페놀화합물 처리 Agrobacterium 및 세포벽 약화 들깨새싹을 이용한 형질전환과 재조합 단백질 발현 (Transformation of Cell Wall-weakened Perilla Seedlings Using Phenolic Compound-treated Agrobacterium Cells and Recombinant Protein Expression)

  • 들깨묘는 재배가 용이하며 건강채소로 애용되고 있다. 본 연구에서는 발아 후 2일째의 들깨묘에 대하여 세포 상해를 일으킬 수 있는 NaOH/SDS 용액을 처리하고 페놀 화합물을 처리한 재조합 Agrobacterium 세포를 이용하여 형질 전환을 수행하였다. 형광분석에 의한 GUS 유전자발현 분석에서 50 mM acetosyringone 처리 및 0.5% NaOH /0.01% SDS 처리의 복합처리에 의하여 가장 높은 GUS활성이 나타났으며 상승효과를 제시할 수 있었다. 이러한 결과는 hepatitis B virus antigen (HBsAg) 단백질 발현으로 성공적으로 확인할 수 있었다.

한국산(韓國産) 복수초성분(福壽草成分)의 $Na^+,\;K^+-ATPase$활성(活性)에 대한 연구(硏究) (Studies on $Na^+,\;K^+-ATPase$ Activity of the Constituents of Adonis amurensis in Korea)

  • Four known cardiotonic steroids were isolated from roots of Adonis amurensis Regel et Radde and identified as digitoxigenin, cymarin, K-strophanthin and convallatoxin by chromatography on Amberlite XAD-2 resin and silica gel, high performance liquid chromatography and gel chromatography on Sephadex LH-20. In order to clarify the structure-activity relationship, thirteen related compounds of digitoxigenin were tested for the inhibitory activities for $Na^+,\;K^+$-adenosine triphosphatase from guinea pig heart. The inhibitory activities of related compounds of digitoxigenin were dependent upon the dicarboxylic acid and amino acid components. The compound having both the arginine and suberic acid moiety showed the higher inhibitory activity. The sulfate and glucuronide of digitoxigenin exhibited much less potency than the parent genin.

점성물질 제거가 다시마 추출액의 휘발성 향미성분에 미치는 영향 (Effect of Viscous Materials Removal from Sea Tangle Extracts on Volatile Flavor Constituents)

  • 다시마 추출액의 향미성분을 분석하기 위하여 steam distillation방법으로 추출한 뒤 GC/MS를 이용하여 분석하였다. 다시마 분말과 100에서 2시간 추출한 대조구 그리고 효소분해와 NaCl용액에서 추출한 뒤 ultrafiltration으로 점성 물질을 제거한 추출액을 비교하였다. 그 결과 다시마 분말의 주요 향미성분은 2-nonen-1-ol, myristic acid, palmitic acid와 oleic acid이었고 $100^{\circ}C$에서 열수추출한 대조구에서는 pentadecane, 1,4-cyclooctadiene과 myristic acid로 나타났으며, 점성물질을 제거한 시료에서는 cyclohexanone, (E,Z)-octa-1,5-dien-3-ol, acetic acid, ethanone, benzaldehyde, 2,6-nonadienal 등으로 나타났다. 또한, 점성물질제거 과정중 분자량이 비교적 큰 지방산들과 3,5-nonadien-2-ol, 1-penton-3-ol 등이 현저히 감소하였다.

새로운 Tetrazole유도체의 합성과 항균활성 (Synthesis and Antibacterial Activity of New Tetrazole Derivatives)

  • 3-Acetyl/Formyl 4-hydroxy-2H(1)-benzopyran-2-one를 malonitrile와 ethyl cyanoacetate로 처리하여 각각 1,1-dicyano-2-[4/-hydroxy-2/H(1)-benzopyran-2/-one-3/-yl] ethene/propene 2a-h와 ethyl-2-cyano-3-[4/-hydroxy-2/H (1)-benzopyran-2/-one-3/-yl] propenoate/butenoate 3a-h를 얻었다. NaN3와 2a-h의 1.3 dipolar 반응에서 4a-h인 tetrazole유도체를 얻었다. 3a-h는 PPA를 이용한 고리화 반응으로 3-cyano-2H,5H-pyrano [3, 2-c] benzopyran-2,5-diones 5a-h를 얻었다. 5a-h는 NaN3와1.3 dipolar 반응으로 -(1/H-tetrazol-5/-yl)-2H,5H-pyrano[3, 2-c] benzopyran-2,5-diones 6a-h얻었다. 화합물의 구조는 스펙트럼과 자료 분석을 기초로 입증했다.모든 화합물은 항균 활성을 검사하였고 의미있는 항균성을 가짐을 밝혔다. 2h화합물과 4h 화합물은 50 킽/mL에서 활성을 보였다

Preferential Intercalation of Organic Anions into Layered Double Hydroxide

  • Intercalation compounds of organic anions into layered double hydroxides (LDH) are synthesized by the coprecipitation route. X-ray diffraction data reveal that the intercalated terephthalate (TP), naphthalene-2,6-disulfonate (NA26), and anthraquinone-2,6-disulfonate (AQ26) are arranged with their molecular planes perpendicular to the hydroxide layer. HPLC data show that 26.2% of TP and 73.8% of AQ26 are cointercalated, whereas NA26 is not intercalated into the Zn/Al-LDH. These results indicate the possibility of a molecular recognition ability of Zn/Al-LDH. The molecular recognition ability of intercalation into Zn/Al-LDH is in the order AQ26 > TP >> NA26.

Analysis of the Components of Guibitang and Fermented Guibi-tang and their Ability to Inhibit Angiotensin-converting Enzyme

  • Guibi-tang is a traditional medicine used for the treatment of colds. We investigated the levels of several compounds in Guibi-tang before and after fermentation with Lactobacillus and tested their ability to inhibit angiotensin-converting enzyme. Six known compounds (decursin, decursinol angelate, nodakenin, liquiritin, formononetin, and 6-gingerol) and 2 unidentified compounds were detected in Guibi-tang (GB) and fermented Guibi-tang (FGB) by an established HPLC-DAD method. The levels of the 6 known compounds were decreased after fermentation. FGB showed more potent inhibition of angiotensin-converting enzyme activity than GB. In conclusion, fermentation with Lactobacillus affects the content of several compounds in GB and improves its angiotensin-converting enzyme inhibitory activity.

산겨릅나무로부터 페놀화합물의 분리 및 항염증 활성의 측정 (Isolation and Identification of Phenol Compounds from Acer tegmentosum and their Anti-inflammatory Activity)

  • The Acer tegmentosum (3 kg) were extracted with boiled water and the freeze dried extract powder was partitioned with $CH_2Cl_2$, EtOAc, n-BuOH and $H_2O$, successively. From the EtOAc and n-BuOH fraction, six phenolic compounds were isolated through the silica gel, octadecyl silica gel and sephadex LH-20 column chromatography. On the basis of spectroscopic methods, such as $^1H$-NMR and $^{13}C$-NMR, and LC/MS, the chemical structures of the compounds as feniculin (1), avicularin (2), (+)-catechin (3), (-)-epicatechin (4), salidroside (5) and 6'-O-galloylsalidroside (6). In this study, compounds 1 and 2 have been first isolated from the A. tegmentosum. To provide insight into the effects of six compounds isolated from A. tegmentosum on inflammation, we investigated its effect on nitric oxide (NO) production in RAW 264.7 cells using lipopolysaccharide (LPS) stimulation. Compounds 1 and 6 slightly repressed NO production. Also, compounds 3 and 4 inhibited NO secretion with statistical significance. However, compounds 2 and 5 did not show any inhibitory effect on NO production.

Recent Progress on Sodium Vanadium Fluorophosphates for High Voltage Sodium-Ion Battery Application

  • Na-ion batteries are being considered as promising cost-effective energy storage devices for the future compared to Li-ion batteries owing to the crustal abundance of Na-ion. However, the large radius of the Na ion result in sluggish electrode kinetics that leads to poor electrochemical performance, which prohibits the use of these batteries in real time application. Therefore, identification and optimization of the anode, cathode, and electrolyte are essential for achieving high-performance Na-ion batteries. In this context, the current review discusses the suitable high-voltage cathode materials for Na-ion batteries. According to a recent research survey, sodium vanadium fluorophosphate (NVPF) compounds have been emphasized for use as a high-voltage Na-ion cathode material. Among the fluorophosphate groups, $Na_3V_2(PO_4)_2F_3$ exhibited the high theoretical capacity ($128mAh\;g^{-1}$) and working voltage (~3.9 V vs. $Na/Na^+$) compared to the other fluorophosphates and $Na_3V_2(PO_4)_3$. Here, we have also highlighted the classification of Fluorophosphates, NVPF composite with carbonaceous materials, the appropriate synthesis methods and how these methods can enhance the electrochemical performance. Finally, the recent developments in NVPF for the application in energy storage devices and its outlook are summarized.

Structural, FTIR and ac conductivity studies of NaMeO3 (Me ≡ Nb, Ta) ceramics

  • Lead-free complex perovskite ceramics $NaMeO_3$ ($Me{\equiv}Nb$, Ta) were synthesized using conventional solid state reaction technique and characterized by structural, FTIR and electrical (dielectric and ac conductivity) studies. The crystal symmetry, space group and unit cell dimensions were determined from the experimental results using FullProf software. XRD analysis of the compound indicated the formation of single-phase orthorhombic structure with the space group Pmmm (47). Dielectric studies showed the diffuse phase transition at $394^{\circ}C$ for $NaNbO_3$ and $430^{\circ}C$ for $NaTaO_3$. Ac conductivity in both the compounds follows Jonscher's power law.
