  • The hydrolysis of sphingomyelin (SM), known as the SM pathway, is induced by the activation of sphingomyelinase (SMase) to generate the second messenger ceramide, which plays a key role in cellular responses such as apoptosis, differentiation, senescence, and inflammation. Here, we identified a 60 kDa membrane-associated, neutral and Mg$\^$2+/-dependent SMase, termed N-SMase $\varepsilon$, from mammalian brains, which was revealed as the heat shock protein 60 (HSP60) through cDNA cloning and mass spectrometrical analysis. Cytochemical Measurement of Lysosomal Responses in the Digestive Cells of Wild Pacific Oyster, Crassostrea gigas on the South Coast of Korea (세포화학적 방법을 이용한 남해안 조간대에 서식하는 참굴(Crassostrea gigas) 소화맹낭 세포 내 리소솜 활동 측정에 관한 연구)

  • Digestive cells of the bivalves have a highly developed lysosomal system and the system is known to be sensitive to changes in environmental qualities. In this study, we measured lysosomal responses of the digestive cells in wild oyster, Crassostrea gigas using frozen section. Oysters were collected in June 2010 from intertidal areas in the inner and outer bay of Gwangyang off the south coast of Korea. From the tissue sections, we measured the digestive cell lysosomal membrane stability (LMS), level of neutral lipids (NL), lipofuscin (LF) and the digestive gland atrophy (DGA). The DGA and condition index of oysters from the inner bay were significantly lower (P<0.05). The statistical test indicated that LMS levels of oysters in the inner bay and the outer bay were not significantly different since a shorter activity was displayed by oysters from the inner bay than that of oysters in outer bay. The LF deposition level of the oysters in the inner bay displayed significantly higher levels than the outer bay (P<0.05). In contrast, the NL accumulation measured from oysters in outer bay was significantly higher than the level observed in the inner bay (P<0.05). Different levels of DGA and LF that were observed in the inner and outer bays were thought to be associated to different level of environmental contamination and these two assays are considered to be good biomarkers.

An Investigation of a Sensibility Evaluation Method Using Big Data in the Field of Design -Focusing on Hanbok Related Design Factors, Sensibility Responses, and Evaluation Terms- (디자인 분야에서 빅데이터를 활용한 감성평가방법 모색 -한복 연관 디자인 요소, 감성적 반응, 평가어휘를 중심으로-)

  • This study seeks a method to objectively evaluate sensibility based on Big Data in the field of design. In order to do so, this study examined the sensibility responses on design factors for the public through a network analysis of texts displayed in social media. 'Hanbok', a formal clothing that represents Korea, was selected as the subject for the research methodology. We then collected 47,677 keywords related to Hanbok from 12,000 posts on Naver blogs from January $1^{st}$ to December $31^{st}$ 2015 and that analyzed using social matrix (a Big Data analysis software) rather than using previous survey methods. We also derived 56 key-words related to design elements and sensibility responses of Hanbok. Centrality analysis and CONCOR analysis were conducted using Ucinet6. The visualization of the network text analysis allowed the categorization of the main design factors of Hanbok with evaluation terms that mean positive, negative, and neutral sensibility responses. We also derived key evaluation factors for Hanbok as fitting, rationality, trend, and uniqueness. The evaluation terms extracted based on natural language processing technologies of atypical data have validity as a scale for evaluation and are expected to be suitable for utilization in an index for sensibility evaluation that supplements the limits of previous surveys and statistical analysis methods. The network text analysis method used in this study provides new guidelines for the use of Big Data involving sensibility evaluation methods in the field of design.

A Study on Measuring the Change of the Response Results in Likert 5-Point Scale Measurement (리커트 5점척도에서 자극에 의한 응답결과의 변화 측정에 관한 연구)

  • This study examines how and which direction respondents who participated in 5-point Likert scale surveys change their initial responses when they are given an identical second survey after certain treatments. The research employs three identical questionnaires (first, second and third surveys) to analyze survey results based on group differences, kinds of treatment, survey purposes, and response change direction and the degree. This paper concludes that, first, it is significant that specialist groups do not change their initial responses compared to a general librarian group. Second, there are no differences by survey purpose; however, participants tend to change their initial responses by others' opinions rather than by previous use experiences. Third, participants who initially answered positively tend not to change their responses, and most participants who answered negatively change their initial responses in a positive direction. Fourth, when there are changes, participants change their initial responses by less than two points, and most of them change by one point. Finally, the hypothesis that middle responses change most and that participants who respond at both ends do not change their opinion was rejected by the finding that participants who answered on the negative end tend to change their initial responses in a positive direction.

Effect of Chemotaxis on Nodulation in Bradyrhizobium-Soybean Symbiosis (근류균의 화학주성이 근류형성에 미치는 영향)

  • To research the effect of chemotaxis of Rhizobia toward the root exudate on nitrogen fixing ability in soybean Rhizobia symbiosis system. Root exudate from seedlings of Glycine max. L was collected aseptic conditions. B. japonicum KCTC 2422 induced the formation of symbiotic nitrogen fixing nodules on the root of soybean plant and possessed motility and chemotaxis toward the 2mM proline. LPN-100 mutant was $Nod^-$, $Che^+$, and LPN-101 was $Che^-$, $Nod^+$ strains. Physiological properties of mutants were similar to parent strain. The crude root exudate was tested for its chemotactic ability using the capillary tube method. Chemotactic responses of RCR 3407 toward crude root exudate were 2.2, 2.6, 2.9, those of KCTC 2422 were 2.3, 2.9, 3.0, respectively. The crude root exudate was fractionated into neutral, cationic and anionic fractions. Chemotactic responses of KCTC 2422 was least with anionic fraction, most with neutral and intermediate with cationic fraction. B. japonicum KCTC 2422 was attracted by carbohydrates, amino acids and carboxylic acid. Carbohydrates and amino acids were good chemoattractants and carboxylic acids were intermediate chemoattractants. The peak concentration was $10^{-3}M$ for ribose, glucose, glutamine, aspartic acid and carboxylic acids, with exception of xylose, arabinose, tryptophan, which elicited maximum responses at $10^{-4}M$. The formation of nodules and nitrogenase activity of soybean inoculated with KCTC 2422 was determined in 7days after inoculation, and those of LPN-101 was detected in 15days after inoculation, but LPN-100 didn't form of nodules in soybean plants.

  • Early wound healing responses in the skin of the Korean fire bellied toad, Bombina orientalis, were examined using histochemical , scanning and transmission electron microscopical techniques. Cutaneous wounds were induced by sharp razor blade , and maintained in special cages for up to 3 days. By this treatment whole parts of epithelium and dermis were profoundly affected. After injury, retraction of damaged surface of the skin was observed. At 3 hours after injury, poly-band was form by mucous substance. The wound skin consisted of mucous gland secreted mucous substance which flows into wound cavity and the wound area was sealed by mucous substance which form thick layer at 6 hours. It appeared that natural disposition showed acid and neutral mucous substance by reaction of PAS and Alcian blue (pH 2.5) staining. After formation of the poly-band, epithelial cells were formed by migration of regenerating epithelial cells. These cells moved over the wound surface by migration at 12 hours after wounding. Poly-band was much increased in thickness and migrating cells were completely formed wound closure at 24 hours post-wounding. After 72 hours regenerated epithelial layer consists of four or five cell layers which are similar to clear, granular, and spinous layer.

  • The special interest should be paid to the analysis of the influences of positive emotions in terms of their possible effects on the dyanmics of autonomic. recovery after the negative affective stimualtion. Taking into account emotion-specific autonomic response patterning and dissociation of parameters of autonomic arousal during experience of both positive and negative emotional states, this problem seems a challenging one. In present study several autonomic parameters were analyzed altogether, namely inedices of electrodermal activity, heart rate and respitation rate during consecutive combination or both IAPS-based visual affective and auditory stimulation. The aim of the study was analysis of patterns of electrodermal and cardiorespiratory responses during emotional states evoked by negative affective visual stimulation followed by positive or neutral auditory one with intention to identify if the latter is able to facilitate post-stress recovery and enhance restoration of pre-arousal levels. The main orientation was dirdcted towards the further application of experimentally induced comfort emotions for dampening the negative consequences of exposure to stressful stimuli.

Effects of CSR Motives on Authenticity and Attitude in the Food and Beverage Franchise Sectors (식음료 프랜차이즈 기업의 CSR 활동 동기에 대한 지각이 진정성 및 태도에 미치는 영향)

  • Purpose: Previous studies show that perceived CSR motives have a significant impact on company evaluations. However, consumer responses to CSR motives vary depending on CSR motives. From this perspective, this study investigates the impact of CSR motives on consumers' responses in the context of food and beverage franchise companies using a scenario. Research design, data, and methodology: For achieving the purposes of the study, an example of a domestic food and beverage franchise company actively carrying out CSR activities was presented. Data was collected from 304 respondents aged 20 or older who were aware of CSR activities. The respondents answered the questionnaire after reading the scenario. The data was analyzed with SPSS 28.0 and SmartPLS 4.0 program. Result: Values-driven motive and strategic motive influence authenticity, while stakeholder-driven motive and egoistic motive did not influence authenticity. Values-driven motive influences on attitude, while stakeholder-driven motive, strategic motive and egoistic motive didn't. Lastly, authenticity influences attitude. Conclusions: Companies need to be aware that consumers may infer different motives for their CSR activities, and pay close attention to consumers' perceived motives from the planning stage of CSR activities. In particular, companies should focus on the values-driven motive and the strategic motive when planning CSR activities.

Effects of Taurine and ${\beta}-Alanine$ on the Responses of Dorsal Horn Cell to Various Stimuli in Cats (Taurine 및 ${\beta}-alanine$이 고양이 척수후각세포의 Activity에 미치는 효과)

  • In 19 cats anesthetized with ${\alpha}-chloralose$ effects of taurine and ${\beta}-alanine$ on the responses of wide dynamic range (WDR) cells to mechanical, chemical and thermal stimuli were investigated in the lumbar spinal cord of the cat. Also studied was an interaction of strychnine with taurine in affecting the activities of WDR cells. Following intravenous administration of taurine, the responses of WDR cells to all types of mechanical stimuli were markedly enhanced, demonstrating that the response to pressure was most sensitive to taurine action. When the receptive field was exposed to thermal stimuli ($50^{\circ}C$) for 20 sec. taurine increased activity of WDR cell to 169.5% of the control value. The $K^{+}$-induced activation of WDR cells was invariably suppressed after taurine administration. Intravenously administered strychnine remarkably reduced the enhanced response of WDR cell to natural stimuli resulting from intravenous administration of taurine. Also ${\beta}-alanine$ markedly activated the response of spinal dorsal horn cell to natural mechanical stimuli. These findings suggest that neutral amino acid and its derivative such as ${\beta}-alanine$ and taurine can enhance the response of WDR cells to different stimuli in cats.

Comparison of the Sensitivity of Human Bronchial Epithelial Cells to Cigarette Smoke-induced Inflammatory Responses (인간 유래 폐 세포주별 담배연기 분획의 염증 반응 민감도 비교)

  • The aim of this study is to compare the sensitivity of both two NCI-H292 and A549 cell types to acute inflammatory responses induced by cigarette smoke. For this, we treated two kinds of smoke fractions derived from 2R4F reference cigarettes: total particulate matter(TPM) collected onto a Cambridge filter pad and gas/vapor phase(GVP) prepared by bubbling through in buffer solution. When we measured cellular cytotoxicity by neutral red uptake assay after treatment for 24 hours, TPM and GVP induced cytotoxic effect in a dose-dependent manner in the range of 10-$100{\mu}g$/mL and 60-$300 {\mu}g$/mL., respectively, in both cell types without any cellular difference. Additionally, when we examined acute inflammatory responses by analyzing cytokines secreted into culture media including tumor necrosis factor-$\alpha$ (TNF-$\alpha$), interleukin-8(IL-8), and transforming growth factor-$\alpha$(TGF-$\alpha$) as well as matrix metalloproteinase-1(MMP-1), the treatment with smoke fractions increased those marker proteins in a dose-dependent manner in NCI-H292. Meanwhile, in A549 cells only MMP-1 was observed to be increased in a dose-dependent fashion. Collectively, our data indicate that NCI-H292 cell type is more sensitive to cigarette smoke-induced inflammatory response than A549 cells. This suggests that NCI-H292 could be useful as an in vitro evaluation tool to assess harmful effects of cigarette smoke.
