Toxin Genes and Antimicrobial Resistance of Clostridium perfringens Strains Isolated from Commercial Jeotgals (시판 젓갈에서 분리한 Clostridium perfringens의 독소 유전자 및 항균제 내성 분석)

  • Clostridium perfringens causes diarrhea and other diseases in humans and animals. We investigated the prevalence, toxin gene profiles, and antimicrobial resistance of C. perfringens isolated from commercial jeotgal sample. C. perfringens was isolated from 11 of 22 commercial jeotgals. All C. perfringens strains were positive for the alpha toxin gene, but not for the beta, epsilon, iota, CPE or NetB toxin genes; therefore, all strains were identified as type A C. perfringens. However, the beta2 toxin gene was identified in 54.5% of isolates. Disk diffusion susceptibility tests showed that most isolates were resistant to kanamycin (90.9%), nalidixic acid (72.7%), oxacillin (54.5%), erythromycin (27.3%), ciprofloxacin (9.1%) and clindamycin (9.1%). However, all strains were susceptible to 14 other antimicrobial including amoxicillin, ampicillin, and chloramphenicol. The average minimum inhibitory concentrations against C. perfringens of clindamycin, kanamycin, and nalidixic acid were 128.0, 128.0, and 54.0 ㎍/mL, respectively. These results provide new insight into the necessity for sanitation of commercial jeotgal, and provide evidence to help reduce the risk of contamination with antimicrobial-resistant bacteria.

  • Eubacterium is one of the predominant bacteria in the human large intestine. currently ES (Eubacterium Selective) medium developed by T. Mitsuoka is commonly used as a selective medium. neomycin sulfate which is one of the selective agents of ES medium inhibited about 50% of the growth of Eubacterium isolated, whereas malidixic acid inhibited only 5% while inhibiting other intestinal bacteria. NES medium which replaced neomycin with nalidixic acid in the ES medium was designed and shown to be better for the isolation and enumeration of Eubacterium sp. than ES medium.

E. faecalis and E. faecium Isolated in Dried Marine Products (시판 건해산물에서 분리한 Enterococcus faecalis와 E. faecium의 미생물학적 특성)

  • Eighty seven strains were isolated from 164 dried marine products(dried squid and dried alaska pollack etc) in Seoul Garak wholesale market. Among 87 isolates, twenty four E. faecalis and 4 E. faecium were identified by API strep kit. Twenty eight strains of E. faecalis, and E. faecium were resistant in streptomycin (95.6%), kanamycin (84.5%), gentamycin (66.7%), cephaloxin (97.8%), ampicillin/sulbactam (88.9%), ticarcillin(66.7%), amikacin (97.8%), sulfonamides (97.8%), ceftriaxone (75.6%), nalidixic acid (100.0%), and cefoxitin (100.0%), and were susceptible in amoxicillin/clavulanic acid(97.8%), chloramphenicol(95.6%), sulfamethoxazole/trimethoprim (97.8%), and tetracycline (71.1%). Also, ten strains of E. faecalis was resistant in $S-K-GM-CF-SAM-TIC-An-S_3-CRO-NA-FOX$ drugs simultaneously. Conclusively, E. faecalis strains from dried marine products were resistant on antibiotic drugs residue.

Characterization of Vibrio ponticus KJS1 Isolated from Larvae of Maroon clownfish, Premnas biaculeatus (Maroon clownfish, Premnas biaculeatus 자어로부터 분리한 Vibrio ponticus KJS1의 특성)

  • Vibriosis is one of the most important diseases affecting marin fish. In this study, a bacterium belonging to the Vibrio were isolated from maroon clownfish (Premnas biaculeatus) larvae (3~7 days after hatching). The bacterium was identified as Vibrio ponticus KJS1 by 16S rRNA gene sequence analysis. The result of biochemical test using the GN2 microplate (BioLog Inc, USA) showed that it has the ability to use 35 substrates including dextrin and sucrose was detected, could not degrade 52 substrates including α-cyclodextrin, and showed the weak ability to use 7 substrates including glycogen. It was sensitive to oxolinic acid, flumequine, ciprofloxacine, ofloxacin, norfloxacin, nalidixic acid, pefloxacin, and doxycycline hydrochloride.

Probiotic Properties of Bifidobacteria Isolated from Feces of Infants (유아 분변에서 분리한 비피도박테리아의 프로바이오틱스 기능성 연구)

  • Bifidobacteria are a prototype probiotic, which normally inhabit the intestinal tract of humans. In the present study, four species of Bifidobacterium isolated from the feces of infants were characterized. The tolerance for acid or bile salt, autoaggregation, and antibiotic resistance of the bacteria were examined. The four species were resistant to low pH, bile salts, and up to 3% bile acid. Autoaggregation rates were as high as 90%. The bacteria were consistently resistant to gentamicin, kanamycin, streptomycin, ciprofloxacin, and nalidixic acid. Due to their tolerance to environmental factors like acid and bile salts, B. longum MG723, B. breve MG729, B. bifidum MG731, and B. animalis subsp. lactis MG741 are potentially valuable as probiotics and may be useful for industrial application.

Production of Standard Sample for Quality Control of Total Coliform (총대장균군 측정의 정도관리에 적합한 균주의 조제)

  • Standard sample for quality control of total coliform measurement was procured by addition of nalidixic acid and cephalexin as bacteriostatic agents to Escherichia coli cultured broth. After making the standard sample, the number of E. coli was measured by fluorescence microscopic count method and plate count method by 12 hr interval. The numbers of E. coli remained unchanged for at least for 48 hr at room temperature which ranged from 3.5 to $4.2{\times}10^5\;cells/ml$ and from 1.0 to $1.2{\times}10^4\;CFU/ml$ by direct fluorescence microscopic count method and plate count method, respectively. This result suggests that microbial standard sample with bacteriostatic agents of nalidixic acid and cephalexin is usable for quantitative quality control.

Transition-Metal-Mediated Cytotoxicity of Quinolones to L1210 Cells

  • Transition metals tested, $Cu^{2+}$, and $Ni^{2+}$, were found effective in the induction of the cytotoxicity of the quinolones tested, nalidixic acid, oxolinic acid, and pipemidic acid, against L1210 leukemia cells in vitro, whereas the alkaline earth metal, $Mg^{2+}$, was not. The differential effect of the metals on the quinolone cytotoxicity can be explained by their different mode of interaction with the quinolones. Our present difference spectroscopic titration data suggest that the transition metals can form DNA-intercalating agents, with the quinolones, which can cause the cytotoxicity.

Effect of temperature on pharmacokinetics of nalidixic acid, piromidic acid and oxolinic acid in olive flounder Paralichthys olivaceus following oral administration (넙치, Paralichthys olivaceus에 nalidixic acid, piromidic acid, oxolinic acid의 경구투여 약물동태에 미치는 수온의 영향)

  • Effects of temperature ($13{\pm}1.5^{\circ}C$, $23{\pm}1.5^{\circ}C$) on the pharmacokinetic properties of nalidixic acid (NA), piromidic acid (PA) and oxolinic acid (OA) were studied after oral administration to cultured olive flounder, Paralichthys olivaceus. Serum concentrations of these antimicrobials were determined after oral administration of a single dosage of 60 mg/kg body weight (average 700 g). At $23{\pm}1.5^{\circ}C$, the peak serum concentrations of NA, PA and OA, which attained at 10 h, 24 h and 30 h post-dose, were 11.55, 3.79 and $1.12{\mu}g/m\ell$, respectively. At $13{\pm}1.5^{\circ}C$, the peak serum concentrations of NA, PA and OA, which attained at 10 h, 15 h and 30 h post-dose, were 6.36, 1.4 and $1.01{\mu}g/m\ell$, respectively. Better absorption of NA and PA was noted at $23{\pm}1.5^{\circ}C$ compared to $23{\pm}13^{\circ}C$. The elimination of NA from serum of olive flounder was considerably faster at $23{\pm}1.5^{\circ}C$ than at $13{\pm}1.5^{\circ}C$. However, both absorption and elimination of OA were not affected significantly by temperature. The kinetic profile of absorption, distribution and elimination of these antimicrobials in serum were analyzed by fitting to a one- and two compartment model, with WinNonlin program. In the one compartment model for NA, AUC, Tmax and Cmax at $23{\pm}1.5^{\circ}C$ were $258.26{\mu}g{\cdot}h/m\ell$, 10.67 h and $8.91{\mu}g/m\ell$, respectively. The AUC, $T_{max}$ and $C_{max}$ at $13{\pm}1.5^{\circ}C$ were $341.45 {\mu}g{\cdot}h/m\ell$, 7.72 h and $6.23{\mu}g/m\ell$, respectively. In the one compartment model for PA, AUC, $T_{max}$ and $C_{max}$ at $23{\pm}1.5^{\circ}C$ were $248.12{\mu}g{\cdot}h/m\ell$, 21.15 h and $3.09{\mu}g/m\ell$, respectively. The AUC, $T_{max}$ and $C_{max}$ at $13{\pm}1.5^{\circ}C$ were $103.89{\mu}g{\cdot}h/m\ell$, 12.89 h and $1.22{\mu}g/m\ell$, respectively. In the two compartment model for OA, AUC, $T_{max}$ and $C_{max}$ at $23{\pm}1.5^{\circ}C$ were $138.20{\mu}g{\cdot}h/m\ell$, 23.95 h and $1.06{\mu}g/m\ell$, respectively. The AUC, $T_{max}$ and $T_{max}$ at $13{\pm}1.5^{\circ}C$ were $159.10{\mu}g{\cdot}h/m\ell$, 28.03 h and $1.02{\mu}g/m\ell$, respectively.

Antibacterial Activity and Probiotic Properties of Lactic Acid Bacteria from Korean Intestine Origin (한국인 장관에서 분리한 유산균의 항균활성 및 프로바이오틱스 특성 연구)

  • The purpose of this study was to investigate the probiotic properties of lactic acid bacterial strains isolated from human feces. The properties were tested on the basis of guideline for probiotic selection protocol such as tolerance for acid or bile salt, autoaggregation, antibiotic resistance, and antimicrobial activity. Total 25 lactic acid bacteria were isolated from human feces, and their antibacterial activity was tested against Staphylococcus aureus, Escherichia coli, E. coli O157:H7, Vibrio parahaemolyticus, V. alginolyticus using an agar diffusion assay. Among them, 4 selected strains were identified by analysis of their 16S rRNA, as Lactobacillus rhamnosus MG316, L. acidophilus MG501, L. reuteri MG505, and L. gasseri MG570. Results show that resistance to low pH and bile salts. Also, the selected strains were resistant to bile acid up to 3% and their autoaggregation rates were as high as 60%. All strains tested were resistance to nalidixic acid and kanamycin.

Pulsed-Field Gel Electrophoresis and Mutation Typing of gyrA Gene of Quinolone-Resistant Salmonella enterica Serovar Paratyphi A Isolated from Outbreak and Sporadic Cases,1998-2002, Korea

  • In early 2002, over 200 people in the city of Pusan. Korea suffered from paratyphoid fever resulting from Salmonella Paratyphi A Infection. Antimicrobial susceptibility tests and Xbal pulsed-field gel electrophoresis (PFCE) were conducted to 54 Salmonella Paratyphi A isolated from humans during the period of 1998 to 2002. Most of the isolates ($83\%$) were only nalidixic acid-resistant and $78\%$ were X 1 PFGE patterns. Also, we measured the MIC of ciprofloxacin and screened gyrA mutation(5) using allele- specific PCR and restriction fragment length polymorphism (AS-PCR-RFLP). The representative 5 isolates in 2002 and 1 isolate in 2000 were $1{\mu}g/ml$ of MIC and had mutation at the 83rd codon in gyrA. These data suggest that the outbreak in the early 2002 might have been due to dissemination of the strain present In 2000. Also, decreased susceptibility to ciprofloxacin was partly due to the mutation at the 83rd codon in gyrA.
