Antioxidant Activities and Whitening Effects of Extracts from Hippophae rhamnoides L. (비타민 나무(Hippophae rhamnoides L.) 열매 추출물의 항산화 활성 및 미백 효과)

  • This study was performed to investigate the antioxidant activities and the melanin inhibitory effects of Hippophae rhamnoides L. fruit extracts. Two in vitro methods were used; the 1,1-diphenyl-2-picrylhydrazyl (DPPH) method to determine antioxidant activity and measurements of the inhibitory effects of tyrosinase activity to determine melanogenesis in B16/F10 melanoma cells. The radical scavenging activity of the extract was 56.0% at $700{\mu}g/mL$, similar to ascorbic acid (56.9%), in the DPPH assay. The tyrosinase inhibitory activity of the extract was 52.1% and 73.4% at 100 and $500{\mu}g/mL$, which is also similar to ascorbic acid. In B16/F10 mouse melanoma cells, the extract inhibited melanin synthesis by 56% at $500{\mu}g/mL$, a more prominent inhibition of melanin synthesis compared to extracts from arbutin. These results suggest that extracts from H. rhamnoides L. have antioxidant activity and skin-whitening effects; allowing their application in cosmetics as a natural product.

Physiological Activity of Aconitum pseudolaeve var erectum Nakai and Its Effect on Perfume on an Electroencephalogram (EEG) (진범(Aconitum pseudolaeve var erectum Nakai)의 생리활성과 진범 향이 뇌파에 미치는 영향)

  • This study analyzed the in vitro physiological activity of Aconitum pseudolaeve var erectum Nakai (AP) and its effect on perfume on an electroencephalogram (EEG). The results indicated that the absolute alpha power spectrum (AA) and the absolute theta power spectrum (AT) decreased significantly during more than before the inhalation of the AP perfume and its reconstruction perfume. Although there were a little different pattern in the induced part of the wave generated by the inhalation of the AP perfume and its reconstruction perfume, alpha- and theta- were shown equally. In addition, there was the same pattern in which the rate of increase reduced. The results suggest that the perfume of the AP and its reconstruction perfume have a stimulating effect on the brain. In terms of the physiological activity of AP, the activity of AP ethanol extract was significantly higher than that of water extract in DPPH, collagenase, and nitric oxide, except for an astringent effect. The AP ethanol extract was about 80% at 500 ppm in collagenase inhibition activity. In addition, the AP water and ethanol extracts were 50% at 100 ppm in the NO inhibition activity. Based on these results, we conclude that this natural substance could be used in cosmetics and in the development of perfumes.

Antioxidant and Whitening Effects of Sorbus commixta HEDL Cortex Extract (정공피 추출물의 항산화 활성 및 미백효과에 관한 연구)

  • This study was performed to assess the antioxidant activities and whitening effects of Sorbus commixta HEDL cortex on melanin synthesis. The whitening effects of Sorbus commixta HEDL cortex water extracts were examined by in vitro mushroom tyrosinase assay and B16BL6 melanoma cells. We assessed inhibitory effects of Sorbus commixta HEDL cortex water extracts on expression of melanogenic enzyme proteins including tyrosinase, tyrosinase-related protein 1 (TRP-1) and tyrosinase-related protein 2 (TRP-2) in B16BL6 cells. Inhibitory effects of Sorbus commixta HEDL cortex onto free radical generation were determined by measuring DPPH and hydroxyl radical scavenging activities. Our results indicated that Sorbus commixta HEDL cortex water extracts effectively inhibited free radical generation. In DPPH radical scavenging activity, Sorbus commixta HEDL cortex water extracts had a potent anti-oxidant activity in a dose-dependent manner. They significantly inhibited tyrosinase activity in vitro and in B16BL6 melanoma cells. Also, Sorbus commixta HEDL cortex suppressed the expression of tyrosinase, TRP-1 and TRP-2 in B16BL6 melanoma cells. These results show that Sorbus commixta HEDL cortex inhibited melanin production on the melanogenesis. The underlying mechanism of Sorbus commixta HEDL cortex on whitening activity may be due to the inhibition of tyrosinase activity and tyrosinase, TRP-1, TRP-2 expression. We suggest that Sorbus commixta HEDL cortex may be contain new natural active ingredients for antioxidant and whitening cosmetics.

Measuring of Anti-oxidant Activity and Skin Improvement Effect using Adenophora remotiflora leaf (모시대잎을 이용한 항산화 및 피부개선 효과 측정)

  • The purpose of this study was to find out the potential of the Mosidae(Adenophora remtriflora) leaf, one of the natural ingredients, for the cosmetics by measuring their antioxidant functions and skin improving effects. The result of the phytonutrient of the Mosidae leaf test shows, that the heat extract of Mosidae leaf DPPH has 9% higher ability than Ethanol extract.(p<0.05), but significant a difference was not found between the hydroxyl Radical of Mosidae leaf heat extract (65.22%) and Ethanol extract(63.50%). Mosidae leaf heat extract(40.82mg/mL) has much lower polyphenol compound than Ethanol extract(47.90mg/mL)(p<0.05). A clinical test of Mosidae leaf powder pack was performed for a group of 16 middle aged women(age between 35-60) from October 20 to November 17, 2011. Control group was used on the right cheek, and test group was used on left cheek for comparison. The procedure was twice a week for 4 weeks to identify and grade level of moisture, roughness, pore size and spot. After 4 weeks of testing, there were significant improvements in moisture enhancement and spot(p<0.05), subtle change in roughness and the size of the pores. Therefore, we confirmed skin improvement effect of Mosidae leaf as new cosmetic material.

Inhibition of Melanoma Differentiation by Melanogenesis Inhibitor Isolated from Yeast (효모에서 분리한 멜라닌 생성 억제 물질의 세포분화 억제)

  • Melanocytes synthesize melanin within discrete organelle termed melanosomes which are transferred to the surrounding keratinocytes and can be produced in varying sizes, numbers and densities. Skin whitening products have become increasingly popular in the past few years. The most successful natural skin whitening agents are: arbutin, vitamin C, kojic acid, and mulberry, which are all tyrosinase inhibitors. In this work, melanoston, a melanogenesis inhibitor isolated from yeast was studied to understand its mechanism of melanogenesis inhibition. It was found that melanoston was not a tyrosinase inhibitor, while when melanoston was applied to the B16 melanoma cell culture media, the intracellular tyrosinase activity was decreased by more than $30\%$. When B16 melanoma was stimulated with $\alpha$-MSH, cell morphololgy was dramatically changed to have lots of dendrites on the cell membrane surface. On the other hand, B16 was treated with $\alpha$-MSH and melanoston, simultaneously, the change of cell morphologv was not so great. This inhibitory effect of melanoston was found to be related to the inhibition of intracellar activation and transportation of tyrosinase, which was observed by irmmunostaining of B16 melanoma using anti-tyrosinase antibody. From these results, melanoston was regarded as an inhibitor to the differentiation of melanoma cells.

  • Particulate matter and ultra-particle matters generally refer to very small floating dust, such as 1/6 to 1/7 and 1/20 to 1/30, respectively, compared to the thickness of human hair, and contain various types of heavy metal ions. In addition to breathing, particle matters (PM) that flows in through the gaps in the pores of the skin can induce health problems in the body's tissues and skin, so it must be removed by blocking the inflow or by washing. Through this study, we confirmed the possibility that heavy metal ions can be adsorbed and removed by using Oryza sativa (Rice) bran oil (OSBO). In addition, the cell viability is much higher than that of grain-derived components through cytotoxicity experiments, and the cytoprotective effect of an external stimulus source can be expected. It was confirmed that the expression amount of COL1A1 mRNA increased, and accordingly, it was believed that wrinkles that might be caused by moisture lost by heavy metal ions in fine dust could be alleviated. Based on the results of these experiments, we tried to present a cosmetics containing OSBO, which is a wash-off formulation, in order to finally remove heavy metals.

  • Nanoparticles are substances that are smaller in size and smaller than cells that make up the skin. Therefore, they are very suitable as mediators for transmitting drugs or genes across cell membranes, and also deliver specific ingredients into the skin.In this study, nanoparticles were extracted from mugwort and particles of around 100 nm were obtained through dynamic light scattering (DLS), and the results of concentration-dependent enhancement of cell viability in fibroblasts were obtained through MTT assay. In addition, it was confirmed that the COL1A1 mRNA expression level was increased and the IL-6 mRNA expression level was decreased through the quantitative real-time PCR analysis method. Moreover, as these nanoparticles were confirmed to be stable, they can be applied not only to cell experiments but also to cosmetic formulations. While the demand for plant-derived ingredients continues to increase, excluding chemical ingredients from the recent cosmetics industry trend, there is a limitation in that there are few research results suggesting the application field of plant-derived nanoparticles. Therefore, in order to overcome the limitations of the cosmetic industry at the present time, the results obtained in this study present nanoparticles derived from Artemisia princeps (NDAP) as a highly functional cosmetic material.

Tyrosinase Inhibitory Activity and Melanin Production Inhibitory Activity of Taraxinic Acid from Taraxacum coreanum (흰민들레(Taraxacum coreanum)에서 분리한 taraxinic acid의 tyrosinase 활성저해 및 melanin 생성저해 효과)

  • This study was to investigate the Taraxinic acid from Taraxacum coreanum on tyrosinase activity and melanogenesis. In B16BL6 cell, Taraxinic acid did not show cytotoxicity even at concentrations of up to 100 ㎍/mL. In addition, tyrosinase inhibitory activity and melanogenesis inhibitory activity were confirmed by stimulation with α-melanocyte stimulating hormone (α-MSH) in the presence of taraxinic acid. Taraxinic acid was added to cells at concentrations of 10, 50 and 100 ㎍/mL and treated for 48 hours to confirm tyrosinase inhibitory activity and melanin production. The tyrosinase inhibitory activity increased in proportion to the amount of the sample, and showed an inhibitory activity of about 54.5% at a concentration of 50 ㎍/mL. Melanin production decreased in proportion to the sample amount, and it was about 62.2% at the concentration of 10 ㎍/mL. From the above results, it was found that Taraxinic acid had higher tyrosinase activity and melanin synthesis inhibitory activity in melanocyte than arbutin. The results suggest that Taraxinic acid can be utilized in natural whitening cosmetics.

Anti-cancer and Anti-microbial Effect of the Fraction Isolated from Pyrus ussuriensis Leaves (산돌배나무(Pyrus ussuriensis) 잎 분획물의 항암 및 항균활성에 관한 연구)

  • This study was conducted to confirm the application as ingredients of cosmetics through an examination of the function for anti-cancer and anti-microbial of the fraction isolated from Pyrus ussuriensis leaves. The dried leaf of P. ussuriensis were extracted with acetone-$H_{2}O$ (6:4, v/v), concentrated and fractionated with the upper layer of acetone on a separatory funnel. Each fraction was freeze dried, then a portion of acetone soluble powder was chromatographed on a Sephadex LH-20 column using a series of aqueous methanol as eluents and also used the MIC-gel using a series of aqueous methanol as developing solvent. The isolated compounds were identified by silica-gel TLC. The growth inhibition activity was measured using the MTT assay by the mouse meltioma (B16F10) cell. The cancer cell growth inhibition rate of fractions isolated from P. ussuriensis leaf was 80%. In anti-microbial activity test, the fraction of P. ussuriensis with 0.25 mg/disc resulted in the clear zone of 1.3 cm and 2 cm for Staphylococcus aureus and S. epidermidis of gram positive bacillus, respectively. In Escherichia coli of gram negative bacillus, the fraction with 0.5 mg/disc resulted in the clear zone of 1.1 cm~1.5 cm each fraction. From these results, we confirmed that acetate fraction of P. ussuriensis has a great potential as a natural ingredients with a anti-cancer and anti-microbial source.

Studies on the Antioxidant and Whitening Effects of Chamaecyparis obtusa Extract (편백나무(Chamaecyparis obtusa) 추출물의 항산화, 미백효과에 관한 연구)

  • This study was conducted as follows in order to find out the possibility of using (Chamaecyparis obtusa) by part as a cosmetic material. Chamaecyparis obtusa was separated leaves, bark, wood, and root, and extracted with 99.9% ethanol, and the antioxidant and whitening effects of the sample were confirmed. For this, the following studies were conducted. Antioxidant evaluation was confirmed by 1 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, 2 2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS+) radical scavenging activity. To evaluate the whitening effect, mushroom tyrosinase inhibitory activity evaluation and cytotoxicity evaluation of Chamaecyparis obtusa extract through MTT assay were conducted, and cellular tyrosinase inhibition rate was measured and melanin contents was confirmed. As a result of antioxidant activity, Bark extract among Chamaecyparis obtusa parts showed the best efficacy, and bark in B16F10 melanoma cell showed tyrosinase activity inhibition and melanin contents inhibitory effects. Therefore, it was confirmed that the ethanol extract of Chamaecyparis obtusa was developed as a natural material for safe and excellent whitening functional cosmetics.
