Genetic Variation in the Selected Populations of Hovenia dulcis var. koreana Nakai. Based on RAPD Analysis

  • This study used RAPD markers to assume genetic diversity and variation in selected populations of Hovenia dulcis var. koreana. Ratio of polymorphic RAPD markers were 93.4% in selected populations of Hovenia dulcis Thunb., difference of genetic structure among populations and within populations showed 16.45%, 83.55%, respectively in amount of total genetic variation of 4 populations. Total gene diversity($H_T$) that show genetic diversity appeared 0.313 and coefficient of gene differentiation($G_{ST}$) that compare genetic differentiation of populations appeared 0.1645, analysis of AMOVA for variation among populations and within populations was significantly different (P<0.001). Genetic diversity of whole populations showed that 12.44% difference among population and 87.56% difference within populations. As a result, difference within populations was larger than difference among populations in genetic diversity. Nei's genetic distance and cluster analysis appeared that mean genetic distance among populations was 0.076, thus dividing two main groups and geographic relationship did not show in populations.

한국 해역에 분포하는 오징어의 집단유전학적 방법에 의한 계군분석 (A Population Genetic Analysis of the Common Squid, Todarodes pacificus Steenstrup in the Korean Waters)

  • 본 연구는 1994년 6월부터 1995년 5월까지 우리 나라 해역에 분포하는 오징어, Todarodes pacificus에 대해 유전자 조성을 관찰하고 군간 비교로 계군을 분석한 것이다. 각기 채집 장소가 다른 9개 지역 집단에 있어서 17개 효소를 사용하여 전기영동법으로 유전적 변이와 분화를 분석하였다. 명확하게 유전자형이 추정된 9개 효소에 대해, 11개의 유전자좌를 표지로 하여, 각기 채집 장소가 다른 9개 지역 집단의 대립유전자빈도와 유전도 변이성의 범위를 조사하였다. 그 결과, 1유전자좌당 평균 대립유전자수는 $1.64\~2.18$개 이었다. 집단의 변이량을 나타내는 다형율 $(P+P^{\ast})$$0.273\~0.546$이었다. 평균 이형접합체율 $(H_e)$$0.038\~0.085$이었다. 그리고 Nei의 유전적 거리는 $0.00019\~0.000814$로 넓음을 알 수 있었다. 각기 채집 장소가 다른 9개 지역집단간의 유전적 유연관계를 알아 본 결과, 세 집단으로 구분되었다. 이상의 결과를 종합하여 보면, 하계군, 추계군 및 동계군에는 각각의 계군으로 구명되었고, 하계군과 추계군에는 해류나 지리적인 해양환경에 의해 독립된 하나의 지역개체군이 존재하고 있음을 알 수 있었다.

Evaluation of Genetic Variation and Phylogenetic Relationship among North Indian Cattle Breeds

  • In the present study, genetic analyses of diversity and differentiation were performed on four breeds of Indian zebu cattle (Bos indicus). In total, 181 animals belonging to Ponwar, Kherigarh, Gangatiri and Kenkatha breeds were genotyped for 20 cattle specific microsatellite markers. Mean number of alleles observed per locus (MNA) varied between 5.75 (Kenkatha) to 6.05 (Kherigarh). The observed and expected heterozygosity for the breeds varied from 0.48 (Gangatiri) to 0.58 (Kherigarh) and 0.65 (Kenkatha) to 0.70 (Kherigarh), respectively. $F_{IS}$ estimates of all the breeds indicated significant deficit of heterozygotes being 28.8%, 25.9%, 17.7% and 17.7% for Gangatiri, Ponwar, Kherigarh and Kenkatha, respectively. The $F_{ST}$ estimates demonstrated that 10.6% was the average genetic differentiation among the breeds. Nei's genetic distance DA and Cavalli- Sforza and Edwards Chord distance ($D_C$) and the phylogenetic tree constructed from these reflected the close genetic relationship of Gangatiri and Kenkatha, whereas Ponwar appears to be more distant.

  • Randomly Amplified Polymorphic DNA (RAPD) was performed to define the genetic variation and relationships of Artemisia capillaris. Fifteen populations by the distributions and habitat were collected to conduct RAPD analysis. RAPD markers were observed mainly between 300bp and 1600bp. Total 72 scorable markers from 7 primers were applied to generate the genetic matrix, and 69 bands were polymorphic and only 3 bands were monomorphic. The genetic dissimilarity matrix by Nei's genetic distance (1972) and UPGMA phenogram were produced from the data matrix. Populations of Artemisia capillaris were clustered with high genetic affinities and cluster patterns were correlated with distributional patterns. Two big groups were clustered as southern area group and middle area group. The closest OTUs were GW2 and GG1 in middle area group, and GB1 from southern area group was clustered with OTUs in middle area group. RAPD data was useful to define the genetic variations and relationships of A. capillaris.

Genetic Diversity and Population Genetic Structure of Black-spotted Pond Frog (Pelophylax nigromaculatus) Distributed in South Korean River Basins

  • The objective of this study was to analyze the genotype of black-spotted pond frog (Pelophylax nigromaculatus) using seven microsatellite loci to quantify its genetic diversity and population structure throughout the spatial scale of basins of Han, Geum, Yeongsan, and Nakdong Rivers in South Korea. Genetic diversities in these four areas were compared using diversity index and inbreeding coefficient obtained from the number and frequency of alleles as well as heterozygosity. Additionally, the population structure was confirmed with population differentiation, Nei's genetic distance, multivariate analysis, and Bayesian clustering analysis. Interestingly, a negative genetic diversity pattern was observed in the Han River basin, indicating possible recent habitat disturbances or population declines. In contrast, a positive genetic diversity pattern was found for the population in the Nakdong River basin that had remained the most stable. Results of population structure suggested that populations of black-spotted pond frogs distributed in these four river basins were genetically independent. In particular, the population of the Nakdong River basin had the greatest genetic distance, indicating that it might have originated from an independent population. These results support the use of genetics in addition to designations strictly based on geographic stream areas to define the spatial scale of populations for management and conservation practices.

Genetic Relationships and Phylogeny of the Asplenium antiquum Makino (Aspleniaceae) and its relative species based on RAPD Analysis

  • This study characterized the genetic variations of 13 populations of Asplenium antiquum and its relative species using randomly amplified polymorphic DNA (RAPD) markers. A total 88 scorable RAPD bands were generated by the 12 random oligo primers and were analyzed by Nei and Li's genetic distance. High genetic variability was detected between A. antiquum and A. nidus, with the range from 0.568 to 0.682. And slightly low genetic variations showed within the populations of same species. Seven populations of A. antiquum showed slight differences (0.000-0.216), and five populations of A. nidus showed similar low genetic variations (0.114 to 0.171). Two individuals from Sup-seom Island which are growing in might be the regenerated one from abroad. A. antiquum were clustered as two groups (Group I, Group II) by UPGMA phenogram. And five populations of A. nidus were clustered as two groups correlated with geographical distribution. The RAPD data was very useful to define the genetic variations and to discuss the phylogenetic relationships among A. antiquum and the related species..

I-SSR PCR을 이용한 한국의 11개 주요 산지에서 채집한 송이의 유전변이에 관한 연구 (A Study on the Genetic Variations of Tricholoma matsutake Collected from Eleven Sites of Korea Using I-SSR PCR)

  • 본 연구는 국내 주요 송이 산지에서 생산되는 송이{Tricholoma matsutake(S. Ito et Imai) Sing}의 개체간, 그리고 지역간 유전적 다양성을 구명하기 위하여 실시하였다. 경상북도의 봉화, 울진, 고령, 청도의 4개 지역, 경상남도의 창녕, 하동, 함양의 3개 지역, 강원도의 양양, 인제의 2개 지역, 충청북도의 괴산, 전라북도의 남원을 포함하여 총 11개의 송이 산지를 대상으로 1994년부터 1997년까지 산지별로 $3{\sim}8$개의 균환에서, 그리고 각 균환마다 2개의 자실체를 채집하여 6개의 primer를 이용하여 Inter Simple Sequence Repeat Polymerase Chain Reaction(I-SSR PCR)법을 이용하여 자실체의 유전적 특성을 조사하였다. 그 결과 총 131개의 DNA band가 재현성 있게 나타났다. 또한 산지간의 유연관계를 살펴보기 위하여 Nei의 genetic distance를 이용하여 UPGMA tree를 작성하였다. I-SSR PCR법을 이용하여 DNA를 분석한 결과, 산지간의 유전적 변이는 12.9%에 불과한 반면 산지 내 개체간 변이가 87.1%로 나타났다. 유집 분석 결과는 11개 송이 산지를 제 I 그룹(양양, 함양, 인제, 하동, 울진), 제 II 그룹(남원, 창녕, 청도), 제 III 그룹(고령), 제 IV 그룹(봉화, 괴산)의 4개 그룹으로 분류하였다. 송이의 유전적 다양성, 집단간의 유전적 차이를 나타내는 수치인 Fst 값이 0.13이므로 집단간의 유전적 변이를 어느 정도 나타낸다고 결론짓는다.

동위효소와 체액단백질 분석에 의한 한국산 멧누에나방의 지역적 특성 (Phylogeny of Bombyx mandarina inhabiting Korea analysing the isozyme and hemolymph protein polymorphism)

  • 한반도에 서식하는 멧누에나방의 서식지역에 따른 유전적 변이와 집누에나방과의 종간 유연관계를 조사하였다. 멧누에나방의 isozyme 및 체액단백질을 대상으로 유전자 빈도를 조사한 결과, 조사지역 중 경북 칠곡과 경남 진주집단 사이의 유전적 유사도가 가장 높았으며, 경남 진주와 강원 고성집단 사이에서 가장 낮게 나타났다. 그러나 서식집단 상호간의 유사도는 매우 높았다. 한국산 멧누에나방과 집누에나방의 isozyme 유전자형은 대부분이 동일하게 발현되었으나, 집누에나방의 한국종에서만 발현된다고 알려진 Bes의 B형, Amy-hc의 F형, Ict-E의 n형 및 Ict-h의 M형과 S형 등의 유전자형이 한반도에 서식하는 멧누에나방에서 발현되었다. 따라서, 한반도 내에서 멧누에나방의 서식지역간 유전적 변이는 인정되지 않으며, 지금까지는 고려되지 않았던 집누에 한국종과 한국산 멧누에나방의 종간 유연관계에 대해 새로운 의문이 야기된다.

Genetic Variability in the Natural Populations of Daba Ecorace of Tasar Silkworm (Antheraea mylitta Drury), as Revealed by ISSR Markers

  • Genetic diversity within the natural populations of Daba ecorace of Antheraea mylitta Drury was studied using individual silkworms collected from the South Singhbhum district of Jharkhand state of India with 21 inter simple sequence repeat (ISSR) primers. A total of 148 bands were produced, of which 79% was polymorphic. The pair wise genetic distance among the individuals varied from 0.186 to 0.329. The dendrogram grouped the individuals into 3 major clusters. Nei's heterozygosity analysis revealed 0.265 ${\times}$ 0.18 variability within the population. The high genetic variability present within the natural population of Daba ecorace of A. mylitta is indicative of their adaptational strategy in nature and have much importance for in situ conservation as well as utilization in breeding programs.

Genetic Relationships among Australian and Mongolian Fleece-bearing Goats

  • Microsatellites (MS) are useful for quantifying genetic variation within and between populations and for describing the evolutionary relationships of closely related populations. The main objectives of this work were to estimate genetic parameters, measure genetic distances and reconstruct phylogenetic relationships between Australian Angora/Angora_Aus/ and Cashmere/Cashmere_Aus/ populations and three Mongolian Cashmere goat (Bayandelger/BD/, Zavkhan Buural/ZB/, and Gobi Gurvan Saikhan/GGS/) populations based on variation at fourteen MS loci. The level and pattern of observed and expected heterozygosity and polymorphic information content of the fourteen loci studied across the populations were quite similar and high. Except for SRCRSP07, all studied microsatellites were in Hardy-Weinberg Equilibrium (p<0.001). Moderate genetic variation (7.5%) was found between the five goat populations with 92.5% of total genetic variation attributable to diversity existing between the individuals within each population. The greatest Nei's genetic distances were found between the Angora and four Cashmere populations (0.201-0.276) and the lowest distances were between the Mongolian Cashmere goat populations (0.026-0.031). Compared with other Cashmere goat populations, the GGS (crossbred with Russian Don Goats) population had the smallest pairwise genetic distance from the Australian Angora population (0.192). According to a three-factorial correspondence analysis (CA), the three different Mongolian Cashmere populations could hardly be distinguished from each other.
