Botanical antimicrobial agent-grapefruit seed extract mixture (BAAG) have an unknown compounds which exhibit the antibiotic activities aganist microorganisms including bacteria and fungi. We have examined the effects of BAAG on the physiological function of Botrytis cinerea which was isolated from necrotic lesions of decayed fruits and vegetables such as cucumbers, grapes, tomatoes, and red peppers during storage. In the results of enzymatic activities related to the energetic metabolism there was no inhibitory effect of BAAG on the activities of several enzymes in vitro including glucose 6-phosphate dehydrogenase and malate dehydrogenase, while there was inhibitory effect of BAAG on the activities of hexokinase and succinate dehydrogenase. O-nitrophenyl-$\beta$-D-galactopyranoside(ONPG), the artificial substrate of $\beta$-galactosidase was hydrolyzed in the presence of BAAG, indicating that the membrane was pertubated by the BAAG. From the results we suggested that the antibiotic activity of BAAG is due to the change of membrane permeability of the cell. BAAG was fractionated and purified by silica gel and sephadex column chromatography. Among active fractions two peaks were identified as naringin and limonin when they were analyzed by by NMR and Fast atomic bombardment.

Response surface methodology was employed to investigate the change of flavonoids components of citrus peel hydrolysate using Viscozyme L as the enzyme. As citrus peels were hydrolyzed by the enzyme, hesperetin and naringenin contents of flavonoids aglycone form increased. The optimal enzyme treatment conditions which were superimposed of the maximized levels for soluble solid, hesperetin, and naringenin contents were enzyme concentration of 1.5% and reaction time of 18 hr. In enzyme-untreated citrus peels (CC), soluble solid content was 48.49% and the content of hesperidin only detected flavonoids was 58.85 mg/g. In the case of optimal enzyme-treated citrus peels (CE), soluble solid content was 72.97% and the contents of naringin, hesperidin, naringenin and hesperetin were 1.56 mg/g, 31.31 mg/g, 2.58 mg/g and 3.90 mg/g, respectively. In the results of electron donating ability and angiotensin converting enzyme inhibition activity, the activity of CE was higher than that of CC.

In this study, we investigated the change of antioxidant activity and flavonoid contents by fermentation of Citrus grandis Osbeck peel (CGP) using the Saccharomyces cerevisiae (KCCM35053), comparing to unfermented CGP. Total flavonoid content in the fermented Citrus grandis Osbeck peel (FCGP) was 3,768 g/100 g sample and higher than that of CGP. The antioxidant activities of FCGP was determined by DPPH, hydroxyl, alkyl radicals, and hydrogen peroxide scavenging assays. FCGP showed higher activities than CGP in all scavenging assays. The $IC_{50}$ values of FCGP were 261.3 ${\mu}g$/mL for DPPH; 1,474 ${\mu}g$/mL for hydroxyl; 90.9 ${\mu}g$/mL for alkyl and 1,195 ${\mu}g$/mL for $H_2O_2$ in respective scavenging assays. Flavonoid compositions of both samples were determined by liquid chromatography/mass spectrometry (LC/MS). In the spectrum FCGP was similar to CGP in the contents of neohesperidin, naringin and an unknown No. 7 compound, but some unknown compounds (No. 1, 2, 4, 5, 6) were higher than CGP in each flavonoid contents. Therefore, the fermentation of CGP could increase the contents of unknown compound and improved antioxidant activities.
Agaricus brasiliensis, belonging to the family Agaricaceae of Agaricales, Basidiomycota, has been used for edible and medicinal purposes. This study was initiated to evaluate the antioxidant, anti-diabetic, and nitric oxide inhibitory activities of fruiting bodies of A. brasiliensis extracted with methanol. The HPLC analysis of phenolic compounds from the mushroom extracts identified 10 phenolic compounds including gallic acid, procatechuic acid, chlorogenic acid, (-)-epicatechin, vanillin, rutin hydrate, naringin, quercetin, formononetin, and biochanin-A. The free radical scavenging activities of methanol extract were lower than that of positive control, BHT. The chelating effects of methanol extract were significantly higher than those of the positive control, BHT at the all concentrations tested. The methanol extract exhibited the lower reducing power activities compared with the positive control at the 0.5~6.0 mg/mL concentration. The mushroom extract inhibited the ${\alpha}$-glucosidase activity by 54.48% and 78.43% at the 1.0 and 2.0 mg/mL while acarbose, the positive control, inhibited the ${\alpha}$-glucosidase activity by 51.77% and 81.81% at the same concentrations, respectively. Nitric oxide (NO) production in lipopolysaccahride (LPS) induced RAW 264.7 cells were inhibited by the methanol extracts in a concentration dependent manner. Therefore, it is concluded that fruiting bodies of A. brasiliensis contained natural antioxidant, anti-diabetic, and anti-inflammatory substances which can be useful for human health.

When I evaluated the component variation of Naesowhajung-tang by the three types of extraction method, and each type's effects on the gastrointestinal tract, I got the following results. 1. The output ratio of extracts made out of Naesowhajung-tang were not significantly different among 13.8% of water extract(Sample-I), 13.5% of 50% ethanol extract(Sample-II), and 15.6% of water extract by spray dryer(Sample-III). 2. magnolol, honokiol, hesperidin, naringin, poncirin and glycyrrhizin Sample II had the largest amount of the following contents: magnolol, honokiol, hesperidin, naringin, poncirin and glycyrrhizin. 3. All the extracts of Naesowhajung-tang showed the inhibitory effect on the smooth muscle contraction of the isolated ileum in mice and fundus-strip in rats induced by acetylcholine chloride and barium chloride. 4. High concentration Sample-II was recognized to be effective in preventing gastric ulcers in Shay's rats. but not in the other rat group. 5. All the extracts of Naesowhajung-tang were recognized to be effective in preventing gastric ulcers induced by Ethanol-HCl in rats. 6. The increase of transport ability in the small intestine was recognized only when the concentration of all the samples was doubled, but not in the other concentrations. 7. The increase of transport ability in the large intestine was recognized only when the concentration of Sample-II was doubled, but not in other concentration. Using the results mentioned above, I suggest that Sample-II has more significant effects on the gastrointestinal tract than the others.
Overproduction of peroxynitrite ($ONOO^-$) causes a variety of disease such as atherosclerosis, hypercholesterolemia, diabetes mellitus or obesity. Peroxynitrite scavenging activities and HPLC analysis on the five Oriental medicinal drugs belonging to the genus Citrus, Aurantium or Poncirus (Rutaceae family) and HPLC analysis were taken to evaluate flavanone glycosides with peroxynitrite scavenging activity. The $IC_{50}s$ of the five crude drugs were shown as follows: Aurantii nobilis Pericarpium (Jinpi, 18.3 ${\mu}g$/ml), Citrii unshiu Pericarpium (Chungpi, 7.50${\mu}g$/ml), Citrii unshiu Semen (Gyulhaek, >50.0${\mu}g$/ml), Aurantii Fructus (Jigak, 18.3${\mu}g$/ml), and Poncirii Fructus (Jisil, >50.0${\mu}g$/ml) where Korean crude drug's names are noted in the parenthesis. Peroxynitrite scavenging effect of flavanones or their glycosides usually contained in Citrus species were observed as follows: hesperetin (1.89 ${\mu}g$/ml), naringenin (7.77 ${\mu}g$/ml), hesperidin (8.44 ${\mu}g$/ml), poncirin (>50.0 ${\mu}g$/ml)and ponciretin(>50.0 ${\mu}g$/ml). The activities of naringin and poncirin with ${\alpha}$-L-rhamnopyranosyl($1{\rightarrow}2$)-${\beta}$-D-glucopyranosyl moiety were weak. HPLC analytical data revealed that Jinpi (the peels of mature fruits of Citrus unshiu) and Chungpi (the peels of immature fruits of C. unshiu) had high quantities of hesperidin as the value of 142.1${\pm}$0.21 and 104.51${\pm}$1.10 mg/g dried weight, respectively. Poncirin was clearly detected in only Jisil and naringenin and naringin were not observed on the HPLC chromatogram of the five crude drugs.

To investigate the usefulness of natural plants, physicochemical characteristics, functional components and physiological activities of dodder(Cuscuta australis R.Brown) were experimented. The extraction of dodder was carried out at 60℃ in shaking bath with 10 times of distilled water for three hours. Harmful metals such as lead, cadmium and arsenic were not detected, but the content of mercury from dodder was 0.0004ppm in acceptable standards. The extraction yield was 23.45%, total content of phenolics equivalent to gallic acid was 699.8 mg/100g and total content of flavonoid equivalent to naringin was 373.8 mg/100g. 28 kinds of free amino acids were detected with the contents of 252mg/100g in the dodder extract, and the contents of anserine and sarcosine were analysed highest in all amino acids. From the comparison of physiological activities of dodder extract with those of ascorbic acid, total reducing power, electron donating ability based on DPPH radical scavenging and ferric reducing antioxidant power was equivalent to 88%, 56% and 40%, respectively, of ascorbic acid. The nitrite scavenging activity of dodder extract was 78% of ascorbic acid on pH 1.5, but the antioxidant index of dodder on perilla oil was about 4 times greater than that of ascorbic acid.
The objective of this study was to investigate the effect of drying methods on the physicochemical characteristics of Ponciri fructus. Immature fruits of Poncirus trifoliata were harvested, dried by shade, infrared, or freeze drying, and powdered to size of 20 mesh. Then, extracted for 3 hrs with hot-water or 40% ethanol(80$\circC$℃). Freeze dried powder showed significantly higher L and -a value. The content of hesperidin was not affected by drying methods. The yield was the highest from freeze drying. Water extracts showed significantly more browning color, and higher acidity and total sugar content than those of ethanol extracts. Acidity and total sugar of water extracts were the highest in infrared drying and freeze drying, respectively. Browning color, pH, acidity and total sugar of ethanol extracts were not affected by the drying methods. The results suggest that freeze drying has a beneficial effects to enhance the quality of Ponciri fructus.
Cooking characteristics of coated rice with water homogenate of citrus fruit peels (1% for rice) were investigated. The color of the coated rice both before and after cooking was dark yellow. The total content of carotenoids, hesperidin and naringin were 10.74, 2173.12 and 1468.40 mg% for citrus fruit peels, 0.46, 108.65 and 73.35 % for its water homogenate, 0.12, 21.73 and 14.62 mg% for coated rin, and 0.05, 8.67 and 5.87 mg% fur cooked coated rice, respectively. Citrus fruits peel contained 94.22 mg% of asparagine, 24.88 mg% of methionine, 19.64 mg% of alanine, and 15.37 mg% of ${\gamma}$-aminoisobutyric acid as the majority free amino acids, accounting for 70% of the total free amino acids present. Total free amino acid content of the cooked coated rice increased by 15% compared to those of cooked uncoated rice. The majority of minerals in the citrus fruit peels were K and Ca, accounting for 56% of total minerals present. The mineral content of cooked coated rice was generally higher than that of the cooked uncoated rice. The cooked coated rice showed comparable hardness, gumminess and brittleness, but higher cohesiveness and springiness than the cooked uncoated rice. There were no differences in sweet and bitter taste between the cooked uncoated and coated rice. However, the cooked coated rice showed higher sensory scores fur color acceptability, savory taste and overall acceptability than the cooked uncoated rice.

We investigated the physiological activities of extracts from Campbell Early and Muscat Bailey A (MBA) grapevine leaves. Total phenol and flavonoid contents were highest in ethanol extracts from MBA grapevine leaves compared to extracts from Campbell Early grapevine leaves. Specific polyphenols higher in ethanol extracts from MBA grapevine leaves include gallic acid, epicatechin, caffeic acid, naringin, and resveratrol. Resveratrol content from MBA grapevine leaves increased when extracted for more than two hours in ethanol and water. The hydroxyl radical scavenging ability of ethanol extracts was higher than the water extract from both strains of grapevine leaves. DPPH and total antioxidants were highest in ethanol extracts from MBA grapevine leaves among the other extracts. Therefore, these results suggest that ethanol extracts from MBA grapevine leaves are a highly valuable resource for the development of natural functional foods.
