Novel reassortant 2.3.4.4B H5N6 highly pathogenic avian influenza viruses circulating among wild, domestic birds in Xinjiang, Northwest China

  • Background: The H5 avian influenza viruses (AIVs) of clade 2.3.4.4 circulate in wild and domestic birds worldwide. In 2017, nine strains of H5N6 AIVs were isolated from aquatic poultry in Xinjiang, Northwest China. Objectives: This study aimed to analyze the origin, reassortment, and mutations of the AIV isolates. Methods: AIVs were isolated from oropharyngeal and cloacal swabs of poultry. Identification was accomplished by inoculating isolates into embryonated chicken eggs and performing hemagglutination tests and reverse transcription polymerase chain reaction (RT-PCR). The viral genomes were amplified with RT-PCR and then sequenced. The sequence alignment, phylogenetic, and molecular characteristic analyses were performed by using bioinformatic software. Results: Nine isolates originated from the same ancestor. The viral HA gene belonged to clade 2.3.4.4B, while the NA gene had a close phylogenetic relationship with the 2.3.4.4C H5N6 highly pathogenic avian influenza viruses (HPAIVs) isolated from shoveler ducks in Ningxia in 2015. The NP gene was grouped into an independent subcluster within the 2.3.4.4B H5N8 AIVs, and the remaining six genes all had close phylogenetic relationships with the 2.3.4.4B H5N8 HPAIVs isolated from the wild birds in China, Egypt, Uganda, Cameroon, and India in 2016-2017, Multiple basic amino acid residues associated with HPAIVs were located adjacent to the cleavage site of the HA protein. The nine isolates comprised reassortant 2.3.4.4B HPAIVs originating from 2.3.4.4B H5N8 and 2.3.4.4C H5N6 viruses in wild birds. Conclusions: These results suggest that the Northern Tianshan Mountain wetlands in Xinjiang may have a key role in AIVs disseminating from Central China to the Eurasian continent and East African.

Effects of Overall Shading and Partial Shading on the Response of Chlorophyll Fluorescence of Soybean (전면적차광과 부분차광이 콩 엽록소 형광 반응에 미치는 영향)

  • The growth experiment under shading condition has been performed to understand the eco-physiological responses of crops to light in terms of photosynthesis. There are two types of shading: overall shading and partial shading. In this study, the chlorophyll fluorescence of soybean was observed under the overall shading of the box made by polyresin and the partial shading at agrivoltaic system. The overall shading condition during vegetative growth induced lower SPAD and Electron transport rate (ETR). These lower values recovered after removal of shading box. However, the Non-photochemical fluorescence quenching (NPQ) became lower under overall shading and higher under partial shading. Such increase in NPQ limited crop photosynthesis even though the ETR was almost same to the control without shading treatment. Under the condition of partial shading, the values of SP AD and ETR for soybean did not change. However, the NPQ was higher than control condition. This suggests that the crop photosynthesis under both types of shading would be decreased by different eco-physiological processes which are the lower ETR in overall shading and the higher NP Q in partial shading despite the reduced light under shading conditions.

Comparative Analysis of the Body Muscle Activity According to the Prop and Different Foot Stability during Pilates Bridge Motion (필라테스 브릿지 동작 시 소도구와 발의 불안정성 차이에 따른 신체 근육의 근활성도 비교 분석)

  • The aim of this study was to examine the comparative analysis of body muscle EMGs according to the prop and different foot stability during Pilates bridge motion. Eighteen adult males(age, 22.3±2.1 years; height, 173.89±4.51 cm; body mass, 72.61±4.13 kg; and BMI, 24.03±1.31 kg/m2) participated in this study as subjects. The Pilates bridge was composed of a total of 9 motions, according to the props (NP, no prop; RG, ring; GB, gym ball) and different foot stability (BS, basic surface; FR, form roller; BOSU, both sides up). We measured the right's muscle activities of the upper rectus abdominis, lower abdominal stabilizers, external oblique, adductor longus, rectus femoris, vastus lateralis, tibialis anterior, and biceps femoris. The research findings were as follows. During Pilates bridge motion, the use of GB was found to be more effective in activating the body muscle. And during Pilates bridge motion, the use of BOSU was found to be most high in activating the body muscle as well. To summarize the research findings, the use of GB and BOSU during Pilates bridge motion were discovered to enormous affect muscle activities. Therefore, the results of this study are expected to be able to present efficient Pilates bridge exercises for strengthening physical strength.

Optimization of Protoplast Isolation and Ribonucleoprotein/Nanoparticle Complex Formation in Lentinula edodes (표고버섯의 원형질체 분리 최적화와 RNPs/나노파티클 복합체 형성)

  • Despite the long history of mushroom use, studies examining the genetic function of mushrooms and the development of new varieties via bio-molecular methods are significantly lacking compared to those examining other organisms. However, owing to recent developments, attempts have been made to use a novel gene-editing technique involving CRISPR/Cas9 technology and genetic scissors in mushroom studies. In particular, research is actively being conducted to utilize ribonucleoprotein particles (RNPs) that can be genetically edited with high efficiency without foreign gene insertion for ease of selection. However, RNPs are too large for Cas9 protein to pass through the cell membrane of the protoplasmic reticulum. Furthermore, guide RNA is unstable and can be easily decomposed, which remarkably affects gene editing efficiency. In this study, nanoparticles were used to mitigate the shortcomings of RNP-based gene editing techniques and to obtain transformants stably. We used Lentinula edodes (shiitake mushroom) Sanjo705-13 monokaryon strain, which has been successfully used in previous genome editing experiments. To identify a suitable osmotic buffer for the isolation of protoplast, 0.6 M and 1.2 M sucrose, mannitol, sorbitol, and KCl were treated, respectively. In addition, with various nanoparticle-forming materials, experiments were conducted to confirm genome editing efficiency via the formation of nanoparticles with calcium phosphate (CaP), which can be bound to Cas9 protein without any additional amino acid modification. RNPs/NP complex was successfully formed and protected nuclease activity with nucleotide sequence specificity.

Analysis on Connecting User Experience of Metaverse Related with Landscape Architecture - Focused on Meta-Everland - (메타버스 조경 공간의 이용자 경험 분석 - 메타 에버랜드를 중심으로 -)

  • As the concept of metaverse has received great attention, interest in metaverse related to landscape architecture is also increasing. The aim of this research is to understand the potential and tasks of applying metaverse in the field of landscape architecture by analyzing the user experience of a metaverse platform. The object of the research is Meta-Everland built in the Roblox platform, which has the most users among landscape architectural metaverses in Korea. NPS of 30 users who have been to Everalnd was investigated after using Meta-Everland with interviews. NPS before the metaverse experience was -16 and NPS after the experience was -24. This result means that the promotion level was lowered after the experience of the metaverse. There were three causes of lowered NPS: lack of users, low-quality graphics and interface, and lack of content. The factor of lack of users was the result of the other two problems. The factor of low technical quality is hard to be improved in a short period of time. Therefore, the main task to improve the metaverse is developing better metaverse content related to landscape architecture. It is more appropriate to develop metaverse-specific content rather than improve reality issues. Applying AR and VR devices, enhancing communication function, and developing potential as a simulation device are needed to be considered.

  • Flash lamp annealing (FLA) of metal nanoparticle (NP) ink has provided powerful strategies to fabricate high-performance electrodes on a flexible substrate because of its rapid processing capability (in milliseconds), low-temperature process, and compatibility with to roll-to-roll process. However, metal NPs [e.g., gold (Au), silver (Ag), copper (Cu), etc.] have limitations such as difficulty in synthesizing fine metal NPs (diameter less than 10 nm), high price, and degradation during ink storage and FLA processing. In this regard, organometallic ink has been proposed as a material that can replace metal NPs due to their low-cost (usually 1/100 times cheaper than metal nano inks), low-temperature processability, and high material stability. Despite these advantages, the fabrication of flexible electrodes through FLA treatment of organometallic compounds has not been extensively researched. In this paper, we experimentally guide how to determine the optimal conditions for forming electrodes on flexible substrates by considering material parameters, and flashlight processing parameters (energy density, pulse duration, etc) to minimize the difficulties that may arise during the FLA of organometallic ink.

Bioaccumulation of Ag and Zn in earthworms (Eisenia fetida) from soil contaminated with Ag and Zn nanoparticles using a radiotracer method (방사성동위원소 추적자 기법을 이용한 제조나노입자로 오염된 토양으로부터 지렁이(Eisenia fetida)의 은과 아연 축적 연구)

  • In a radiotracer study, the bioaccumulation and efflux of metals in earthworms (Eisenia fetida) exposed to soil spiked with ZnO and Ag nanoparticles (AgNP) were compared to those exposed to soil spiked with ionic Zn and Ag. Additionally, the bioavailability and chemical mobility of nano- and ionic metals in the soil were estimated using the sequential extraction method and compared to the bioaccumulation factor(BAF). The BAF for ZnO (0.06) was 31 times lower than that for Zn ions (1.86), suggesting that ZnO was less bioavailable than the ionic form in contaminated soil. In contrast, the BAFs for two types of AgNPs coated with polyvinylpyrrolidone (0.12) or citrate (0.11) were comparable to those of ionic Ag (0.17). The sequential extraction of metals from the soil suggests that the chemically mobile fractions in the Zn ion treatment were higher(35%) than those (<20%) in the Ag ion treatment, which was consistent with the greater BAFs in the former than the latter. However, the chemical mobility in the ZnO treatments did not predict bioavailability. The efflux rates of Ag (3.2-3.8% d-1) in the worms were 2-3×those(1.2-1.7% d-1) for Zn.

Association between the severity of hypodontia and the characteristics of craniofacial morphology in a Chinese population: A cross-sectional study

  • Objective: To investigate craniofacial differences in individuals with hypodontia and explore the relationship between craniofacial features and the number of congenitally missing teeth. Methods: A cross-sectional study was conducted among 261 Chinese patients (males, 124; females, 137; age, 7-24 years), divided into four groups (without hypodontia: no teeth missing, mild: one or two missing teeth, moderate: three to five missing teeth, severe: six or more missing teeth) according to the number of congenitally missing teeth. Differences in cephalometric measurements among the groups were analyzed. Further, multivariate linear regression and smooth curve fitting were performed to evaluate the relationship between the number of congenitally missing teeth and the cephalometric measurements. Results: In patients with hypodontia, SNA, NA-AP, FH-NA, ANB, Wits, ANS-Me/N-Me, GoGn-SN, UL-EP, and LL-EP significantly decreased, while Pog-NB, AB-NP, N-ANS, and S-Go/N-Me significantly increased. In multivariate linear regression analysis, SNB, Pog-NB, and S-Go/N-Me were positively related to the number of congenitally missing teeth. In contrast, NA-AP, FH-NA, ANB, Wits, N-Me, ANS-Me, ANS-Me/N-Me, GoGn-SN, SGn-FH (Y-axis), UL-EP, and LL-EP were negatively related, with absolute values of regression coefficients ranging from 0.147 to 0.357. Further, NA-AP, Pog-NB, S-Go/N-Me, and GoGn-SN showed the same tendency in both sexes, whereas UL-EP and LL-EP were different. Conclusions: Compared with controls, patients with hypodontia tend toward a Class III skeletal relationship, reduced lower anterior face height, flatter mandibular plane, and more retrusive lips. The number of congenitally missing teeth had a greater effect on certain characteristics of craniofacial morphology in males than in females.

Antioxidant and Anti-inflammatory Activities of Artemisia annua L. According to Extract Methods (개똥쑥 용매추출 방법에 따른 항산화 활성 및 항염증 효과 )

  • This study was to investigate the antioxidant and anti-inflammatory activity of leaf and stem of Artemisia annua L. extract by hot water and 70% ethanol. The total polyphenol content was significantly higher in the 70% ethanol extract of Artemisia annua L. than in the hot water extract, but there was no significant difference in the total flavonoid content between the hot water extract and the 70% ethanol extract. The DPPH radical and ABTS radical scavenging ability was the highest in the vitamin C treatment group at the concentration of 62.5~500 ㎍/mL, followed by the 70% ethanol extract and the hot water extract. As a result of measuring the NO production inhibitory effect at 125 ~ 500 ㎍/mL, the hot water extract showed strong NO production inhibition at the concentration of 500 ㎍/mL, and showed the inhibition of NO production with concentration-dependent pattern. In addition to inhibitory activity of NP production, the 70% ethanol extract also showed an inhibitory effect on inflammatory cytokines production. It is thought that it can be widely used in the treatment and improvement of inflammatory diseases because it shows antioxidant effects and significantly reduces the expression of inflammatory cytokines.

Electrochemical Characteristics of Pencil Graphite Electrode Through Surface Modification and its Application of Non-enzymatic Glucose Sensor (표면 개질된 샤프심 전극의 전기화학적 특성 고찰 및 비효소적 글루코스 센서 활용)

  • Most medical sensors are disposable products. In order to reduce inspection and diagnosis costs, it is more important to develop the inexpensive electrode materials. We fabricated the CuO NPs/PANI/E-PGE as an electrode material for disposable electrochemical sensors and applied it to a non-enzymatic glucose sensor. For surface activation of PGE, pretreatment was performed using chemical and electrochemical methods, respectively. Electrochemical properties according to the pretreatment method were analyzed through chronoamperometry (CA), cyclic voltammetry (CV) and electrochemical impedance (EIS). From these analytical results, the electrochemically pretreated PGE (E-PGE) was finally adopted. The non-enzymatic glucose sensor based on CuO NPs/PANI/E-PGE shows sensitivity of 239.18 mA/mM×cm2 (in a linear range of 0.282~2.112 mM) and 36.99 mA/mM×cm2 (3.75423~50 mM), detection limit of 17.6 μM and good selectivity. Based on the results of this study, it was confirmed that the modified PGE is a high-performance electrode material. Therefore, these electrodes can be applied to a variety of disposable sensors.
