Efficient Transdermal Penetration and Improved Stability of L-Ascorbic Acid Encapsulated in an Inorganic Nanocapsule

  • Encapsulation of L-ascorbic acid (vitamin C) within a bio-compatible layered inorganic material was achieved by coprecipitation reaction, in which the layered inorganic lattice and its intercalate of vitamin C are simultaneously formed. The nano-meter sized powders of vitamin C intercalate thus prepared was again encapsulated with silica nano-sol to form a nanoporous shell structure. This ternary nanohybrid of vitamin Clayered inorganic core-$SiO_2$ shell exhibited an enhanced storage stability and a sustained releasing of vitamin C. Furthermore, the nano-encapsulation of vitamin C with inorganic mineral was very helpful in delivering vitamin C molecules into skin through stratum corneum, facilitating transdermal penetration of vitamin C in topical application.

Pharmacokinetic Characterization of Nano-emulsion Vitamin A, D and E (LaVita) in Rats

  • BACKGROUND: Bioavailability enhancement by solubilization of lipophilic drugs in nano-emulsion has been reported and it may be useful in pharmaceutical and nutraceutical products. This study was performed to compare in vivo bioavailability of nano-emulsion formulation with that of the general product as control. METHODS AND RESULTS: The pharmacokinetics assessment of Vitamin A, D and E complex of nanoemulsion formulation (LaVita), in comparison to the general product, was performed in the male rat plasma by a single oral dose at 20 mL/kg body weight (n=3/group). For nano-emulsion formulation (LaVita), $C_{max}$ of vitamin A and E in plasma were much higher and the area under the curve (AUC) of vitamin A, D and E were 14-63% higher, and the half-life of vitamin E was 2-fold longer than the general product. According to statistical analysis, each $C_{max}$ of vitamin A, D & E was significantly higher (p<0.01, 0.05 and 0.01, respectively) than that of general product. Half-life of vitamin A was significantly higher (p<0.01) and AUC of vitamin A and D were also significantly higher than the general product. CONCLUSION(s): Considering significant increment of $C_{max}$ and AUC, LaVita made of nano-emulsion could be more effective the absorption rate and extent for bioavailability of vitamin A, D & E than those of general product.

THE STUDY ON THE EFFECT OF NANOEMULSION FOR THE PREVENTION AND TREATMENT OF GINGIVAL INFLAMMATION (나노 에멀젼이 잇몸 염증의 예방 및 치료에 미치는 효과에 대한 연구)

  • We have found out the relationship of nanoemulsion containing nano vitamin C, E and propolis and gingival disease. We've confirmed effect of nanoemulsion through the experiment of in vivo and in vitro. We tested cell viability of gingival fibroblast cells by MTT assay and mRNA appearance of interleukin-$1{\beta}$, using mouse that was guided inflammation. Anti-microbacterial activity for Antibacterial effect's experiment was carried out by using S.aureus and E.coli. In addition, inflammation tissue has been observed with scanning electrical microscopy. In this study, expression of interleukin-$1{\beta}$ was decreased after adding nanoemulsion containing nanovitamin C, E and propolis. We've also obtained good results from the test of Antibacterial effect against S.aureus and E.coli. Also, swelling of inflammation tissues observed by scanning electrical microscopy has gone down. In conclusion, we have gained confidence that nanoemulsion containing nano vitamin C, E and propolis has very high Antibacterial effect against bacteria in oral. And it made us guess that inflammation of gingival reduces after decreasing interleukin-$1{\beta}$. Thus, we expect that nanoemulsion containing nano vitamin C, E and propolis gives good effects to patient having gingival disease.

  • 산업화 과정에 방출된 할로겐 화합물 및 프레온 가스가 오존층을 파괴하면서 많은 양의 자외선이 지표에 도달하여 생태계에 적지 않은 영향을 끼치고 있다. 자외선은 살균, 소독작용 및 Vitamin D 합성을 위해 생명체에 필수적이지만 과다할 경우 색소침착, 피부노화, 피부암 발생, 백내장의 증가, 면역기능저하 등의 피해를 준다. 자외선은 파장 영역에 따라 UV-A(장파장 자외선), UV-B(중파장 자외선)및 UV-C(단파장 자외선)으로 나뉜다. 이 중 인체에 가장 유해한 것으로 알려진 단파장의 UV-C는 파장이 짧아 침투력이 미약해 오존층과 성층권에서 대부분 흡수되므로 생물학적으로 큰 의미는 없다. 그러나 자외선 중 UV-B와 UV-A 영역의 자외선은 인체에 영향을 가장 많이 미치므로 이들 영역의 자외선을 차단하는 것이 필요하다. Vitamin C는 아스코르브산(ascorbic acid)이라고도 불리며 미백효과, 주름 개선, 자외선 차단 및 항산화 작용을 하지만 섬유가공 시 Vitamin C는 쉽게 산화되며 세탁 내구성이 떨어지기 때문에 가교제를 필요로 한다. Vitamin C를 이용하여 자외선 차단성 섬유제품을 개발하기 위해, 포름알데히드를 유리하지 않는 폴리카르복시산 가교제 중 하나인 Citric Acid(CA)를 사용하여 고온에서 면직물에 처리했을 때 에스테르 형 가교를 형성하여 내세탁성을 가질 수 있었다. 자외선 차단 효과를 알아보기 위하여 KS K 0850 섬유제품의 자외선 차단율 및 차단지수 시험방법으로 평가하였다.

Nano-capsulation of L-Ascorbic Acid in Nonaqueous System (L-Ascorbic Acid의 비수계 나노 캡슐화)

  • In this work, the basic research for nano-capsulation of L-ascorbic acid (Vitamin C) in nonaqueous system was carried out. 500 nm-sized nano-capsules were prepared in nonaqueous system, and the emulsified capsule had mean size of 410 nm. The stability test on the temperature and the storage periods was performed at 4, 20, and $30^{\circ}C$ for 30 days. After 5 days, L-ascorbic acid was extricated 5.1, 9.3, and 12.5% at each temperature, but only 1~2 % was extricated after the time span. Likewise, the results of the skin susceptibility on women and men, each 10 persons, revealed that the very thin allergy was shown from only a woman after 2 days, but it was not shown from the others.

  • This paper was described that the preparation of polymetylmethacrylate (PMMA) microsphere and coating of vitamin C onto surface of the prepared PMMA microsphere for application of cosmetic materials. The PMMA microsphere with various sizes can be obtained by change of reaction condition such as reaction temperature and reaction time. The coating of vitamin C on the surface of PMMA microsphere by using cyclodextrin as binder can be achieved to 30 wt-% in water/ethanol mixture. The vitamin C coated with cyclodextrin was stabilized during 56 days at $40^{\circ}C$. The color of the coated Vitamin C was changed from white to dark yellow after 14 days at $40^{\circ}C$. The vitamin C coated with cyclodextrin on the surface of PMMA microsphere can be sufficiently used for cosmetic materials.

To evaluate the skin permeation, experiments on VEA permeation to the skin of the ICR outbred albino mice (12 weeks, about 50 g, female) and on differences of solubility as a function of receptor formulations was performed. The analysis of nano-emulsions containing VEA 0.07 % showed that the higher ethanol contents the larger emulsions were formed, while the higher surfactant contents the size became smaller.In this study, vitamin E acetate (VEA, tocopheryl acetate), a lipid-soluble vitamin which is widely used A certain contents of ethanol in receptor phase increased VEA solubility on the nano-emulsion. When the ethanol contents were 10.0 % and 20.0 %, the VEA solubility was higher than 5.0 % and 40.0 %, respectively. The type of surfactant in receptor solution influenced to VEA solubility. The comparison between three kind surfactants whose chemical structures and HLB values are different, showed that solubility of VEA was increased as order of sorbitan sesquioleate (Arlacel 83; HLB 3.7) > POE (10) hydrogenated castor oil (HCO-10; HLB 6.5) > sorbitan monostearate (Arlacel 60; HLB 4.7). VEA solubility was also shown to be different according to the type of antioxidant. In early time, the solubility of the sample including ascorbic acid was similar to those of other samples including other types of antioxidants. However, the solubility of the sample including ascorbic acid was 2 times higher than others after 24 h. Franz diffusion cell experiment using mouse skin was performed with four nano-emulsion samples which have different VEA contents. The emulsion of 10 wt% ethanol was shown to be the most permeable at the amount of 128.8 ${\mu}g/cm^2$. When the result of 10 % ethanol content was compared with initial input of 220.057 ${\mu}g/cm^2$, the permeated amount was 58.53 % and the permeated amount at 10 % ethanol was higher 45.0 % and 15.0 % than the other results which ethanol contents were 1.0 and 20.0 wt%, respectively. Emulsion particle size used 0.5 % surfactant (HCO-60) was 26.0 nm that is one twentieth time smaller than the size of 0.007 % surfactant (HCO-60) at the same ethanol content. Transepidermal permeation of VEA was 54.848 ${\mu}g/cm^2$ which is smaller than that of particlesize 590.7 nm. Skin permeation of nano-emulsion containing VEA and difference of VEA solubility as a function of receptor phase formulation were determined from the results. Using these results, optimal conditions of transepidermal permeation with VEA were considered to be set up.

Effects of dietary supplementation of high-dose folic acid on biomarkers of methylating reaction in vitamin $B_{12}$-deficient rats

  • Folate is generally considered as a safe water-soluble vitamin for supplementation. However, we do not have enough information to confirm the potential effects and safety of folate supplementation and the interaction with vitamin $B_{12}$ deficiency. It has been hypothesized that a greater methyl group supply could lead to compensation for vitamin $B_{12}$ deficiency. On this basis, the present study was conducted to examine the effects of high-dose folic acid (FA) supplementation on biomarkers involved in the methionine cycle in vitamin $B_{12}$-deficient rats. Sprague-Dawley rats were fed diets containing either 0 or $100{\mu}g$ (daily dietary requirement) vitamin $B_{12}/kg$ diet with either 2 mg (daily dietary requirement) or 100 mg FA/kg diet for six weeks. Vitamin $B_{12}$-deficiency resulted in increased plasma homocysteine (p<0.01), which was normalized by dietary supplementation of high-dose FA (p<0.01). However, FA supplementation and vitamin $B_{12}$ deficiency did not alter hepatic and brain S-adenosylmethionine (SAM) and S-adenosylhomocysteine (SAH) concentrations and hepatic DNA methylation. These results indicated that supplementation of high-dose FA improved homocysteinemia in vitamin $B_{12}$-deficiency but did not change SAM and SAH, the main biomarkers of methylating reaction.

Effects of vitamin C and E supplementation on oxidative stress and liver toxicity in rats fed a low-fat ethanol diet

  • We compared the preventive capacity of high intakes of vitamin C (VC) and vitamin E (VE) on oxidative stress and liver toxicity in rats fed a low-fat ethanol diet. Thirty-two Wistar rats received the low fat (10% of total calories) Lieber-DeCarli liquid diet as follows: either ethanol alone (Alc group, 36% of total calories) or ethanol in combination with VC (Alc + VC group, 40 mg VC/100 g body weight) or VE (Alc + VE group, 0.8 mg VE/100 g body weight). Control rats were pair-fed a liquid diet with the Alc group. Ethanol administration induced a modest increase in alanine aminotransferase (ALT), aspartate aminotransferase (AST), conjugated dienes (CD), and triglycerides but decreased total radical-trapping antioxidant potential (TRAP) in plasma. VE supplementation to alcohol-fed rats restored the plasma levels of AST, CD, and TRAP to control levels. However, VC supplementation did not significantly influence plasma ALT, AST, or CD. In addition, a significant increase in plasma aminothiols such as homocysteine and cysteine was observed in the Alc group, but cysteinylglycine and glutathione (GSH) did not change by ethanol feeding. Supplementing alcohol-fed rats with VC increased plasma GSH and hepatic S-adenosylmethionine, but plasma levels of aminothiols, except GSH, were not influenced by either VC or VE supplementation in ethanol-fed rats. These results indicate that a low-fat ethanol diet induces oxidative stress and consequent liver toxicity similar to a high-fat ethanol diet and that VE supplementation has a protective effect on ethanol-induced oxidative stress and liver toxicity.

CASE REPORTS: TREATMENT OF ORAL SOFT TISSUE LESIONS AND WOUNDS WITH HIGH FUNCTIONAL TOOTH PASTE MADE FROM NANOEMULSION GEL (나노에멀젼 젤로 만든 고기능성치약을 이용한 구강 연조직 창상 및 병소에 대한 치험례)

  • It is a gel type high functional toothpaste containing vitamin C, E, propolis extract and the rest of herb with a nanoemulsion state. Vitamin C, E is known as the material with an eminent anti-oxidation effect. Propolis is known as the material with an antimicrobial and anti-inflammatory effect. We have been succeeding in making nanoemulsion of vitamin C, E and propolis through the high pressure homogenizer using stable oil and lecithin and the gel type high functional tooth paste were made from nanoemulsion of vitamin C, E and propolis. We observed the process of wound protecting effect and cure effect for a wound of soft tissue, gingival tissue and mucous membrane showing ulcer and inflammation in oral cavity after applying a gel type high functional toothpaste to patient. As a result, the wound were healed very fast and any side effects were not shown. We confirmed that a gel type high functional toothpaste with nanoemulsion of vitamin C, E and propolis extract has good effect not only for wound healing but also for treatment of ulcer-like lesion in oral cavity. So we report our cases with review of literatures.
