To investigate heavy metals (Hg, Pb and Cd) and their potential health risks in commercial dried laver (Porphyra sp.), we collected 45 samples from the major production areas on the western and southern coasts of Korea (Hwaseong, Seocheon, Gunsan, Muan, Shinan, Jindo, Haenam, Wando, Jangheung, Goheung and Busan). The Hg, Pb and Cd concentrations were measured using inductively coupled plasma spectrometry (ICP-MS) or a mercury analyzer. The average Hg, Pb and Cd concentrations in the dried laver were $0.006{\pm}0.0017$, $0.196{\pm}0.0614$ and $0.894{\pm}0.4882$ mg/kg, respectively. Based on the 2007 Korean Public Nutrition Report, these levels are 0.02, 0.11 and 2.47% of the provisional tolerable weekly intake (PTWI) for Hg, Pb and Cd, respectively, established by the FAO/WHO. The hazard quotient (HQ) determined from the ratio of exposure and safe levels were less than 1.0. Therefore, the levels of overall exposure to Hg, Pb and Cd for dried laver were below the recommended JECFA (Joint FAO/WHO Expert Committee on Food Additives) levels, which indicate safe levels for public health.

Asparagus oligoclonos is one of the endemic halophytes used folk medicine in Korea. We isolated the main compound rutin from methanol extracts of A. oligoclonos based on nuclear magnetic resonance and TOF ESI-MS data. We have investigated the quantitative analysis method of main compound using HPLC and the results exhibit that rutin content of A.oligoclonos were 1.816%. To explore anti-oxidant from A. oligoclonos ethanol extracts (AOE), we investigated the antioxidant effects of AOE on $H_2O_2$-induced oxidative stress in RAW 264.7 cell. AOE were reduced $H_2O_2$-induced oxidative stress via enhancement of cell viability, and AOE significantly decreased ROS production depending on concentration. Next, to screen for anti-inflammatory activity of AOE, we investigated the inhibitory effects of AOE in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. AOE had no effect on cell viability at a concentration of $500{\mu}g/mL$. Nitric oxide (NO) production inhibited in a dose-dependent manner. These results suggest that AOE may be a useful antioxidant and anti-inflammatory agent.
This study was conducted to confirm the possible use of female Yangwonjam as a host for synnemata production of Isaria tenuipes in eight local areas in Korea. Silkworm pupation rate, infection rate and synnemata characteristics of I. tenuipes were examined. Normal silkworms had a higher pupation rate than silkworms inoculated with I. tenuipes. The pupae survival percentage of normal silkworm in cocoons was 92.5~97.6%, whereas it ranged from 91.1~95.6% in silkworms sprayed with I. tenuipes. Female Yangwonjam showed the highest survival percentage at 97.6% among the silkworm varieties tested. I. tenuipes infection rate of larvae of 5th instar newly-exuviated silkworms was 89.2~90.7% in the spring rearing season and 98.2~99.3% in the autumn rearing season. Synnemata production of I. tenuipes was excellent in female Yangwonjam with an incidence rate of 98.0% followed by male Yangwonjam (94.1%) and Baegokjam (93.3%) in the spring rearing season. Synnemata living weight ranged from 1.44~0.94 g in the spring rearing season. The female Yangwonjam had the heaviest synnemata weight (1.44 g) in the spring rearing season. The synnemata of I. tenuipes produced on pupae were white or milky-white in color, and were similar in shape and color to wild synnemata collected in Korea.

Injection inoculation protocols for fruit body formation of Cordyceps militaris (C. militaris) were investigated to improve the incidence of infection in the silkworm species Bombyx mori (B. mori). Injection, with suspensions of C. militaris hyphal bodies into living silkworm pupae, was used to test for fruit body production. Use of Daeseungjam rather than Baegokjam or Keumokjam varieties of B. mori is thought to be suitable for infection by C. militaris. From mounting, nine-day-old to 11-day-old pupae showed the best incidence of infection with a $100\;{\mu}L$ injection volume. Silkworm pupae injected with a hyphal suspension concentration of more than $2\;{\times}\;10^5$ colony-forming unit (cfu) recorded a greater than 96% incidence of infection. Also, fruit bodies of C. militaris were induced and produced at a light intensity between 500 and 1,000 lx.

The ${\gamma}$-oryzanol, ferulic acid esters, which are well-known for their function on cholesterol lowering and prevention of inflammation, diabetes and cancer, are found in the seeds of cereal crops such as rice, corn, wheat and rye. Among them, rice has been reported to contain the highest ferulic acid esters. Since rice cultivation with green manure as a N source is an environmental friendly agricultural practice, it is necessary to identify and quantify as well as evaluate the variations in these compounds in rice samples as affected by different green manure conditions. A total of ten components of ${\gamma}$-oryzanol were isolated and cycloartenyl ferulate, 24-methylenecycloartanyl ferulate, campesteryl ferulate and sitosteryl ferulate were identified as the major components in Korean rice cultivars, 'Unkwang' and 'Hopum'. Comparing the ${\gamma}$-oryzanol contents of these varieties, 'Unkwang' showed clearly similar pattern with conventional type. With the PLS-DA (partial least squares of discriminant analysis) using SIMCA 11.0 ver., the specific pattern and cluster of ${\gamma}$-oryzanol scores with green manure conditions were confirmed, and thus distinguishing green manure effects were possible.
Five mutants were investigated at the molecular level to determine the factors responsible for mutated endosperm types. They were classified as high (HA) or low amylose (LA) phenotypes based on the amylose content in endosperm. The five were previously produced from Ilpum and Shindongjin cultivar treated with N-methyl-N-nitrosourea and gamma-ray irradiation, respectively. Analysis of the genomic structure and expression of Granule-bounded Starch Synthase I (GBSSI) genes revealed that mutants generally showed a higher incidence of nucleotide transition than transversion, and the $A:T{\rightarrow}G:C$ transition was particularly prevalent. The rates of nucleotide substitution in HA mutants were generally higher than those in the LA mutants, leading to higher substitutions of amino acid in the HA mutants. Neither nucleotide substitutions interfering with intron splicing or causing early termination of protein translation were found, nor any large-sized deletions or additions were found in all the mutants. In principle, amylose content can be regulated by three factors: internal alterations of GBSSI protein, the strength of gene expression, and other unknown external factors. Our results indicate that the endosperm mutants from Shindongjin arose from internal alterations of GBSSI proteins, which may be the result of amino acid substitutions. On the other hand, the Ilpum mutants might be principally caused by the alteration of gene expression level. Analysis of another three glutinous cultivars revealed that the major factor leading to glutinous phenotypes is the 23-bp duplicative motif (5'-ACGGGTTCCAGGGCCTCAAGCCC-3') commonly found in exon 2, which results in the premature termination of protein translation leading to the production of a non-functional GBSSI enzyme.

In a previous work, we performed de novo RNA sequencing of Allomyrina dichotoma using next generation sequencing and identified several antimicrobial peptide candidates based on transcriptome analysis. Among them, a cationic antimicrobial peptide, allomyrinasin, was selected bioinformatically based on its physicochemical properties. Here, we assessed the antimicrobial and anti-inflammatory activities of allomyrinasin against microorganisms and mouse macrophage Raw264.7 cells. Allomyrinasin showed antimicrobial activities against various microbes and decreased the nitric oxide production of the lipopolysaccharide-induced Raw264.7 cells. Furthermore, quantitative RT-PCR and ELISA revealed that allomyrinasin reduced cytokine expression levels in the Raw264.7 cells. We also identified inducible nitric oxide synthase, cyclooxygenase-2 expression, and $PGE_2$ production through western blot analysis and ELISA. We confirmed that allomyrinasin bound to bacterial cell membranes via a specific interaction with lipopolysaccharides. Taken together, these data indicate that allomyrinasin has antimicrobial and anti-inflammatory activities as exemplified in lipopolysaccharide-induced Raw264.7 cells. We have provided a potentially useful antimicrobial peptide candidate that has both antimicrobial and anti-inflammatory activities.
There is an increasing frequency in the occurrence of abnormal weather phenomena such as sharp increases and decreases in temperature. Under these weather conditions, the heading time of rice changes unexpectedly, which poses problems in agriculture. Therefore, we investigated the effect of temperature on the heading response at different growth stages in rice. During the period from transplanting to heading, the plants were subjected to different temperature treatments, each for a 9-day period, to observe the heading response. For the heading date analysis, "heading date" was defined as the number of days from transplanting to the appearance of the first spikelet. We found that the influence of temperature increased in the order of rooting stage, followed by meiosis, early tillering, spikelet differentiation, and panicle initiation stage in all ecological types and cultivars. In particular, unlike the results reported previously, the effect of temperature on heading during the photo-sensitive period was very small. Meanwhile, the influence of temperature on vegetative growth response at different growth stages was not consistent with heading response. These results can be used as basic data for predicting the variation in heading date owing to temperature variation at each growth stage. In addition, we propose that the concept of day length should be included in determining the influence of temperature on the photo-sensitive period.
A wide range of techniques have been developed to separate X or Y- chromosome-bearing sperm. In particular, bovine semen sex-sorted by using flow cytometry based on differences in the amount of DNA between X and Y chromosome bearing sperm is used in dairy farms. The first piglets were produced using sex-sorted sperm 30 years ago. However, sexed sperm have not been commercially available in pigs because the flow cytometry technique is not capable of sorting the high number of sperm required for porcine artificial insemination (AI), and the prolonged exposure to an electrical filed might damage to the DNA in sperm. The purpose of this study was to evaluate a boar sperm sorting method based on magnetic nanoparticles. A flow cytometer assay verified the efficacy of the magnetic nanoparticles (> 90% of sex-sorted sperm). In addition, a duplex polymerase chain reaction (PCR) assay using sex chromosome specific genes including SRY (sex-determining region Y; male), ZFY (zinc finger protein Y-linked; male), and ZFX (zinc finger protein X-linked; female) showed that in vitro fertilized porcine embryos by X and Y-chromosome bearing sperm were 100% female (40/40) and 72% female (35/48), respectively, at 8-cell or morula stages, suggesting that the sex-sorted sperm were fertile. In conclusion, our findings suggest that the sex-sorted method based on magnetic nanoparticles can be utilized for porcine sex-sorted AI.
