조팝나무(Spiraea prunifolia) 잎을 80% MeOH 수용액으로 추출한 뒤, 감압 농축한 추출물을 EtOAc, n-BuOH과 $H_2O$층으로 계통 분획을 실시하였다. n-BuOH분획에 대하여 $SiO_2$, ODS column chromatograph 및 중압분취 장비를 반복실시하여 2종의 phenolic glucoside화합물을 분리 및 정제하였다. NMR 및 Mass데이터를 해석하여, isosalicin (1)와 crenatin (2)로 구조 동정 하였다. 분리한 2종의 화합물에 대하여 UPLC-MS/MS 질량 분석기를 기반으로 다중반응검지 (MRM mode) 분석법을 이용하여 정량분석 하였다. 그 결과, crenatin은 9.53 mg/g 및 isosalicin은 0.65 mg/g이 조팝나무 잎 추출물에 함유된 것을 확인하였다. 조팝나무 잎에서 분리된 화합물이 $H_2O_2$에 의해 유발된 산화스트레스로부터 신경세포 보호효과를 검증한 결과, 신경모세포종인 SK-N-MC 세포에 대해서 화합물 1 및 2 (100 mM) 모두 세포 독성은 보이지 않았으며, 특히 화합물 2 (crenatin)는 $H_2O_2$처리에 따른 신경세포의 외형적 변화, 세포 손상, 세포 사멸을 억제하여 산화스트레스로부터 신경세포를 보호하는 효과가 있음을 확인하였다.
Background: The incidence of ischemic cerebrovascular disease is increasing in recent years and has been one of the leading causes of neurological dysfunction and death. Ginsenoside Rg1 has been found to protect against neuronal damage in many neurodegenerative diseases. However, the effect and mechanism by which Rg1 protects against cerebral ischemia-reperfusion injury (CIRI) are not fully understood. Here, we report the neuroprotective effects of Rg1 treatment on CIRI and its possible mechanisms in mice. Methods: A bilateral common carotid artery ligation was used to establish a chronic CIRI model in mice. HT22 cells were treated with Rg1 after OGD/R to study its effect on [Ca2+]i. The open-field test and poleclimbing experiment were used to detect behavioral injury. The laser speckle blood flowmeter was used to measure brain blood flow. The Nissl and H&E staining were used to examine the neuronal damage. The Western blotting was used to examine MAP2, PSD95, Tau, p-Tau, NOX2, PLC, p-PLC, CN, NFAT1, and NLRP1 expression. Calcium imaging was used to test the level of [Ca2+]i. Results: Rg1 treatment significantly improved cerebral blood flow, locomotion, and limb coordination, reduced ROS production, increased MAP2 and PSD95 expression, and decreased p-Tau, NOX2, p-PLC, CN, NFAT1, and NLRP1 expression. Calcium imaging results showed that Rg1 could inhibit calcium overload and resist the imbalance of calcium homeostasis after OGD/R in HT22 cells. Conclusion: Rg1 plays a neuroprotective role in attenuating CIRI by inhibiting oxidative stress, calcium overload, and neuroinflammation.
연구목적: 치수는 신경과 혈관을 포함하고 있는 부위로 다양한 자극이나 세균에 의해 염증이 생기면 이를 치수염이라고 한다. 치수염은 말초 신경조직 변화와 심한 통증을 유발하는 질환으로 만성적 통증을 유발하나, 삼차신경절의 신경세포 활성화와 특정 나트륨 채널(Nav1.7) 발현 사이의 관계는 잘 알려져 있지 않다. 이에 본 연구에서는 실험적으로 유도된 치수염이 말초신경에서 Nav1.7 나트륨 채널의 발현을 활성화시켜, 삼차신경절의 뉴런을 활성화함으로써 통증을 유발한다는 사실을 발견하고 이를 조절하는 신호기전을 규명하고자 하였다. 연구방법: 실험동물(Male C57BL/6 생쥐, 6주, 20-25 g)의 상악 제1대구치 치수에 AITC를 처리하여 급성 치수염을 유발하고, 3일 후 실시간 광영상 이미지를 이용하여 삼차신경절의 뉴런 활성화를 측정 및 비교 분석하였다. 단백질 분석을 통해 뇌간(SpVc)에서 치수염 통증유발 신호조절기전에 관여하는 여러가지 단백질들(p-ERK, c-FOS, TRPA1, p-CRMP2)의 발현을 관찰하였다. 연구결과: 시공간적 광영상 이미지를 통해 삼차신경절의 뉴런세포들은 대조군과 비교 시 급성 치수염 모델에서 흥분성 활성화가 유도되어 신경학적 변화가 일어남을 관찰하였다. 또한 조직학 및 분자생물학적 결과를 통해 치수염으로 인한 특정 나트륨 채널(Nav1.7)의 증가가 통증을 유발한다는 사실을 확인하였다. 또한, ProTxII(Nav1.7의 선택적 억제 약물) 처리를 통해 뉴런의 과흥분성 활성이 억제됨에 따라, 치수염이 삼차신경절에서 나트륨 채널(Nav1.7)의 증가를 유도하고 이러한 특정 나트륨 채널을 효과적으로 제어하면 치수염의 통증을 줄이는 방법이 될 것이라 생각된다. 결론: 본 연구의 결과를 통해 과 발현된 특정 나트륨 채널(Nav1.7)을 억제하면 삼차신경절에서 통각 신호 처리를 조절하고 치수염에 의해 유발되는 통증을 효과적으로 조절할 수 있음을 시사한다.
알츠하이머 질환(Alzheimer's disease)은 대표적인 신경퇴행성 질환이며, 뇌 내 amyloid beta (Aβ) 축적에 의한 산화적 스트레스 및 염증반응은 대표적인 AD의 원인으로 알려져 있다. 본 연구에서는 kaempferol (K), kaempferol-3-O-glucoside (KG), quercetin (Q) 및 quercetin-3-β-ᴅ-glucoside (QG)와 같은 4가지 flavonoids의 C6 신경교세포에서 Aβ로 인한 신경독성으로부터의 보호 효능을 살펴보고자 하였다. C6 신경교세포에 Aβ를 처리하였을 때 세포 생존율이 감소한 반면, 4가지 flavonoids 중에서 Q와 QG의 처리 시 세포 생존율 증가를 통해 신경교세포 보호 효과를 확인하였다. 또한, Aβ를 처리한 control군의 경우 reactive oxygen species (ROS) 생성을 유도한 반면, flavonoids의 처리 시 ROS 생성이 감소하였다. 특히 Aβ를 처리한 control군은 133.39%의 ROS 생성을 나타내었으며, 1 μM의 KG와 QG를 각각 처리시 107.44, 113.10%의 수치를 나타내어 ROS 생성 감소를 확인하였다. Flavonoids의 Aβ에 대한 신경교세포 보호 기전을 확인하기 위해 염증 관련 단백질 발현을 측정하였다. Aβ로 신경독성이 유도된 control군은 염증 관련 단백질 발현이 증가하였다. 그러나, flavonoids를 처리한 군의 경우 염증 매개 인자인 inducible nitric oxide synthase, cyclooxygenase-2 and interleukin-1β의 발현 감소를 확인하였다. 특히, KG와 QG를 처리한 군은 aglycone 형태인 K와 Q를 처리한 군에 비해 효과적으로 염증 매개 인자 발현을 감소시켰다. 본 연구는 flavonoids의 일종인 K, Q와 그 배당체인 KG, QG의 Aβ로 신경독성이 유도된 신경교세포에서 산화적 스트레스 및 염증반응 조절을 통한 보호 효과를 나타냄을 알 수 있었으며, 이들 생리활성성분은 AD 예방 및 치료 소재로써의 가능성이 있을 것으로 사료된다.
Main strategy for a treatment of Parkinson's disease (PD), due to a progressive degeneration of dopaminergic neurons, is a pharmaceutical supplement of dopamine derivatives or ceil replacement therapy. Both of these protocols have pros and cons; former exhibiting a dramatic relief but causing a severe side effects on long-term prescription and latter also having a proven effectiveness but having availability and ethical problems Embryonic stem (ES) cells have several characteristics suitable for this purpose. To investigate a possibility of using ES cells as a carrier of therapeutic gene(s), human ES (hES, MB03) cells were transfected with cDNAs coding for tyrosine hydroxylase (TH) in pcDNA3.1 (+) and the transfectants were selected using neomycin (250 $\mu /ml$). Expression of TH being confirmed, two of the positive clone (MBTH2 & 8) were second transfected with GTP cyclohydrolase 1 (GTPCH 1) in pcDNA3.1 (+)-hyg followed by selection with hygromycin-B (150 $\mu /ml$) and RT-PCR confirmation. By immune-cytochemistry, these genetically modified but undifferentiated dual drug-resistant cells were found to express few of the neuronal markers, such as NF200, $\beta$-tubulin, and MAP2 as well as astroglial marker GFAP. This results suggest that over-production of BH4 by ectopically expressed GTPCH I may be involved in the induction of those markers. Transplantation of the cells into striatum of 6-OHDA- denervated PD animal model relieved symptomatic rotational behaviors of the animals. Immunohistochemical analyses showed the presence of human cells within the striatum of the recipients. These results suggest a possibility of using hES cells as a carrier of therapeutic gene(s).

Alzheimer's disease (AD) is a neurodegenerative disorder associated with progressive memory loss and neuronal cell death. Although numerous previous studies have been focused on disease progression or reverse pathological symptoms, therapeutic strategies for AD are limited. Alternatively, the identification of traditional herbal medicines or their active compounds has received much attention. The aims of the present study were to characterize the ameliorating effects of spinosin, a C-glucosylflavone isolated from Zizyphus jujuba var. spinosa, on memory impairment or the pathological changes induced through amyloid-${\beta}_{1-42}$ oligomer ($A{\beta}O$) in mice. Memory impairment was induced by intracerebroventricular injection of $A{\beta}O$ ($50{\mu}M$) and spinosin (5, 10, and 20 mg/kg) was administered for 7 days. In the behavioral tasks, the subchronic administration of spinosin (20 mg/kg, p.o.) significantly ameliorated $A{\beta}O$-induced cognitive impairment in the passive avoidance task or the Y-maze task. To identify the effects of spinosin on the pathological changes induced through $A{\beta}O$, immunohistochemistry and Western blot analyses were performed. Spinosin treatment also reduced the number of activated microglia and astrocytes observed after $A{\beta}O$ injection. In addition, spinosin rescued the $A{\beta}O$-induced decrease in choline acetyltransferase expression levels. These results suggest that spinosin ameliorated memory impairment induced through $A{\beta}O$, and these effects were regulated, in part, through neuroprotective activity via the anti-inflammatory effects of spinosin. Therefore, spinosin might be a useful agent against the amyloid ${\beta}$ protein-induced cognitive dysfunction observed in AD patients.
조직을 이용한 역전사 (RT)-PCR법을 이용하면 원하는 특정유전자의 발현을 비교적 정확하게 알 수 있지만 조직의 RNA를 이용하므로 세포단위의 정확한 유전자 발현을 알기에는 한계가 있다. 특히 그 기능과 성질이 다른 세포가 무수하게 많이 혼재하는 두뇌와 같은 조직은 신경계의 각종 뉴런(신경세포), 글리어 (glial cell) 등이 서로 얽혀 있다. 대표적인 신경세포의 degeneration 질병으로는 파킨슨병 (Parkinson's disease; PD)이 있다. 파킨슨병은 사람의 신경세포 관련 질병에 있어서 가장 일반적인 질병의 하나이다. PD의 가장 중요한 원인은 도파민 생성 신경세포의 퇴행 혹은 사멸에 기인하여 도파민 (dopamine)이라는 신경전달물질이 감소하는 것이 그 원인이다. 도파민과 같은 카테콜아민의 생합성에 관련된 효소는 타이로신 하이드록실레이스 (TH), 도파 데카르복실레이스 (DDC) 등이 알려져 있다. 그러나 그런 효소들의 생화학적 연구는 많이 되어 있음에도 불구하고 단일 흑질 신경세포에서의 이들 관련 유전자의 발현 양상에 대해서는 알려진 바가 거의 없다. PD와 관련된 유전자의 발현 정도를 밝히기 위하여, 레이저 다이섹터 (laser micro-dissector)에 의한 단일 신경세포의 분리에 착수하였다. 정해진 방법에 따라 정상 대조구 (비PD)와 PD 환자에서 각각 한 개 또는 여러 개를 성공적으로 분리한 흑질 신경세포를 이용하여 유전자 특이적 프라이머를 사용하여 RT-PCR을 행하였다. 그 결과, 단 한 개의 신경세포에서도 여러 개의 세포를 사용한 것과 같은 동일한 결과를 얻는 데 성공하였다. PD환자의 뇌에서 분리한 10개의 독립적인 세포의 예에서는 각 세포간의 발현차이가 인정되었으며, 특히 TH 유전자의 발현은 상당히 높은 확률로 검출되지 않았다. 이 결과로 단일 신경세포에서의 mRNA양을 검출하기 위해서는 본 본문의 RT-PCR법이 효과적인 방법임을 알 수 있다.

Heme oxygenase-1 (HO-1) degrades heme to carbon dioxide, biliverdin, and $Fe^{2+}$, which play important roles in various biochemical processes. In this study, we examined the protective function of HO-1 against oxidative stress in SH-SY5Y cells and in a Parkinson's disease mouse model. Western blot and fluorescence microscopy analysis demonstrated that PEP-1-HO-1, fused with a PEP-1 peptide can cross the cellular membranes of human neuroblastoma SH-SY5Y cells. In addition, the transduced PEP-1-HO-1 inhibited generation of reactive oxygen species (ROS) and cell death caused by 1-methyl-4-phenylpyridinium ion ($MPP^+$). In contrast, HO-1, which has no ability to transduce into SH-SY5Y cells, failed to reduce $MPP^+$-induced cellular toxicity and ROS production. Furthermore, intraperitoneal injected PEP-1-HO-1 crossed the blood-brain barrier in mouse brains. In a PD mouse model, PEP-1-HO-1 significantly protected against 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced toxicity and dopaminergic neuronal death. Therefore, PEP-1-HO-1 could be a useful agent in treating oxidative stress induced ailments including PD.

Parkinson's disease (PD) is a neurodegenerative disease characterized by progressive degeneration of dopaminergic (DAergic) neurons, particularly in the substantia nigra (SN). Although circadian dysfunction has been suggested as one of the pathophysiological risk factors for PD, the exact molecular link between the circadian clock and PD remains largely unclear. We have recently demonstrated that $REV-ERB{\alpha}$, a circadian nuclear receptor, serves as a key molecular link between the circadian and DAergic systems. It competitively cooperates with NURR1, another nuclear receptor required for the optimal development and function of DA neurons, to control DAergic gene transcription. Considering our previous findings, we hypothesize that $REV-ERB{\alpha}$ may have a role in the onset and/or progression of PD. In the present study, we therefore aimed to elucidate whether genetic abrogation of $REV-ERB{\alpha}$ affects PD-related phenotypes in a mouse model of PD produced by a unilateral injection of 6-hydroxydopamine (6-OHDA) into the dorsal striatum. $REV-ERB{\alpha}$ deficiency significantly exacerbated 6-OHDA-induced motor deficits as well as DAergic neuronal loss in the vertebral midbrain including the SN and the ventral tegmental area. The exacerbated DAergic degeneration likely involves neuroinflammation-mediated neurotoxicity. The $REV-erb{\alpha}$ knockout mice showed prolonged microglial activation in the SN along with the over-production of interleukin $1{\beta}$, a pro-inflammatory cytokine, in response to 6-OHDA. In conclusion, the present study demonstrates for the first time that genetic abrogation of $REV-ERB{\alpha}$ can increase vulnerability of DAergic neurons to neurotoxic insults, such as 6-OHDA, thereby implying that its normal function may be beneficial for maintaining DAergic neuron populations during PD progression.
Objectives : The aim of this study was to investigate the protective effect of extract of Thuja orientalis leaves (TOFE) against 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced neurotoxicity by inhibition of inflammation in in vitro and in vivo models of Parkinson's disease (PD). Methods : We evaluated the effect of TOFE against lipopolysaccharide (LPS)/1-methyl-4-phenylpyridinium ($MPP^+$) toxicity using nitric oxide (NO) assay, inducible NO synthase and cyclooxygenase 2 western blot, tyrosine hydroxylase and microglia activation immunohistochemistry (IHC) in BV2 cell, primary rat mesencephalic neurons, or C57BL/6 mice. We also evaluated the effect of TOFE in mice PD model induced by MPTP. C57BL/6 mice were treated with TOFE 50 mg/kg for 5 days and were injected intraperitoneally with four administrations of MPTP on the last day. We conducted behavioral tests and IHC analysis to see how TOFE affect MPTP-induced neuronal loss of dopaminergic neurons in substantia nigra pars compacta (SNpc) and striatum (ST) of mice. To assess the anti-inflammation effects, we carried out glial fibrillary acidic protein and macrophage-1 antigen integrin alpha M in IHC in SNpc and ST of mice. Results : In an in vitro system, TOFE decreasesd NO generations in BV2 cells. TOFE protected dopaminergic cells against LPS or $MPP^+$-induced toxicity in primary mesencephalic dopaminergic neurons. In vivo system, TOFE at 50 mg/kg treated group showed improved motor deteriorations than the MPTP only treated group and TOFE significantly protected striatal dopaminergic damage from MPTP-induced neurotoxicity in mice. Moreover, TOFE inhibited activation of astrocyte and microglia in SNpc and ST of the mice. Conclusions : We concluded that TOFE showed anti-parkinsonian effect by protection of dopaminergic neurons against MPTP toxicity through anti-inflammatory actions.

서비스를 일시 중단할 수 있으며, 예정되어 있는 작업으로 인한 서비스 일시중단은 당 사이트 홈페이지를
통해 사전에 공지합니다.
② 당 사이트는 서비스를 특정범위로 분할하여 각 범위별로 이용가능시간을 별도로 지정할 수 있습니다. 다만

제 13 조 (홈페이지 저작권)
① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)

제 16 조 (서비스 이용제한)

② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
