Natural products have been used as drugs and cosmetics due to their bioactivity and their biochemical diversity. Natural products usually refer to secondary metabolites produced by various living organisms including marine animals, insects, microbes, amphibians, and plants. These secondary metabolites, which usually have molecular weights less than 2,000 amu, are unnecessary for survival, development, growth, and reproduction but play major roles in plant defense systems against other species. These secondary metabolites such as lignans, flavonoids, monoterpenes, and phenylethanoid glycosides showed various biological activities like anti-oxidant behavior, anti-cancer properties, neuroprotective properties, and so forth. Thus, isolation and elucidation of secondary metabolites from living organisms is of great significance to human life.
To identify genes implicated in the control of pluripotency as well as characteristics of stem cells, we analyzed expression profiles of genes derived from mouse morulas, blastocysts, embryonic stem cells, mesenchymal stem cells, and uterus tissue using cDNA microarray. Comparative analyses of their expression profiles identified putative clones that expressed specifically in specific samples or not in a specific sample. The expression pattern of these candidate clones was analyzed using RT-PCR and non-radioactive in situ hybridization. Functional annotation of these clones on pluripotency and stem cell plasticity is in ongoing. These studies may further our understanding on the nature of the stem cells and molecular mechanisms underlying many facets of mammalian development and differentiation.
Purpose: The purpose of this study was to explore the meanings of 'Natural childbirth' from experiences of Korean women who gave birth to a baby in the midwifery using a feminist approach. Methods: This paper is a qualitative research study and applies a feminist epistemology and methodology to the experiences of women who gave birth in midwifery. The data were collected by individual in-depth interviews with eleven participants. Results: Two main themes emerged from the feminist content analyses and each main theme had three sub themes. A. transformation of control and knowledge on childbirth and the body 1) refusing coercive medicalization and building a new normality, 2) specific expectations about biological health and maternity rather than a return to nature, 3) the subject of pregnancy and childbirth, B. 'natural childbirth' practice as a new embodied discipline 1) helpers to support mothers, midwives, 2) helping the body to do 'natural childbirth', 3) from isolated labor to cooperative reproduction. Conclusion: These results indicate that women desired to practice being a subject, consultation with professionals, self-discipline and named actors except for women as 'other subjects' in childbirth.
The incidence of both obesity and Type 2 Diabetes Mellitus( DM) is increasing proportionately so that causes of deaths from these has overtaken from that of malnourishment. Hence it has been recommended to treat the 2 in parallel considering the role of diabesity on health. Important causes of T2DM are insulin resistance (IR) and /or inadequate insulin secretion. Protein tyrosine phosphatase 1B(PTPIB) has a negative impact in insulin signaling pathways and hence plays crucial role inT2DM,since its overexpression might induce IR. Thus PTPIB is considered a therapeutic target for both obesity and T2DM, there has been a search for novel ,promising natural inhibitors. We conducted a pubmed search for articles related to PTPIB inhibitors from natural causes be it marine sources or other natural sources. Out of 988 articles we selected 100 articles for review. Thus various bioactive molecules isolated from marine organisms that can acts as PTPIB Inhibitors and thus possess antidiabetic activity both in vitro/ in vivo studies ,besides products from fruits like Chinese raspberry or curcumin used as routine spices are described with their chemical classes, structure-activity relationships and potency as assessed by IC 50 values are discussed. More work is required to make this a reality.
Safflower, a natural dye representing red, is the dye that materials and dyeing method are recorded in the literature, including materials and dyeing. Although the safflower is the same, the ash used as a mordant is recorded differently in each literature, which greatly affects the aesthetic perspective in realizing the traditional safflower red. Therefore, the optimal conditions for realizing the traditional safflower red were sought. The experiment was conducted by pH investigation, dyeing and color analysis by dyeing solution water, concentration, and temperature by ash, and the unique color of red was confirmed. As a result of the test, the pH point of time when the uniq ue color was expressed was 11.53 as goosefoot ash (natural bedrock water), which was 1:100 for concentration and 70℃ for temperature, which was easier to extract red pigment than other ash, indicating that it is suitable for safflower dyeing. The analysis of the ash showed that K and Si play an important role in dyeing, especially Si, which is an element that inhibits carthamon. The color of red was similar to that of KS Standard vivid purplish red in the ash of the goosefoot, and the other ash was deep purplish pink. In the light of findings, it was possible to quantify the dyeing method through traditional materials and find the standard color of red color, and it is judged to be a basic data for studying the unique color of natural materials.
Genomic DNA (gDNA) extracted from two populations of natural and cultured river pufferfish (Takifugu obscurus) was amplified by polymerase chain reaction (PCR). The complexity of the fragments derived from the two locations varied dramatically. The genetic distances (GDs) between individuals numbered 15 and 12 in the cultured population was 0.053, which was the lowest acknowledged. The oligonucleotide primer OPC-11 identified 88 unique loci shared within each population reflecting the natural population. The OPC-05 primer identified 44 loci shared by the two populations. The average band-sharing (BS) values of individuals in the natural population (0.683±0.014) were lower than in those derived from the cultured population (0.759±0.009) (p<0.05). The shortest GD demonstrating a significant molecular difference was found between the cultured individuals # 15 and # 12 (GD=0.053). Individual # 02 of the natural population was most distantly related to cultured individual # 22 (GD=0.827). A cluster tree was built using the unweighted pair group method with arithmetic mean (UPGMA) Euclidean GD analysis based on a total of 578 various fragments derived from five primers in the two populations. Obvious markers identified in this study represent the genetic structure, species security, and proliferation of river pufferfish in the rivers of the Korean peninsula.
This study investigated the effect of variation in the number of somatic-cell-cloned embryos and their developmental stage at transfer on pregnancy, as well as the influence of the estrus status of recipient pigs on in vivo development of cloned porcine embryos after embryo transfer. For somatic cell nuclear transfer (SCNT), fibroblast cells were obtained from a male porcine fetus. Recipient oocytes were collected from prepubertal gilts at a local abattoir and then cultured. After SCNT, reconstructed embryos of different numbers and developmental stages were transferred into recipient pigs. The developmental stage of the cloned embryos and the number of transferred embryos per surrogate showed no significant differences in terms of the resulting cloning efficiency. However, the pregnancy rate improved gradually as the number of transferred cloned embryos was increased from 100-150 or 151-200 to 201-300 per recipient. In pre-, peri-, and post-ovulation stages, pregnancy rates of 28.6%, 41.8%, and 67.6% and 16, 52, and 74 offspring were recorded, respectively. The number of cloned embryos and estrus status of the recipient pig at the time of transfer of the cloned embryo affect the efficiency of pig production; therefore, these variables should be particularly considered in order to increase the efficiency of somatic cell pig cloning.
In this study, we analyzed signal transduction by equine follicle-stimulating hormone receptor (eFSHR) on sti- mulation with recombinant $eelFSH{\beta}/{\alpha}$ ($rec-eelFSH{\beta}/{\alpha}$), natural porcine FSH (pFSH), and natural human FSH (hFSH). cAMP stimulation in CHO-K1 cells expressing eFSHR was determined upon exposure to different doses (0-1450 ng/mL) of these hormones. The $EC_{50}$ value of $rec-eelFSH{\beta}/{\alpha}$ was 53.35 ng/mL. The Rmax values of $rec-eelFSH{\beta}/{\alpha}$ and pFSH were 28.12 and 2.88 ng/mL, respectively. The activity of $rec-eelFSH{\beta}/{\alpha}$ was much higher than that of natural pFSH. However, signal transduction in CHO PathHunter Parental cells expressing eFSHR was not enhanced by stimulation with natural hFSH. Thus, $rec-eelFSH{\beta}/{\alpha}$ was completely active in cells expressing eFSHR. However, natural hFSH did not invoke a signal response in cells expressing eFSHR. Particularly, natural pFSH was weakly active in the same cells. These results showed that $eelFSH{\beta}/{\alpha}$ has potent activity in cells expressing eFSHR. Thus, $rec-eelFSH{\beta}/{\alpha}$ may efficiently bind to eFSHR, where as natural hFSH does not bind to eFSHR.
