Reconstruction of basement membrane in the artificial skin (인공피부 제조시 기저막 재건의 효과)

  • We attempted to reconstruct basement membrane (BM) in between the epidermal compartment and dermal compartment in the artificial skin preparation and examine its effect on the skin architecture as well as on the epidermal differentiation. Laminin, one of the component of BM, stimulate the migration of the basal cells but type IV collagen which is a major component of the mechanical network of BM did not stimulate epidermal migration. However laminin in the presence of type IV collagen at a 1:1 molar ratio did not stimulate epidermal migration but provide nice demarcation between epidermis and dermis. This mixture of laminin and type IV collagen enhanced epidermal differentiation in the artificial skin based on the morphological observation as well as biochemical criteria. The epidermal acquirement of migratory ability on the laminin-rich substrate suggest that this type of unbalance in the expression of the components of BM may prevail in the area of healing tissue and the invasive transition of the tumor. The result in this study provide the technical improvement in the artificial skin preparation and further application of this technique for the reconstruction of other bio-artificial organ.

Improvement of Methanogenic activity in the Anaerobic Treatment of Wastewater with High Sulfate (고농도 황산염을 함유한 폐수의 혐기성 처리시 메탄균의 효율 향상을 위한 연구)

  • UASB reactor was operated for treating wastewater containing high sulfate to assess their performance, competition between sulfate-reducing bacteria(SRB) and methane-producing bacteria(MPB), and the change in the characteristics of microbial granules according to change of hydraulic retention time(HRT) in the reactor. The reactor was fed with a synthetic moderate strength wastes(glucose, 2000 mgCOD/l) containing high sulfate($2400mgSO_4{^{2-}}/l$). The organic loading rate(OLR) ranged from 1.5 to 3.0 gCOD/l.d as HRT maintained 15 to 30 hrs in the stage I. The COD removal efficiency was between 80 to 92%. During this period, methane yield rapidly decreased from 0.3 to 0.1 1 $CH_4$/gCODremoved. While sulfide concentration in the effluent increased from 80 to 200 mgS/l. This indicates that SRB becomes dominant over MPB at a relatively long HRT in the excess sulfate. When OLR of reactor maintained from 5 to 8 gCOD/l.d in the stage II, methane yield increased from 0.1 to 0.17 1 $CH_4$/gCODremoved regardless of decrease of COD removal efficiency. This indicates that SRB is more sensitive to the change of a short HRT than MPB. In the competition between SRB and MPB, about 30% of the removed COD was utilized by SRB at HRT of 30 hrs during the start-up period, while about 73% was used by SRB at HRT of 15hrs at the final step of second experimental stage. Whereas after shock exposure of OLR about 62% was utilized by SRB at HRT of 5hrs. It indicates that SRB is strongly suppressed by the wash-out of significant dispersed SRB since a large electron flow is distributed to the MPB. In addition, the granulation in the presence of high sulfate is unfavoured at a long HRT because of substrate transport limitations into MPB like Methanothrix spp. which is an important factor in the composition of the granules. Accordingly, granule sizes in the UASB reactor decreased with time due to weak network frame of granules by the decreased activity of MPB.

  • Recently, pattern recognition methods have been widely used by researchers for fault diagnoses of mechanical systems. A pattern recognition method determines the soundness of a mechanical system by detecting variations in the system's vibration characteristics. Hidden Markov models (HMMs) and artificial neural networks (ANNs) have recently been used as pattern recognition methods in various fields. In this study, a HMM-ANN hybrid method for the fault diagnosis of a mechanical system is introduced, and a rotating wind turbine blade with a crack is selected for fault diagnosis. The existence, location, and depth of said crack are identified in this research. For improving the diagnostic accuracy of the method in spite of the presence of noise, a moment with a few specific frequencies is applied to the structure.

Estrogen modulates serotonin effects on vasoconstriction through Src inhibition

  • Estrogen has diverse effects on cardiovascular function, including regulation of the contractile response to vasoactive substances such as serotonin. The serotonin system recently emerged as an important player in the regulation of vascular tone in humans. However, hyperreactivity to serotonin appears to be a critical factor for the pathophysiology of hypertension. In this study, we examined the modulatory mechanisms of estrogen in serotonin-induced vasoconstriction by using a combinatory approach of isometric tension measurements, molecular biology, and patch-clamp techniques. $17{\beta}$-Estradiol (E2) elicited a significant and concentration-dependent relaxation of serotonin-induced contraction in deendothelialized aortic strips isolated from male rats. E2 triggered a relaxation of serotonin-induced contraction even in the presence of tamoxifen, an estrogen receptor antagonist, suggesting that E2-induced changes are not mediated by estrogen receptor. Patch-clamp studies in rat arterial myocytes showed that E2 prevented Kv channel inhibition induced by serotonin. Serotonin increased Src activation in arterial smooth muscle required for contraction, which was significantly inhibited by E2. The estrogen receptor-independent inhibition of Src by E2 was confirmed in HEK293T cells that do not express estrogen receptor. Taken together, these results suggest that estrogen exerts vasodilatory effects on serotonin-precontracted arteries via Src, implying a critical role for estrogen in the prevention of vascular hyperreactivity to serotonin.

  • In this paper, a broadband power line communication modem and network technology to develop tests with actual characteristics of the underground power line communication channel to be interpreted. Length and structure of the underground distribution line due to underground distribution line to the broadband powerline communication channels to measure the amount of attenuation on the signal generator, spectrum analyzer, such as contactless coupler was used to configure the channel measurement system. Demonstration tests in the test line as a single line by selecting the longest high-voltage lines where the two-channel measurement and signal attenuation on the noise level was measured. Measuring results degrade communication performance can be quite sure that was the presence of impulse noise, long lines, the line length was confirmed by the high-frequency attenuation appears.

Utilizing Purely Symmetric J Measure for Association Rules (연관성 규칙의 탐색을 위한 순수 대칭적 J 측도의 활용)

  • In the field of data mining technique, there are various methods such as association rules, cluster analysis, decision tree, neural network. Among them, association rules are defined by using various association evaluation criteria such as support, confidence, and lift. Agrawal et al. (1993) first proposed this association rule, and since then research has been conducted by many scholars. Recently, studies related to crossover entropy have been published (Park, 2016b). In this paper, we proposed a purely symmetric J measure considering directionality and purity in the previously published J measure, and examined its usefulness by using examples. As a result, it is found that the pure symmetric J measure changes more clearly than the conventional J measure, the symmetric J measure, and the pure crossover entropy measure as the frequency of coincidence increases. The variation of the pure symmetric J measure was also larger depending on the magnitude of the inconsistency, and the presence or absence of the association was more clearly understood.

  • Limitation of the TCP/IP network technology included in the vehicle communication is due to the frequent mobility of the vehicle, the increase in intermittent connection requirements, and the constant presence of the possibility of vehicle hacking. VNDN technology enables the transfer of the name you are looking for using textual information without the need for vehicle identifiers like IP/ID. In addition, intermittent connectivity communication is possible rather than end-to-end connection communication. The data itself is the subject of communication based on name-based forwarding using two types of packets: Interest packet and Data packet. One of the issues to be solved for the realization of infotainment services under the VNDN environment is the traffic explosion caused by data broadcasting. In this paper, we analyze and compare the existing technologies to reduce the data broadcast storm. Through this, we derive and analyze the requirements for presenting the best data mitigation technique for solving the data explosion phenomenon in the VNDN environment. We expect this paper can be utilized as prior knowledge in researching improved forwarding techniques to resolve the data broadcast explosion in connected vehicles over NDN.

  • In this paper, inductive couplers realizing broadband powerline communication (PLC) are fabricated using Fe-based nanocrystalline alloy and their performance is analyzed. As a result of the field tests using the distribution automation system (DAS), the couplers achieve comparatively excellent data communication in the principal frequency band of broadband PLC although there is a difference in communication rate depending on the presence or absence of a branch. In addition, it has been confirmed that the communication speed is maintained for a real-time transmission even in a high current environment although there is a difference in the transmission rate depending on the distance. Hence, it is considered that the inductive couplers can be used as a core device to realize the intelligent power network by exploiting them for the monitoring and remote controlling of the power plant equipments for the DAS located in the inaccessible areas.

Menin Enhances Androgen Receptor-Independent Proliferation and Migration of Prostate Cancer Cells

  • The androgen receptor (AR) is an important therapeutic target for treating prostate cancer (PCa). Moreover, there is an increasing need for understanding the AR-independent progression of tumor cells such as neuroendocrine prostate cancer (NEPC). Menin, which is encoded by multiple endocrine neoplasia type 1 (MEN1), serves as a direct link between AR and the mixed-lineage leukemia (MLL) complex in PCa development by activating AR target genes through histone H3 lysine 4 methylation. Although menin is a critical component of AR signaling, its tumorigenic role in AR-independent PCa cells remains unknown. Here, we compared the role of menin in AR-positive and AR-negative PCa cells via RNAi-mediated or pharmacological inhibition of menin. We demonstrated that menin was involved in tumor cell growth and metastasis in PCa cells with low or deficient levels of AR. The inhibition of menin significantly diminished the growth of PCa cells and induced apoptosis, regardless of the presence of AR. Additionally, transcriptome analysis showed that the expression of many metastasis-associated genes was perturbed by menin inhibition in AR-negative DU145 cells. Furthermore, wound-healing assay results showed that menin promoted cell migration in AR-independent cellular contexts. Overall, these findings suggest a critical function of menin in tumorigenesis and provide a rationale for drug development against menin toward targeting high-risk metastatic PCa, especially those independent of AR.

Machine Learning-based Process Condition Selection Method to Prevent Defects in Korean Traditional Brass Casting (한국 전통 유기 제작에서 결함을 방지하기 위한 기계 학습 기반의 공정 조건 선택 방안)

  • In the present study, in order to prevent the misrun defects that occur during traditional brass casting, a method for selecting the proper casting process conditions is proposed. A learning model was developed and demonstrated to be able to learn the presence or absence of defects according to the casting process conditions and to predict the occurrence of defects depending on the certain process given. Appropriate process conditions were determined by applying the proposed method, and the determined conditions were verified through a comparison of different simulation results with additional conditions. With this method, it is possible to determine the casting process conditions that will prevent defects in the desired sand model. This technology is expected to contribute to realization of smart traditional brass farming workshops.
