한 저항성 특성 (Resistance Characteristics of Watermelon Cultivars to Fusarium oxysporum f. sp. niveum)

  • Fusarium oxysporum f. sp. niveum (Fon)에 의한 덩굴쪼김병은 수박 재배지에서 심각한 피해를 주는 병해 중 하나이다. 국내에서 대부분의 수박 품종들은 덩굴쪼김병에 대해 감수성이다. 덩굴쪼김병균은 4개의 수박 품종에 대한 병원성을 기준으로 4개의 레이스(0, 1, 2, 3)가 알려져 있다. 본 연구에서는 7개의 덩굴쪼김병균을 수집하고 이 병원균들의 레이스를 확인하였다. 그 결과, KACC 40902와 HA 균주는 race 0, NW1, NW2, CW 그리고 KACC 40901 균주들은 race 1로, KACC 40905 균주는 race 2로 동정되었으며, race 3 균주는 확인되지 않았다. 또한, 수박의 3개의 감수성 품종에서 7개의 덩굴쪼김병균의 병원력을 비교하였다. 실험에 사용한 Fon 균주들은 균주들 간에 서로 다른 병원력을 나타냈다. 그리고 중도 저항성이거나 저항성으로 알려진 수박 품종 3개, 수박 대목 1개, 감수성 품종 1개를 선발하여 Fon에 대한 수박의 저항성 특성을 조사하였다. 수박 대목은 7개 Fon 균주들에 대해 높은 저항성 반응을 나타냈고, 'Calhoun gray'는 KACC 40905 균주를 제외한 6개 균주에 대해서는 높은 저항성 반응을 나타낸 반면, 나머지 2개의 수박 품종('속노란꿀'과 '초당꿀')은 실험한 7개 균주들의 병원력에 비례하여 수박 덩굴쪼김병이 발생이 증가하였다. 이상의 결과로부터 Fon에 대한 수박의 저항성은 병원균의 레이스 분화가 아닌 병원균의 병원력에 의해 결정된다고 생각되었다.

고추 풋마름병에 대한 효율적인 저항성 검정법 확립 (Development of an Efficient Bioassay Method to Evaluate Resistance of Chili Pepper Cultivars to Ralstonia solanacearum)

  • Ralstonia solanacearum에 의한 고추 풋마름병은 고추의 생산량 감소에 영향을 미치는 주요 병해 중 하나이다. 그리고 병 저항성 작물을 재배하는 것은 고추의 풋마름병 방제에 가장 효과적인 방제 방법이다. 본 연구는 고추의 풋마름병 저항성을 효율적으로 검정하기 위한 방법을 개발하기 위하여 수행되었다. 풋마름병에 대한 저항성 및 감수성 6개 고추 품종을 선발하고, 다양한 조건에서 이들 품종의 풋마름병 발생을 조사하였다. 접종 방법에 따른 고추 품종들의 풋마름병 발생 정도를 실험한 결과, 상처없이 토양관주하는 방법과 접종원에 뿌리를 침지하는 방법보다 뿌리 절단 후 접종원을 관주하는 방법이 더 간단하고 효율적이었다. 풋마름병 저항성 계통인 'MC4'는 파종 후 21일부터 28일 재배한 고추 유묘에 $1{\times}10^8cfu/ml$ 농도의 세균 현탁액을 포트 당 20 ml 접종하였을 때 가장 높은 저항성을 나타냈다. 한편 감수성 품종들은 이들 조건에서 높은 감수성을 보였다. 이들 결과는 우리가 고추 품종의 풋마름병에 대한 효율적인 저항성 검정방법을 개발하였다는 것을 나타낸다. 그리고 선발한 발병조건을 사용하여 시판중인 140개 고추 품종의 풋마름병에 대한 저항성 정도를 평가하였다.

배추 품종들의 검은썩음병에 대한 저항성 특성 (Resistance Characteristics of Chinese Cabbage Cultivars to Black Rot)

  • Xanthomonas campestris pv. campestris (Xcc)에 의한 검은썩음병은 배추 재배에서 가장 심각한 피해를 입히는 병해 중 하나이다. 본 연구는 Xcc에 대한 배추의 저항성 특성을 조사하기 위하여 실험하였다. 먼저, Xcc 8개 균주의 병원력을 비교하기 위하여, 감수성인 배추 4개 품종에서 이들 균주에 의한 배추 검은썩음병 발생을 조사하였다. 실험에 사용한 Xcc 균주들은 균주들 간에 서로 다른 병원력을 보였다. 그리고 Xcc에 대한 저항성 및 중도저항성 22개 배추 품종을 선발하고, Xcc 8개 균주를 사용하여 이들 배추 품종의 검은썩음병 발생을 조사한 결과, 배추 품종들의 평균 검은썩음병 병반면적률은 Xcc 균주들의 병원력에 따라 증가하였다. 게다가 실험한 각 배추 품종들의 검은썩음병 발생은 실험한 균주들의 병원력에 비례하여 증가하였으며, Xcc 균주들에 대해 저항성인 배추 품종의 수는 균주들의 병원력에 반비례하게 감소하였다. 이상의 결과로부터 Xcc에 대한 배추의 저항성은, 병원균의 레이스 분화가 아니라 병원균의 병원력에 영향받는다고 생각되었다.

Inheritance of Cyst Nematode Resistance in a New Genetic Source, Glycine max PI 494182

  • Worldwide, cyst nematode(Heterodera glycines Ichinohe) is the most destructive pathogen of cultivated soybean. In the USA, current annual yield losses are estimated to be nearly a billion dollars. Crop losses are primarily reduced by the use of resistant cultivars. Nematode populations are variable and have adapted to reproduce on resistant cultivars over time because resistance primarily traces to two soybean accessions. It is important to use diverse resistance sources to develop new nematode resistant cultivars. Soybean PI 494182 is a recent introduction from Japan and found to be resistant to multiple nematode populations. It is yellow seeded and maturity group 0. We have determined inheritance of resistance in PI 494182 using $F_{2:3}$ families derived from cross PI 494182 X cv. Skylla. Skylla is a susceptible parent. Three nematode populations, races 1, 3, and 5, corresponding to HG types 2.5.7, 0, and 2.5.7 were used to bioassay 162 $F_{2:3}$ families in greenhouse experiments. Based on Chi-square tests, a two-gene model is proposed for resistance to race 1 and a three-gene model is proposed for conditioning resistance to both races 3 and 5. Correlation coefficient analysis indicated that some genes conditioning resistance to races 1, 3, and 5 are shared or closely linked with each other. These results will be useful to soybean breeders for developing soybean cultivars for broad resistance to nematodes.

Differential Responses of Soybean Cultivars to Cercospora sojina Isolates, the Causal Agent of Frogeye Leaf Spot in Korea

  • During the summer of 2005, specimens of soybean cultivars (Daepung, Daewon, Hwanggeum and Taegwang) showing frogeye leaf spot (FLS) were obtained from various areas in Korea. Eight isolates identified as Cercospora sojina were inoculated on the adaxial leaf surfaces of 63 Korean soybean cultivars; the disease responses to each isolate were evaluated 14 days and 21 days after inoculation. Based on the disease responses (resistant or susceptible) of the cultivars by the isolates, a set of cultivars (Anpeong, Bogwang, Cheongdu No. 1, Cheongja No. 3, Dachae, Daemang, Jangwon, Namhae, Sowon, Taegwang) were selected and inoculated with seven isolates for further testing pathogenic variance. Interestingly, 6 out of 7 tested C. sojina isolates revealed differential ability in infecting different soybean cultivars. This result may indicate the possibility of new race occurrence or pathogenic variation; this also presents evidence for prevalent FLS occurrence during humid and hot weather in Korea.

헤어리 베치의 품종별 생육 특성과 생산성 비교 (Comparison of Growth Characteristics and Productivities of Hairy Vetch (Vicia villosa) Varieties)

  • 본 연구는 2005년과 2006년 축산과학원 2개 지역의 시험포장에서 각각 수행되었다. 1년차는 수원(중부)과 남원(남부)에서 2년차는 천안(중부)과 남원(남부)에서 수행되었으며 공시 품종은 헤어리 베치 11품종 도입품종 9품종(Penn-02, Sander, Latigo, Welta, Ostsaat, VV4712, Minnie, Barlosa 및 Capello), 국내품종 2품종(Cold green과 Cheong pa)를 이용하였다. 시험에 사용된 품종을 개화시기를 기준으로 평가한 결과 도입품종 Minnie, Barlosa, Capello 및 국내 품종 Cold green이 조생종이었고, 도입 품종 Penn-02, Sander, Latigo, Welta, Ostsaat, VV4712 및 국내품종 Cheong pa는 중만생종에 속하였다. 월동상태는 조생종 Minnie, Barlosa 및 Capello를 제외하고 대부분 우수하였다. 2년간 시험한 결과 11개 품종중 VV4712가 가장 수량이 높은 품종이었다. 그리고 중부지방에서는 중만생종의 수량이 남부지방에서는 조생종의 수량이 많았다. 두 지역에서 국내 육성품종인 중만생종 Cheong pa는 도입품종들에 비하여 뒤떨어지지 않았고 조생종 Cold green은 도입 품종들에 비하여 생육과 수량이 우수하였다. 헤어리 베치의 NDF, ADF, IVDMD 및 CP 등 사료가치는 조생종보다 중만생종이 좋았고, 그 중에서 NDF는 중만생종 Latigo, ADF는 중만생종 Welta, 소화율과 조단백질 함량은 중만생종 Sander가 가장 좋았다. 이 시험의 결과 헤어리 베치중 VV4712 품종의 생산성이 가장 높았고 국내 육성품종인 Cheong pa와 Cold green은 유망한 품종으로 판단된다.

Analysis of Genetic Diversity of Korean Wheat Cultivars Using Microsatellite DNA Polymorphisms

  • Genetic background and phylogenetic relationships among 20 Korean wheat cultivars were assessed using microsatellites after amplifying with 13 SSR primer pairs. Average allele number per primer pair was 3.36. Genetic similarities for every pair of cultivars ranged from 0.42 to 0.97, with 0.69 of overall average. Korean cultivars were divided into two major groups based on microsatellite DNA polymorphisms. Group I consisted of relatively old cultivars developed until 1970s, and group II contained the recent cultivars developed during 1980s and 1990s. Amongst old elite cultivars/lines, ‘Yukseung 3’, ‘Norin 12’ and ‘Norin 72’ contributed most to the genetic background of cultivars belonging to group I, and ‘Norin 4’, ‘Norin 12’, ‘Norin 43’ and ‘Norin 72’ to group II, respectively. The phylogenetic relationship of Korean wheat cultivars was in accordance with the genealogical data of each cultivar. The genetic background of each cultivar was assessed from the point of breeding and germplasm management such as variety identification and duplicated accessions for assisting in developing a system for the registration of new variety based on the molecular characterization in future.

Comparison of NERICA and Asian rice among traits relevant to drought resistance in the field and the effects of compost

  • Recently NERICA (New Rice for Africa) was developed by a crossing of African rice (Oryza glaberrima Steud.) and Asian rice (Oryza sativa L.) in West Africa, and is considered to be drought resistant, but drought resistance of NERICA and differences between Asian rice are not clarified enough. In this research, NERICA (four cultivars and two lines), Asian rice (three cultivars and sativa parent of NERICA) and African rice (glaberrima parent of NERICA) were cultivated in the field in Shizuoka University under drought and traits of each cultivar and line relevant to drought resistance, stomatal conductance by porometer, soil water content of individual depths by TDR method, SPAD values by SPAD meter and leaf thickness by micrometer, were measured and compared with dry matter production and yield. Effects of compost were also compared among sativa parent, one NERICA cultivar and two NERICA lines. Glaberrima parent showed highest top dry weight. One NERICA line, one drought resistant Asian rice cultivar and sativa parent, showed higher top dry weight and yield (ear weight) than other Asian rice cultivars and NERICA cultivars and line tested. Compost tended to increase top dry weight and yield in one of NERICA line and sativa parent. But in one NERICA cultivar and line, top dry weight and yield were not increased. In one of Asian rice, one of NERICA line and sativa parent that showed high top dry weight and yield, stomatal conductance was high. On the contrary the glaberrima parent and in other NERICA cultivars and line it was low. In sativa parent compost increased stomatal conductance but in NERICA cultivar and lines it was not. Among cultivars and lines that showed high top dry weight and yield sativa parent and one of NERICA line SPAD value and leaf thickness were high but in one of Asian rice and glaberrima parent they were low. Cultivar and line differences in yield and top dry weight among Asian rice and NERICA were significantly correlated with those in stomatal conductance ($r=0.778^{**}$ and $r=0.654^*$, respectively) and those in leaf thickness ($r=0.600^*$ and $r=0.640^*$, respectively). In Asian rice cultivars average soil water content was significantly correlated with yield ($r=0.886^*$) but in NERICA cultivars and lines it was not significant correlated (r= -0.256). Cultivar and line differences in leaf thickness were significantly correlated with SPAD value ($r=0.773^{**}$). In Asian rice cultivars it was significantly correlated ($r=0.962^{**}$), but in NERICA cultivars and lines it was not significantly correlated (r=0.559). Asian rice cultivars tended to consume soil water to increase yield but in NERICA cultivars and lines the tendency was not clear. Correlation between SPAD value and leaf thickness was different between Asian rice and NERICA cultivars and lines, and in Asian rice cultivars it was significantly correlated but in NERICA cultivars and lines it was not significant. Importance of maintaining high stomatal conductance by high leaf thickness was clarified.
