  • The objectives of this research were to study the effect of salt-shrinkage finishing of silk on shrinkage degree and dyeing property of cochineal, and to investigate the efficacy of obtained results for imparting surface design effect on silk fabric. Ultimately it was aimed to suggest a technical process for developing natural dyed silk products with diverse design. Premordanted silk fabric was treated with $Ca(NO_3)_2$ solution(gravity: 1.45) at $90^{\circ}C$ for 1 min, washed, dried for further evaluation. The shrinkage of salt-treated fabric was calculated. The effects of salt treatment on the dye uptake of cochineal and colorfastness were investigated. The degrees of shrinkage were 10% and 3% for warp and weft, respectively. The salt treatment resulted in improving dye uptake of cochineal slightly. In addition, it improved colorfastness to washing and light. On the basis of the results, a technical process composed of premordanting, salt treatment and natural dyeing was suggested and using the process, two examples of textile design were presented. It can be concluded to impart various three dimensional surface design effect on silk fabric by applying salt-shrinkage finishing with combination of natural dyeing and mordanting.

  • The comparison of Corylopsis coreana populations between natural groups and deforestation groups is carried in five regions such as Seungju, Woldeung, Hwangjeon in Suncheon, Myeongseok in Jinju and Mt. Baegun in Pocheon. The number of sprouting per both $100m^2$ and each individual stump were appeared more in deforestation than in natural groups. In contrast, the height and the diameter of individuals by the number of sprouting are higher and larger in the natural groups. The relationship between the floristic composition and vegetation in the two groups of C. coreana populations was not appeared. Corylopsis coreana are propagated sprouting as well as seedling in deforestation groups located an opening area. Because of the powerful environmental adaptability through growth of the sprouting and the seedling in deforestation groups, the population of C. coreana are not extinction of species oneself, and on the contrary are showed expansion of C. coreana population after temporary disturbance.

  • In order to get basic data for flood plain restoration, we surveyed the woody vegetation in Korean natural rivers and analyzed the species' characteristics with regards to patterns of richness and diversity. These characteristics were higher in hard wood forests than those in soft wood forests, such as Salix spp. community. Futhermore, they were the highest in the Prunus sargentii-Pinus densiflora community(H' 1.095), and the lowest in the Carpinus laxiflora community(H' 0.118) among the hard wood forests. Species' richness diversity were the highest in the Salix gracilistyla community, but the lowest in the S. koriyangi community or S. koreensis community among the soft wood forests. With regards to the dominant index, just one community is over 0.9, 13 communities are between 0.3-0.7 and 15 communities are less than 0.3. The Salix koreensis community was the highest at 0.931, and Prunus sargentii-Pinus densiflora community was the lowest at 0.13. Species' richness and diversity was significantly correlated with tree layer coverages and degree of slope. These results mean that in order to increase plant species diversity in flood plains planted hard woody trees, such as oaks and fir, are needed to suit environmental conditions with steeper slope and lower canopy coverage.

  • Phragmites australis is the dominant and constructive species among plant communities in the wetlands of the downstream of Yellow River, China. Its morphological characters were high variable in different habitats. Studies on Morphological and RAPD variation of 15 P. australis populations from this region showed that soil salinity was the dominant ecological factor that affected the morphological characters of P. australis. The basal diameter, height, leaf length, leaf width, internode length, internode accounts, panicle length were negatively related to salinity. 194 loci were amplified by RAPD, of which 9 loci was highly negative-related to salinity, and showed a tendency to prefer the habitats with fresh water. 4 loci were positively related to the salinity, and showed a tendency to prefer the salinized habitats. Most loci were neutral to salinity. The morphological and genetic characters of BZH were special, and the speciality should not be determined by salinity. The morphological characters were affected by genetic information and environment. The morphological characters should change gradually and continuously along environmental gradient under plasticity, but should changed continuously or not in genetic control. The relevancies among quantitive characters, ecological factors and genetic variation in natural populations still will still be a focus and difficulty of ecological genetics of P. australis in the future.

  • There is a host of habitat assessment systems available to evaluate and grade tidal flat ecosystems in Korea. Nevertheless, we recognize the need to develop a better ecological scheme to improve reliability and accuracy given the importance of using an empirical approach in assigning grades to indicators and the limitation of current systems which evaluate tidal flats on a regional basis. A preferable system would be one that enables habitat mapping within a tidal flat, provides a diagnosis of ecological stress/health, has a high level of ecological relevance, communicability, and statistical robustness, and enables evaluation of diverse habitats within a tidal flat. The following points should be considered in framing such a system. Indicators should discriminate habitat quality into five to seven grades accomodating diverse characteristics and conservation value of a habitat, and the grading should be made from frequency distribution of indicators based on nationwide data.We suggest the following tasks in crafting such a system: i) The decision on selection of indicators should be made based on accuracy of assessment and practical application; ii) effects of physical habitat conditions on variability of indicators should be reflected; and, iii) further works on stress/health indicators should be addressed for adopting a multiple-indicator approach which reduces misdiagnosis.

  • Freshly harvested seed of Chinese milk vetch (Astragalus sinicus L.; CMV) was strongly dormant because of hardseedness. Seeds of freshly harvested germinated only 8% while clipping the seed coat completely overcome the innate dormancy, which indicates inhibition of germination of the seed is mainly due to seed coat (87%). The dormant (intact) hard seeds did not imbibe water whereas the non-dormant (clipped) seeds took up rapidly. In natural environment condition, the hard seed coat dormancy was broken only after 5 months after seed harvest. To break such a strong seed coat dormancy, the chemical and heat treatments were effective. Concentrated sulfuric acid was more effective than dry heat and hot water treatments. Hot water treatment improved germination but the germination percentage was less than 41%. Treatments increased germination due to its effect on the seed coat integrity. A scanning electron microscope reveled that disruption of seed coat layers and subsequent development of numerous crack in the hilum region of the seed and on the seed coat surface of concentrated sulfuric acid treatment and formation of cracks in the dry heat treatments, respectively, were observed in the seed coat surface, which served as water entry points.

  • Recently, pest-resistant living modified (LM) crops developed using RNA interference (RNAi) technology have been imported into South Korea. However, the potential adverse effects of unintentionally released RNAi-based LM crops on non-target species have not yet been reported. Coccinella septempunctata, which feeds on aphids, is an important natural enemy insect which can be exposed to the double-stranded RNA (dsRNA) produced by RNAi-based LM plants. To assess the risk of ingestion of Snf7 dsRNA by C. septempunctata, we first identified the species through morphological analysis of collected insects. A method for species identification at the gene level was developed using a specific C. septempunctata 12S rRNA. Furthermore, an experimental model was devised to assess the risk of Snf7 dsRNA ingestion in C. septempunctata. Snf7 dsRNA was mass-purified using an effective dsRNA synthesis method and its presence in C. septempunctata was confirmed after treatment with purified Snf7 dsRNA. Finally, the survival rate, development time, and dry weight of Snf7 dsRNA-treated C. septempunctata were compared with those of GFP and vATPase A dsRNA control treatments, and no risk was found. This study illustrates an effective Snf7 dsRNA synthesis method, as well as a high-concentration domestic insect risk assessment method which uses dsRNA to assess the risk of unintentional released of LM organisms against non-target species.

  • In this study, carbon storage in the aboveground biomass, litter layer, and soil layer was calculated for abandoned mining restoration areas to determine the level of carbon storage after the restoration project through comparison with the ecological reference. Five survey sites were selected for each abandoned mining restoration area in Boryeong-si, Chungcheongnam-do, and the ecological reference that can be a goal and model for the restoration project. The carbon storage in the restoration area was 0~21.3Mg C ha-1, the deciduous layer 3.3~6.0Mg C ha-1, and the soil layer(0-30cm) 8.3~35.1Mg C ha-1, showing a significant difference in carbon storage by target site. The total carbon storage was between 6.1 and 35.3% of the ecological reference, with restoration area ranging from 14.0 to 62.4 Mg C ha-1. The total carbon storage in the restoration area and the ecological reference differed the most in the aboveground biomass and was less than 12%. Based on these results, forest restoration area need to improve the carbon storage of forests through continuous management and monitoring so trees can grow and restore productivity in the early stages of the restoration project. The results of this study can be used as primary data for preparing future forest restoration indicators by identifying the storage of abandoned mining restoration areas.

  • Bats serve as vectors and natural reservoir hosts for various infectious viruses, bacteria, and fungi. These pathogens have also been detected in bat feces and can cause severe illnesses in hosts, other animals, and humans. Because pathogens can easily spread into the environment through bat feces, determining the bacterial communities in bat guano is crucial to mitigate potential disease transmission and outbreaks. This study primarily aimed to examine bacterial communities in the feces of insectivorous bats living in South Korea. Fecal samples were collected after capturing 84 individuals of four different bat species in two regions of South Korea, and the bacterial microbiota was assessed through next generation sequencing of the 16S rRNA gene. The results revealed that, with respect to the relative abundance at the phylum level, Myotis bombinus was dominated by Firmicutes (47.24%) and Proteobacteria (42.66%) whereas Miniopterus fuliginosus (82.78%), Rhinolophus ferrumequinum (63.46%), and Myotis macrodactylus (78.04%) were dominated by Proteobacteria. Alpha diversity analysis showed no difference in abundance between species and a significant difference (p < 0.05) between M. bombinus and M. fuliginosus. Beta-diversity analysis revealed that Clostridium, Asaia, and Enterobacteriaceae_g were clustered as major factors at the genus level using principal component analysis. Additionally, linear discriminant analysis effect size was conducted based on relative expression information to select bacterial markers for each bat species. Clostridium was relatively abundant in M. bombinus, whereas Mycoplasma_g10 was relatively abundant in R. ferrumequinum. Our results provide an overview of bat guano microbiota diversity and the significance of pathogenic taxa for humans and the environment, highlighting a better understanding of preventing emerging diseases. We anticipate that this research will yield bioinformatic data to advance our knowledge of overall microbial genetic diversity and clustering characteristics in insectivorous bat feces in South Korea.
