We employed dual color Fluorescence Cross Correlation Spectroscopy (FCCS) to measure the interaction between PKA regulatory (RII) and catalytic subunits (CAT) in living cells. Elevation of intracellular cAMP with forskolin decreased the cross-correlation amplitude between RFP-fused RII (RII -mRFP) and GFP-fused CAT (CAT-EGFP) by 50%, indicating that cAMP elevation leads to dissociation of RII-CAT complexes. Moreover, diffusion coefficient analysis showed that the diffusion rate of CAT-EGFP was significantly increased, suggesting that the decreased RII-CAT association caused by cAMP generated free CAT subunits. Our study demonstrates that in vivo FCCS measurements and their quantitative analysis permit one not only to directly quantify protein-protein interactions but also to estimate changes in the intracellular cAMP concentration.

In this letter, we propose a dual autostereoscopic display platform employing a natural interaction method, which will be useful for sharing visual data with users. To provide 3D visualization of a model to users who collaborate with each other, a beamsplitter is used with a pair of autostereoscopic displays, providing a visual illusion of a floating 3D image. To interact with the virtual object, we track the user's hands with a depth camera. The gesture recognition technique we use operates without any initialization process, such as specific poses or gestures, and supports several commands to control virtual objects by gesture recognition. Experiment results show that our system performs well in visualizing 3D models in real-time and handling them under unconstrained conditions, such as complicated backgrounds or a user wearing short sleeves.
The natural convection heat transfer in a vertical plate with discrete heat sources was studied experimentally. The particular interest was the thermal interaction of the heat sources. In this study, the radiative and conductive heat transfer were considered as heat loss, Thus, the net convective heat transfer rate was presented as adiabatic temperature and thermal wake function. As a results, for non-uniform heating condition, heat input ratio(q1/q2) was most dominant parameter for the thermal wake function. The convective heat transfer rate is decreased with the increasing of channel ratio. For the range of $7.50{\times}10^5<Rac<8.66{\times}10^6$, a useful correlation was proposed as a function of channel Rayleigh number.
This paper studies the effect of vibrational characteristics of the various tubes analyzed though experiment. By an experiment analysis we found out that the factor of system vibration is fluid-structure interaction of tube line. In fluid-filled tube system we study on the influence that the natural frequency of system and the frequency of wave motion produce upon through three experiments. Three experiments are modal test on each tube, FRF in continuous system, and vibrating tests when the system is driving on. From the results of the experimental studies, we obtained that the natural frequencies of system are very important than wave induced vibrations. and according to the variation of configuration, the frequencies are different each other. And we found that though fluid passed away through the tube, the tendency of system vibration level was similar with the mode shape at the simple system.
Recent experimental evidence that fluorinated graphene creates local magnetic moments around F adatoms has not been supported by semilocal density-functional theory (DFT) calculations where the adsorption of an F adatom induces no magnetic moment in graphene. Here, we show that such an incorrect prediction of the nonmagnetic ground state is due to the self-interaction error inherent in semilocal exchange-correlation functionals. The present hybrid DFT calculation for an F adatom on graphene predicts not only a spin-polarized ground state with a spin moment of ${\sim}1{\mu}_B$, but also a long-range spin polarization caused by the bipartite nature of the graphene lattice as well as the induced spin polarization of the graphene states. The results provide support for the experimental observations of local magnetic moments in fluorinated graphene.
In order to elucidate conformational properties of bilayer semiflexible amphiphile molecules, we derive a expression of free energy separation with respect to bilayer width, and segment density profiles on the basis of cubic lattice model. Our result shows that at the moderate surface coverage region (i.e., ${\sigma}$ < 0.35), bilayer system tends to have thermodynamically favorable bilayer width corresponding to free energy minimum condition resulting from the major contribution of attractive interaction between chain segments. However such a favorable bilayer width do not occur in the region of high surface converage (i.e., ${\sigma}$> 0.4) where repulsive interaction between chain segments is considered to be dominant.
The objective of present study is to investigate the effect of granulated blast furnace slag (GBS) as partial substitution of natural sand on behaviour of cement mortar. For this, the methods of factorial design with water cement (w/c) ratio and incorporation percentages of GBS as replacement of natural fine aggregate i.e., GBS(%) as factors are followed. The levels of factor w/c ratio are fixed at 0.4, 0.45, and 0.5 and the levels of factor GBS(%) are kept fixed as 0%, 20%, 40%, 60%, 80% and 100%. The compressive strength (CS) of mortar after 3, 7, 14, 28, 56 and 90 days, and water absorption (WA) are chosen as responses of the study. Analysis of variance (ANOVA) of experimental results has been carried out and those are illustrated by ANOVA tables, main effect and interaction plots. The results of study depict that the selected factors have substantial influence on the strength and WA of mortar. However, the interaction of factors has no substantial impact on CS and WA of mixes.
In this study, on-site testing was carried out to investigate the vibration performance of a composite steel-bar truss slab with steel girder system. Ambient vibration was performed to capture the primary vibration parameters (natural frequencies, damping ratios, and mode shapes). The composite floor possesses low frequency (< 10 Hz) and damping (< 2%). Based on experimental, theoretical, and numerical analyses on natural frequencies and mode shapes, the boundary condition of SCSC (i.e., two opposite edges simply-supported and the other two edges clamped) is deemed more reasonable for the composite floor. Walking excitations by one person (single excitation), two persons (dual excitation), and three persons (triple excitation) were considered to evaluate the vibration serviceability of the composite floor. The measured acceleration results show a satisfactory vibration perceptibility. For design convenience and safety, a crest factor ${\beta}_{rp}$ describing the ratio of peak acceleration to root-mean-square acceleration induced from the walking excitations is proposed. The comparisons of the modal parameters determined by ambient vibration and walking tests reveal the interaction effect between the human excitation and the composite floor.
This paper is concerned with the numerical calculation of mixed-mode stress intensity factors (SIFs) of 2-D isotropic functionally graded materials (FGMs) by the natural element method (more exactly, Petrov-Galerkin NEM). The spatial variation of elastic modulus in non-homogeneous FGMs is reflected into the modified interaction integral ${\tilde{M}}^{(1,2)}$. The local NEM grid near the crack tip is refined, and the directly approximated strain and stress fields by PG-NEM are enhanced and smoothened by the patch recovery technique. Two numerical examples with the exponentially varying elastic modulus are taken to illustrate the proposed method. The mixed-mode SIFs are parametrically computed with respect to the exponent index in the elastic modulus and external loading and the crack angle and compared with the other reported results. It has been justified from the numerical results that the present method successfully and accurately calculates the mixed-mode stress intensity factors of 2-D non-homogeneous functionally graded materials.
Resveratrol (RES) and genistein (GEN) are the dietary natural products known to possess chemopreventive property and also the ability to repair DNA damage induced by mutagens/carcinogens. It is believed that the therapeutic activity of these compounds could be primarily due to their interaction with nucleic acids but detailed reports are not available. We here explore the interaction of these drugs with nucleic acids considering DNA and RNA as a potential therapeutic target. The interaction of RES and GEN has been analysed in buffered solution with DNA [saline sodium citrate (SSC)] and RNA [tris ethylene diammine tetra acetic acid (TE)] using UV-absorption and Fourier transform infrared (FTIR) spectroscopy. The UV analysis revealed lesser binding affinity with nucleic acids at lower concentration of RES (P/D = 5.00 and 10.00), while at higher drug concentration (P/D = 0.75, 1.00 and 2.50) hyperchromic effect with shift in the ${\lambda}_{max}$ is noted for DNA and RNA. A major RES-nucleic acids complexes was observed through base pairs and phosphate backbone groups with K = $35.782\;M^{-1}$ and K = $34.25\;M^{-1}$ for DNA-RES and RNA-RES complexes respectively. At various concentrations of GEN (P/D = 0.25, 0.50, 0.75, 1.00 and 2.50) hyperchromicity with shift in the ${\lambda}_{max}$ from 260 $\rightarrow$ 263 om and 260 $\rightarrow$ 270 nm is observed for DNA-GEN and RNA-GEN complexes respectively. The binding constant (from UV analysis) for GEN-nucleic acids complexes could not be obtained due to GEN absorbance overlap with that of nucleic acids at 260 nm. Nevertheless a detailed analysis with regard to the interaction of these drugs (RES/GEN) with DNA and RNA could feasibly be understood by FTIR spectroscopy. The NH band of free DNA and RNA which appeared at $3550-3100\;cm^{-1}$ and $3650-2700\;cm^{-1}$ shifted to $3450-2950\;cm^{-1}$ and $3550-3000\;cm^{-1}$ in DNA-RES and RNA-RES complexes respectively. Similarly shifts corresponding to $3650-3100\;cm^{-1}$ and $3420-3000\;cm^{-1}$ have been observed in DNA-GEN and RNA-GEN complexes respectively. The observed reduction in NH band of free nucleic acids upon complexation of these drugs is an indication of the involvement of the hydroxyl (OH) and imino (NH) group during the interaction of the drugs and nucleic acids (DNA/RNA) through H-bonded formation. The interaction of RES and GEN with bases appears in the order of G $\geq$ T > C > A and A > C $\geq$ T > G. Further interaction of these natural compounds with DNA and RNA is also supported by changes in the vibrational frequency (shift/intensity) in symmetrical and asymmetrical stretching of aromatic rings of drugs in the complex spectra. No appreciable shift is observed in the DNA and RNA marker bands, indicating that the B-DNA form and A-family conformation of RNA are not altered during their interaction with RES and GEN.
