Background: It was to develop an air purification system (APS) using an underground air purification layer to verify the effect of basalt forest's underground air (sumgol) on a volcanic Jeju. Finally, it is necessary to analyze these purified air components and their usefulness to the human body in an air experience center. Purpose: It was to collect basalt forest air, analyze its composition, and explore its effect on the human body. Methods: We APS devices installed at four points in the Papaville area of Jeju. The air discharged from the APS was collected and analyzed the recycling components. An installed experience room filled with negative ions is about 5,000 ions/m3. After allowing the participants to stay for 60 to 120 minutes, we investigated the state of blood vessels. Results: In the analysis of the underground air, the O2 concentration was 21.18%, which was higher than the average oxygen concentration of 20.94% in the atmosphere. However, Formaldehyde was not detected, and the CO2 was 419 ppm, which was lower than that of indoor air. The PM2.5 concentration was less than 24 ㎍/m3 and detected anions over 5.000 /m3. The experiencer's vascular states improved, and the increase in pulse rate and stress relief were high. Conclusions: The valuable ingredients identified by analyzing the air were precious for natural healing. The experience results showed that it effectively improved the pulse rate, blood vessels, and stress. These conditions may be highly beneficial as a new area for expanding the basalt lava forest in the Jeju area into the natural healing and wellness industry.
We screened many species from a wide variety of bacterial genera for a new type II restriction endonuclease. The purification and characterization of SolI from a soil isolate, Streptoverticillium olivoverticillatum are described here. The enzyme turned out to be an isoschizomer of BamHI. It recognized the hexanucleotide sequence of 5'-G$\downarrow$GATCC-3' and cleaved as in dicated by the arrow, generating a 4 base 5' extension. Unlike its isoschizomer, BamHI, the activity was sensitive to dam methylation within the recognition sequence. Following ammonium sulfate fractionation of the crude extract, heparin-agarose and Affi-gel Blue column chromatography were employed to purify the enzyme. SolI required at least 0.2 mM of $MgCl_2$ for the cleavage to occur. The enzyme exhibited its maximal activity in the absence of NaCl, but was inhibited completely in the presence of 120 mM NaCl. The pH and temperature optima for activity were pH 8.6 and $40^{\circ}C$, respectively. The molecular weight of SolI was estimated to be 43,000 Da by Superose-12 gel filtraion chromatography.

Distribution of biota and removal efficiency of organic matter in natural wetland systems were examined from June to October 1998. The aquatic macrophyte communities were consisted of 13 families and 22 species. The composition of occurrence species were as follows: Phragmites communis Trinius are 35%, Persicaria thunbergii Nakai are 19%, Typha angustifolia L. are 17%, Bidens tripartita L. are 16%, Echinochloa crus-galli (L.) Beauv. are 12% and others are l%. The fauna collected from 5 sites consisted of total 6 classes of macroinvertebrates. The composition of occurrence classes were as follows: Insecta are 76%, O1igochaeta are 19%, Gastrapoda are 4% and others are 1%. These included 18 families, 7 orders of Insects. The larve and mosquito and midges were found in wetlands. Preventive strategies are needed for the suppression of mosquito at the stage of wetland planning and design. In wetland, removal efficiency of dissolved organic carbon is low because of primary production and the background level of DOC in the wetland discharge seems to be 5 mgC/1, but those of BOD was ca. 50%. Wetlands receiving water of low concentration can not work as purification field.

The generally known important functions of forests include air purification and the health benefits that humans can receive when relaxing and enjoying recreation in the forests. In recent years, people have appreciated the value of the natural environment but it is not easy to answer the question how much monetary value a natural environment has. Because environmental property is public property, which is not traded on the market, market prices cannot be established, so it is not easy to assess the currency value. Methods for estimating environmental property value have been studied by economists. The representative method for measuring environmental property value is a contingent valuation method, or CVM. Various methods have been researched and attempted along with the development and fusion of mathematics, statistics, and economics. Representative methods of CVM are single-bound and double-bound logit and probit methods. This study has been carried out to compare four estimates. Estimates are as follows: the lowest estimate is derived from a single-bound logit WTPmedian while the highest estimate is from double-bound probit WTPmean. While there are some preceding studies on price estimation and methods of measurement through CVM, they offer only partial comparisons. This study suggests four analytic methods and prices through 1,123 questionnaires. The results can be used for the subsequent comparison of estimate prices and the methods of measurement

Marine natural products isolated from the sponge Fascaplysinopsis cf reticulata, in French Polynesia, were investigated as an alternative to antibiotics to control pathogens in aquaculture. The overuse of antibiotics in aquaculture is largely considered to be an environmental pollution, because it supports the transfer of antibiotic resistance genes within the aquatic environment. One environmentally friendly alternative to antibiotics is the use of quorum sensing inhibitors (QSIs). Quorum sensing (QS) is a regulatory mechanism in bacteria which control virulence factors through the secretion of autoinducers (AIs), such as acyl-homoserine lactone (AHL) in gram-negative bacteria. Vibrio harveyi QS is controlled through three parallel pathways: HAI-1, AI-2, and CAI-1. Bioassay-guided purification of F. cf reticulata extract was conducted on two bacterial species, i.e., Tenacibaculum maritimum and V. harveyi for antibiotic and QS inhibition bioactivities. Toxicity bioassay of fractions was also evaluated on the freshwater fish Poecilia reticulata and the marine fish Acanthurus triostegus. Cyclohexanic and dichloromethane fractions of F. cf reticulata exhibited QS inhibition on V. harveyi and antibiotic bioactivities on V. harveyi and T. maritimum, respectively. Palauolide (1) and fascaplysin (2) were purified as major molecules from the cyclohexanic and dichloromethane fractions, respectively. Palauolide inhibited QS of V. harveyi through HAI-1 QS pathway at 50 ㎍ ml-1 (26 μM), while fascaplysin affected the bacterial growth of V. harveyi (50 ㎍ ml-1) and T. maritimum (0.25 ㎍). The toxicity of fascaplysin-enriched fraction (FEF) was evaluated and exhibited a toxic effect against fish at 50 ㎍ ml-1. This study demonstrated for the first time the QSI potential of palauolide (1). Future research may assess the toxicity of both the cyclohexanic fraction of the sponge and palauolide (1) on fish, to confirm their potential as alternative to antibiotics in fish farming.
This research has developed the criteria and index for systematically and objectively assessing the quality of river water by fixing the various factors that affect Gyeongin ARA waterway's water quality through analysis with the Delphi Technique and analytic hierarchy program (AHP) Method. Based on the results, the highest criteria are, in order of importance, physical and environmental factors 28%, administrative factors 26%, natural fixed factors 26% and finally, cultural and social factors 20%. The three dimensions of the criteria show that for the internal physical and environmental factors, the most important are the loss of self-purification capacity, and the external factors are Gulpocheon and the sludge deposit due to Gyulhweon-weir the bridge. The facility factor in management was affected by the coagulation and waste water disposal facilities. The problem for the policy and institutional factors was seen in the regulatory area. The aquatic ecology/ point pollution source for the natural fixed factors show that it is due to the polluted water of Gulpo-cheon and the living environment/ non-point pollution source is shown through the inflow water from other rivers. Cultural and social factors show that the economical causes were due to the cargo and passenger flight operations and the external factors of having a lack of sewage treatment equipment have an importance effect. In order to estimate the order of priority through logical evidence and objectivity, future research must be continued on the evaluation indexes to measure the specific methodology and technique needed to improve the Gyeongin ARA Waterway.
Using the PCR(polymerase chain reaction method), gone 1 of phage K11 coding for K11 phage RNA polymerase has been cloned and expressed under the control of lac promoter. K11 phage RNA polymerase was conventionally purified through the DEAE-sephacel and Affigel blue column chromatographies. The 0.2-0.3 M $NH_4Cl$ fractions of DAEA-sephacel column chromatography showed K11 phage RNA polymerase activity and further purification with Affigel blue column chromatography showed nearly single protein band on SDS-polyacryl amide gel. K11 phage RNA polymerase, which is one of the T7 group phage RNA polymerase (E. coil phage T7, T3 and Salmonella tyhimurium phage SP6 RNA polymerase), shares high degrees of homology with the other T7 group phage RNA polymerase. Previously we constructed T7 and SP6 promoter variants and revealed promoter specificity of T7 and SP6 RNA polymerase (Lee and Kang, 1993). To investigate the promoter specificity of K11 RNA polymerase in vitro K11 promoter activity was measured with SP6 promoter variants. The SP6 promoter variant share highest degrees of sequence homology with K11 promoter sequence show strongest promoter activity.
Nowadays, there are increasing demands of golf courses and it is necessary to make more golf courses than the present. To do this, we need to improve the environmental problems with the regional inhabitants, and it is said that the first thing to be considered in developing any golf course in Korea is to preserve the environment. In this context, the purpose of this study is to set forth several design factors to lessen the negative impacts which are accompanied with the development of golf courses. 1. The present conditions of golf courses in Korea Many new golf courses have come into being, particularly since the late 1980s, and now, in the year of 1997, over one hundred of golf courses are doing their business, yet the number of golf course is still less than required. So far, over a half of them have been made in the vicinity of Seoul on account of various reasons, and this has adversely affected on our natural environment. This unreasonable development of golf courses has caused serious water pollution, landslides and the other problems. Also, the topography of Korea is not good for golf courses. Although the demands of golf courses are increasing, the suitable sites for them are very limited, and therefore it is sometimes unavoidable to make golf courses on steep hills. The liquid culture extract of Coriolus versicolor was prepared by directly boiling the whole culture broth 7 days after incubation in 12% citrus extract medium. After removal of mycelial debris through filtration, this extract was further extracted with equal volume of ethyl acetate (1 : 1, v/v). The ethyl acetate extracts showed significant antibacterial activities against Stapylococcus aureus CCARM3230 and Psudomonas aeruginosa CCARM2171, which are resistant to several antibiotics. The most active fraction was eluted from a silica gel column with a mixture of dichloromethane and methanol (9 : 1, v/v) and the purity of this active substance was confirmed by HPLC analysis. The results suggest that the purified active substance could be a good source for the development of a new antimicrobial agent, especially for the treatment of antibiotic resistant bacteria.
Keratinase is widely used in certain industrial applications. The present study sought to improve the culture conditions of Bacillus subtilis SMMJ-2 to facilitate mass production of keratinase. Strain SMMJ-2 was irradiated by ultraviolet light and the resulting isolates were tested for keratinase activity. Isolates displaying elevated keratinase activity were selected and used to determine the optimum temperature (24, 30, 37, 45, $55^{\circ}C$) for bacterial keratinase production during a 4 day incubation period. The highest enzyme activity (55 units/mL/min), from a Bacillus subtilis SMMJ-2 mutant (mutant No. 2) was demonstrated following incubation at $30^{\circ}C$. The effects of carbon and nitrogen sources on keratinase production were confirmed by measuring the enzyme activity from the culture broth of the mutant strain cultured in various media containing different carbon source and nitrogen sources during a 4 day period. The optimal medium composition for producing keratinase consisted of 1% glucose, 0.7% $K_2HPO_4$, 0.2% $K_2HPO_4$, and 1.2% soybean meal. Optimal initial pH and temperature for producing keratinase were 7.0 and $30^{\circ}C$, respectively. Keratinases produced by B. subtilis SMMJ-2 and the mutant No. 2 were purified from the culture broth which used soybean meal as a nitrogen source. Membrane ultrafiltration, DEAE-sephacel ion exchange and Sephadex G-100 gel chromatography were used to purify the enzymes. The purified keratinases from both B. subtilis SMMJ-2 and the mutant No. 2 showed single bands and their molecular weights were estimated as 28 kDa and 42 kDa, respectively on SDS-polyacrylamide gel electrophoresis.
