Effects of Nitrogen Application Levels on Grain Yield and Yield-related Traits of Rice Genetic Resources (질소비료 시비 수준이 벼의 수량 및 수량구성요소에 미치는 영향)

  • Nitrogen is a major and essential macronutrient for plant growth and development. However, excessive nitrogen application can lead to ecological pollution or greenhouse gas emissions, consequently resulting in climate change. In this study, we used 153 genetic resources of rice to evaluate the effects of the levels of nitrogen application on grain yield and yield-related traits. Significant differences were noted in the yield and yield-related traits of genetic resources between two nitrogen application levels, namely, 4.5 kg/10a (NN: normal nitrogen condition) and 9.0 kg/10a (LN: low-nitrogen condition). Among the tested traits, days to heading (DTH), clum length (CL), grain yield per plant (GYP), number of panicles per plant (NPP), and number of spikelets per panicle (NSP) decreased by 1.8 to 17.9% when the nitrogen application levels decreased from NN to LN. The 1,000-grain weight (TWG) and percentage of ripened grain (PRG) increased by 2.6 to 11.2% under these conditions. Based on nitrogen application levels, two-way analysis of variance (ANOVA) demonstrated significant differences in GYP, NPP, and PRG but not in NSP and TGW. NPP exhibited negative correlations with NSP (-0.44) and TGW (-0.44), and TGW displayed a negative correlation with PRG (-0.34), whereas, GYP exhibited a positive correlation with PRG (0.37) and NSP (0.38). A similar pattern was recorded under the LN condition. NPP, TGW, and PRG were clustered as PA (principle axis) 1 under the LN condition by factor analysis. NSP and GYP were clustered as PA (principle axis) 2. These results demonstrated NPP and NSP as the primary factors contributing to the decrease in grain yield under LN conditions. In conclusion, we selected eight genetic resources that exhibited higher GYP under both NN and LN conditions with higher NPP or NSP. These genetic resources can be considered valuable breeding materials for the adaptation of plants to nitrogen deficiency.

Anti-inflammatory Activities of Ethanol Extracts from Leaf, Seed, and Seedpod of Nelumbo nucifera (연잎, 연자육, 연자방 에탄올 추출물의 항염증 활성)

  • Nelumbo nucifera, also known as sacred lotus, has mainly been used as a food throughout the Asian countries. In the present study, we prepared ethanol extracts from leaf (NL), seed (NS), and seedpod (NSP) of Nelumbo nucifera and investigated their anti-inflammatory activities in mouse macrophage RAW 264.7 cells. To evaluate the anti-inflammatory activities of NL, NS, and NSP, nitric oxide (NO) production was measured in LPS-stimulated RAW 264.7 cells. NL, NS, and NSP significantly reduced NO production in a dose-dependent manner without affecting cell viabilities. NL, NS, and NSP dramatically decreased the protein expression of pro-inflammatory genes such as iNOS and COX-2. NL, NS, and NSP also suppressed phosphorylation of MAPKs and the nuclear translocation of $NF-{\kappa}B$ p65 indicating they have their anti-inflammatory activities via regulating mitogen-activated protein kinases (MAPKs) and nuclear factor kappa B ($NF-{\kappa}B$) pathways. In addition, we analyzed the production of reactive oxygen species (ROS) by the treatment of NL, NS, and NSP. All extracts reduced ROS production in a dose-dependent manner. And also, they increased heme oxygenase-1 (HO-1) protein expression and the nuclear translocation of nuclear respiratory factor 2 (Nrf2). In conclusion, our results suggest that Nelumbo nucifera has its anti-inflammatory activity via regulating MAPKs, $NF-{\kappa}B$, and Nrf2/HO-1 pathways.

Effects of Enzyme Supplementation on Growth, Intestinal Content Viscosity, and Digestive Enzyme Activities in Growing Pigs Fed Rough Rice-based Diet

  • The purpose of the present study was to investigate the effects of exogenous non-starch polysaccharides (NSP) enzymes on performance, intestinal content viscosity and digestive enzyme activities of growing pigs fed a rough rice-based diet. A total of 60 crossbred barrows with an initial body weight of 35.16 kg (SD = 0.82) were blocked by body weight and randomly assigned to two treatments with three replications. Each group was fed the diet based on rice with or without exogenous NSP enzymes (2 g/kg of diet). During the 70 days of the feeding trial, all pigs were given free access to feed and water. At the end of the feeding trial, six pigs from each treatment were randomly selected and slaughtered to collect intestinal digesta, intestinal mucosa, and pancreas. The addition of NSP enzymes improved average daily gain (p<0.05) and feed:gain (p<0.05), and decreased viscosity of digesta in the jejunum (p<0.001) and ileum (p<0.01) of pigs. The supplementation of NSP enzymes increased activities of protease (p<0.01), trypsin (p<0.01) and ${\alpha}$-amylase (p<0.05) in duodenal contents. However, digestive enzymes in the pancreas, jejunal and ileal mucosa were unaffected by the supplemental NSP enzymes (p>0.10). The results indicate that the addition of NSP enzymes to rough rice-based diets improved performance of pigs, reduced viscosity and increased digestive activity in the small intestine.

  • Synthesis of a deuterium-labeled derivative of nitrospirobenzopyran (NSP), one of representative photochromic compounds, has been described. Four deuteriums were successfully introduced on 1-methyl and ${\alpha}$-methyne relative to spiro-carbon in the title compound with more than 95atom%D purity. Main photodegraded products of NSP were two oxindoles in acetonitrile, and additional products were formed in poly(isobutyl-methacrylate) films possibly due to restricted molecular motion in polymer matrix. Quantitative HPLC analysis revealed that partial introduction of deuterium to NSP brought a noticeable isotope effect, recognizable enhancement in photo-resistivity of NSP, i.e.,8.3% in solutions and 29% in polymeric films.

구제역 감염 확인을 위한 비구조단백질(NSP) 항체 검출에 대한 고찰

  • 구제역은 급성 발열, 파행을 일으키며, 혀, 발굽, 콧등, 유두 등에 수포형성이 특징인 질병으로 소와 양, 염소는 지속감염(캐리어)동물이 될 수 있으며, 감염된 소의 경우 2-3년 동안 바이러스를 체내에 보유할 수 있다. 따라서 구제역의 정확한 진단 및 예찰은 구제역 청정화를 유지하고 관리하는 데 있어서 매우 중요한 일이다. 우리나라는 2010년 구제역이 발생하여 전국적으로 확산되는 상황에서 백신을 접종했다. 구제역 감염항체로 알려져 있는 비구조단백질 (non-structural protein : NSP) 항체를 검출하면 감염동물과 백신접종 동물을 감별하는 것이 가능하다고 알려져 있으나, 전국적인 백신접종으로 복잡한 축산현장에서는 NSP 검사결과를 해석하는 것이 어려운 경우가 생길 수 있다. 따라서 역학 등과 진단 결과를 종합하여 그 의미를 적절히 파악하는 것은 중요하다. 본 글에서는 구제역 방역에 있어서 중요한 실험법인 NSP 검사법의 실체와 결과의 의미에 대한 이해를 돕고자 하였다.

Application of cotton rope to detect foot-and-mouth disease virus in the pigs of farms in which nonstructural protein (NSP) antibody were detected in 2016 (2016년 구제역 비구조단백질(NSP) 항체 지속 검출농가에서 구제역바이러스 검출을 위한 로프법 적용)

  • The objective of this study was to assess the possibility of detecting Foot-and-Mouth Disease Virus (FMDV) from the herd-based oral fluids specimens collected by the cotton ropes from pig farms that were found as FMDV nonstructural protein (NSP) antibodies positive. The cotton ropes were applied to detect FMDV in the selected pig farms which NSP antibodies were continuously detected in 2016, including the one pig farm which FMDV antigen were detected at the specimens from the pigsty environment. As the result, FMDV antigen were not detected in the oral fluid specimens collected by the cotton ropes. Theoretically, to detect FMDV antigen from the pigs with NSP antibodies has very low possibility because FMDV antigen disappeared at the time when NSP antibodies were produced by FMDV. Therefore, in order to detect FMDV antigen from the oral fluids using the cotton rope, it would be more effective to be applied to target the FMDV infected pigs rather than the NSP antibodies positive pigs. The collected oral fluids using cotton rope could be useful test specimens to monitor high-density pig populations for FMDV infection. Then, oral fluids sampling using cotton rope will be used for the efficient FMDV surveillance to detect FMDV antigen.

Molecular characterization of avian rotavirus isolated in Korea

  • An avian rotavirus (AvRV-2) was isolated from feces of broilers suffering from acute gastroenteritis in 2011. It was the first avian rotavirus isolated in Korea. To investigate the molecular characteristics of AvRV-2, the VP4, VP6, VP7 and NSP4 gene nucleotide sequences were determined and compared with those of rotavirus strains available in the GenBank database. The phylogenetic tree of VP7 gene showed that AvRV-2 had a high degree of nucleotide sequence homology (93.4% to 94.7%) with those of rotaviruses belonging to genotype G19 cluster. The phylogenetic tree of the VP4 gene revealed a high degree of nucleotide sequence homology (95.8% to 95.9%) with genotype P[30] rotaviruses isolated from chickens. The VP6 and NSP4 gene nucleotide sequences showed the highest identities with those of avian strains with 95.3% to 96.4% and 90.3% to 92.2%, respectively. Genetic characterization of the VP4, VP6, VP7 and NSP4 showed that AvRV-2 strain was most closely related to chicken rotavirus strains from Germany and Japan. Comparative nucleotide sequences and phylogenetic analysis indicated that avian rotavirus isolated from broilers belonged to genotype G19P[30] and it was the first report on avian rotavirus infection in Korea.

Biochemical Characterization of Exoribonuclease Encoded by SARS Coronavirus

  • The nsp14 protein is an exoribonuclease that is encoded by severe acute respiratory syndrome coronavirus (SARS-CoV). We have cloned and expressed the nsp14 protein in Escherichia coli, and characterized the nature and the role(s) of the metal ions in the reaction chemistry. The purified recombinant nsp14 protein digested a 5'-labeled RNA molecule, but failed to digest the RNA substrate that is modified with fluorescein group at the 3'-hydroxyl group, suggesting a 3'-to-5' exoribonuclease activity. The exoribonuclease activity requires $Mg^{2+}$ as a cofactor. Isothermal titration calorimetry (ITC) analysis indicated a two-metal binding mode for divalent cations by nsp14. Endogenous tryptophan fluorescence and circular dichroism (CD) spectra measurements showed that there was a structural change of nsp14 when binding with metal ions. We propose that the conformational change induced by metal ions may be a prerequisite for catalytic activity by correctly positioning the side chains of the residues located in the active site of the enzyme.

  • Determination of dietary fiber contents in some Korean foods was attempted by Englyst's gas chromatographic and colorimetric methods which measure the nonstarch polysaccharide (NSP) contents NSP values, except seaweeds, by gas chromatography (x) and colorimetry (y) were very closely correlated (y=1.01x-0.52, r=0.997, n=9), NSP values by gas chromatography were lower than total dietary fiber (TDF) Haloes by AOAC method. By adding lignin to NSP values, relation between the two methods was improved. But, TDF values (y) by adding lignin to NSP values and TDF values (x) by AOAC method were related as y=0.68x-0.64 and F=0.903(n= 12), which was not closely related.

Heuristics for Selecting Nodes on Cable TV Network (케이블 TV 망에서 노드 선택을 위한 휴리스틱 연구)

  • The cable TV network has delivered downward broadcasting signals from distribution centers to subscribers. Since the traditional coaxial cable has been upgraded by the Hybrid Fiber Coaxial(HFC) cable, the upward channels has expanded broadband services such as Internet. This upward channel is vulnerable to ingress noises. When the noises from the children nodes accumulated in an amplifier exceeds a certain level, that node has to be cut off to prevent the noise propagation. The node selection problem(NSP) is defined to select nodes so that the noise in each node does not exceed the given threshold value and the sum of Profits of selected nodes can be maximized. The NSP has shown to be NP-hard. In this paper, we have proposed heuristics to find the near-optimal solution for NSP. The experimental results show that interval partitioning is better than greedy approach. Our heuristics can be used by the HFC network management system to provide privileged services to the premium subscribers on HFC networks.
