Configuration and Ground Tests of Solar Cell and Fuel Cell Powered System for Long Endurance UAV (장기체공 무인기용 태양전지-연료전지를 활용한 동력원 구성 및 지상시험)

  • Each of power systems of solar cell and fuel cell were configured and validated for long endurance UAV, as the preliminary research for the integration of power systems. Solar power system consisted of solar modules fabricated by solar cells of Sunpower's C60, commercial solar MPPT controller and Li-po battery, and then was validated. The re-start characteristics of hydrogen production from $NaBH_4$ hydrolysis was validated for operating the commercial fuel cell. The average voltage drop of Li-po battery in solar power system was -2.9 V/hour. The performance of re-start characteristics of $NaBH_4$ hydrolysis was stable in sequence mode of mission profile. Each of single systems were satisfied for the proposed mission profile.

Color quality evaluation of composite resins used for splinting teeth (동요치 고정에 사용되는 복합레진의 색품질 평가)

  • Purpose: The purpose of this study was to evaluate the color stability of composite resins used for splinting teeth by comparing a self-cure resin cement (Superbond C&B, SB), a flowable composite resin (G-aenial Universal Flo A2, GU), and a composite resin exclusively used for splinting teeth (G-Fix, GF) before and after aging in NaOCl. Methods: Resin samples were fabricated to a size of 2 mm thickness and 8.5 mm diameter and light-cured with an LED light curing unit (G-Light, n = 12). Immediately after fabrication, CIE L*a*b* values of the resin samples were measured with a spectrophotometer (CM-5) as baseline. Then, the resin samples were immersed in 5% NaOCl at $60^{\circ}C$ and the color was measured after 23 hours every day for 4 days. Data were analyzed with one-way ANOVA followed by Duncan post hoc test (p<0.05), and color differences (${\Delta}E$) of resin samples before and after NaOCl aging were also calculated. Results: For SB, there were no changes in L* and a* values but changes were observed in b* values. For GU, there were no changes in L* and b* values but in a* values. For GF, changes were observed in L*, a*, and b* values. All resin samples showed highest ${\Delta}E$ between baseline and the 1st day of NaOCl immersion. ${\Delta}E$ of SB, GU, and GF was 4.6 - 5.8, 4.9 - 7.9, and 9.9 - 16.9, respectively. GF showed highest color change during NaOCl aging. Conclusion: The results of this color quality evaluation showed that the composite resin exclusively used for splinting teeth might be more vulnerable to color change during intraoral service.

Comparison of Environmental Stress Tolerance Between Lactobacillus fermentum Strains with High and Low Cell Surface Hydrophobicity

  • Previous studies have suggested a possible correlation between cell surface hydrophobicity (CSH) and stress tolerance in Bifidobacterium. In this study, the relationship was examined between CSH and environmental stress tolerance in Lactobacillus spp. By measuring the adhesion to hexadecane, 2 Lactobacillus fermentum strains- KLB 261 and KLB 231 were found to have high and low CSH, respectively. To measure their tolerance to various stresses, cells were subjected to salt (2 M NaCl), acid (pH 2), $H_2O_2$ (0.01 %, v/v), ethanol (20%, v/v), heat ($60^{\circ}C$), and cold ($-20^{\circ}C$). Compared with KLB 231, the hydrophobic KLB 261 was found to be much more resistant to the various stresses examined. After being subjected to different stresses for a period of time, KLB 261 and KLB 231 showed 50 and 0% survivability in 2 M NaCl, 108.2 and 0.6% in 0.01 %(v/v) $H_2O_2$, 40.2%(v/v), and 3.7% at $60^{\circ}C$ incubation, 4 and 0.6% at $-20^{\circ}C$, 12.9 and 0.1 % in pH 2, 33.8 and 0.2% in 20%(v/v) ethanol, respectively. Autoaggregation test and morphological observation were also conducted in an attempt to explain these differences. These results suggested that high CSH could strengthen the stress tolerance of lactobacilli.

Purification and Characterization of Xylanase from Bacillus sp. A-6 (Bacillus sp. A-6의 Xylanase의 정제와 특성)

  • A xylanase was purified from the culture supernatant of Bacillus sp. A-6 by using ultrafiltration and ion exchange chromatography on the column of SP-Sepharose using 5 mM acetate buffer, pH 5.0. The xylanase was eluted from the column at the concentration less than 0.05 M NaCl. The eluted xylanase was shown to be a single protein band in SDS-PAGE. Zymogram analysis indicated that the protein band in SDS-PAGE had the enzyme activity to hydrolyze oat spelt xylan. The molecular weights of the xylanase were 15,000 based on SDS-PAGE and 14,100 based on gel filtration chromatography. Thin layer chromatography showed that the xylanase hydrolyzed oat spelt xylan into xylobiose and high-molecular-weight xylooligosaccharides. The relative activities of the heated xylanase decreased to 80% at $40^{\circ}C$ after 7 hr and less than 40% at $60^{\circ}C$ after 1 hr.

  • Some enzymatic characteristics of the aflatoxin degrading factor produced extraceIlularly by Aspergillus awamori var. fumeus were investigated. When aflatoxin B1 was incubated with the culture filtrate of A. awamori var. fumeus. 60% of it was degraded within an hour. The degradation rate decreased with time and there was virtually no degradation after one hour. The apparent Michaelis constant ($K_m$) determined by Lineweaver-Burk plot was $10.2{\mu}M$. The optimum degradation was observed at $30^{\circ}C$ and pH 5. For the degradation, molecular oxygen seemed to be required. The degradation was enhanced by the $Co^{2+}$. but was inhibited by many other ions like $Fe^{2+}$, $Ca^{2+}$. $Mg^{2+}$, $Zn^{2+}$,$Cu^{2+}$, and $Ba^{2+}$, The presence of either KeN or metyrapone inhibited the reaction while that of $NaI0_4$ cytochrome C or NADPH showed no effect.

A study on the Rapid Processing of Hydrolyzed Anchovy Paste and Its Quality Stability (효소분해법에 의한 페이스트형 속성 멸치젓의 제조 및 품질에 관한 연구)

  • A study on the processing method of anchovy hydrolysate paste (AHP) was carried out to improve the sensory quality of salted and fermented fish. Homogenized whole anchovy was hydrolyzed using commercial pretenses, Complex enzyme-2000 (CE, Pacific Chem. Co.) and Alcalase (AL, Novo), in a cylindrical vessel with 4 baffle plates and 6-bladed turbine impeller. Optimal pH, temperature, and enzyme concentration for the hydrolysis with CE and AL were $7.0,\;52^{\circ}C,\;7\%$, and $8.0,\;60^{\circ}C,\;6\%$, respectively. The rational amount of water for homogenization, agitation speed, and hydrolyzing time were $100\%\;(w/w)$, 100 rpm, and 210 min, respectively. To make the hydrolysate to paste type, it was effective to mix the additives, such as starch, soybean protein, agar, and carrageenan gum to the hydrolysate 5 min before the end of boiling at $100^{\circ}C$ for 30 min. Minimal NaCl concentration for long-term preservation was $15\%$, and this could be reduced to $12\%$ by adding $5\%$ of KCl. yield of the AHP based on the total nitrogen content was $94.6\~97.0\%,\;and\;86.0\~89.2\%$, of the nitrogen was amino nitrogen. Salinity, pH and histamine content of the AHP prepared with $12\%$ NaCl and $5\%$ KCl were $9.3\~9.9\%,\;6.1\~6.2$, and below 13 mg/100 g, respectively. The AHP was stable at $26{\pm}3^{\circ}C$ for 60 days on bacterial growth, and addition of $0.05\%$ of rosemary (Herbalox) extract was effective to inhibit the lipid oxidation of the AHP during storage.

Optimization of Culture Conditions for Extracellular Lipid Production from Rhodotorula glutinis K-501 (Rhodotorula glutinis K-501로부터 세포외지질 생산을 위한 배양 조건의 최적화)

  • An extracellular lipid-producing strain, Rhodotorula glutinis K-501, was isolated from soil. The extracellular lipid produced by the cell was found to have good and stable emulsifying activity. Factors affecting the extracellular lipid production were studied to optimize culture conditions for maximum production. Sucrose and ammonium sulfate were selected as best carbon and nitrogen sources, respectively because they gave the highest concentration of product. The optimum C/N ratio was 50. Optimum concentrations of $KH_2PO_4,\; Na_2HPO_4, \;and\; CaCl_2 $were 3.5, 1.0, 0.75, and 0.1g/L, respectively. The optimum temperature and initial pH were determined as $22^{\circ}C$ and 7.0, respectively. When the batch culture was conducted with a sucrose concentration of 60g/L under the optimized conditions, extracellular lipid was produced to a concentration of 8.1g/L with a high production yield of 51.9% based on dry cell weight.

Isolation of a Potent Protease Producing Bacillus subtilis from Kimchi (김치로부터 단백질 분해 효소활성이 우수한 Bacillus subtilis 균주의 분리)

  • Microbial strains exhibiting proteolytic activity were isolated from kimchi, one of traditional fermented foods in Korea. Eight strains formed clear zones around their colonies when grown on TSA plates supplemented with skim milk. MBE/L865 exhibited 2.6-fold higher protease activity than that of control strain (Bacillus subtilis KCTC13112). MBE/L865 was identified as B. subtilis and deposited in the Korean Collection for Type Cultures under the accession number of KCCM43059. The optimum growth conditions for B. subtilis KCCM43059 were determined to be $37^{\circ}C$ and pH 8. The strain showed maximum protease activity ($429.37{\pm}18.65U/mg$ protein) at $60^{\circ}C$ and pH 6. Further, B. subtilis KCCM43059 had a higher salt (NaCl) tolerance than that of the control strain.

Microbiological Studles of Red pepper paste Fermentation (II) (고추장의 발효미생물에 관한 연구 2)

  • Among 50 yeasts of red pepper paste isolated and identified in the previous report, 12 strains of yeast which possess osmophilic and non-film characteristics were examined their growth rates and alcohol fermentation ability in red pepper medium. 5 strains of yeast possessing powerful growth rates and excellent alcohol fermentation ability were selected as excellent yeasts in this experiment. And their cultural conditions, salt resistance and red pepper concentration upon the growth were examined in this report. The results obtained were as follows ; 1) Among 12 strains of yeast, Saccharomyces cerevisiae group II(strain $D_1$), Saccharomyces oviformis($D_2$), Saccharomyces steineri($M_3$), Saccharomyces rouxii($D_9$) and Saccharomyces mellis($G_8$) were selected as excellent yeasts. 2) Among 5 strains of yeast selected, the strain $D_1$ and $D_2$showed the best alcohol fermentation ability and the best fermentative flavour. 3) The optimum temperature for growth of the strain $D_1$ and $D_2$ was $25^{\circ}C$$M_3$, $D_9$ and $G_8$ $30^{\circ}C$$D_1$ and $D_2$ was $62^{\circ}C$$M_3$,$D_9$ and $G_8$ ws 60.deg.C with treatment for 10 minutes. 4) The optimum pH for growth of all strains was around 5.0. 5) The strain $D_1$ and $M_3$were grown exceedingly well on the emdia containing 3 percent of NaCl and $D_2$,$D_9$ and $G_8$ on the media containing 6 percent of NaCl. 6) In the case of 5 percent of red pepper liquor added 5 percent of glucose was more nutritional than in the case of 5 percent glucose only. However, the growth rates of yeasts were restrained by the increase of red pepper concentration.

Improved Viability and Proteome Analysis of Lactobacillus fermentum KLB12 upon Heat Stress (Lactobacillus fermentum KLB12의 열 전처리에 따른 열 스트레스 내성 증진 및 프로테옴 변화)

  • In the previous study, we have isolated several vaginal lactobacilli from Korean woman and selected one of them (KLB12) for further study, which was indentified as Lactobacillus fermentum by sequence analysis of 16S rRNA gene. Formulated L. fermentum KLB12 can be used for ecological treatment of bacterial vaginosis. For pharmaceutical formulation, the spray-drying process is required where stress such as high temperature is routinely applied. In this study, we found that heat stress at 60$^{\circ}C$ for 20∼30min reduced the viable cell population of KLB12 by 10$\sub$6/~10$\sub$9/. However, adaptation of KLB12 cells at 52$^{\circ}C$ made them more thermotolerant upon exposure to 60$^{\circ}C$. The level of thermal protection in RSM (reconstituted skim milk) by adaptation in acid (pH 5), cold (4$^{\circ}C$), ethanol (3%), NaCI (0.3M) was also examined. Although not as efficient as the homologous stress, adaptations in both cold and NaCI gave considerable cross protection against heat stress. When chloramphenicol was added during heat adaptation, adaptation effect was abolished. This suggests that de novo protein synthesis is necessary during the adaptation process. Important changes in proteome during heat adaptation was examined with two-dimensional gel electrophoresis.
