Effects of Ethanol on $Na^+-dependent$ Solute Uptake in Rabbit Renal Brush-Border Membrane Vesicles

  • This study was undertaken to examine the effect of ethanol on $Na^+-dependent$ transport systems (glucose, phosphate, and dicarboxylate) in renal brush-border membrane vesicles (BBMV). Ethanol inhibited $Na^+-dependent$ uptakes of glucose, phosphate, and succinate in a dose-dependent manner, but not the uptakes of $Na^+-dependent.$ The $H^+/TEA$ antiport was reduced by 8% ethanol. Kinetic analysis showed that ethanol caused a decrease in $V_{max}$ of three transport systems, leaving $K_m$ values unchanged. Ethanol decreased phlorizin binding, which was closely correlated with the decrease in $V_{max}$ of $Na^+-glucose$ uptake. These results indicate that ethanol inhibits $Na^+-dependent$ uptakes of glucose, phosphate, and dicaboxylate and that the reduction in $V_{max}$ of $Na^+-glucose$ uptake is caused by a decrease in the number of active carrier proteins in the membrane.

  • This study is concerned with coemulsifier which is better emulsifiable properties than emulsifier itselves' use only. It's compound was synthesized by esterification of propylene glycol and succinic acid. The structure of them was comfirmed with FT-IR and $^1H$-NMR. Surface active properties with surface tension, cmc, emulsing power were tested respectively at given conditions. Their surface tensions in the aqueous solution was decreased to 33-35 dyne/cm and their cmc was evaluated by surface tension method. And it's emulsifying power was excellent in jojoba oil. As results, the synthesized dipropylene glycol succinate is expected to apply as O/W coemulsifier.

Heterogeneity of Mammalian Plasma Albumin (포유류 혈장알부민의 이질성)

  • Plasma albumin was purified from the fresh bovine blood using a minor modification of the polyethyleneglycol and ethanol procedure. The resulting protein solution was tested for its purity by both electrophoretic and immunochemical methods and found to contain only the albumin molecules. Each of the four thiol reagents, maleate, iodoacetate, iodoacetamide and glutathione, was incubated with the purified plasma albumin. The electrophoresis on cellulose acetate of those complexes in various buffers with different component and pH demonstrated that the albumin-glutathione complex was separated into two zones in all buffers used except the barbital and sodium acetate buffers, that the complexes of albumin-iodoacetate and albumin-iosoacetamide also into two zones only in pH 4.8 citrate buffer and in pH 4.8 succinate buffer and that the new zone had more positive net charge compared to the native protein in any case. These results might suggest a possibility that the electrophoretic albumin fraction is composed of at least two molecular species with different conformation.

Continuous PTFE Coating Process on Basalt Sewing Thread (현무암 재봉사의 연속식 테프론 코팅 공정)

  • On the basis of our previous research results concerning a batch Teflon coating process on the surface of basalt fiber which has superior fire-resistance and chemical resistance, we have tried to set up suitable operating conditions for continuous polytetrafluoroethylene(PTFE) coating process. The basalt fiber was continuously pre-treated with 7.5 wt%(6.5% of DPU) of triethoxytrifluoropropylsilane(TMTFPS) and then coated with 20 wt% of PTFE dispersions containing 0.25 wt% of penetrating agent sodium bis(2-ethylhexyl)sulfo succinate (DOS-Na) to get the highest tensile and loop strengths. After dipping process, the PTFE coated basalt fiber was dried under 2 m drying chamber at $120^{\circ}C$ with 12 m/min of winding speed and consequently sintered under 2 m sintering chamber at $380^{\circ}C$ for 40 s. Conclusively, PTFE coated basalt fiber whose tensile and loop strengths were to $3.4g_f/D$ and $2.3g_f/D$, respectively, applicable to high temperature sewing thread could be continuously prepared with our pilot scale process under optimum conditions.

  • This study was performed to investigate the formation process of calcium sulfate ${\alpha}$-hemihydrate from FGD gypsum produced at thermal power plant burning bituminous coal. The experimental results showed that calcium sulfate $\alpha$-hemihydrate with a large aspect ratio was produced in the temperature range of $120^{\circ}C$ and $140^{\circ}C$ in the absence of additives through dissolution-recrystallization mechanism. It was also observed that addition of Na-succinate as a catalyst changed crystal shape from acicular to prismatic, resulting in decreased water/powder ratio down to 33%. Optimum concentration of Na-succinate was 20mM. It was confirmed that the optimum moulding pressure and moisture content of moulded body from FGD gypsum were $30kg_f/cm^2$ and between 10% and 15% respectively, which prevent moulded body from collapsing and maximize the capillary effect by given pore volume while autoclaving.

  • The effects of salts which was used as a catalysis in formation of $\alpha$-calciumusulfate hemihydrate from dicalcium sulfate hydrate were investigated on the hydration of $\alpha$-calciumsulfate hemihydrate. The hydration of $\alpha$-calciumsulfate hemihydrate was studied by the measurements of crystalline water, heat evolution. Also the hydrates were analyzed by XRD, DSC and SEM. The promotive effect each salts on the hydration was as follows: NaCl>NH4Cl>NaNO3>NH4NO3, and the hydration rate was accelerated with concentration of salts. The effect of Al2(SO4)3 and potassium sodium tartrate on the hydration was slmilar to water, whereas sodium succinate and gelatin retarded the hydration in comparision with water. These salts affected the hydration time but total heat evoution was similar.

  • A tylosin-hyperproductive mutant of S. fradiae MNU20 was isolated among 3500 strains obtaincd from either MNNG- or UV-treated Streptomyces fradiae NRRL2702. The composition of optimal medium for tylosin production was formulated as followed as: 4 g soluble starch, 1 g glucose, 1 g corn steep liquor, 7.5 ml soy bean oil, 0.2 g KH$_{2}$PO$_{4}$, 1 g Na$_{2}$S$_{2}$O$_{3}$$\cdot $5H$_{2}$O, 2 g CaCO$_{3}$, 2 g NaCl, 0.001 g CoCl$_{2}$$\cdot $6H$_{2}$O in 1 liter of distilled water. With the optimal medium, S. fradiae MNU20 was able to produce 159 mg tylosin (g biomass)$^{-1}$, indicating that tylosin productivity of Streptomyces fradiae NRRL2702 was increased 14 times higher by mutation. When the effect of valine, succinate, and natural zeolite on tylosin production was investigated by using the optimal medium, these substances essentially enhanced tylosin production and their addition time during culture period appeared to be critical for the increase of tylosin production.

The Effects of Acupuncture at LR3 Acupoint on Mitochondrial Complex IV Oxidase activity in Liver (태충 침자가 간 미토콘드리아 내 Complex IV에 미치는 영향)

  • Objectives : The liver is rich in mitochondria and it plays a key role in whole-body energy homeostasis. Mitochondria is double membrane-bound organelle that supplies energy for intracellular metabolism including Krebs cycle and beta-oxidation. Acupuncture is known to stimulate and regulate the flow of energy. To explore the effect of acupuncture on the mitochondrial respiratory chain activity in the rats' livers, the activity of mitochondrial respiratory chain complexes I to IV was observed. Methods : The rats were divided into 4 groups; Normal 1 (no acupuncture treatment and anesthesia for 5 min), Normal 2 (no acupuncture treatment and anesthesia for 10 min), MA1 (acupuncture treatment at bilateral LR3 under anesthesia for 5 min), and MA2 (acupuncture treatment at bilateral LR3 under anesthesia for 10 min). All rats were sacrificed and the livers were examined for respiratory chain change. Results : There was no difference in ubiquinon oxidoreductase, succinate dehydrogenase, and ubiquinol cytochrome C oxidoreductase after acupuncture at LR3. Acupuncture at LR3 for 10 min increased the activity of cytochrome C oxidase compared with no acupuncture groups. Conclusions : Acupuncture at LR3 mediated mitochondrial respiratory chain activity via the cytochrome C oxidase signaling pathway in the livers of rats.

The Effects of Acupuncture at Heart and Kidney Meridian on Mitochondrial Respiratory Chain Complexes Activities in Rats (심경, 신경의 오수혈 침자가 Mitochondrial Respiratory Chain Complexes에 미치는 영향)

Also, acupuncture has been known to stimulate the flow of energy. To explore the effect of acupuncture on the mitochondrial respiratory chain activities in rat's heart and kidneys, the activities of mitochondrial respiratory chain complexes I to IV were observed. Methods : The rats were divided into 11 groups; Normal (no acupuncture treatment and under anesthesia for 10 min), heart meridian five-transport-points (acupuncture treatment at HT9, HT8, HT7, HT4 and HT3 under anesthesia for 10 min), and kidney meridian five-transport-points (acupuncture treatment at KI1, KI2, KI3, KI7 and KI10 under anesthesia for 10 min). All rats were sacrificed and the heart and kidneys were examined for the changes of respiratory chain activities. Results : Acupuncture at HT7 increased the activity of succinate dehydrogenase; acupuncture at KI2 increased the activity of ubiquinol cytochrome C oxidoreductase; and acupuncture at HT9, HT8, HT3 and KI1 increased activities of cytochrome C oxidase. Conclusions : Acupuncture assists mitochondrial repiratory chain activity via the Cytochrome C oxidase signaling pathway in heart and kidney of rats.

The Effect of Penetrating Agent and Fluorosilane on High Temperature Teflon Coating (침투제와 불소화실란이 고온용 테프론 코팅에 미치는 영향)

  • Although the basalt fiber has superior fire-resistance and chemical resistance, it has many disadvantages in its applications. Generally, the tensile and loop strengths of basalt fiber were decreased with generated frictional heat during industial appplications. To solve this problem, polytetrafluoroethylene (PTFE) coating system was introduced and a sutable coating condition was evaluated. The basalt fiber was pre-treated with triethoxytrifluoropropylsilane (TMTFPS) at various pHs and then coated with PTFE dispersions with penetrating agent sodium bis(2-ethylhexyl)sulfo succinate (DOS-Na) to increase the tensile and loop strengths as well as to reduce the fibril during working. A universial testing machine (Instron Model 3366) was used to measure tensile and loop strengths. When the PTFE dispersion with 0.25 wt% of DOS-Na was coated on the surface of basalt fiber after pre-treating with 5 wt% of PTFE, the highest tensile and loop strengths were reached to 3.5 gf/D and 2.4 gf/D, respectively.
