Anti-hypertensive Activities of Lactobacillus Isolated from Kimchi (김치에서 분리한 유산균의 항고혈압 활성)

  • This study was conducted to evaluate the anti-hypertensive effect of Lactobacillus sp. isolated from Kimchi by examining its effects on renal angiotensin-converting enzyme (ACE) inhibitory activity, lipid components and blood pressure using the spontaneously hypertensive rat (SHR) system. Most Lactobacillus sp. extracts (lysozyme, sonication and ethyl acetate extracts) showed higher capacities for the inhibition of ACE activity than those of cultured media. Particularly, LG 7, 8 and 42 of Lactobacillus sp. showed the strongest inhibitory activity among the Lactobacillus sp. extracts. The concentrations of total cholesterol and triglycerides in the serum were lower in the Lactobacillus sp. administration groups than in the control group, but these differences were not significant. The HDL-cholesterol concentrations of the LG 42 administration groups (IX, X) were significantly higher than that of the control group. At 4 weeks, the systolic blood pressure (SBP) in the LG 42 Lactobacillus sp. ($1{\times}10^9$ cfu/mL) group (XI) was about 27% lower than that of the control group (V). No adverse effects were observed on the liver and there was no difference in the aspartate aminotransferase (AST) and alanine aminotransferase (ALT) values among groups. The results of this study suggest that long term consumption of LG 42 Lactobacillus sp. may be beneficial to the prevention of high blood pressure.

  • The methylotrophic yeast Hansenula polymorpha has been extensively studied as a model organism for methanol metabolism and peroxisome biogenesis. Recently, this yeast has also attracted attention as a promising host organism for recombinant protein production. Here, we describe the fabrication and evaluation of a DNA chip spotted with 382 open reading frames (ORFs) of H. polymorpha. Each ORF was PCR-amplified using gene-specific primer sets, of which the forward primers had 5'-aminolink. The PCR products were printed in duplicate onto the aldehyde-coated slide glasses to link only the coding strands to the surface of the slide via covalent coupling between amine and aldehyde groups. With the partial genome DNA chip, we compared efficiency of direct and indirect cDNA target labeling methods, and found that the indirect method, using fluorescent-labeled dendrimers, generated a higher hybridization signal-to-noise ratio than the direct method, using cDNA targets labeled by incorporation of fluorescence-labeled nucIeotides during reverse transcription. In addition, to assess the quality of this DNA chip, we analyzed the expression profiles of H. polymorpha cells grown on different carbon sources, such as glucose and methanol, and also those of cells treated with the superoxide­generating drug, menadione. The profiles obtained showed a high-level induction of a set of ORFs involved in methanol metabolism and oxidative stress response in the presence of methanol and menadione, respectively. The results demonstrate the sensitivity and reliability of our arrays to analyze global gene expression changes of H. polymorpha under defined environmental conditions.

The Nuclear Security Summit Achievements, Limitations, and Tasks against Nuclear Terrorism Threat (핵테러리즘 위협에 대한 핵안보정상회의 성과, 한계 및 과제)

  • In April 2009, in the wake of President Obama's Prague speech, the international community held four nuclear sec urity summits from 2010 to 2016 to promote nuclear security and prevent nuclear terrorism. The Nuclear Security S ummit has made significant progress in preventing terrorists from attempting to acquire nuclear weapons or fissile materials, but it still has limitations and problems. To solve this problem, the international community should resume the joint efforts for strengthening bilateral cooperation and multilateral nuclear security regime, and the participating countries should strive to protect their own nuclear materials and fulfill their commitments to secure nuclear facilitie s. Second, the United Nations(UN), the IAEA(International Atomic Energy Agency), International Criminal Police Or ganization(INTERPOL), the Global Initiative to Combat Nuclear Terrorism(GICNT), and the Global Partnership(G P) must continue their missions to promote nuclear security in accordance with the five action plans adopted at the Fourth Nuclear Security Summit. Third, the participating countries should begin discussions on the management and protection of military nuclear materials that could not be covered by the Nuclear Security Summit. Fourth, the intern ational community must strive to strengthen the implementation of the Convention on the Physical Protection of Nuc lear Material(CPPNM) Amendment and International Convention for the Suppression of Acts of Nuclear Terrori sm(ICSANT), prepare for cyber attacks against nuclear facilities, and prevent theft, illegal trading and sabotage invo lving nuclear materials.

Cholesterol Improvement Effects of Fermented Defatted Soybean Grits Added to Corchorus olitorius (몰로키아 첨가 탈지대두grit(defatted soybean grit) 발효물의 콜레스테롤 개선 효과)

  • This study was performed to investigate cholesterol improvement effects of adding fermented defatted soybean grit (FD) and FD added to 5 or 10% Corchorus olitorius (FDC). Cholesterol adsorption in the FD and FDC group was more than 70%. Apolipoprotein AI and CIII improved in HepG2 cells, and a greater improvement effect was shown in FDC than that in FD. We also investigated the effect of FDC on body lipid metabolism and a high-fat diet for 4 weeks. Rats were divided into control (Con), high-fat (HF), HF treated with 20% FD (HF-FD), and HF treated with 20% FDC (HF-FDC) groups. Plasma total cholesterol and low density lipoprotein-cholesterol concentrations, hepatic total cholesterol, and triglyceride contents were significantly lower in the HF-FDC group than those in the HF group. Additionally, fecal total cholesterol and triglyceride contents increased in rats treated with FDC. Hepatic 3-hydroxy-3-methylglutaryl-coenzyme A reductase activities were significantly lower in the HF-FDC group than those in the HF group.

Antioxidant Effects of Extracts from Fermented Red Ginseng Added with Medicinal Herbs in STZ-induced Diabetic Rats (약용식물 첨가 발효홍삼 추출물의 Streptozotocin 유발 당뇨쥐에 대한 항산화 효과)

  • Antioxidant and anti-hyperglycemic activities of fermented red ginseng added with 5 kinds of medicinal herbs (FRGM) were investigated in vitro. Total polyphenol and total flavonoid contents in FRGM extracts were $22.41{\pm}3.51$ and $16.80{\pm}4.22{\mu}g/mg$, respectively. FRGM extracts were capable of directly scavenging DPPH free radicals ($RC_{50}=95.57{\pm}7.40{\mu}g/mL$), and then showed higher inhibitory activities for ${\alpha}$-glucosidase. This study was also conducted to evaluate the effects of FRGM extracts in streptozotocin (STZ)-induced diabetic (DM) rats. The activities with regards to serum aspartate aminotransferase and alanine aminotransferase were significantly decreased by FRGM extracts compared to those from the STZ group. The hepatic glutathione content depleted by STZ was significantly increased by FRGM extracts, but elevation of lipid peroxide content induced by STZ was significantly decreased by FRGM extracts. The decreased activities of superoxide dismutase, catalase, glutathione peroxidase and glutathione reductase after STZ-treatment were increased through the treatment of FRGM extracts. These results indicated that fermented red ginseng added with medicinal herbs can protect against STZ-induced diabetic rats through its antioxidant properties.

A Kinetic Study on Aminolysis of 2-Pyridyl X-Substituted Benzoates: Effect of Changing Leaving Group from 4-Nitrophenolate to 2-Pyridinolate on Reactivity and Mechanism

  • Second-order rate constants ($k_N$) have been measured spectrophotometrically for nucleophilic substitution reactions of 2-pyridyl X-substituted benzoates 8a-e with a series of alicyclic secondary amines in $H_2O$ at $25.0{\pm}0.1^{\circ}C$. The $k_N$ values for the reactions of 8a-e are slightly smaller than the corresponding reactions of 4-nitrophenyl X-substituted benzoates 1a-e (e.g., $kN^{1a-e}/k_N^{8a-e}$ = 1.1 ~ 3.1), although 2-pyridinolate in 8a-e is ca. 4.5 $pK_a$ units more basic than 4-nitrophenolate in 1a-e. The Br$\o$nsted-type plot for the aminolysis of 8c (X = H) is linear with $\beta_{nuc}$ = 0.77 and $R^2$ = 0.991 (Figure 1), which is typical for reactions reported previously to proceed through a stepwise mechanism with breakdown of a zwitterionic tetrahedral intermediate $T^{\pm}$ being the rate-determining step (RDS), e.g., aminolysis of 4-nitrophenyl benzoate 1c. The Hammett plot for the reactions of 8a-e with piperidine consists of two intersecting straight lines (Figure 2), i.e., $\rho$ = 1.71 for substrates possessing an electron-donating group (EDG) while $\rho$ = 0.86 for those bearing an electron-withdrawing group (EWG). Traditionally, such a nonlinear Hammett plot has been interpreted as a change in RDS upon changing substituent X in the benzoyl moiety. However, it has been proposed that the nonlinear Hammett is not due to a change in RDS since the corresponding Yukawa-Tsuno plot exhibits excellent linear correlation with $\rho$ = 0.85 and r = 0.62 ($R^2$ = 0.995, Figure 3). Stabilization of substrates 8a-e in the ground state has been concluded to be responsible for the nonlinear Hammett plot.

Purification and Cellular Localization of Extracellular Nuclease of Serratia marcescens Expressed in Escherichia coli (대장균에 발현된 Serratia marcescens의 Nuclease의 정제와 세포내 분포)

  • Nuclease was secreted to the environmental media from the Escherichia coli JM107 tranformant harboring the extracellular nuclease gene of Serratia marcescens in the plasmid of pNUC4. Under the growth conditions, the amount of secreted enzyme was increased in parallel with bacterial growth conditions, the amount of secreted enzyme was increased in parallel with bacterial growth. The enzyme was purified using chromatofraphic procedures of Matrex green gel and heparin agarose affinity gel, resulted in 50-fold purification with 15% recovery of the enzyme. The apparent molecular weight of the enzyme was estimated to be 29Kda by sodium dodecylsulfate denaturing gel electrophoresis. Using the purified enzyme, polyclonal antibody was obtained from the rabbit. The specificity of the antibody was confirmed by immunoblotting and immunoprecipitaion. For the investigation of cellular distribution of the enzyme, cells were fractionated into three fractions; cytoplasm, periplasm and extracellular fluid. While more than 80% of the enzymatic activity was detected in the extracellular fluid and periplasm, a little was found in the cytoplasm, indicating that the enzyme was likely to be immediately exported to the membrane for excretion after biosynthesis. These results were confirmed again by immunocytochemistry technique using the antibody.

A Study of the Scene-based NUC Using Image-patch Homogeneity for an Airborne Focal-plane-array IR Camera (영상 패치 균질도를 이용한 항공 탑재 초점면배열 중적외선 카메라 영상 기반 불균일 보정 기법 연구)

  • The detector of a focal-plane-array mid-wave infrared (MWIR) camera has different response characteristics for each detector pixel, resulting in nonuniformity between detector pixels. In addition, image nonuniformity occurs due to heat generation inside the camera during operation. To solve this problem, in the process of camera manufacturing it is common to use a gain-and-offset table generated from a blackbody to correct the difference between detector pixels. One method of correcting nonuniformity due to internal heat generation during the operation of the camera generates a new offset value based on input frame images. This paper proposes a technique for dividing an input image into block image patches and generating offset values using only homogeneous patches, to correct the nonuniformity that occurs during camera operation. The proposed technique may not only generate a nonuniformity-correction offset that can prevent motion marks due to camera-gaze movement of the acquired image, but may also improve nonuniformity-correction performance with a small number of input images. Experimental results show that distortion such as flow marks does not occur, and good correction performance can be confirmed even with half the number of input images or fewer, compared to the traditional method.

Comparative Genomic Analysis of Pathogenic Factors of Pectobacterium Species Isolated in South Korea Using Whole-Genome Sequencing

  • In this study, we conducted whole-genome sequencing with six species of Pectobacterium composed of seven strains, JR1.1, BP201601.1, JK2.1, HNP201719, MYP201603, PZ1, and HC, for the analysis of pathogenic factors associated with the genome of Pectobacterium. The genome sizes ranged from 4,724,337 bp to 5,208,618 bp, with the GC content ranging from 50.4% to 52.3%. The average nucleotide identity was 98% among the two Pectobacterium species and ranged from 88% to 96% among the remaining six species. A similar distribution was observed in the carbohydrate-active enzymes (CAZymes) class and extracellular plant cell wall degrading enzymes (PCWDEs). HC showed the highest number of enzymes in CAZymes and the lowest number in the extracellular PCWDEs. Six strains showed four subsets, and HC demonstrated three subsets, except hasDEF, in type I secretion system, while the type II secretion system of the seven strains was conserved. Components of human pathogens, such as Salmonella pathogenicity island 1 type type III secretion system (T3SS) and effectors, were identified in PZ1; T3SSa was not identified in HC. Two putative effectors, including hrpK, were identified in seven strains along with dspEF. We also identified 13 structural genes, six regulator genes, and five accessory genes in the type VI secretion system (T6SS) gene cluster of six Pectobacterium species, along with the loss of T6SS in PZ1. HC had two subsets, and JK2.1 had three subsets of T6SS. With the GxSxG motif, the phospholipase A gene did locate among tssID and duf4123 genes in the T6SSa cluster of all strains. Important domains were identified in the vgrG/paar islands, including duf4123, duf2235, vrr-nuc, and duf3396.

성장율 및 사료효율에 있어서 키토산의 첨가급여가 육용계 두 품종에 나타나는 상관반응에 관한 연구

  • Two experiments were conducted to investigate the response of chitosan supplementation in diet on the major economic traits of broiler in two different breeds. In the both experiments, the Arbor Acres and Ross breeds were used as experimental stocks and two groups were assigned in each breed. The control group birds(CON) were fed with basal diet only and the experimental group birds(EXP) were fed with basal diet added with 10.5mg chitosan/bird/day. The chitosan was supplied to birds from day-old in experiment 1 and from 15-day-old in experiment 2. In experiment 1, the mean body weight at 35-day-old were significantly(P〈0.05) heavier by 121.2 g and 243.7 g in the EXP groups than in the CON groups of Arbor Acres and Ross, respectively. Whereas, the mean body weights at 35-day-old in experiment 2 were lighter by 91.7 g and 70.2 g in the EXP groups than in the CON groups of Arbor Acres and Ross, respectively : however, the comparisons between breeds in the mean body weight at 35-day-old did not show significant difference in each other in both breeds. In the mean feed conversion ratio of Arbor Acres from 14 to 35-day old in experiment 1, it did not show significant difference between EXP and CON groups although the feed conversion ratio of the EXP group of Ross was significantly higher(P〈0.05) than the CON group. In experiment 2, the feed conversion ratios from 14 to 35-day-old did not show significant differences between the two breeds. The percentage of mean abdominal fat depositions of EXP groups in both breeds In experiment 1 were significantly(P〈0.05) higher than those of CON groups. And the percentage of mean abdominal fat deposition of Ross was significantly(P〈0.05) lower than that of Arbor Acres. In experiment 2, the percentage of mean abdominal fat depositions did not show significant difference between EXP and CON groups in both breeds. Whereas, the interaction effects between breed and experimental groups on the above economic traits did not show significant in both experiments.
