Ursolic acid (UA) a bio-active ingredient found in a variety of fruits and vegetables, and it has potent antioxidant activity. However, the role of UA in mouse embryonic stem (ES) cells is poorly understood. This study investigated the functional role of UA in regulating the development of mouse ES cells under hypoxia. Hypoxia did not exert a significant effect on the undifferentiated state of mouse ES cells. However, it induced reactive oxygen species (ROS) generation and increased the level of lactate dehydrogenase (LDH) production at 48 h of hypoxic exposure. Conversely, oxidative stress induced by hypoxia was significantly inhibited by UA ($30{\mu}M$) pretreatment. Hypoxia significantly decreased cell survival and the level of [$^3H$] thymidine incorporation, both of which recovered following pretreatment of UA. In addition, UA decreased the apoptotic effect of hypoxia by attenuating caspase-3 cleavage or by recovering cellular inhibition of the apoptotic protein (cIAP)-2 and Bcl-2 expression. We further found that UA decreased senescence-associated beta-galactosidase activity. We suggest that UA is a natural antioxidant and one of the functional modulators of hypoxia-induced survival, apoptosis, proliferation, and aging in mouse ES cells.
Macrophages play a pivotal role in the innate and adaptive immune systems. This study investigated the immuno-modulatory activities of polysaccharides separated from Chrysanthemum zawadskii var. latilobum (CZPS) in macrophages. Polysaccharides from Chrysanthemum zawadskii var. latilobum were extracted by the ethanol precipitation method. RAW 264.7 mouse macrophage cell line was treated with CZPS (4 to $128{\mu}g/mL$), and there was no cytotoxicity at a dose below $32{\mu}g/mL$. The levels of nitric oxide (NO) and pro-inflammatory cytokines (tumor necrosis factor (TNF)-${\alpha}$ and interleukin (IL)-6, IL-$1{\beta}$) production in the CZPS treated group ($32{\mu}g/mL$) were $6.5{\pm}0.12{\mu}m$ (NO), $1252.8{\pm}79.85$ (TNF-${\alpha}$), $305.4{\pm}29.41$ (IL-6), and $683.3{\pm}59.71$ (IL-$1{\beta}$), respectively, and they were significantly increased when compared to the control group; $2.2{\pm}0.03{\mu}m$ (NO), $452.3{\pm}38.34$ (TNF-${\alpha}$), $31.7{\pm}5.75$ (IL-6), and $184.1{\pm}11.52$ (IL-$1{\beta}$). Additionally, protein expression of inducible nitric oxide synthase (iNOS) and phosphorylation of MAPKs and NF-${\kappa}B$ expression were significantly increased upon CZPS treatment. Therefore, these results indicated that polysaccharides separated from Chrysanthemum zawadskii var. latilobum (CZPS) may have a potential immunomodulatory activity in macrophages through MAPKs and NF-${\kappa}B$ signaling, and this information is useful for the development of immune enhancing adjuvant materials using a natural ingredient.

In this research, water and ethanol extract of Momordica charantia shells, fruits and seeds were tested to see possibility as natural functional cosmetic agent. Water and ethanol extract showed 69.45 mg/g and 70.87 mg/g polyphenol concentration, respectively. Momordica charantia water and ethanol extracts did not indicate cell toxicity up to $1,000{\mu}g/ml$ concentration in MTT assay. Tyrosinase inhibition effects of water and ethanol extract were lower than arbutin, however, ethanol extract showed better DOPA oxidation inhibition effect than arbutin. Elastase inhibition effects of ethanol extract displayed similar efficacy with adenosine at higher concentrations. Solution formulations (5% extract) were stable for 28 days in both extracts, however, lotion formulation (1% extract) showed considerable variation in viscosity whereas ethanol extraction indicated relative stability. In conclusion, water and ethanol extract of Momordica charantia shells, fruits and seeds indicated strong possibility for whitening and antiwrinkle functional cosmetic ingredient.
Objectives: This study was carried out in order to analyze the composition of the leaves and stems of Abeliophyllum distichum Nakai, with the aim of obtaining basic data for utilizing the plant as a food ingredient, as well as for processing. Methods: Leaves and stems from Abeliophyllum distichum Nakai were harvested at Cheongcheon-myeon, Geosan-gun, Chungcheongbuk-do, and were subsequently freeze-dried and ground to a fine powder for chemical component analysis and safety evaluation. Results: The moisture contents of Abeliophyllum distichum Nakai leaves and stems were respectively 65.07% and 40.97%, and the crude ash contents were 1.32% and 0.91%. In addition, the crude protein contents were 11.97% and 3.77%, and the crude fat contents were 2.52% and 0.36%, respectively. The fructose and glucose contents were 32.13 mg/g and 56.17 mg/g for leaves, and 11.38 mg/g and 10.59 mg/g for stems. The major fatty acids of the leaves were palmitic acid (31.79%) and stearic acid (14.79%), and those for stems were linolenic acid (32.78%) and palmitic acid (26.75%). The ascorbic acid contents of leaves and stems were 1.32 mg/g and 0.30 mg/g respectively. The calcium content was found to be the highest among the minerals tested, both in the leaves and stems, with the levels being 166.17 mg/100 g for leaves and 592.34 mg/100 g for stems. The content of organic acid was greater in leaves than in stems, with that of malic acid accounting for more than 75% of total organic acids for both samples. The total phenolic compounds and flavonoid contents of Abeliophyllum distichum Nakai were 50.64 mg/g and 13.53 mg/g in leaves and 96.47 mg/g and 18.53 mg/g in stems, respectively. No changes were shown in the number of micronucleated polychromatic erythrocytes (MNPCE) among 2,000 polychromatic erythrocytes compared to the negative control. Abeliophyllum distichum Nakai was administered orally to rats in order to investigate acute toxicity. The $LD_{50}$ values in rats were above 2,000 mg/kg. Conclusion: These results indicate that the leaves and stems of Abeliophyllum distichum Nakai can be used as natural ingredients in the development of nutritional and functional materials.
A pavilion is located in the place with beautiful scenery, which maximizes the poetical features of nature. So a scholar in the Chosun Dynasty had the dream of possessing it. This trends of the Chosun scholar resulted in producing lots of literary works related with the pavilion. In this thesis, I researched the characters of Pavilion Si-Jo in accordance with the function of pavilion. First, the pavilion, which is surrounded by the beautiful scenery, is a good place to see the beauty of nature. It is needless to walk around to see the scenery thanks to its open structure. So the feeling in the pavilion Si-Jo is distinguished from other sightseeing Si-Jos: those show the way of seeing the scenery walking here and there, but this shows the way of seeing the scenery only just looking up, looking down, and looking in the distance. Second, the pavilion functions as an academic place. In this case, it can be said that it functions like a lecture hall. It is more effective to study in the pavilion than in the closed structure, a lecture hall. Furthermore, the scholar studying in the pavilion understand the principle of nature with seeing the beautiful mountain and river, and it can be a place for growing a vast-flowing spirit. This kind of poetry has been handed down with the Chinese poetry, and the character who operated the pavilion often appears in the work. Third, the pavilion functions as a space for a fraternity or poetry circles. The poetic exchange was conducted by host of pavilion. which is the key ingredient for organizing a poetry circles. Finally, the pavilion functions as an entertainment. Although it is a space for an entertainment, there is no deviation as a man of novel birth. If the pavilion has the feature of closeness, there are sometimes works which shows the deviation of them.

To develop a new natural whitening agent for cosmetics, we investigated effects of Acanthopanax sessiliflorum Seeman on melanogenesis. We prepared phenolic acid-rich extract including two phenolic acids, chlorogenic acid and caffeic acid, as predominant constituents from Acanthopanax sessiliflorum Seeman. Phenolic acid-rich extract showed ROS scavenging activities in 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical and xanthine/xanthine oxidase system with the $IC_{50}$ values of $3.43{\pm}0.35{\mu}g/mL$ and $158.91{\pm}1.57{\mu}g/mL$, respectively. Phenolic acid-rich fraction reduced melanin contents of B16 melanoma cells dose-dependantly and the decrease was $27.27{\pm}2.66%$ at a concentration of $100{\mu}g/mL$. And the phenolic acid-rich fraction reduced intracellular tyrosinase activity about $53.67{\pm}8.55%$ at a concentration of $100{\mu}g/mL$. Phenolic acid-rich extract inhibited tyrosinase and TRP-2 expression at protein level. These results suggest that phenolic acid-rich fraction reduced melanin formation by the inhibitions of tyrosinase activity and expression in B16 melanoma cells. Therefore, we suggest that phenolic acid-rich extract could be used as a whitening ingredient in cosmetics.

The functions of anti-oxidation and anti-inflammation were investigated with the crude 80% methanol extract, subfractions and henryin isolated from Isodon inflexus (Thunb.) Kudo (I. inflexus (Thunb.) Kudo). Antioxidative ability was evaluated by bioassays using 2, 2-diphenyl-1-1-picrydrazyl (DPPH) free radical scavenging activity, xanthine oxidase inhibition, and superoxide radical scavenging effects. Ethyl acetate and butanol fractions exhibited free radical scavenging activity on superoxide with $IC_{50}$ values of $0.9{\mu}g/mL$, $0.2{\mu}g/mL$, respectively, which were stronger activity than that of allopurinol ($2.2{\mu}g/mL$) as reference. For the inhibition of anti-inflammatory activity in RAW 264.7 cell, the ethyl acetate fraction showed strong inhibition activity NO production, and henryin isolated from its subfraction reduced the activity in a dose-dependent manner. Ethyl acetate fraction and henryin suppressed not only mRNA expression of iNOS and COX-2, but also the mRNA expression of pre-inflammatory cytokines such as, TNF-${\alpha}$, 1L-$1{\beta}$, IL-6, in a dose-dependent manner. These results suggested that ethyl acetate fraction of I. inflexus (Thunb.) Kudo has considerable potential as a cosmetics ingredient with an antioxidant and anti-inflammatory effects and henryin can be applied as an functional reference.

Objectives: This study was performed to set easily applicable portion sizes by sex and age for children at the Community Child Centers (CCC) in Korea. Methods: Considering the age and gender specific energy level at Target Patterns for children aged 6-18 years, which were suggested as a part of the 2010 Korean Food Guidance System (KFGS), we set three meal sizes. We reclassified the recommended daily servings of Grains, Meat fish eggs beans and Vegetables group at Target Patterns into three meal sizes, and then calculated the recommended serving per meal. Each proposed amount of food per meal was calculated based on serving size of foods commonly eaten at KFGS, which was then allocated to five meal components; rice, soup stew, protein and vegetable side-dishes and Kimchi. Each proposed amount of food per meal was applied to 173 menus' recipes from CANpro 3.0 as main ingredient's amounts. We cooked the 173 menus at the medium size and measured their weights after cooking. Results: Each recommended serving per meal was 0.75, 0.9 and 1.2 for Grains; 1.2, 1.6 and 2.4 for Meat fish eggs beans; 2, 2.4 and 2.8 for Vegetables by meal sizes. Among five meal components, the ratio of small and large to medium size was 1/5 less and 1/3 more for rice and 1/3 less and 1/3 more for soup stew, protein side-dish and Kimchi, respectively. We suggested the same amount for a vegetable side-dish to encourage vegetable intake. Proper portion sizes per meal of medium were rice 190 g, soup stew 210 g (solid ingredients 60 g), protein side-dish 100 g (meat eggs beans) and 70 g (fish), vegetable side-dish 80g and Kimchi 30 g. Conclusions: Proper portion size per meal suggested in this study may be useful at the CCC where dietitians are not available and the approach could be applicable to the other types of meal services.

The aim of this study is to investigate the effect of Hovenia dulcis (oriental raisin tree) extract on ganodermanondiol (GN) contents in Ganoderma lucidum (G. lucidum) mycelia. GN has a triterpenoid structure and is one of the major active components of G. lucidum. Furthermore, we previously proved its inhibitory effects on tyrosinase activity and melanin biosynthesis in B16F10 melanoma cells. In this study, we observed significantly increased GN contents in G. lucidum mycelial extracts supplemented with 15% (v/v) oriental raisin tree extract (ORTE) by HPLC analysis. In addition, melanogenesis was significantly inhibited by G. lucidum extract supplemented with 15% ORTE when compared to G. lucidum extract without ORTE supplementation. Furthermore, mycelial growth of G. lucidum was increased by ORTE supplementation in both solid and liquid cultivation. These results suggest that the oriental raisin tree is useful as natural ingredient for increasing GN biosynthesis as well as whitening effect of G. lucidum.
Onion peels is a natural source of high-value functional ingredients produced in the onion industry without suitable processing. The objective of this study was to evaluate characteristics of onion peel extract (OPE), including its biological activities, obtained from solvent extraction in 3 times pilot scales (Lot A, B, and C). Mineral analysis showed that K was present in the largest amount (13,767.56-15,506.78 ppm), followed by Na and Ca at 8,602.44-9,796.00 ppm and 4,255.78-4,903.33 ppm, respectively. The amounts of total phenol, total flavonoid, and quercetin in the OPE were in the ranges of 598.57~626.73, 211.73~233.64, and 93.78~107.29 mg/g, respectively. The biological activities such as antioxidant and effects of fibrinolysis increased in parallel with the concentration of OPE. The $IC_{50}$ value of DPPH radical scavenging activity was in the range of 517.58~557.32 ppm in the OPE. The $IC_{50}$ value for superoxide dismutase (SOD)-like activity was in the range of 11,900.91~12,690.35 ppm. A clear zone of OPE (20,000 ppm) in fibrinolysis test was three times higher than the plasmin as a reference. In conclusion, OPE could be used as a good source of antioxidants and fibrinolytic activities.
