  • Quantitative trait locus (QTL) is a particular region of the genome containing one or more genes associated with economically important quantitative traits. This study was conducted to identify QTL regions for body weight and growth traits in purebred Korean native chicken (KNC). F1 samples (n = 595) were genotyped using 127 microsatellite markers and 8 single nucleotide polymorphisms that covered 2,616.1 centi Morgan (cM) of map length for 26 autosomal linkage groups. Body weight traits were measured every 2 weeks from hatch to 20 weeks of age. Weight of half carcass was also collected together with growth rate. A multipoint variance component linkage approach was used to identify QTLs for the body weight traits. Two significant QTLs for growth were identified on chicken chromosome 3 (GGA3) for growth 16 to18 weeks (logarithm of the odds [LOD] = 3.24, Nominal p value = 0.0001) and GGA4 for growth 6 to 8 weeks (LOD = 2.88, Nominal p value = 0.0003). Additionally, one significant QTL and three suggestive QTLs were detected for body weight traits in KNC; significant QTL for body weight at 4 weeks (LOD = 2.52, nominal p value = 0.0007) and suggestive QTL for 8 weeks (LOD = 1.96, Nominal p value = 0.0027) were detected on GGA4; QTLs were also detected for two different body weight traits: body weight at 16 weeks on GGA3 and body weight at 18 weeks on GGA19. Additionally, two suggestive QTLs for carcass weight were detected at 0 and 70 cM on GGA19. In conclusion, the current study identified several significant and suggestive QTLs that affect growth related traits in a unique resource pedigree in purebred KNC. This information will contribute to improving the body weight traits in native chicken breeds, especially for the Asian native chicken breeds.

  • Food and agricultural production sector, especially livestock production is vital for Mongolia's economic and social development. Domestic sheep play key roles for Mongolians, providing food (meat, milk) and raw materials (wool, sheepskin), but genetic diversity, origin of sheep populations in Mongolia have not been well studied. Studies of population genetic diversity is important research field in conservation and restoration of animal breeds and genetic resources. Therefore, this study aimed to investigate genetic characteristics and estimate origin through the analysis of mitochondrial DNA control region D-loop and Cytochrome b of Mongolian indigenous sheep (Mongolian native, Orkhon and Altanbulag) and one Europe sheep (Suffolk). As a result of there were found, 220 SNPs (Single nucleotide polymorphism) in the D-loop region, 28 SNPs in the Cytochrome B region, furthermore, 77 Haplotypes. The nucleotide diversity was only found in D-loop region (n = 0.0184). Phylogenetic analysis showed that 3 (A, B, and C) of 5 haplogroups of sheep have been identified in our research. Haplogroup C was only found in Mongolian indigenous sheep. Haplogroup D and E were not observed. As a result of haplogroups, haplogroup A was dominant (n = 46 of 94 sheeps), followed by haplogroup B (n = 36) and haplogroup C (n = 12). Sequence analysis showed that T deletion, insertion and heteroplasmy in D-loop region occurred at a high rate in Mongolian indigenous sheep population (T insertion = 47, T deletion = 83). The heteroplasmy, which has never been found in Mongolian sheep, has been newly discovered in this study. As a result, the Mongolian sheep varieties, which mainly derived from Asia, were in hybridization with European sheep varieties.

  • The peroxisome proliferator-activated receptor gamma coactivator-1 alpha protein, encoded by the PPARGC1A gene, plays an important role in energy homeostasis. The genetic variations within the PPARGC1A gene promoter region were scanned in 808 Chinese native bovines belonging to three cattle breeds and yaks. A total of 6 SNPs and one 4 bp insertion variation in the promoter region of the bovine PPARGC1A gene were identified: SNP -259 T>A, -301_-298insCTTT, -915 A>G, -1175 T>G, -1590 C>T, -1665 C>T and -1690 G>A, which are in the binding sites of some important transcription factors: sex-determining region Y (SRY), myeloid-specific zinc finger-1 (MZF-1) and octamer factor 1(Oct-1). It is expected that these polymorphisms may regulate PPARGC1A gene transcription and might have consequences at a regulatory level.

  • The International Union for Conservation of Nature and the Korean Red Data Book both categorize the dragon swallowtail Sericinus montela Grey, 1852 (Lepidoptera: Papilionidae) as a vulnerable insect. In South Korea, the main reason for this categorization is habitat destruction and deterioration, along with limited distribution mainly to the eastern region of South Korea, particularly to Gangwon-do and Gyeongsangbuk-do. Considering much of the information is older than ten years the renewed survey to estimate distributional stability is required, particularly under global warming. In addition, the population genetic diversity and isolation of S. montela, which is important information when seeking to evaluate its vulnerability, is not yet known. In this study, we visited 38 habitable sites, consisted of 15 sites with the previous record for the sightings of the species and 23 sites without such record. Moreover, nuclear ribosomal internal transcribed spacer region was sequenced for 56 individuals collected from seven sites. We observed S. montela at 20 sites, consisted of 11 recorded and 9 new sites in seven provinces, excluding Jeollanam-do and Jeju-do. Finding of the species in 9 new sites and location of the 4 in western region, which has a rare previous record are promising. However, failure to observe in 4 recorded sites and an extremely low observed number in new sites (5.3 vs. 17.6 individuals per site on average) is not optimistic. Population genetic data partially corroborate with the field observation data, showing that the populations in western region have lower genetic diversity and intermittent genetic isolation compared to those in eastern region. Taken these together, we suggest that S. montela should be remained as a vulnerable insect. However, more extensive field monitoring and co-dominant molecular data are required to verify this conclusion.

  • Patanga japonica Bolívar, 1898 (Orthoptera: Acrididae) is listed as a climate-sensitive indicator species in South Korea and is called southern group of insects in that the main distributional range is southern region of South Korea and Asian continent. In South Korea, thus, the species was distributed mainly in southern region of South Korea including southward a remote Jeju Island, but recently the species has often been detected in mid to northern region of South Korea, implying northward range expansion in response to climate change. Understanding the characteristics of the changes in genetic diversity during range expansion in response to climate change could be a foundation for the understanding of future biodiversity. Thus, in this study, we attempted to understand the changing pattern of the genetic diversity of the P. japonica in newly expanded regions. For the purpose of study, we collected 125 individuals from seven localities throughout South Korea including two newly distributed regions (Pyeongtaek and Yeongwol at ~37° N). These were sequenced for a segment of mitochondrial cytochrome oxidase subunit I (COI) and analyzed for genetic diversity, haplotype frequency, and population genetic structure among populations. Interestingly, northward range expansion accompanied only haplotypes, which are most abundant in the core populations, providing a significant reduction in haplotype diversity, compared to other populations. Moreover, genetic diversity was still lower in the expanded regions, but no genetic isolation was detected. These results suggest that further longer time would take to reach to the comparable genetic diversity of preexisting populations in the expanded regions. Probably, availability of qualified habitats at the newly expanded region could be pivotal for successful northward range expansion in response to climate change.

  • This study was conducted to investigate novel genetic variations of MCIR^*3 allele. In general, white spotting or white belt on a black backgroud in pigs is determined by the E$^p$ allele at the MCIR/Extention locus. E$^p$ shares a frameshift mutation with the E$^{D2}$ allele for dominant black color. An oligonucleotide primer set was designed to amplify complete coding sequence of the porcine MCIR gene. The MCIR coding sequences obtained from five breeds those were Landrace(white). Yorkshire(white), Hampshire(belt), Berkshire(spot) and Jeju native black pigs(black), were used for this study. A multiple sequence alignment of the MCIR coding region using Clustal W was performed. The total length of the MCIR coding sequence ranged from 963 to 966 base pairs(bp) among the selected breeds. The sequence analysis of the complete coding region of MCIR was revealed that Hampshire and Jeju native black pig have 3 cytosines deletion and Birkshire has 2 cytosines deletion at codon 23(nt68) in Extention loci. Besides the finding, there were three different missense mutations and a frameshift mutation in the MCIR coding region.

  • This study is aimed at establishing the categories and items for ecological assessment and evaluation of the environmental friendliness of golf courses in the capital region of Korea. The categories and items for the assessment have been derived based on the existing literature and interviews with golf experts. This study covers 32 golf courses in the capital region of Korea that are available in terms of data and on-site surveys. In order to conduct a comprehensive assessment of the environmental friendliness of the golf courses, the assessment area was divided into 4 categories that include a total of 14 sub-categories. The 4 categories encompass 1) location, 2) topography, 3) vegetation, and 4) construction. As its sub-categories, the location category includes current land use and zoning in the National Land Use Management Law. Topography has 2 sub-categories in the damage ratio of existing topography, gradient, cut area, and slope height. The assessment of vegetation is largely based on site surveys in the categories of preservation of the existing vegetation, the use of natural resources and existing trees, the component ratio of native tree species, the multi-layered structure of vegetation, and the utilization of water purification plants. In the aspect of construction, afforestation on tile slopes and the utilization of existing surface soil were evaluated. The examination of comparative analysis among the 10 items as a ratio measure showed that the scores were low in the sub-categories of current land we, the use of existing trees, and the multi-layered structure of vegetation. However, the rating results were satisfactory in the 2 sub-categories including cut area, and the utilization of native tree species. Those proved to be contributing factors in the ecological health of the golf courses. According to correlation analysis of the 10 items to the overall ecological rating of each golf course, the sizes of the 32 golf courses were mainly affected by the damage ratio of existing topography, gradient, preservation of vegetation and slope height. This study has the initiative to conduct an ecological assessment of golf courses in the country based on site surveys. The study results revealed that location factors such as current land use, damage ratio of topography and gradient and topographical factors were the main factors affecting the environmental friendliness of golf courses. This indicates indicating the significance of these factors in the future construction practices of golf courses. Furthermore, this study raises the need for follow-up studies to establish more detailed assessment criteria and to develop assessment techniques for areas such as slope afforestation and water purification plants that need a qualitative approach.

  • This experiment was carried out to obtain the maximum quantity of essential oil on the basis of harvesting time, cultivation regions & types, and drying methods. We have selected 6 native and introduced herbs viz. $Thymus$ $quinquecostatus$, $Agastache$ $rugosa$, $Chrysanthemum$ $indicum$, $Rosmarinus$ $officinalis$, $Chamaemelum$ $nobile$, and $Lavandula$ $stoechas$. The quantity of essential oil ranged from 0.06% to 3.46% in all six herbs. Native herbs produce 30.5% higher quantity of oil in comparison to introduced herbs. The quantity of essential oil obtained from $Lavandula$ $stoechas$ was 3.46%, followed by $Rosmarinus$ $officinalis$ 2.89%, while minimum in $Agastache$ $rugosa$ 0.60%. Higher quantity of essential oil obtained if they are cultivated under rain shelter culture (PE film) in comparison to open field conditions. Recovery of essential oil is always high if herbs flower in July. Semi-alpine region was found better than the plain region for herbs production. Quantity of oil is high if oil is extracted from fresh herbs followed by freezed herbs, shade dry and hot wind dry herbs. As far as plant part is concern flower produced maximum oil than any other part.
